| 1 | Efficacy and safety of Xinglouchengqi decoction for acute ischemic stroke with constipation:study protocol for a randomized con-trolled trial显示文摘OBJECTIVE: To investigate the efficacy and safety of Xinglouchengqi(XLCQ) decoction in treatment of acute ischemic stroke with constipation.METHODS: In this prospective, multicenter, assessor-blinded, randomized controlled trial, 360 eligible participants will be randomized to the XLCQ group or the control group. Participants in the XLCQ group will receive XLCQ decoction, while those in the control group will undergo clysis therapy using glycerin enemas or oral administration of lactulose solution. Both groups will undergo a treatment period of(5 ± 2) d and a 1-month follow-up.The primary outcome measure will be the Constipation Scale score. The secondary outcome measures will include scores on the National Institutes of Health Stroke Scale, the Traditional Chinese Medicine(TCM) Stroke Scale, the Diagnostic Scale for TCM Syndromes of Ischemic Stroke and TCM Scale for Syndrome of Phlegm-heat and Fu-organ Excess.Therapeutic mechanism outcomes and safety outcomes will also be assessed. Assessments will be conducted at baseline, at the end of the treatment period, and at the follow-up. | 2 | Therapeutic window of Qingkailing injection for focal cerebral ischemia/reperfusion injury显示文摘The time window in which a drug is effective varies between drugs. The present study investigated the therapeutic window of Qingkailing injection for focal cerebral ischemia/reperfusion in mice. Animals underwent middle cerebral artery occlusion and were injected with Qingkailing (1.5, 3, 6 mL/kg). Infarct volume and neurological function were assessed after 24 hours of ischemia. In addition, to establish the therapeutic time window, mice were injected with 3 mL/kg Qingkailing at 0, 1, 3, 4, 6, 9 and 12 hours after occlusion. | 4 | Estimation of transpiration coefficient and aboveground biomass in maize using time-series UAV multispectral imagery显示文摘Estimating spatial variation in crop transpiration coefficients(CTc) and aboveground biomass(AGB)rapidly and accurately by remote sensing can facilitate precision irrigation management in semiarid regions. This study developed and assessed a novel machine learning(ML) method for estimating CTc and AGB using time-series unmanned aerial vehicle(UAV)-based multispectral vegetation indices(VIs)of maize under several irrigation treatments at the field scale. Four ML regression methods: multiple linear regression(MLR), support vector regression(SVR), random forest regression(RFR), and adaptive boosting regression(ABR), were used to address the complex relationship between CTcand VIs. AGB was then estimated using exponential, logistic, sigmoid, and linear equations because of their clear mathematical formulations based on the optimal CTcestimation model. The UAV VIs-derived CTcusing the RFR estimation model yielded the highest accuracy(R^(2)= 0.91, RMSE = 0.0526, and n RMSE = 9.07%). The normalized difference red-edge index, transformed chlorophyll absorption in reflectance index, and simple ratio contributed significantly to the RFR-based CTcmodel. The accuracy of AGB estimation using nonlinear methods was higher than that using the linear method. The exponential method yielded the highest accuracy(R^(2)= 0.76, RMSE = 282.8 g m, and n RMSE = 39.24%) in both the 2018 and 2019 growing seasons. | 19 | miR-335-5p regulates the proliferation,migration and phenotypic switching of vascular smooth muscle cells in aortic dissection by directly regulating SP1显示文摘Uncontrolled proliferation,migration and phenotypic switching of vascular smooth muscle cells(VSMCs)are important steps in the development and progression of aortic dissection(AD).The function and potential mechanism of miR-335-5p in the pathogenesis of AD are explored in this study.Specifically,the biological function of miR-335-5p is explored in vitro through CCK-8,Transwell,immunofluorescence,EdU,wound-healing,RT-qPCR and western blotting assays.In addition,an AD model induced by angiotensin II is used to investigate the function of miR-335-5p in vivo.A dual-luciferase assay is performed to verify the targeting relationship between miR-335-5p and specificity protein 1(SP1).Experiments involving the loss of SP1 function are performed to demonstrate the function of SP1 in the miR-335-5p-mediated regulation of human aortic-VSMCs(HA-VSMCs).AD tissues and platelet-derived growth factor BB(PDGF-BB)-stimulated HA-VSMCs show significant downregulation of miR-335-5p expression and upregulated SP1 expression.Overexpression of miR-335-5p effectively suppresses cell proliferation,migration and synthetic phenotype markers and enhances contractile phenotype markers induced by PDGF-BB treatment.Additionally,SP1 is identified as a target gene downstream of miR-335-5p,and its expression is negatively correlated with miR-335-5p in AD.Upregulation of SP1 partially reverses the inhibitory effect of miR-335-5p on HA-VSMCs,whereas the downregulation of SP1 has the opposite effect.Furthermore,Ad-miR-335-5p clearly suppresses aorta dilatation and vascular media degeneration in the AD model.Our results suggest that miR-335-5p inhibits HA-VSMC proliferation,migration and phenotypic switching by negatively regulating SP1,and indicate that miR-335-5p may be a potential therapeutic target in AD. | Runwei Ma Dayong Zhang Yi Song Jichang Kong Chunjie Mu Pin Shen Wenting Gui | 2022 | Acta Biochimica et Biophysica Sinica2022,54,7: | 0 |
| 20 | Effects of nitrogen and phosphorus additions on soil microbial community structure and ecological processes in the farmland of Chinese Loess Plateau显示文摘Microorganisms regulate the responses of terrestrial ecosystems to anthropogenic nutrient inputs.The escalation of anthropogenic activities has resulted in a rise in the primary terrestrial constraining elements,namely nitrogen(N)and phosphorus(P).Nevertheless,the specific mechanisms governing the influence of soil microbial community structure and ecological processes in ecologically vulnerable and delicate semi-arid loess agroecosystems remain inadequately understood.Therefore,we explored the effects of different N and P additions on soil microbial community structure and its associated ecological processes in the farmland of Chinese Loess Plateau based on a 36-a long-term experiment.Nine fertilization treatments with complete interactions of high,medium,and low N and P gradients were set up.Soil physical and chemical properties,along with the microbial community structure were measured in this study.Additionally,relevant ecological processes such as microbial biomass,respiration,N mineralization,and enzyme activity were quantified.To elucidate the relationships between these variables,we examined correlation-mediated processes using statistical techniques,including redundancy analysis(RDA)and structural equation modeling(SEM).The results showed that the addition of N alone had a detrimental effect on soil microbial biomass,mineralized N accumulation,andβ-1,4-glucosidase activity.Conversely,the addition of P exhibited an opposing effect,leading to positive influences on these soil parameters.The interactive addition of N and P significantly changed the microbial community structure,increasing microbial activity(microbial biomass and soil respiration),but decreasing the accumulation of mineralized N.Among them,N24P12 treatment showed the greatest increase in the soil nutrient content and respiration.N12P12 treatment increased the overall enzyme activity and total phospholipid fatty acid(PLFA)content by 70.93%.N and P nutrient contents of the soil dominate the microbial community structure and the corresponding changes in hydrolytic enzymes.Soil microbial biomass,respiration,and overall enzyme activity are driven by mineralized N.Our study provides a theoretical basis for exploring energy conversion processes of soil microbial community and environmental sustainability under long-term N and P additions in semi-arid loess areas. | KOU Zhaoyang LI Chunyue CHANG Shun MIAO Yu ZHANG Wenting LI Qianxue DANG Tinghui WANG Yi | 2023 | Journal of Arid Land2023,15,8: | 0 |
