1Benzene containing polyhydroxyalkanoates homo-and copolymers synthesized by genome edited Pseudomonas entomophila显示文摘Microbial synthesis of functional polymers has become increasingly important for industrial biotechnology.For the first time,it became possible to synthesize controllable composition of poly(3-hydroxyalkanoate)(P3HA)consisting of 3-hydroxydodecanoate(3HDD)and phenyl group on the side-chain when chromosome of Pseudomonas entomophila was edited to weaken itsβ-oxidation.Cultured in the presence of 5-phenylvaleric acid(PVA),the edited P.entomophila produced only homopolymer poly(3-hydroxy-5-phenylvalerate)or P(3HPhV).While copolyesters P(3HPhV-co-3HDD)of 3-hydroxy-5-phenylvalerate(3HPhV)and 3-hydroxydodecanoate(3HDD)were synthesized when the strain was grown on mixtures of PVA and dodecanoic acid(DDA).Compositions of 3HPhV in P(3HPhV-co-3HDD)were controllable ranging from 3%to 32%depending on DDDA/PVA ratios.Nuclear magnetic resonance(NMR)spectra clearly indicated that the polymers were homopolymer of P(3HPhV)and random copolymers of 3HPhV and 3HDD.Their mechanical and thermal properties varied dramatically depending on the monomer ratios.Our results demonstrated the possibility to produce tailor-made,novel functional PHA using the chromosome edited P.entomophila.SHEN Rui CAI LongWei MENG DeChuan WU LinPing GUO Kai DONG GuoXing LIU Lei CHEN JinChun WU Qiong CHEN GuoQiang 2014Science China(Life Sciences)2014,57,1:4
2A Filter-Based Pattern Search Method for Unconstrained Optimization显示文摘We discuss a filter-based pattern search method for unconstrained optimization in this paper. For the purpose to broaden the search range we use both filter technique and frames, which are fragments of grids, to provide a new criterion of iterate acceptance. The convergence can be ensured under some conditions. 3Cation-vacancy induced Li+ intercalation pseudocapacitance at atomically thin heterointerface for high capacity and high power lithium-ion batteries显示文摘It is challenging to create cation vacancies in electrode materials for enhancing the performance of rechargeable lithium ion batteries (LIBs). Herein, we utilized a strong alkaline etching method to successfully create Co vacancies at the interface of atomically thin Co_(3−x)O_(4)/graphene@CNT heterostructure for high-energy/power lithium storage. The creation of Co-vacancies in the sample was confirmed by high-resolution scanning transmission electron microscope (HRSTEM), X-ray photoelectron spectroscopy (XPS) and electron energy loss near-edge structures (ELNES). The obtained Co_(3−x)O_(4)/graphene@CNT delivers an ultra-high capacity of 1688.2 mAh g^(−1) at 0.2 C, excellent rate capability of 83.7% capacity retention at 1 C, and an ultralong life up to 1500 cycles with a reversible capacity of 1066.3 mAh g^(−1). Reaction kinetic study suggests a significant contribution from pseudocapacitive storage induced by the Co-vacancies at the Co_(3−x)O_(4)/graphene@CNT interface. 4Effects of initial particle gradation and rock content on crushing behaviors of weathered phyllite fills——A case of eastern Ankang section of Shiyan—Tianshui highway,China显示文摘The objective of this paper is to investigate the effects of initial particle gradation and rock content on the crushing behavior(i.e.grain size before and after crushing) of weathered phyllite fills.Compaction tests were conducted on weathered phyllite fills with rock contents of 35%,45%,55%,65% and 75%(by weight).First,the particle size distributions(PSDs) were observed before and after compaction,and then the particle breakage of weathered phyllite fills was analyzed by fractal dimension.Relative fractal dimension was proposed to evaluate the effects of initial rock content and initial gradation on the particle breakage.It was found that the fractal dimension method can well characterize the crushing behaviors of the weathered phyllite fills.The finer the fills were,the more they were compacted.That is,after the first compaction,the relative fractal dimension of the weathered