1Advances in research of the mid-deep South China Sea circulation显示文摘The South China Sea(SCS)is a large marginal sea connecting the Indian and Pacific oceans.Under the factors of monsoons, strait transport, and varied bathymetry, the SCS presents a three-layer structure and strong diapycnal mixing which is far greater than that in the open ocean.Theoretical analysis and observations reveal that internal tides, internal solitary waves, and strong winds are the sources of the strong mixing in the northern SCS.A major consequence of the strong mixing is an active mid-deep circulation system. This system promotes exchange of water between the SCS and adjacent oceans, and also regulates the upper layer of wind-driven circulation, making the 3 dimensional SCS circulation clearly different from that in other tropical and subtropical marginal seas. The mass transport capacity of the mid-deep circulation has a substantial impact on marine sedimentation, the biogeochemical cycle, and other processes in the SCS.This paper summarizes the recent advances in middeep sea circulation dynamics of the SCS,and discusses the opportunities and challenges in this area.Dongxiao WANG Qiang WANG Shuqun CAI Xiaodong SHANG Shiqiu PENG Yeqiang SHU Jingen XIAO Xiaohui XIE Zhiwei ZHANG Zhiqiang LIU Jian LAN Dake CHEN Huijie XUE Guihua WANG Jianping GAN Xinong XIE Rui ZHANG Hui CHEN Qingxuan YANG 2019Science China Earth Sciences2019,62,12:8
2CD147 antibody specifically and effectively inhibits infection and cytokine storm of SARS-CoV-2 and its variants delta, alpha, beta, and gamma显示文摘SARS-CoV-2 mutations contribute to increased viral transmissibility and immune escape, compromising the effectiveness of existing vaccines and neutralizing antibodies. An in-depth investigation on COVID-19 pathogenesis is urgently needed to develop a strategy against SARS-CoV-2 variants. Here,we identified CD147 as a universal receptor for SARS-CoV-2 and its variants.Meanwhile,Meplazeumab,a humanized anti-CD147 antibody, could block cellular entry of SARS-CoV-2 and its variants-alpha,beta,gamma,and delta, with inhibition rates of 68.7,75.7,52.1,52.1,and 62.3%at 60μg/ml,respectively.Furthermore,humanized CD147 transgenic mice were susceptible to SARS-CoV-2 and its two variants, alpha and beta. When infected, these mice developed exudative alveolar pneumonia, featured by immune responses involving alveoli-infiltrated macrophages, neutrophils, and lymphocytes and activation of IL-17 signaling pathway.Mechanistically,we proposed that severe COVID-19-related cytokine storm is induced by a'spike protein-CD147-CyPA signaling axis':Infection of SARS-CoV-2 through CD147 initiated the JAK-STAT pathway, which further induced expression of cyclophilin A(CyPA);CyPA reciprocally bound to CD147 and triggered MAPK pathway.Consequently, the MAPK pathway regulated the expression of cytokines and chemokines, which promoted the development of cytokine storm.Importantly,Meplazumab could effectively inhibit viral entry and inflammation caused by SARS-CoV-2 and its variants. Therefore, our findings provided a new perspective for severe COVID-19-related pathogenesis.Furthermore, the validated universal receptor for SARS-CoV-2 and its variants can be targeted for COVID-19 treatment.Jiejie Geng Liang Chen Yufeng Yuan Ke Wang Youchun Wang Chuan Qin Guizhen Wu Ruo Chen Zheng Zhang Ding Wei Peng Du Jun Zhang Peng Lin Kui Zhang Yongqiang Deng Ke Xu Jiangning Liu Xiuxuan Sun Ting Guo Xu Yang Jiao Wu Jianli Jiang Ling Li Kun Zhang Zhe Wang Jing Zhang Qingguo Yan Hua Zhu Zhaohui Zheng Jinlin Miao Xianghui Fu Fengfan Yang Xiaochun Chen Hao Tang Yang Zhang Ying Shi Yumeng Zhu Zhuo Pei Fei Huo Xue Liang Yatao Wang Qingyi Wang Wen Xie Yirong Li Mingyan Shi Huijie Bian Ping Zhu Zhi-Nan Chen 2021Signal Transduction and Targeted Therapy2021,6,10:6
