| 1 | Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29 |

| 3 | A Pilot Study of the Therapeutic Efficacy and Mechanism of Artesunate in the MRL/lpr Murine Model of Systemic Lupus Erythematosus显示文摘Recent evidence indicates that artesunate has immunomodulatory properties that might be useful for treating autoimmune disease.In this study,we conducted a pilot study and explored the effect and mechanism of artesunate on the treatment of systemic lupus erythematosus using an MRL/lpr murine model.MRL/lpr mice were divided into control,cyclophosphamide(CTX)and artesunate treatment groups.Blood was collected to measure serum levels of creatinine,antinuclear antibody(ANA)and anti-double-stranded DNA(anti-dsDNA)antibody.Twenty-four-hour urine was collected to measure levels of proteinuria.The concentration of monocyte chemotactic protein-1(MCP-1)in serum and urine was measured.The expression of MCP-1 in kidney was detected by Western blot and immunohistochemistry assay,respectively.The expression of B cell activating factor(BAFF)in spleen was determined by real time-PCR and immunoblotting.We found that artesunate significantly increased the survival rate,body weight and blood leukocyte counts,and reduced the serum levels of ANA and anti-dsDNA antibody titer,24 h urinary protein,and serum creatinine.Our results indicated that artesunate could decrease MCP-1,major pro-inflammation cytokine,in serum,urine and kidney.We also found that the level of BAFF,the major B cell activation factor,was decreased in artesunate treated MRL/lpr mice.Its efficacy was comparable with that of CTX in this study.Taken together,we have demonstrated that artesunate can inhibit the progression of disease and reverse the pathologic lesion of lupus nephritis. | Ouyang Jin Huayong Zhang Zhifeng Gu Shengnan Zhao Ting Xu Kangxing Zhou Bo Jiang Jie Wang Xiaofeng Zeng Lingyun Sun | 2009 | Cellular & Molecular Immunology2009,6,6: | 20 |
| 4 | Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 7 | Effects of Wei Chang An on expression of multiple genes in human gastric cancer grafted onto nude mice显示文摘AIM:To investigate the expression of multiple genes in Chinese jianpi herbal recipe Wei Chang An (WCA) in human gastric cancer cell line SGC-7901. METHODS:A human gastric adenocarcinoma cell line SGC-7901 grafted onto nude mice was used as the animal model. The mice were randomly divided into 3 groups, one control and the two representing experimental conditions. Animals in the two experimental groups received either WCA over a 34-d period or 5-fluorouracil (5-FU) over 6-d period starting at 8th d after grafting. Control animals received saline on an identical schedule. Animals were killed 41 d after being grafted. The expression profiles in paired WCA treated gastric cancer samples and the N.S. control samples were studied by using a cDNA array representing 14 181 cDNA clusters. The alterations in gene expression levels were confirmed by Real-time Quantitative polymerase chain reaction (qPCR). RESULTS:When compared with controls, the average tumor inhibitory rate in WCA group was 44.32% ± 5.67% and 5-FU 47.04% ± 11.33% (P < 0.01, respectively). The average labeling index (LI) for PCNA in WCA group and 5-FU group was significantly decreased compared with the control group. Apoptotic index (AI) was significantly increased to 9.72% ± 4.51% using the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate fluorescence nick end labeling (TUNEL) method in WCA group compared with the controls 2.45% ± 1.37%. 5-FU group was also found to have a significantly increased AI compared with the controls. The expression of cleaved Caspase-3 in WCA group and 5-FU group was significantly increased compared with the control group respectively. There were 45 different expressed sequence tags (ESTs) among the control sample pool and WCA sample pool. There were 24 ESTs up-regulated in WCA samples and 21 ESTs down-regulated. By using qPCR, the expression level of Stat3, rap2 interacting protein x (RIPX), regulator of differentiation 1 (ROD1) and Bcl-2 was lower in WCA group than that in control group respectively. | 8 | Mode-locked 2.8-μm fluoride fiber laser:from soliton to breathing pulse显示文摘The mode-locked fluoride fiber laser(MLFFL)is an exciting platform for directly generating ultrashort pulses in the mid-infrared(mid-IR).However,owing to difficulty in managing the dispersion in fluoride fiber lasers,MLFFLs are restricted to the soliton regime,hindering pulse-energy scaling.We overcame the problem of dispersion management by utilizing