phyllite fills increased as the rock content increased,reaching the values of 0.013,0.016,0.024,0.037 and 0.08,respectively.After the second compaction,these relative fractal dimension values,dominated by the initial particle gradation,became 0.059,0.072,0.052,0.095 and 0.118,respectively.In conclusion,the weathered phyllite fills with 55% rock content exhibited the least breakage and were most suitable for filling the subgrade.Findings in this paper will provide significant guidance for the construction of weathered phyllite filling subgrade in future projects.Feifei Liu Xuesong Mao Yushuo Fan Linping Wu Wei Victor Liu 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,2:2
5Is 1.28 parts per million biomarker specific for neural progenitor cells?显示文摘Nuclear magnetic resonance-visible mobile lipid,at 1.28 parts per million(ppm),is thought to be due to mobile lipid droplets formed in cells and has been considered unique for neural progenitor cells.However,this idea remains controversial.The present study examined the 1.28 ppm biomarker in other stem cells and non-stem cells,and explored the relationship between 1.28 ppm biomarker and mobile lipid droplets.1H nuclear magnetic resonance spectroscopy of EC109 cells,mesenchymal stem cells(MSCs) and adipogenic cells differentiated from MSCs was performed.Results show that 1.28 ppm biomarker was observed in human MSCs,but was absent from EC109 cells.Following adipogenic differentiation induced for 2 weeks,the 1.28 ppm biomarker climbed remarkably,with mobile lipid droplet generation,suggesting that the 1.28 ppm biomarker is not specific for neural progenitor cells because it is also observed in MSCs and adipogenic-induced differentiated cells.Moreover,it is possible to monitor MSCs differentiation following cell transplantation,using 1.28 ppm biomarker changes.Zhifeng Xu Chongyang Shen Haiqiang Zhu Yaowen Chen Linping Wu Peng Huang YeyuXiao Zhiwei Shen Li Pang Xiuqin Guo Lian Ma Renhua Wu 2010Neural Regeneration Research2010,5,15:2

9A novel PD-L1 targeting peptide self-assembled nanofibers for sensitive tumor imaging and photothermal immunotherapy in vivo显示文摘Programmed death 1(PD-1)and its ligand PD-L1 are two typical immune checkpoints.Antibody-based immune checkpoint blockade(ICB)strategy targeting PD-1/PD-L1 achieved a significant therapeutic effect on cancer.However,due to the impenetrability of antibody drugs and the occurrence of immune-related adverse events,only a minority of patients benefit from this treatment.Peptides multimerization has been widely proved to be an effective method to improve receptor binding affinity through a multivalent synergistic effect.In this study,we report a novel peptide-aggregation-induced emission(AIE)hybrid supramolecular TAP,which can self-assemble into nanofibers through non-covalent interactions such as hydrogen bonds,with a specific nanomolar affinity to PD-L1 in vivo and in vitro.Combined with near-infrared agents,it can be used for tumor imaging and photothermal therapy,which enables photothermal ablation of cancer cells for generating tumor-associated antigen(TAA)and triggering a series of immunological events.Collectively,our work suggests that synthetic self-assembled peptide nanofibers can be developed as attractive platforms for active photothermal immunotherapies against cancer.Linping Fu Jianhu Zhang Chenchen Wu Weizhi Wang Dong Wang Zhiyuan Hu Zihua Wang 2022Nano Research2022,15,8:0