3Safety and efficacy of meplazumab in healthy volunteers and COVID-19 patients: a randomized phase 1 and an exploratory phase 2 trial显示文摘Recent evidence suggests that CD147 serves as a novel receptor for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.Blocking CD147 via anti-CD147 antibody could suppress the in vitro SARS-CoV-2 replication.Meplazumab is a humanized anti-CD147 IgG_(2) monoclonal antibody, which may effectively prevent SARS-CoV-2 infection in coronavirus disease 2019(COVID-19)patients.Here,we conducted a randomized,double-blinded,placebo-controlled phase 1 trial to evaluate the safety, tolerability, and pharmacokinetics of meplazumab in healthy subjects, and an open-labeled,concurrent controlled add-on exploratory phase 2 study to determine the efficacy in COVID-19 patients. In phase 1 study,59 subjects were enrolled and assigned to eight cohorts, and no serious treatment-emergent adverse event(TEAE)or TEAE grade≥3 was observed. The serum and peripheral blood Cmax and area under the curve showed non-linear pharmacokinetic characteristics. No obvious relation between the incidence or titer of positive anti-drug antibody and dosage was observed in each cohort. The biodistribution study indicated that meplazumab reached lung tissue and maintained>14 days stable with the lung tissue/cardiac blood-pool ratio ranging from 0.41 to 0.32.In the exploratory phase 2 study,17 COVID-19 patients were enrolled, and 11 hospitalized patients were involved as concurrent control. The meplazumab treatment significantly improved the discharged(P=0.005)and case severity(P=0.021),and reduced the time to virus negative(P=0.045)in comparison to the control group. These results show a sound safety and tolerance of meplazumab in healthy volunteers and suggest that meplazumab could accelerate the recovery of patients from COVID-19 pneumonia with a favorable safety profile.Huijie Bian Zhao-Hui Zheng Ding Wei Aidong Wen Zheng Zhang Jian-Qi Lian Wen-Zhen Kang Chun-Qiu Hao Jing Wang Rong-Hua Xie Ke Dong Jie-Lai Xia Jin-Lin Miao Wen Kang Guoquan Li Di Zhang Mingru Zhang Xiu-Xuan Sun Likun Ding Kui Zhang Junfeng Jia Jin Ding Zhiqin Li Yanyan Jia Lin-Na Liu Zhe Zhang Zhao-Wei Gao Hong Du Na Yao Qing Wang Ke Wang Jie-Jie Geng Bin Wang Ting Guo Ruo Chen Yu-Meng Zhu Li-Juan Wang Qian He Rui-Rui Yao Ying Shi Xiang-Min Yang Jian-Sheng Zhou Yi-Nan Ma Ya-Tao Wang Xue Liang Fei Huo Zhe Wang Yang Zhang Xu Yang Ye Zhang Lu-Hua Gao Ling Wang Xiao-Chun Chen Hao Tang Shuang-Shuang Liu Qing-Yi Wang Zhi-Nan Chen Ping Zhu 2021Signal Transduction and Targeted Therapy2021,6,6:2

7Formation of an anticyclonic eddy and the mechanism involved: a case study using cruise data from the northern South China Sea显示文摘Data from satellite altimetry and in situ observations together with the Hybrid Coordinate Ocean Model (HYCOM) reanalysis data were used to investigate the mechanism and formation of an anticyclonic eddy in the northeastern South China Sea (SCS). Analysis of water mass using cruise data indicated that the water captured in the eddy diff ers from those in the SCS, the Kuroshio intrusion, and the eddy-forming region. Data from sea surface height (SSH) and sea level anomaly (SLA) indicate that the eddy formed due both to the Kuroshio intrusion and the local circulation in the SCS. The Kuroshio intrusion is present at the start of the eddy growth (March 5-9) before Kuroshio leaps the Luzon Strait. The eddy then becomes larger and stronger in the absence of the Kuroshio intrusion. From the eddy budget of the HYCOM reanalysis data, the formation of the eddy goes in three steps. By the third step, the eddy had become aff ected by variations of local SCS circulation, which is more strongly than in the fi rst step in which it is aff ected more by the Kuroshio intrusion. The variability of the temperature and salinity inside the eddy provide a support to this conclusion. The water in the SCS intruded into the eddy from the southeast, which decrease the salinity gradually in the southern part of the eddy during the growth period.HOU Huaqian XIE Qiang XUE Huijie SHU Yeqiang NAN Feng YIN Yuqi YU Fei 2019Journal of Oceanology and Limnology2019,37,5:1

10Atomistic Simulations of the Mechanical Deformation of Irradiation-amorphized Silicon Carbide显示文摘Irradiation-induced atomic-scale defects and lattice disorder in Silicon Carbide(SiC) can significantly affect the material's mechanical properties.Currently there lacks a unified physical model capable of describing the law in which the properties of SiC scale with the accumulation of defects, especially in terms of the underlying physical mechanism. To develop fundamental models that are capable of describing the various physical properties of SiC as a function of microstructural change, molecular dynamics simulations of uniaxial tension were performed on a series of irradiation-amorphized SiC(a-SiC) samples with a range of imposed chemical disorder, which is defined as the ratio between the number of homonuclear bonds and heteronuclear bonds(χ≡ N C-C N Si-C).With