7Near-infrared fluorescence sentinel lymph node detection in gastric cancer: A pilot study显示文摘AIM: To investigate feasibility and accuracy of nearinfrared fluorescence imaging using indocyanine green: nanocolloid for sentinel lymph node(SLN) detection in gastric cancer.METHODS: A prospective, single-institution, phaseI feasibility trial was conducted. Patients suffering from gastric cancer and planned for gastrectomy were included. During surgery, a subserosal injection of 1.6 m L ICG:Nanocoll was administered around the tumor. NIR fluorescence imaging of the abdominal cavity was performed using the Mini-FLARE? NIR fluorescence imaging system. Lymphatic pathways and SLNs were visualized. Of every detected SLN, the corresponding lymph node station, signal-to-background ratio and histopathological diagnosis was determined. Patients underwent standard-of-care gastrectomy. Detected SLNs outside the standard dissection planes were also resected and evaluated.RESULTS: Twenty-six patients were enrolled. Four patients were excluded because distant metastases were found during surgery or due to technical failure of the injection. In 21 of the remaining 22 patients, at least 1 SLN was detected by NIR Fluorescence imaging(mean 3.1 SLNs; range 1-6). In 8 of the 21 patients, tumor-positive LNs were found. Overall accuracy of the technique was 90%(70%-99%; 95%CI), which decreased by higher p T-stage(100%, 100%, 100%, 90%, 0% for respectively Tx, T1, T2, T3, T4 tumors). All NIR-negative SLNs were completely effaced by tumor. Mean fluorescence signal-to-background ratio of SLNs was 4.4(range 1.4-19.8). In 8 of the 21 patients, SLNs outside the standard resection plane were identified, that contained malignant cells in 2 patients.CONCLUSION: This study shows successful use of ICG:Nanocoll as lymphatic tracer for SLN detection in gastric cancer. 9N-terminal truncated RHT-1 proteins generated by translational reinitiation cause semi-dwarfing of wheat Green Revolution alleles显示文摘The unprecedented wheat yield increases during the Green Revolution were achieved through the introduc-tion of the Reduced height(Rht)-B1b and Rht-D1b semi-dwarfing alleles.These Rht-1 alleles encode growth-repressing DELLA genes containing a stop codon within their open reading frame that confers gibberellin(GA)-insensitive semi-dwarfism.In this study,we successfully took the hurdle of detecting wild-type RHT-1 proteins in different wheat organs and confirmed their degradation in response to GAs.We further demonstrated that Rht-B1b and Rht-D1b produce N-terminal truncated proteins through trans-lational reinitiation.Expression of these N-terminal truncated proteins in transgenic lines and in Rht-D1c,an allele containing multiple Rht-D1b copies,demonstrated their ability to cause strong dwarfism,resulting from their insensitivity to GA-mediated degradation.N-terminal truncated proteins were detected in spikes and nodes,but not in the aleurone layers.Since Rht-B 1b and Rht-D1b alleles cause dwarfism but have wild-type dormancy,this finding suggests that tissue-specific differences in translational reinitiation may explain why the Rht-1 alleles reduce plant height without affecting dormancy.Taken together,our findings not only reveal the molecular mechanism underlying the Green Revolution but also demonstrate that trans-lational reinitiation in the main open reading frame occurs in plants.Karel Van De Velde Stephen GThomas Floor Heyse Rim Kaspar Dominique Van Der Straeten Antje Rohde 2021Molecular Plant2021,14,4:8
10Near-infrared fluorescence imaging of a solitary fibrous tumor of the pancreas using methylene blue显示文摘A 67-year-old female presented with unexplained abdominal pain. A contrast-enhanced computed tomography scan of the abdomen incidentally revealed a mass in the uncinate process of the pancreas. This mass was resected and based on histopathological findings, diagnosed as a solitary fibrous tumor (SFT) of the pancreas. A SFT is an extremely rare benign mesenchymal tumor that in 65% of cases affects the visceral pleura but can also affect extra-pleural sites. The intraoperative demarcation of pancreatic tumors, such as SFTs, can bechallenging. 11New evidence for the links between the local water cycle and the underground wet sand layer of a mega-dune in the Badain Jaran Desert, China显示文摘Scientists and the local government have great concerns about the climate change and water resources in the Badain Jaran Desert of western China. A field study for the local water cycle of a lake-desert system was conducted near the Noertu Lake in the Badain Jaran Desert from 21 June to 26 August 2008. An underground wet sand layer was observed at a depth of 20–50 cm through analysis of datasets collected during the field experiment. Measurements unveiled that the near surface air humidity increased in the nighttime. The sensible and latent heat fluxes were equivalent at a site about 50 m away from the Noertu Lake during the daytime, with mean values of 134.4 and 105.9 W/m2 respectively. The sensible heat flux was dominant at a site about 500 m away from the Noertu Lake, with a mean of 187.7 W/m2, and a mean latent heat flux of only 26.7 W/m2. There were no apparent differences for the land surface energy budget at the two sites during the night time. The latent heat flux was always negative with a mean value of –12.7 W/m2, and the sensible heat flux was either positive or negative with a mean value of 5.10 W/m2. A portion of the local precipitation was evaporated into the air and the top-layer of sand dried quickly after every rainfall event, while another portion seeped deep and was trapped by the underground wet sand layer, and supplied water for surface psammophyte growth. With an increase of air humidity and the occurrence of negative latent heat flux or water vapor condensation around the Noertu Lake during the nighttime, we postulated that the vapor was transported and condensed at the lakeward sand surface, and provided supplemental underground sand pore water. 