11Effects of electrostimulation and administration of succinylcholine on the expression of Fos protein in mesencephalic periaqueductal gray matter of rats after simulated weightlessness显示文摘BACKGROUND: Expression of Fos in neurons of periaqueductal gray (PAG) is used to reflect the excitability. However, changes of expression of Fos in neurons of PAG are caused by injured electrostimulation after simulated weightlessness, and the relationship between pretreatment and injection of succinylcholine has not been determined yet. OBJECTIVE: To investigate the changes of expression of Fos in PAG induced by injured electrostimulation, pretreatment and injection of succinylcholine at 2 weeks after simulated weightlessness. DESIGN: Observational and controlled animal study. SETTING: Department of Physiology, Medical School, Xi’an Jiaotong University; Key Laboratory of Environment and Genes Related to Diseases, Ministry of Education. MATERIALS: A total of 24 adult female SD rats, of clean grade and weighing 180-220 g, were selected in this study. METHODS: The experiment was completed in the Experimental Animal Center of Xi’an Jiaotong University. ① All rats were randomly divided into 2 groups according to body mass: simulated weightlessness group and control group with 12 in each group. And then, each group was also divided into 3 subgroups: electrostimulation group, succinylcholine-pretreatment group and succinylcholine-injection group with 4 in each subgroup. ② The model of weightlessness was simulated by tail-suspended female rats, which were described and modified by Cheng Jie. Rats in normal control group were given the same interventions as simulated weightlessness group except for tail-suspended. ③ Experimental method: The rats in electrostimulation group were given nociceptive stimulus by a pair of subcutaneous electrodes inserted into 1 and 5 claw of left hindlimb. The stimulus (current: 10 mA; duration: 1 ms; interval: 1 s) lasted for 30 minutes. The rats in succinylcholine-pretreatment group received stimulus after intravenous administration of succinylcholine, rats in succinylcholine-injection group were not given stimulus, just received succinylcholine. ④ All rats were perfused and fixed after 2 hours from the end of stimulation. The brains were removed, and serial frozen sections of midbrain were stained using immunocytochemical method, observed and taken photos under light-microscope. The number and morphological characters of Fos-immunoreactive (Fos-IR) neurons in ventrolateral part of PAG were investigated. MAIN OUTCOME MEASURES: The alterations in number and morphological characters of Fos-IR neurons in ventrolateral PAG of all rats. RESULTS: A total of 24 rats were involved in the final analysis. ① The morphological changes of Fos-IR neurons: The expressions of Fos in ventrolateral part of PAG were observed in both control and simulated weightlessness groups rats after being given nociceptive stimulus. As compared with control group, Fos-IR neurons in simulated weightlessness group were dyed lightly, cellular integrity was impaired, and cellular verge was unclear. ② The numbers of Fos-IR neurons: In control group, the numbers of Fos-IR neurons in ventrolateral part of PAG in simulated weightlessness group were obviously lower than succinylcholine-pretreatment group, but obviously higher than succinylcholine-injection group (46.94±3.38, 71.06±8.96 and 35.04±4.62, respectively, P < 0.05). In 14-day simulated weightlessness group, the numbers of Fos-IR neurons in electrostimulation group were also obviously lower than succinylcholine-pretreatment group, and obviously higher than succinylcholine-injection group (27.77±3.27, 32.91±2.99 and 11.75±1.00, respectively, P < 0.05). The numbers of Fos-IR neurons in all subgroups in control group were obviously higher than those subgroups in simulated weightlessness group. Compared with electrostimulation group, the percentage of expression of Fos in ventrolateral part of PAG responsed to nociceptive stimulus after administration of succinylcholine (SCH) was increased to 51.83% in control group and 18.51% in simulated weightlessness group. CONCLUSION: ① The expression of Fos in neurons in ventrolateral part of PAG were increased by the pretreatment of SCH before nociceptive stimulus. ② Nociceptive stimulus could increase the expression of Fos in neurons in ventrolateral part of PAG. ③ The numbers of Fos-IR neurons in ventrolateral part of PAG were decreased obviously after 2-week simulated weightlessness.Yongjin Zhu Sudi Wu Xiaoli Fan Xinai Song Linping Xu 2006Neural Regeneration Research2006,1,1:0
