| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Down-regulation of Hsp90 could change cell cycle distribution and increase drug sensitivity of tumor cells显示文摘:AIM To construct Hsp90 antisense RNAeukaryotic expression vector, transfect it intoSGC7901 and SGC7901/VCR of MDR-type humangastric cancer cell lines, HCC7402 of humanhepatic cancer and Eel09 of human esophagealcancer cell lines, and to study the cell cycledistribution of the gene transected cells andtheir response to chemotherapeutic drugs.METHODS A I .03kb cDNA sequence of Hsp90Pwas obtained from the primary plasmid phHsp90by EcoR 1 and BamH I nuclease digestion andwas cloned to the EcoR 1 and BamH 1 site ofthe pcDNA by T4DNA ligase and an antisenseorientation of Hsp900 expression vector wasconstructed. The constructs were transfectedwith lipofectamine and positive clones wereselected with G418. The expression of RNA wasdetermined with dot blotting and RNaseprotection assay, and the expression of Hsp90protein determined with Western blot. Cell cycledistribution of the transfectants was analyzedwith flow cytometry, and the drug sensitivity ofthe transfectants to adriamycin (ADR ),vincrinstine (VCR ), mitomycin (MMC ) andcyclophosphamide (CTX ) with MTT andintracellular drug concentration of thetransfectants was determined with flowcytometry.RESULTS In EcoR 1 and BamH I restrictionanalysis, the size and the direction of the clonedsequence of Hsp900 remained what had beendesigned and the gene constructs were namedpcDNA-Hsp90. AH^SGC7901, AH^SGC7901/ VCR,AH-HCC7402 and AH-Eel09 cell clones allexpressed Hsp90 anti--sense RNA. Theexpression of Hsp90 was down--regulated in AHSGC7901, AH--SGC7901/ VCR, AH-HCC7402 andAH--Eel09 cell clones. Cell cycle distribution waschanged differently. In AH-SGC7901/ VCR andAH-Ec109 cells, G, phase cells were increased; Sphase and G, phase cells were decreased ascompared with their parental cell lines. In AHSGC7901 cell, G, phase cells were decreased, Qphase cells increased and S phase cells were notchanged, and in AH-HCC7402 cells G,, S and qphase cells remained unchanged as comparedwith their parental cell lines. | 4 | Humification characterization of biochar and its potential as a composting amendment显示文摘Biochar has received increasing attention due to its applications as a soil amendment. Here, the chemical properties of solid and water-extractable fractions of four biochar samples were investigated. The results showed that wood biochar and bamboo biochar samples were 60%–80% more hydrophobic than those of rice husk biochar and rice husk ash. In addition, the acidity was 3.88 mmol/g from the total functional groups and 1.03 mmol/g from the carboxyl groups/lactones/phenols found in the wood biochar sample, which were about 1.5 times greater than those of the bamboo biochar sample. These functional groups could be used to determine the sorptive capacity of biochar for ionic solutes and water content and to increase the degradation of compost organics. The wood biochar sample was found to have the most humification materials(fulvic acid-like material + humic acid-like material) in the water-extractable fraction, which was 3–10 times higher than that in the rice husk biochar and rice husk ash; humified materials were not detected in the bamboo biochar sample. Humification materials in biochar may be involved in increasing the proportion of humic acid-like materials in humic-like substances within the compost product. | 7 | Lake sediment resuspension and caused phosphate release——a simulation study显示文摘Intact sediment cores and wet/dried surficial sediments sampled from the two sublakes in Taihu Lake, Meiliang Bay and Wuli Lake, were incubated in the laboratory to determine the effects of resuspension on internal phosphorus loading by simulating different resuspension events. Soluble reactive phosphorus (SRP) release from undisturbed core sediment sampled in the Meiliang Bay and Wuli Lake in July 1998 was 1 53 mg/m 2 and 2 24 mg/m 2 within 4 days, respectively. However during one hour experimental simulation of resuspension, SRP increased by 0 041 mg/L and 0 077 mg/L in the above cores, which indicate that a typical resuspension event in the lake would be accompanied by the release of 10 77 mgSRP/m 2 and 23 1 mgSRP/m 2, respectively. The internal