13Impact of Long-Term Alfalfa Cropping on Soil Potassium Content and Clay Minerals in a Semi-Arid Loess Soil in China显示文摘Alfalfa cropping has been considered an efficient method of increasing soil fertility.Usually nitrogen increase in root nodules is considered to be the major beneficial effect.A 21-year time series (five sampling periods) of alfalfa cultivation plots on a loess soil,initially containing illite and chlorite,in Lanzhou of northwestern China was selected to investigate the relationships among alfalfa cropping,soil potassium (K) content and soil clay minerals.The results indicated that soil K significantly accumulated after cropping,with a peak value at about 15 years,and decreased afterwards.The accumulated K was associated with the K increase in the well-crystallized illite,which was not extracted by the traditional laboratory K extraction methods in assessing bioavailability.The steep decline in soil K content after 15-year cropping was in accord with the observed fertility loss in the alfalfa soil.Plant biomass productivity peaked at near 9 years of culture,whereas soil K and clay minerals continued to increase until cropping for 15 years.This suggested that K increased in the topsoil came from the deep root zone.Thus alfalfa continued to store K in clays even after peak production occurred.Nitrogen did not follow these trends,showing a general decline compared with the native prairie soils that had not been cropped.Therefore,the traditional alfalfa cropping can increase K content in the topsoil.LI De-Cheng B. 15IVF或ICSI周期卵巢过度刺激获取大量卵子并不会导致妊娠率降低显示文摘Objective: To investigate the possible negative effects of a strong ovarian response on oocyte quality.Design: Retrospective study.Setting: Tertiary academic center.Patient(s): A total of 1, 894 women (IVF, n = 1, 544; ICSI, n = 350) who underwent their first ovarian stimulation cycle during the period 1995- 2002.Intervention(s): Standardized controlled ovarian stimulation with urinary FSH or recombinant FSH after pituitary down- regulation, followed by IVF/intracytoplasmic sperm injection (ICSI) and ET.Main Outcome Measure(s): Fertilization rate, fraction of high- quality embryos, and implantation rate.Result(s): Using multivariate logistic regression, we analyzed the effect, expressed as an odds ratio (OR), of the number of oocytes obtained (i.e., ovarian response) on the outcome measures.No effect of the response on the ongoing implantation rate or fraction of high- quality embryos was observed.Both in IVF (OR = 0.81, 95% confidence interval [CI] 0.75- 0.87) and in ICSI (OR = 0.88, 95% CI 0.76- 1.00), a negative effect of increasing ovarian response was seen on the fertilization rate per oocyte obtained.However, no negative effect was observed on the fertilization rate per oocyte injected in ICSI (OR = 1.00, 95% CI 0.87- 1.14).The fraction of immature oocytes rises from 3.9% , in women with ≤ 3 oocytes, to 26% in women with > 20 oocytes.Conclusion(s): Oocytes from high responders contain a greater fraction of immature oocytes, but pregnancy outcome is not impaired.Kok J.D. 17Reverse cholesterol transport: From classical view to new insights显示文摘Cholesterol is of vital importance for the human body. It is a constituent for most biological membranes, it is needed for the formation of bile salts, and it is the pre- cursor for steroid hormones and vitamin D. However, the presence of excess cholesterol in cells, and in particular in macrophages in the arterial vessel wall, might be harmful. The accumulation of cholesterol in arteries can lead to atherosclerosis, and in turn, to other cardiovascular diseases. The route that is primarily thought to be responsible for the disposal of cholesterol is called reverse cholesterol transport (RCT). Therefore, RCT is seen as an interesting target for the development of drugs aimed at the prevention of atherosclerosis. Research on RCT has taken off in recent years. 18Fuel Consumption Potential of the Pushbelt CVT显示文摘The efforts to further reduce fuel consumption of vehicles equipped with a pushbelt type Continuously Variable Transmission(CVT) focus on different sources of loss.In this paper the magnitude of these losses and their potential for reduction is described.Inside the CVT,the variator,its control strategy and the hydraulic actuation circuit can be distinguished as the main potentials.A major opportunity is offered by a new control strategy that takes the actual slip between belt and pulley as the control parameter.The resulting decrease of clamping forces on the pushbelt leads to a reduction of variator and actuation losses.Further potential is found in the hydraulic actuation circuit by an improved tuning of the power supply to the actual power requirement.Outside the CVT additional potential is found in start-stop functionality as supported by measures inside the transmission.The paper describes the theoretical background as well as practical fuel savings of up to 5.5% that were obtained in tests on vehicle level.Slip control adds an inherent robustness to the operation of the pushbelt and opens up the fuel saving potential of the CVT thus reinforcing its position as the benchmark for the near future.Van der Sluis, Francis Van Dongen, Tom Van Spijk, Gert-Jan Van der Velde, Arie Van Heeswijk, Ad 2011重庆理工大学学报(自然科学)2011,25,7:5