the huge normal dispersion generated near the absorption edge of an infrared-bandgap semiconductor and promoted MLFFL from soliton to breathing-pulse mode-locking.In the breathing-pulse regime,the accumulated nonlinear phase shift can be significantly reduced in the cavity,and the pulse-energy-limitation effect is mitigated.The breathing-pulse MLFFL directly produced a pulse energy of 9.3 nJ and pulse duration of 215 fs,with a record peak power of 43.3 kW at 2.8μm.Our work paves the way for the pulse-energy and peak-power scaling of mid-IR fluoride fiber lasers,enabling a wide range of applications. | Zhipeng Qin Guoqiang Xie Hongan Gu Ting Hai Peng Yuan Jingui Ma Liejia Qian | 2019 | Advanced Photonics2019,1,6: | 9 |
| 9 | Association between Plasma Metal Levels and Diabetes Risk: a Case-control Study in China显示文摘Objective Many metals, some of which have been classified as environmental endocrine disruptors, are used extensively in everyday consumer products and are ubiquitous in our living environment. In the present study, we aimed to explore the associations between the prevalence risk of type 2 diabetes and plasma levels of 20 trace elements as well as those of heavy metals in a Han Chinese population. Methods We conducted a case-control study to investigate the associations between plasma concentrations of 20 metals and diabetes in Jiangsu province. A total of 122 newly diagnosed cases of type 2 diabetes and 429 matched controls were recruited from community physical examinations in Suzhou City of Jiangsu Province. Plasma metal levels were measured by inductively-coupled plasma mass spectrometry. Results After adjusting for confounders, plasma vanadium, chromium, manganese, copper, zinc, arsenic, selenium, strontium, palladium, cadmium, cesium, and barium were associated with diabetes risk(P < 0.05). The adjusted OR increased with increasing concentration of vanadium, manganese, copper, zinc, and cesium. Conclusion Many metals, including manganese, copper, zinc, arsenic, selenium, and cadmium in plasma, are associated with the morbidity of diabetes. | 10 | H∞ control of novel active steering integrated with electric power steering function显示文摘Based on the traditional active steering system, a novel active steering system integrated with electric power steering function was introduced, which can achieve the functions of both active steering and electric power steering. In view of the interference from road random signal and sensor noise in the novel active steering system, the H∞ control model of the novel active steering system was built. With satisfying steering feel, good robust performance and steering stability being the control objectives, the H∞ controller for the novel active front steering (AFS) system was designed. The simulation results show that the novel AFS system with H∞ control strategy can attenuate the road interference quickly, and there is no resonance peak in the bode diagram. It can make the driver obtain more useful information in the low frequency range, and attenuate the road interference better in the high frequency range, thus the driver can get more satisfying road feeling. | 12 | Tetrandrine citrate eliminates imatinib-resistant chronic myeloid leukemia cells in vitro and in vivo by inhibiting Bcr-Abl/β-catenin axis显示文摘Objective:To evaluate the effects of tetrandrine citrate, a novel tetrandrine salt with high water solubility, on the growth of imatinib (IM)-resistant chronic myeloid leukemia (CML) in vitro and in vivo, and reveal action molecular mechanisms. Methods:Cell viability in vitro was measured using methyl thiazolyl tetrazolium (MTT) assay. CML cell growth in vivo was assessed using a xenograft model in nude mice. Bcr-Abl and β-catenin protein levels were determined using Western blotting. Bcr-Abl messenger RNA (mRNA) was measured by reverse transcription polymerase chain reaction (RT-PCR). Flow cytometry (FCM) was used to determine cell cycle status. Results:Tetrandrine citrate inhibited the growth of IM-resistant K562 cells, primary leukemia cells, and primitive CD34 + leukemia cells, and their inhibition concentration that inhibited 50% of target cells (IC 50 ) ranged from 1.20 to 2.97 μg/ml. In contrast, tetrandrine citrate did not affect normal blood cells under the same conditions, and IC 50 values were about 10.12-13.11 μg/ml. Oral administration of tetrandrine citrate caused complete regression of IM-resistant K562 xeno-grafts in nude mice without overt toxicity. Western blot results revealed that treatment of IM-resistant