1Undrained shear strength evaluation for hydrate-bearing sediment overlying strata in the Shenhu area, northern South China Sea显示文摘The undrained shear strength of shallow strata is a critical parameter for safety design in deep-water operations.In situ piezocone penetration tests(CPTU) and laboratory experiments are performed at Site W18-19 in the Shenhu area, northern South China Sea, where China's first marine hydrate exploitation operation is due to be located. The validation of the undrained shear strength prediction model based on CPTU parameters. Different laboratory tests, including pocket penetrometer, torvane, miniature vane and unconsolidated undrained triaxial tests, are employed to solve empirical cone coefficients by statistical and mathematical methods. Finally, an optimized model is proposed to describe the longitudinal distribution of undrained shear strength in calcareous clay strata in the Shenhu area. Research results reveal that average empirical cone coefficients based on total cone resistance, effective resistance, and excess-pore pressure are 13.8, 4.2 and 14.4, respectively. The undrained shear strength prediction model shows a good fit with the laboratory results only within specific intervals based on their compaction degree and gas-bearing conditions. The optimized prediction model in piecewise function format can be used to describe the longitudinal distribution of the undrained shear strength for calcareous clay within all depth intervals from the mud-line to the upper boundary of hydrate-bearing sediments(HBS). The optimized prediction result indicates that the effective cone resistance model is suitable for very soft to firm calcareous clays,the excess-pore pressure model can depict the undrained shear strength for firm to very stiff but gas-free clays,while the total cone resistance model is advantageous for evaluating the undrained shear strength for very stiff and gassy clays. The optimized model in piecewise function format can considerably improve the adaptability of empirical models for calcareous clay in the Shenhu area. 2Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. 3QTL analysis of ear leaf traits in maize(Zea mays L.)under different planting densities显示文摘Modem maize varieties have become more productive than ever, owing largely to increased tolerance of high plant density. However, the genetics of ear leaf traits under different densities remains poorly understood. In this study, Zhongdan 909 recombinant inbred lines(RILs) derived from a cross between Z58 and HD568 were genotyped for 3072 single-nucleotide polymorphisms(SNPs), and phenotyped for leaf length(LL), leaf width(LW), and leaf angle(LA) of the uppermost ear leaf under three planting densities(52,500,67,500, and 82,500 plants ha-1, respectively). A genetic map was then constructed using1358 high-quality SNPs. The total length of the linkage map was 1985.2 cM and the average interval between adjacent markers 1.46 cM. With increasing density, LL and LW decreased from 63.68 to 63.02 cm and from 8.56 to 8.21 cm, respectively, while LA increased from19.42° to 19.66°. All three traits had high heritabilities, of 0.75, 0.78, and 0.84, respectively.Using inclusive composite interval mapping, 23, 25, and 17 quantitative trait loci(QTL) were detected for LL, LW, and LA, respectively. Of these, 35 were simultaneously detected under two or three plant densities, while 30 were detected under only one. 4Synchrotron radiation X-ray powder diffraction techniques applied in hydrogen storage materials--A review显示文摘Synchrotron radiation is an advanced collimated light source with high intensity. It has particular advantages in structural characterization of materials on the atomic or molecular scale. Synchrotron radiation X-ray powder diffraction(SR-XRPD) has been successfully exploited to various areas of hydrogen storage materials. In the paper, we will give a brief introduction on hydrogen storage materials, X-ray powder diffraction(XRPD), and synchrotron radiation light source. The applications of ex situ and in situ time-resolved SR-XRPD in hydrogen storage materials, are reviewed in detail. 5Conversion of a normal maize hybrid into a waxy version using in vivo CRISPR/Cas9 targeted mutation activity显示文摘Waxy maize is a specialty maize that produces mainly amylopectin starch with special food or industrial values. The objective of this study was to overcome the limitations of wx mutant allele acquisition and breeding efficiency by conversion of parental lines from normal to waxy maize. