| 2 | Carbon Turnover in a Crop Rotation Under Free Air CO_2 Enrichment (FACE)显示文摘. Mostly based on assumptions derived from controlled-environment studies, predicted future atmospheric CO2 concen- trations [CO2] are expected to have considerable impacts on carbon (C) turnover in agro-ecosystems. In order to allow the in situ examination of C-transformations in the plant-soil system of arable crop rotations under future [CO2], a free air carbon dioxide enrichment (FACE) experiment (550 μmol mol-1 CO2) was started at Braunschweig, Germany in 1999. The crop rotation under investigation comprised winter barley, a cover crop (ryegrass), sugar beets and winter wheat. Assessments of CO2 effects included the determination of above- and belowground biomass production, measurements of canopy CO2- and H2O- fluxes, soil microbial biomass and in situ soil respiration. The results obtained during the 1st crop rotation cycle (3 years) showed that for the selected crops elevated [CO2] entailed significant positive effects (P < 0.05) on aboveground (6%-14% stimulation) and belowground biomass production (up to 90% stimulation), while canopy evapotranspiration was reduced. This resulted in increased soil water content. Also, depending on crop type and season, high CO2 stimulated in situ soil respiration (up to 30%), while soil microbial biomass did not show significant responses to elevated [CO2] during the first rotation cycle. | H. | 5 | HASTY modulates miRNA biogenesis by linking pri-miRNA transcription and processing显示文摘Post-transcriptional gene silencing mediated by microRNAs(miRNAs)modulates numerous developmental and stress response pathways.For the last two decades,HASTY(HST),the ortholog of human EXPORTIN 5,was considered to be a candidate protein that exports plant miRNAs from the nucleus to the cytoplasm.Here,we report that HST functions in the miRNA pathway independent of its cargo-exporting activity in Arabidopsis.We found that Arabidopsis mutants with impaired HST shuttling exhibit normal subcellular distribution of miRNAs.Interestingly,protein-protein interaction and microscopy assays showed that HST directly interacts with the microprocessor core component DCL1 through its N-terminal domain.Moreover,mass spectrometry analysis revealed that HST also interacts independently of its N-terminal domain with the mediator complex subunit MED37.Further experiments revealed that HST could act as a scaffold to facilitate the recruitment of DCL1 to genomic MIRNA loci by stabilizing the DCL1-MED37 complex,which in turn promotes the transcription and proper processing of primary miRNA transcripts(primiRNAs).Taken together,these results suggest that HST is likely associated with the formation of the miRNA biogenesis complex at MIRNA genes,promoting the transcription and processing of primiRNAs rather than the direct export of processed miRNAs from the nucleus. | Damian A.Cambiagno Axel J.Giudicatti Agustin L.Arce Delfina Gagliardi Lei Li Wei Yuan Derek S.Lundberg Detlef Weigel Pablo A.Manavella | 2021 | Molecular Plant2021,14,3: | 5 |

| 10 | Understanding the physiological functions of the host xenobiotic-sensing nuclear receptors PXR and CAR on the gut microbiome using genetically modified mice显示文摘Pharmacological activation of the xenobiotic-sensing nuclear receptors pregnane X receptor(PXR) and constitutive androstane receptor(CAR) is well-known to increase drug metabolism and reduce inflammation. Little is known regarding their physiological functions on the gut microbiome. In this study, we discovered bivalent hormetic functions of PXR/CAR modulating the richness of the gut microbiome using genetically engineered mice. The absence of PXR or CAR increased microbial richness, and absence of both receptors synergistically increased microbial richness. PXR and CAR deficiency increased the pro-inflammatory bacteria Helicobacteraceae and Helicobacter. Deficiency in both PXR and CAR increased the relative abundance of Lactobacillus, which has bile salt hydrolase activity, corresponding to decreased primary taurine-conjugated bile acids(BAs) in feces, which may lead to higher internal burden of taurine and unconjugated BAs, both of which are linked to inflammation, oxidative stress, and cytotoxicity. The basal effect of PXR/CAR on the gut microbiome was distinct from pharmacological and toxicological activation of these receptors. Common PXR/CAR-targeted bacteria were identified, the majority of which were suppressed by these receptors. h PXR-TG mice had a distinct microbial profile as compared to wild-type mice.
