| 6 | Seven-senescence-associated gene signature predicts overall survival for Asian patients with hepatocellular carcinoma显示文摘BACKGROUND Cellular senescence is a recognized barrier for progression of chronic liver diseases to hepatocellular carcinoma(HCC). The expression of a cluster of genes is altered in response to environmental factors during senescence. However, it is questionable whether these genes could serve as biomarkers for HCC patients.AIM To develop a signature of senescence-associated genes(SAGs) that predicts patients' overall survival(OS) to improve prognosis prediction of HCC.METHODS SAGs were identified using two senescent cell models. Univariate COX regression analysis was performed to screen the candidate genes significantly associated with OS of HCC in a discovery cohort(GSE14520) for the least absolute shrinkage and selection operator modelling. Prognostic value of this seven-gene signature was evaluated using two independent cohorts retrieved from the GEO(GSE14520) and the Cancer Genome Atlas datasets, respectively.Time-dependent receiver operating characteristic(ROC) curve analysis was conducted to compare the predictive accuracy of the seven-SAG signature and serum α-fetoprotein(AFP).RESULTS A total of 42 SAGs were screened and seven of them, including KIF18 B, CEP55,CIT, MCM7, CDC45, EZH2, and MCM5, were used to construct a prognostic formula. All seven genes were significantly downregulated in senescent cells andupregulated in HCC tissues. Survival analysis indicated that our seven-SAG signature was strongly associated with OS, especially in Asian populations, both in discovery and validation cohorts. Moreover, time-dependent ROC curve analysis suggested the seven-gene signature had a better predictive accuracy than serum AFP in predicting HCC patients' 1-, 3-, and 5-year OS.CONCLUSION We developed a seven-SAG signature, which could predict OS of Asian HCC patients. | 8 | Evaluation of the fishbone model in simulating the seismic response of multistory reinforced concrete moment-resisting frames显示文摘The fishbone model is a simplified numerical model for moment-resisting frames that is capable of modelling the effects of column-beam strength and stiffness ratios. The applicability of the fishbone model in simulating the seismic responses of reinforced concrete moment-resisting frames of different sets of column-beam strength and stiffness ratios are evaluated through nonlinear static, dynamic and incremental dynamic analysis on six prototype buildings of 4-, 8-and 12-stories. The results show that the fishbone model is practically accurate enough for reinforced concrete frames, although the assumption of equal joint rotation does not hold in all cases. In addition to the ground motion characteristics and the number of stories in the structures, the accuracy of the model also varies with the column-beam stiffness and strength ratios. The model performs better for strong column-weak beam frames, in which the lateral drift patterns are better controlled by the continuous stiffness provided by the strong columns. When the inelastic deformation is large, the accuracy of the model may be subjected to large record-to-record variability. In the present study, viability of primary cultured cerebellar granule neurons (CGNs), combined with sample extraction, chemical fractionation and identification, was applied for screening acid-resistant neuronal toxic substances in environmental samples. River sediments and agricultural soils along the river near a brominated flame retardant (BFR) manufacturing plant in South China were collected to screen the key neuronal toxicants. The results indicated that the manufacturing plant was a source of neuronal toxicity risks. In the sediment and soil near the plant, one of the causative toxicants was identified as tris-(2,3-dibromopropyl) isocyanurate (TBC) using HPLC-MS/MS. | 12 | Comparison of measurements of anterior chamber angle via anterior segment optical coherence tomography and ultrasound biomicroscopy显示文摘BACKGROUND Glaucoma is an irreversible optic neuropathy with the loss of visual field and decrease of vision.The variable clinical manifestations may result in differential diagnostic difficulties.The early screening and diagnosis of glaucoma are currently experiencing a demand for anterior segment analysis tools that can gather more information with one short measurement.Therefore,we