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. 6Microstructure and properties of indium tin oxide thin films deposited by RF-magnetron sputtering显示文摘Tin-doped indium oxide (ITO) thin films were prepared using conventional radio frequency (RF) planar magne-tron sputtering equipped with IR irradiation using a ceramic target of In2O3/SnO2 with a mass ratio of 1:1 at various IR irradiation temperatures TI (from room temperature to 400°C). The refractive index, deposited ratio, and resistivity are functions of the sputtering Ar gas pressure. The microstructure of ITO thin films is related to IR TI, the crystalline seeds appear at TI = 300°C, and the films are amorphous at the temperature ranging from 27°C to 400°C. AFM investigation shows that the roughness value of peak-valley of ITO thin film (Rp-v) and the surface microstructure of ITO thin films have a close relation with TI. The IR irradiation results in a widening value of band-gap energy due to Burstein-Moss effect and the maximum visible transmittance shifts toward a shorter wavelength along with a decrease in the film’s refractive index. The plasma wavelength and the refractive index of ITO thin films are relative to the TI. XPS investigation shows that the photoelectrolytic properties can be deteriorated by the sub-oxides. 7A phase 3,double-blind,randomized placebo-controlled efficacy and safety study of abiraterone acetate in chemotherapynaı¨ve patients with mCRPC in China,Malaysia,Thailand and Russia显示文摘Objective:This double-blind,placebo-controlled phase 3 study was designed to compare efficacy and safety of abiraterone acetate+prednisone(abiraterone)to prednisone alone in chemotherapy-naı¨ve,asymptomatic or mildly symptomatic metastatic castrationresistant prostate cancer(mCRPC)patients from China,Malaysia,Thailand and Russia.Methods:Adult chemotherapy-naı¨ve patients with confirmed prostate adenocarcinoma,Eastern Cooperative Oncology Group(ECOG)performance status(PS)grade 0e1,ongoing androgen deprivation(serum testosterone<50 ng/dL)with prostate specific antigen(PSA)or radiographic progression were randomized to receive abiraterone acetate(1000 mg,QD)t prednisone(5 mg,BID)or placebo t prednisone(5 mg,BID),until disease progression,unacceptable toxicity or consent withdrawal.Primary endpoint was improvements in time to PSA progression(TTPP).Results:Totally,313 patients were randomized(abiraterone:n Z 157;prednisone:n Z 156);and baseline characteristics were balanced.At clinical cut-off(median follow-up time:3.9 months),80% patients received treatment(abiraterone:n Z 138,prednisone:n Z 112).Median time to PSA progression was not reached with abiraterone versus 3.8 months for prednisone,attaining 58%reduction in PSA progression risk(HR=0.418;p<0.0001).Abirateronetreated patients had higher confirmed PSA response rate(50%vs.21%;relative odds=2.4;p<0.0001)and were 5 times more likely to achieve radiographic response than prednisonetreated patients(22.9%vs.4.8%,p=0.0369).Median survival was not reached.Most common(≥10% abiraterone vs.prednisone-treated)adverse events:bone pain(7%vs.14%),pain in extremity(6%vs.12%),arthralgia(10%vs.8%),back pain(7%vs.11%),and hypertension(15%vs.14%).Conclusion:Interim analysis confirmed favorable benefit-to-risk ratio of abiraterone in chemotherapy-naı¨ve men with mCRPC,consistent with global study,thus supporting use of abiraterone in this patient population.Dingwei Ye Yiran Huang Fangjian Zhou Keji Xie Vsevolod Matveev Changling Li Boris Alexeev Ye Tian Mingxing Qiu Hanzhong Li Tie Zhou Peter De Porre Margaret Yu Vahid Naini Hongchuan Liang Zhuli Wu Yinghao Sun 2017Asian Journal of Urology2017,4,2:3
8Drift potential of UAV with adjuvants in aerial applications显示文摘The reduction of pesticide aerial spraying drift is still one of the major challenges in modern agriculture.The aim of this study was to evaluate the drift potential of different types of unmanned aerial vehicle(UAV)and adjuvant products for reducing spray drift in aerial applications.Three types of UAV(3WQF120-12 and 3CD-15 fuel oil powered single-rotor UAV and HY-B-15L battery powered single-rotor UAV)were selected in this study with regular application parameters to compare each spray drift,and 3WQF120-12 fuel oil powered UAV was selected to quantify spray drift of 6 adjuvants dissolved in water under field conditions.Solutions were marked with brillant sulfoflavin dye(BSF)at 0.1%.Petri dishes and rotary impactors were