1Human umbilical cord Wharton's Jelly-derived mesenchymal stem cells differentiation into nerve-like cells显示文摘Background The two most basic properties of mesenchymal stem cells (MSCs) are the capacities to self-renew indefinitely and differentiate into multiple cells and tissue types. The cells from human umbilical cord Wharton’s Jelly have properties of MSCs and represent a rich source of primitive cells. This study was conducted to explore the possibility of inducing human umbilical cord Wharton’s Jelly-derived MSCs to differentiate into nerve-like cells.Methods MSCs were cultured from the Wharton’s Jelly taken from human umbilical cord of babies delivered after full-term normal labor. Salvia miltiorrhiza and β-mercaptoethanol were used to induce the human umbilical cord-derived MSCs to differentiate. The expression of neural protein markers was shown by immunocytochemistry. The induction process was monitored by phase contrast microscopy, electron microscopy (EM), and laser scanning confocal microscopy (LSCM) .The pleiotrophin and nestin genes were measured by reverse transcription-polymerase chain reaction (RT-PCR). Results MSCs in the Wharton’s Jelly were easily attainable and could be maintained and expanded in culture. They were positive for markers of MSCs, but negative for markers of hematopoietic cells and graft-versus-host disease (GVHD)-related cells. Treatment with Salvia miltiorrhiza caused Wharton’s Jelly cells to undergo profound morphological changes. The induced MSCs developed rounded cell bodies with multiple neurite-like extensions. Eventually they developed processes that formed networks reminiscent of primary cultures of neurons. Salvia miltiorrhiza and β-mercaptoethanol also induced MSCs to express nestin, β-tubulinⅢ, neurofilament (NF) and glial fibrillary acidic protein (GFAP). It was confirmed by RT-PCR that MSCs could express pleiotrophin both before and after induction by Salvia miltiorrhiza. The expression was markedly enhanced after induction and the nestin gene was also expressed.Conclusions MSCs could be isolated from human umbilical cord Wharton’s Jelly. They were capable of differentiating into nerve-like cells using Salvia miltiorrhiza or β-mercaptoethanol. The induced MSCs not only underwent morphologic changes, but also expressed the neuron-related genes and neuronal cell markers. 32018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 4Prevalence of Atrial Fibrillation in China and Its Risk Factors显示文摘Objective To study the prevalence of atrial fibrillation(AF)and the relation with its risk factors in China.Methods A total of 19 363 participants(8635 males and 10 728 females) aged ≥35 years in geographically dispersed urban and rural regions of China were included in this cross-sectional survey.All participants received questionnaire,physical and blood examination.Echocardiography were performed for AF patients found in the survey.Results Of the 19 363 participants,199 were diagnosed with AF.The estimated age-standardized prevalence of AF was 0.78% in men and 0.76% in women.The prevalence of AF in participants aged <60 years was 0.41% in men and 0.43% in women,and was 1.83% in both men and women aged ≥60 years.About 19.0% of males and 30.9% of females with AF were diagnosed with valve disease.Age-and sex-adjusted multivariable logistic regression analysis revealed that myocardial infarction,left ventricular hypertrophy(LVH),obesity,and alcohol consumption were associated with a increased risk of AF(P<0.05).Conclusion The age standardized prevalence of AF is 0.77% in the participants enrolled in the present study.The number of AF cases aged ≥35 years is 5.26 million according to 2010 Chinese Census.Most risk factors for AF,identified mainly in Western countries,are also detected in China.LI Ying WU Yang Feng CHEN Ke Ping LI Xian ZHANG Xing XIE Gao Qiang WANG Fang Zheng ZHANG Shu 2013Biomedical and Environmental Sciences2013,26,9:48
5Visualization of the complex structure and stress field inside rock by means of 3D printing technology显示文摘Accurate characterization and visualization of the complex inner structure and stress distribution of rocks are of vital significance to solve a variety of underground engineering problems. In this paper, we incorporate several advanced technologies, such as CT scan, three-dimensional(3D) reconstruction, and 3D printing, to produce a physical model representing the natural coal rock that inherently contains complex fractures or joints. We employ 3D frozen stress and photoelastic technologies to characterize and visualize the stress distribution within the fractured rock under uniaxial compression. The 3D printed model presents the fracture structures identical to those of the natural prototype. The mechanical properties of the printed model,including uniaxial compression strength, elastic modulus,and Poisson's ratio, are testified to be similar to those of the prototype coal rock. The frozen stress and photoelastic tests show that the location of stress concentration and the stress gradient around the discontinuous fractures are in good agreement with the numerical predictions of the real coalsample. The proposed method appears to be capable of visually quantifying the influences of discontinuous,irregular fractures on the strength, deformation, and stress concentration of coal rock. 7Clinicopathological and prognostic analysis of 429 patients with intrahepatic cholangiocarcinoma显示文摘AIM:To understand the clinicopathological characteristics and treatment selections and improve survival and provide valuable information for patients with intrahepatic cholangiocarcinoma(ICC). METHODS:We retrospectively evaluated 5311 liver cancer patients who received resection between October 1999 and December 2003.Of these,429(8.1%)patients were diagnosed with ICC,and their clinicopathological, surgical,and survival characteristics were analyzed. RESULTS:Upper abdominal discomfort or pain(65.0%), no symptoms(12.1%),and hypodynamia(8.2%)were the major causes for medical attention.Laboratory tests showed 198(46.4%)patients were HBsAg positive, 90(21.3%)hadα-fetoprotein>20μg/L,50(11.9%) carcinoembryonic antigen>10μg/L,and 242(57.5%) carbohydrate antigen 19-9(CA19-9)>37 U/mL.Survival data was available for 329(76.7%)patients and their mean survival time was 12.4 mo.The overall survival of the patients with R0,R1 resection and punching exploration were 18.3,6.6 and 5.6 mo,respectively. Additionally,CA19-9>37 U/mL was associated with lymph node metastases,but inversely associated withcirrhosis.Multivariate analysis indicated that radical resection,lymph node metastases,macroscopic tumor thrombi and size,and CA19-9 were associated with prognosis. 