increasing chemical disorder, significant alternation of mechanical response of a-SiC has been detected in terms of increasingly pronounced plastic flow.Meanwhile relevant mechanical properties, including Young's modulus, strength, yield stress and strain,as well as failure strain scale monotonically with chemical disorder while in distinct manners.Specifically slight chemical disorder(χ = 0.045) could induce substantial reduction of Young's modulus up to 2%,whereas strength basically linearly varies with chemical disorder until χ≈ 0.5 upon which the variations in mechanical properties tend to saturate.Further examination of the evolution of atomic structure of a-SiC reveals a crossover of deformation mechanisms from homogeneous elastic deformation to localized plastic flow, which accounts for the strong chemical disorder dependence of the mechanical properties as well as mechanical responses of amorphous SiC.This crossover is also manifested in switching of fracture mode from brittle failure dominated by lattice instability in the ligaments between topological disordered clusters to nanoductile failure preceded by percolation of nanocavities.Employing chemical disorder to measure the defect concentration of a-SiC could contribute to the quantification of the correlation between mechanical properties and the corresponding defective a-SiC structure. Moreover the distinct scale laws shown by Young's modulus and strength with chemical disorder and the proposed critical chemical disorder threshold could benefit the quantitative evaluations of the mechanical performances of SiC components in different irradiation environments.XUE Kun NIU Lisha SHI Huiji 2010Chinese Journal of Mechanical Engineering2010,23,6:1

18Interannual Variability of Shelf and Slope Circulations in the Northern South China Sea显示文摘The northern South China Sea(NSCS) is a dynamically complex region whose shelf and slope currents are driven by different mechanisms. In this study, we used field measurements to identify clear interannual variations in the circulation related to the El Ni?o-Southern Oscillation cycle. To investigate the modulation mechanisms, we used a high-resolution numerical model that covers the shelf and slope regions of the NSCS. The results indicate that the stronger southwestward slope current during La Ni?a and stronger northeastward shelf current during El Ni?o in summer and winter are largely related to changes in wind forcing. The Kuroshio intrusion into the NSCS does not appear to significantly affect the circulation in the southwestern shelf region.LIU Na GENG Bingxu XUE Huijie XIU Peng WANG Qiang WANG Dongxiao 2020Journal of Ocean University of China2020,19,5:0
19Saco-Casco Bays Inundation Modeling of Five Winter Storms显示文摘Coastal inundation along the northeast coast of the United States is usually caused by strong winter storms(WS).However, the accurate prediction of coastal inundation due to the WS is challenging. Therefore, our study aims to develop a unique high-resolution modeling system to accurately predict the coastal inundation in the ungauged coastal areas of Saco-Casco Bays and map the flood risk zones to potential sea level rise due to these storms.Hindcasts of five classic WS in 2014-2015 were studied. The inundation models are based on FVCOM that uses unstructured grid to capture the minor to significant flooding near the shallow areas of the bays, harbor entrance and river banks. In this study, topography has been generated from the NOAA’s integrated dataset of Portland,ME 1/3 arc-second MHW digital elevation model. The model runs were driven by two different sets of meteorological(NECOFS WRF and NOAA’s NAM WRF)forcing to examine the effect of spatial resolution on the predicted inundation. The study reveals that among the five storm surge cases,WS-III produces a maximum surge of 0.7 m and WS-II cause a minimum surge of 0.3 m.In all scenarios, southward wind-driven coastal current flowing towards Biddeford Pool,Pine Point and Camp Ellis forms a small-scale eddy which causes significant inundation however strength of the current varies accordingly.Sensitivity experiments have been carried out using NECOFS WRF simulation products with varying parameters of marshland elevation and bottom friction to understand the influence of intertidal storage on the predicted flooding.Saswati Deb Huijie Xue Shivanesh Rao 2021Journal of Shipping and Ocean Engineering2021,11,2:0