used to collect airborne and sediment drift,respectively.Drift deposits were evaluated by spectrophotometry in order to quantify deposits.The results showed that when the flight height was 1.5-2.0 m above the crop at the flight speed of 4-5 m/s and the average wind speed of 1.63-1.73 m/s,3WQF120-12 fuel oil powered UAV had lower drift potential than the other two types;DV0.5 and percentage of droplets with diameter≤75μm had very significant effects on spray drift percentage(p=0.01);the risk of drift in agricultural spraying could be significantly decreased not only by reducing the percentage of fine droplets but also by changing droplet spectra.Compared to water,Silwet DRS-60,ASFA+B,T1602,Break-thru Vibrant,QF-LY and Tmax could reduce by 65%,62%,59%,46%,42%,and 19%spray drift,respectively.when water without adjuvants were sprayed,90%of drift droplets were located within a range of 10.1 m of the target area while with 0.8%Silwet DRS-60 adjuvant in water,the distance was shortened to 6.4 m.Xiaonan Wang Xiongkui He Jianli Song Zhichong Wang Changling Wang Shilin Wang Ruochen Wu Yanhua Meng 2018International Journal of Agricultural and Biological Engineering2018,11,5:3
9Genetic analysis and QTL mapping of stalk cell wall components and digestibility in maize recombinant inbred lines from B73 × By804显示文摘The cell wall composition and structure of the maize stalk directly affects its digestibility and in turn its feed value.Previous studies of stem quality have focused mostly on common maize germplasm,and few studies have focused on high-oil cultivars with high grain and straw quality.Investigation of the genetic basis of cell wall composition and digestibility of maize stalk using high-oil maize is desirable for improving maize forage quality.In the present study,a high-oil inbred line(By804)was crossed as male parent with the maize inbred line B73 to construct a population of 188 recombinant inbred lines(RILs).The phenotypes of six cell-wall-related traits were recorded,and QTL analysis was performed with a genetic map constructed with SNP markers.All traits were significantly correlated with one another and showed high broad-sense heritability.Of 20 QTLs mapped,the QTL associated with each trait explained 10.0%–41.1%of phenotypic variation.Approximately half of the QTL each explained over 10%of the phenotypic variation.These results provide a theoretical basis for improving maize forage quality by marker-assisted selection.Qi Wang Kun Li Xiaojiao Hu Huimin Shi Zhifang Liu Yujin Wu Hongwu Wang Changling Huang 2020The Crop Journal2020,8,1:2
10Relationship Between Soil Properties and Different Fractions of Soil Hg显示文摘Correlation and path analysis methods were used to study the relationship between soil properties and the distribution of different soil Hg fractions with nine representative soils from Chongqing, China. Results showed that clay (< 2 m) could increase water-soluble Hg (r = 0.700*). Soil organic matter (OM) could enhance the increase of elemental Hg (r = 0.674*). The higher the base saturation percentage (BSP), the more the residual Hg (T = 0.684*). Organic Hg, the sum of acid-soluble organic Hg and alkali-soluble Hg, was positively affected by silt (2-20 μm) but negatively affected by pH, with the direct path coefficients amounting to 1.0487 and 0.5121, respectively. The positive effect of OM and negative effect of BSP on organic Hg were the most significant, with the direct path coefficients being 0.7614 and -0.8527, respectively. The indirect effect of clay (< 2 μm) via BSP (path coefficient = 0.4186) was the highest, showing that the real influencing factor in the effect of clay (< 2 μm) on acid-soluble organic Hg was BSP. 12Ribonuclease H-like gene SMALL GRAIN2 regulates grain size in rice through brassinosteroid signaling pathway显示文摘Grain size is a key agronomic trait that determines the yield in plants.Regulation of grain size by brassinosteroids(BRs)in rice has been widely reported.However,the relationship between the BR signaling pathway and grain size still requires further study.Here,we isolated a rice mutant,named small grain2(sg2),which displayed smaller grain and a semi-dwarf phenotype.The decreased grain size was caused by repressed cell expansion in spikelet hulls of the sg2 mutant.Using map-based cloning combined with a Mut Map approach,we cloned SG2,which encodes a plant-specific protein with a ribonuclease H-like domain.SG2 is a positive regulator downstream of GLYCOGEN SYNTHASE KINASE2(GSK2)in response to