phosphorus loading induced by resuspension is estimated to be 8-10 times greater than the release from undisturbed sediment. SRP release from the dried sediments during simulation of resuspension was mainly dependent on the disturbing intensity. | 8 | Quantitative trait loci(QTLs)for quality traits related to protein and starch in wheat显示文摘Quality traits in wheat (Triticum aestivum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99–40.52% of phe- notypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. | 11 | Epidemiologic study on human immunodeficiency virus infection among children in a former paid plasma donating community in China显示文摘Background Illegal plasma collecting activities in mid 1990s caused a large number of human immunodeficiency virus (HIV) infections in rural areas of central-eastern China. Although most of these activities have been stopped, there were few reports on secondary transmission from infected former plasma donors to their spouses and from infected mothers to their children. This study was to determine the extent of HIV infections among young children in a rural community with a large proportion of plasma donors.Methods A survey was conducted among children aged under 7 years in a former plasma donating community in September 2000: finger blood was collected for HIV antibody testing. Another survey was repeated among children aged under 8 years and their families in the same community in April 2001: urine samples were collected for HIV testing. HIV positive children and samples of HIV negative children, whose mothers were positive based on 2001 survey, were followed up until September 2002 to investigate HIV seroconversion, disease progression and HIV strain analysis. Questionnaires were administered to collect information on children’s delivery, breast feeding, medical history and their parents’ commercial blood donation history and HIV status.Results Among 169 children surveyed in 2000, 10 (5.9%) were HIV positive. Of 224 children, 11 were positive in 2001. The overall prevalence rate in the two surveys was 5.0% (17/337) when counting 56 repeated children only once. Of children born to HIV positive mothers, 28.9% were infected. A seroconversion rate of 2.5 per 100 child-years was observed by following up 28 HIV negative children. No statistically significant associations were found between children’s HIV infection and their histories of blood transfusion, surgery, immunization injection or medical injections. All infections were HIV-1 subtype B’ strain, the average dispersion rate is 7.4%. DNA sequence analysis showed a close relationship between the seroconverted children and their infected mothers.Conclusions HIV vertical transmissions in the rural former plasma donating community was significant. Intervention measures should be taken to prevent further transmission. It was estimated that the HIV spread in this community occurred in 1994 or even earlier. This study used restaurant FW leachates flowing through a 200 tons-waste/day biological treatment plant as a window to investigate the fate of antibiotics and antibiotic-resistance genes (ARGs) during the acceptance and treatment of FW.Sulfonamides (sulfamethazine,sulfamethoxazole) and quinolones (ciprofloxacin,enrofloxacin,ofloxacin) were detected during FW treatment,while tetracyclines,macrolides and chloramphenicols were not observable.ARGs encoding resistance to sulfonamides,tetracyclines and macrolides emerged in FW leachates.Material flow analysis illustrated that the total amount of antibiotics (except sulfamethazine) and ARGs were constant during FW treatment processes.Both the concentration and total amount of most antibiotics and ARGs fluctuated during treatment,physical processes (screening,centrifugation,solid-liquid and oil-water separation) did not decrease antibiotic or ARGs concentrations or total levels permanently;the affiliated wastewater treatment plant appeared to remove sulfonamides and most ARGs concentrations and total amount.Heavy metals Ni,Co and Cu were important for disseminating antibiotics concentrations and MGEs for distributing ARGs concentrations.Humic substances (fulvic acids,hydrophilic fractions),C-associated and N-associated contents were essential for the distribution of the total amounts of antibiotics and ARGs.Overall,this study implied that human food might not be free of antibiotics and ARGs,and FW was an underestimated AR pool with various determinants.Nonetheless,derived hazards of FW could be mitigated through biological treatment with well-planned daily operations. | Pinjing He Zhuofeng Yu Liming Shao Yizhou Zhou Fan Lü | 2019 | Journal of Environmental Sciences2019,31,11: | 7 |