analyzed the agreement,difference,and correlation of chamber angle parameters such as angel opening distance at 500μm(AOD500)and trabeculo-iris space area at 500μm^2(TISA500)measured by anterior segment optical coherence tomography(ASOCT)and ultrasound biomicroscopy(UBM).AIM To compare the differences,correlation,and agreement in measuring AOD500 and TISA500 by AS-OCT and UBM.METHODS Both AS-OCT and UBM were performed to measure AOD500 and TISA500 in 45 subjects(72 eyes).All subjects without glaucoma were collected from October 2015 to August 2016 at the Ophthalmology Department of the Fourth Affiliated Hospital of China Medical University.Data of the two groups(AOD500 and TISA500)were compared by nonparametric tests.Pearson correlative analysis and Bland-Altman analysis were used to compare the correlation and agreement.RESULTS There were no significant differences between AS-OCT and UBM in measuring AOD500(P1=0.110,P2=0.633,P3=0.078,and P4=0.474)and TISA500(P1=0.584,P2=0.889,P3=0.297,and P4=0.550)of the four quadrants of the anterior chamber angle.There was a high correlation in measuring AOD500(r1=0.562,r2=0.671,r3=0.635,and r4=0.720;P<0.001)and TISA500(r1=0.584,r2=0.889,r3=0.297,and r4=0.550;P<0.001).There was a good agreement in measuring AOD500 and TISA500 by the two modalities.CONCLUSION There is a high correlation and agreement between AOD500 and TISA500 measurements by AS-OCT and UBM.They are interchangeable under some circumstances.AS-OCT proves to be a better early screening tool for glaucoma. | Zi-Yan Yu Ting Huang Lu Lu Bo Qu | 2020 | World Journal of Clinical Cases2020,8,15: | 2 |
| 13 | Thrombocytopenia for prediction of hepatocellular carcinoma recurrence: Systematic review and meta-analysis显示文摘AIM: To investigate the association between thrombocytopenia and relapse after treatment for hepatocellular carcinoma(HCC).METHODS: We searched the Pub Med, EMBASE, and Web of Science databases to obtain eligible studies. The hazard ratios(HRs) values and 95% confidence intervals(CIs) were pooled by random effects model. Subsequently, we estimated the heterogeneity, performed a sensitivity analysis, determined the publication bias, and performed subgroup and meta-regression analyses. Study quality was assessed by using the Oxford Center for Evidence Based Medicine tool.RESULTS: We identified 18 eligible studies by retrieval(published during 2000-2014). Out of the 4163 patients with HCC who were recruited, 2746(66.0%) experienced recurrence. In general, our meta-analysis suggested that low platelet count(PLT) before therapy significantly increased the probability of postoperative recurrence(HR = 1.53, 95%CI: 1.29-1.81). PLT was also valuable in the prediction of intrahepatic distant recurrence(HR = 1.49, 95%CI: 1.25-1.77). Subgroupand meta-regression analyses identified various therapeutic modalities as the source of a high degree of heterogeneity. The pooled HR values showed no obvious change when a single study was removed, but otherwise, an opposite-effects model was used. | 20 | Development of pot-cover effect apparatus with freezing-thawing cycles显示文摘‘Pot-cover effect’ refers to the phenomenon of moisture accumulation beneath the pavement under condensation or desublimation because the vapor transfer in the soil is blocked by the pavement. To study this phenomenon in the laboratory, we have developed the pot-cover effect apparatus with freezing-thawing cycles(PEAFC), which consists of three parts, namely, the vapor transfer system, the temperature control system, and the temperature and water content monitoring system. The major functions of this apparatus include: simulation of both vapor transfer and freezing-thawing cycling in the soil, real-time monitoring of the temperature and the water content in soil samples, and one-dimensional heat and moisture transfer in the samples. The FDR(frequency domain reflectometry) sensors of the apparatus are calibrated and a calibration formula is proposed to eliminate the water content measurement errors induced by temperature changes. Constant temperature difference tests and a freezing-thawing cycling test are conducted with the apparatus. Results indicate that the apparatus can control the water replenishment in the soil samples, monitor the vapor transfer in the samples in real time, and simulate the vapor transfer process under the condition of the freezing-thawing cycling in the soil.