BR signaling,and its mutation causes insensitivity to exogenous BR treatment.Genetical and biochemical analysis showed that GSK2 interacts with and phosphorylates SG2.We further found that BRs enhance the accumulation of SG2 in the nucleus,and subcellular distribution of SG2 is regulated by GSK2 kinase activity.In addition,Oryza sativa OVATE family protein 19(OsOFP19),a negative regulator of grain shape,interacts with SG2 and plays an antagonistic role with SG2 in controlling gene expression and grain size.Our results indicated that SG2 is a new component of GSK2-related BR signaling response and regulates grain size by interacting with Os OFP19.Yunshuai Huang Hui Dong Changling Mou Ping Wang Qixian Hao Min Zhang Hongmin Wu Fulin Zhang Tengfei Ma Rong Miao Kai Fu Yaping Chen Ziyan Zhu Cheng Chen Qikai Tong Zhuoran Wang Shirong Zhou Xi Liu Shijia Liu Yunlu Tian Ling Jiang Jianmin Wan 2022Journal of Integrative Plant Biology2022,64,10:1
13The Variation of Microbial(Methanotroph)Communities in Marine Sediments Due to Aerobic Oxidation of Hydrocarbons显示文摘Methanotrophs in marine sediments and overlying water attenuate the emissions of methane into the atmosphere and thus play an important role for the global cycle of this greenhouse gas.However,gas released from natural hydrocarbon seeps are not pure methane but commonly mixed hydrocarbons.Currently,how methanotrophic bacteria behave in the co-presence of methane and heavier hydrocarbons remains unknown.In this paper,the bacteria were cultured aerobically in fresh sediment samples(collected from Bohai Bay in eastern China)at 28℃under the atmospheres of pure methane and methane+ethane+propane mixed gas,respec-tively.The prevailing terrigenous n-alkanes and fatty acids in the original sediment samples varied consistently after incubations,confirming the proceeding of aerobic bacterial activities.The real-time quantitative PCR assay and sequencing of the 16S rRNA and particulate methane monooxygenase(pmoA)genes revealed the changes of microbe communities to a methanotroph-dominating structure after incubations.Particularly,after incubations the family Methylococcaceae(typeⅠmethanotrophs)became dominant with proportions higher than 40%,whereas Methylocystaceae(typeⅡmethanotrophs)nearly disappeared in all incubated samples.More-over,the species of methanotrophs from the samples treated with pure methane were dominated by Methylobacter luteus,whereas Methylobacter whittenburyi took the predominant proportion in the samples treated with mixed gas.The phenomenon suggests that some methanotrophs may also utilize ethane and propane.Collectively,this study may help to gain a better understanding of the ef-fects and contributions of microbial activities in marine hydrocarbon seep ecosystems.LI Jing LIU Changling WU Nengyou HE Xingliang HAO Xiluo LI Feng CHEN Ye MENG Qingguo 2021Journal of Ocean University of China2021,20,3:1

17Primary gastric adenosquamous carcinoma: a case report and literature review显示文摘Primary gastric adenosquamous carcinoma (GASC) is exceedingly rare. It accounts for less than 1% of all gastric cancers. In this report, we describe our pathological findings along with a review of the literature to improve our understanding of the disease and reduce misdiagnosis, as well as to provide evidence for its treatment and prognosis. A 49-year-old male patient was admitted to our hospital (Dalian Municipal Central Hospital, Dalian, China) with a complaint of epigastric pain that had persisted for half a month. Physical examination, regular laboratory blood tests, and computed tomography revealed no obvious abnormalities. Gastroscopy revealed ulcers in the lower part of the stomach, and pathological assessment revealed adenocarcinoma. Radical gastrectomy was performed, and the folinic acid, fluorouracil, oxaliplatin (FOLFOX) chemotherapy regimen was administered postoperatively. Pathological assessment of the mass revealed a protruding tumor measuring 1.5 × 1.5 × 0.7 cm in the lower part of the stomach. The tumor infiltrated through the full wall of the stomach. This was confirmed by immunohistochemical (IHC) staining for cytokeratin (CK)(+), villin (-), p63 (++), and high-molecular-weight CK (+++). The patient remains alive with no recurrence more than seven years after surgery. Primary GASC is a rare malignant neoplasm. The diagnostic criteria for GASC mainly depend on the clinical, radiographic, and histopathological findings. Pathological assessment and IHC staining can be utilized to confirm the diagnosis. 