| 14 | Importance of storage time in mesophilic anaerobic digestion of food waste显示文摘Storage was used as a pretreatment to enhance the methanization performance of mesophilic anaerobic digestion of food waste. Food wastes were separately stored for 0, 1,2, 3, 4, 5, 7, and 12 days, and then fed into a methanogenic reactor for a biochemical methane potential(BMP) test lasting up to 60 days. Relative to the methane production of food waste stored for 0–1 day(285–308 m L/g-added volatile solids(VSadded)), that after2–4 days and after 5–12 days of storage increased to 418–530 and 618–696 m L/g-VSadded,respectively. The efficiency of hydrolysis and acidification of pre-stored food waste in the methanization reactors increased with storage time. The characteristics of stored waste suggest that methane production was not correlated with the total hydrolysis efficiency of organics in pre-stored food waste but was positively correlated with the storage time and acidification level of the waste. | 16 | Observation on the Effect of Acupoint Injection plus Massage Therapy on Primary Dysmenorrhea显示文摘Objective: To observe the clinical effects of acupoint injection therapy plus massage on primary dysmenorrhea(PD). Methods: Ninety patients with PD were randomly divided into a treatment group or a control group, 45 cases in each group. The treatment group was treated by injection of Vitamin K3 into Sanyinjiao(SP 6) plus massage on Diji(SP 8). The control group was treated by oral administration of Ibuprofen sustained-release capsule. Before and after the treatment, visual analogue scale(VAS) was adopted to assess pain degree of the patients. The therapeutic effects were observed after continuous treatment of three cycles of menstruation. Results: After treatment, VAS scores were obviously decreased in both groups and the differences were statistically significant(all P<0.05). The difference in VAS score after treatment was more significant in the treatment group than that in the control group(P<0.01). | 18 | Occurrence and molecular characterization of Potato spindle tuber viroid(PSTVd) isolates from potato plants in North China显示文摘China is the largest potato producing country worldwide,with this crop representing the fourth largest staple food crop in China.However,the steady presence of Potato spindle tuber viroid(PSTVd) over the past five decades has a significant economic impact on potato production.To determine why PSTVd control measures have been ineffective in China,more than 1 000 seed potatoes collected between 2009 and 2014 were subjected to PSTVd detection at the Supervision and Testing Center for Virus-free Seed Potatoes Quality,Ministry of Agriculture,China.A high PSTVd infection rate(6.5%) was detected among these commercial seed potatoes.Some breeding lines of potato collected from 2012 to 2015 were also tested for PSTVd infection,revealing a high rate of PSTVd contamination in these potato propagation materials.Furthermore,comparison of the full-length sequences of 71 different Chinese PSTVd isolates revealed a total of 74 predominant PSTVd variants,which represented 42 different sequence variants of PSTVd.Comparative sequence analysis revealed 30 novel PSTVd sequence variants specific to China.Comprehensive phylogenetic analysis uncovered a close relationship between the Chinese PSTVd sequence variants and those isolated from Russia.It is worth noting that three intermediate strains and six mild strains were identified among these variants.These results have important implications for explaining the ineffective control of PSTVd in China and thus could serve as a basic reference for designing more effective measures to eliminate PSTVd from China in the future. | QIU Cai-ling ZHANG Zhi-xiang LI Shi-fang BAI Yan-ju LIU Shang-wu FAN Guo-quan GAO Yan-ling ZHANG Wei ZHANG Shu Lü Wen-he Lü Dian-qiu | 2016 | Journal of Integrative Agriculture2016,15,2: | 5 |