13Magma mixing of granites at Lianghe:In-situ zircon analysis for trace elements, U-Pb ages and Hf isotopes显示文摘In this paper, we report an integrated study of trace element, U-Pb age and Hf isotopic composition of zircons from alkali feldspar granites, granodiorites and diorite enclaves in a recently discovered ring complex at Lianghe in western Yunnan, Chi na. The granitoids showed identical U-Pb ages of 127, 115 and 122 Ma, from felsic to mafic, but had different zircon trace el ements and Hf isotopic compositions. Trace element content decreased with a gradual increase in εHf(t) values of ?9.1 to ?5.4, ?4.5 to 0, and 3.6 to 6.2, respectively. Results indicate that changes in zircon trace elements generally correlate with changes in Hf isotope signatures within single samples and among various granitoids. These relationships reflect the mixing of felsic and mafic magmas. Evidence indicates that depleted mantle-derived mafic magma underplating caused ancient crustal melting, and then formed large-scale granites in Lianghe during the Early Cretaceous. 14Pilose antler aqueous extract promotes the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells by stimulating the BMP-2/Smad1, 5/Runx2 signaling pathway显示文摘Peptides from Pilose antler aqueous extract(PAAE) have been shown to stimulate the proliferation and differentiation of bone marrow mesenchymal stem cells(BMSCs). However, the underlying molecular mechanisms are not well understood. Here, PAAE was isolated and purified to explore the molecular mechanisms underlying PAAE’s effects on BMSCs as well as its osteoprotective effects in ovariectomized rats. Our results showed that PAAE promoted proliferation and differentiation of BMSCs to become osteoblasts by enhancing ALP activity and increasing extracellular matrix mineralization. The trabecular microarchitecture of ovariectomized rats was also found to be protected by PAAE. Quantitative reverse transcription-polymerase chain reaction(Quantitative RT-PCR) results suggest that PAAE also increased the expression of osteogenic markers including, alkaline phosphatase(ALP), runt-related transcription factor 2(Runx2), osteocalcin(OCN), bone morphogenetic protein-2(BMP-2), and collagen I(COL-I). Immunoblotting results indicated that PAAE upregulated the levels of BMP-2 and Runx2 and was associated with Smad1/5 phosphorylation. PAAE A at the concentration of 200μg·mL^-1 showed the strongest effect on proliferation and osteogenic differentiation of BMSCs after 48 h. Using matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF MS), we identified the molecular weight of PAAE A and found that it is less than 3000 Da and showed several significant peaks. In conclusion, PAAE activates the BMP-2/Smad1, 5/Runx2 pathway to induce osteoblastic differentiation and mineralization in BMSCs and can inhibit OVX-induced bone loss. 18Changes in global potential vegetation distributions from 1911 to 2000 as simulated by the Comprehensive Sequential Classification System approach显示文摘Vegetation classification models play an important role in studying the response of the terrestrial ecosystem to global climate change. In this paper, we study changes in global Potential Natural Vegetation (PNV) distributions using the Comprehensive Sequential Classification System (CSCS) approach, a technique that combines geographic information systems. Results indicate that on a global scale there are good agreements among maps produced by the CSCS method and the globally well-accepted Holdridge Life Zone (HLZ) and BIOME4 PNV models. The potential vegetation simulated by the CSCS approach has 6 major latitudinal zones in the northern hemisphere and 2 in the southern hemisphere. In mountainous areas it has obvious altitudinal distribution characteristics due to topographic effects. The distribution extent for different PNV classes at various periods has different characteristics. It had a decreasing trend for the tundra and alpine steppe, desert, sub-tropical forest and tropical forest categories, and an increasing trend for the temperate forest and grassland vegetation categories. The simulation of global CSCS-based PNV classes helps to understand climate-vegetation relationships and reveals the dynamics of potential vegetation distributions induced by global changes. 20Osteoimmunomodulatory effects of biomaterial modification strategies on macrophage polarization and bone regeneration显示文摘Biomaterials as bone substitutes are always considered as foreign bodies that can trigger host immune responses.Traditional designing principles have been always aimed at minimizing the immune reactions by fabricating inert biomaterials.However,clinical evidence revealed that those methods still have limitations and many of which were only feasible in the laboratory.Currently,osteoimmunology,the very pioneering concept is drawing more and more attention-it does not simply regard the immune response as an obstacle during bone healing but emphasizes the intimate relationship of the immune and skeletal system,which includes diverse cells,cytokines,and signaling pathways.Properties of biomaterials like topography,wettability,surface charge,the release of cytokines,mediators,ions and other bioactive molecules can impose effects on immune responses to interfere with the skeletal system.Based on the bone formation mechanisms,the designing methods of the biomaterials change from immune evasive to immune reprogramming.Here,we discuss the osteoimmunomodulatory effects of the new modification strategies—adjusting properties of bone biomaterials to induce a favorable osteoimmune environment.Such strategies showed potential to benefit the development of bone materials and lay a solid foundation for the future clinical application.Yajuan Xie Cheng Hu Yi Feng Danfeng Li Tingting Ai Yulei Huang Xiaodan Chen Lijia Huang Jiali Tan 2020Regenerative Biomaterials2020,7,3:22