18Barocaloric Material with High Thermal Conductivity for Room-Temperature Refrigeration显示文摘Barocaloric refrigeration technology,one of the caloric-effect refrigeration technologies,is drawing more and more attention.Neopentyl glycol(NPG)was reported to have a giant barocaloric effect,making it a potential barocaloric material.However,the high solid-solid(S-S)phase transition temperature and low thermal conductivity hinder the application of NPG in barocaloric refrigeration.This work lowers the S-S phase transition temperature and improves the thermal conductivity of the NPG-based barocaloric material.An NPG/TMP(TMP:Trimethylolpropane)binary system with an S-S phase transition temperature of 283.15 K is prepared,in which the mass ratio of TMP is 20%.Graphene is then added to the binary system to enhance thermal conductivity,and the optimal mass ratio of graphene was determined to be 5%.The thermal conductivity of this composite is 0.4 W/(m·K),increased by 110%compared to the binary system.To predict the effect of enhanced thermal conductivity on the cold-extraction process of the barocaloric refrigeration cycle,a numerical model is developed.The results show that the cold-extraction time of the barocaloric refrigeration cycle utilizing the composite with 5%graphene as the refrigerant is shortened by 50%compared with that using the binary system.ZHU Liutao DAI Zhaofeng GAO Yuanzhi WU Dongxu WANG Changling ZHAO Dongliang SHE Xiaohui DING Yulong ZHANG Xiaosong 2023Journal of Thermal Science2023,32,6:0
19CRISPR-powered optothermal nanotweezers:Diverse bio-nanoparticle manipulation and single nucleotide identification显示文摘Optothermal nanotweezers have emerged as an innovative optical manipulation technique in the past decade,which revolutionized classical optical manipulation by efficiently capturing a broader range of nanoparticles.However,the optothermal temperature field was merely employed for in-situ manipulation of nanoparticles,its potential for identifying bio-nanoparticles remains largely untapped.Hence,based on the synergistic effect of optothermal manipulation and CRIPSR-based bio-detection,we developed CRISPR-powered optothermal nanotweezers(CRONT).Specifically,by harnessing diffusiophoresis and thermo-osmotic flows near the substrate upon optothermal excitation,we successfully trapped and enriched DNA functionalized gold nanoparticles,CRISPR-associated proteins,as well as DNA strands.Remarkably,we built an optothermal scheme for enhancing CRISPR-based single-nucleotide polymorphism(SNP)detection at single molecule level,while also introducing a novel CRISPR methodology for observing nucleotide cleavage.Therefore,this innovative approach has endowed optical tweezers with DNA identification ability in aqueous solution which was unattainable before.With its high specificity and feasibility for in-situ bio-nanoparticle manipulation and identification,CRONT will become a universal tool in point-of-care diagnosis,biophotonics,and bio-nanotechnology.Jiajie Chen Zhi Chen Changle Meng Jianxing Zhou Yuhang Peng Xiaoqi Dai Jingfeng Li Yili Zhong Xiaolin Chen Wu Yuan Ho-Pui Ho Bruce Zhi Gao Junle Qu Xueji Zhang Han Zhang Yonghong Shao 2023Light(Science & Applications)2023,12,12:0
20Microstructure and properties of indium tin oxide thin films deposited by RF-magnetron sputtering显示文摘Tin-doped indium oxide (ITO) thin films were prepared using conventional radio frequency (RF) planar magnetron sputtering equipped with IR irradiation using a ceramic target of In2O3/SnO2 with a mass ratio of 1∶1 at various IR irradiation temperatures T1 (from room temperature to 400?℃). The refractive index,deposited ratio,and resistivity are functions of the sputtering Ar gas pressure. The microstructure of ITO thin films is related to IR T1,the crystalline seeds appear at T1=300?℃,and the films are amorphous at the temperature ranging from 27?℃ to 400?℃. AFM investigation shows that the roughness value of peak-valley of ITO thin film (R p-v ) and the surface microstructure of ITO thin films have a close relation with T1. The IR irradiation results in a widening value of band-gap energy due to Burstein-Moss effect and the maximum visible transmittance shifts toward a shorter wavelength along with a decrease in the film’s refractive index. The plasma wavelength and the refractive index of ITO thin films are relative to the T1. XPS investigation shows that the photoelectrolytic properties can be deteriorated by the sub-oxides.