K562 cells with tetrandrine citrate resulted in a significant decrease of both p210 Bcr-Abl and β-catenin proteins, but IM did not affect the Bcr-Abl protein levels. Proteasome inhibitor, MG132, did not prevent tetrandrine-mediated decrease of the p210 Bcr-Abl protein. RT-PCR results showed that tetrandrine treatment caused a decrease of Bcr-Abl mRNA. FCM analysis indicated that tetrandrine induced gap 1 (G 1 ) arrest in CML cells. Conclusions:Tetrandrine citrate is a novel orally active tetrandrine salt with potent anti-tumor activity against IM-resistant K562 cells and CML cells. | 13 | Correlation Between Birth Defects and Dietary Nutrition Status in a High Incidence Area of China显示文摘Objective To investigate the association between birth defects and dietary nutrient intake in a high risk area of China. Methods A dietary survey was performed and serum folic acid was measured in women whose pregnancy was affected by neural tube defects (NTDs) or unaffected by any birth defects (BDs) in Zhongyang and Jiaokou Counties in Shanxi Province of China. Results The local average consumption of foods including dark green vegetables, fruits, fat and meat, and nutrient intake (e.g. energy, protein, retinol, riboflavin, vitamin E, and selenium) were lower than the national average level. In women of childbearing age, these regions, the intake of nutrients was much lower than the recommended nutrient intake (9%-77%). The case-control dietary nutrition study of women whose pregnancy was affected by BDs (including NTDs and congenital heart defects) demonstrated that, in early pregnancy, adequate nutrition (i.e. eating meat, fresh vegetables, fruit more than once a week) was a protective factor, while eating germinated potatoes was a risk factor. The geometrical mean (p5-p95) of serum folic acid in women with NTD birth defects was 9.6 nmol/L (3.6, 23.03), which was significantly lower than that in normal women (14.03 nmol/L). Conclusion Women of childbearing age in the two counties of Shanxi Province, China, have a marked insufficient intake of some nutrients, especially folic acid, zinc, vitamins A and B12. This nutrient deficiency may be an important risk factor for the high prevalence of birth defects in these regions. | 14 | Evaluating the grassland net primary productivity of southern China from 2000 to 2011 using a new climate productivity model显示文摘Grassland is the important component of the terrestrial ecosystems. Estimating net primary productivity(NPP) of grassland ecosystem has been a central focus in global climate change researches. To simulate the grassland NPP in southern China, we built a new climate productivity model, and validated the model with the measured data from different years in the past. The results showed that there was a logarithmic correlation between the grassland NPP and the mean annual temperature, and there was a linear positive correlation between the grassland NPP and the annual precipitation in southern China. All these results reached a very significant level(P<0.01). There was a good correlation between the simulated and the measured NPP, with R2 of 0.8027, reaching the very significant level. Meanwhile, both root mean square errors(RMSE) and relative root-mean-square errors(RRMSE) stayed at a relatively low level, showing that the simulation results of the model were reliable. The NPP values in the study area had a decreasing trend from east to west and from south to north, and the mean NPP was 471.62 g C m^(-2) from 2000 to 2011. Additionally, there was a rising trend year by year for the mean annual NPP of southern grassland and the tilt rate of the mean annual NPP was 3.49 g C m^(-2) yr^(-1) in recent 12 years. | 15 | Bi2WO6 doped with rare earth ions:Preparation,characterization and photocatalytic activity under simulated solar irradiation显示文摘Rare-earth modification Bi2WO6 composites(RE/Bi2WO6)were studied by experimental performance and theory computation based on the different 4 f orbits of selected rare earth elements(La,Ce,Gd,and Yb).The prepared RE/Bi2WO6 was characterized by XRD,SEM/TEM,XPS,UV-vis DRS,and N2 adsorption to learn their physical-chemical properties.Azo dye Rhodamine B(RhB)was photodegraded as a target pollutant to investigate the photocatalytic activity of prepared RE/Bi2WO6 composites.The results of experiment and computation show that four rare earth elements with different electron configurations retain the phase and morphology of Bi2 WO6 and enhance the removal efficiency of RhB under simulated solar irradiation.The optimum doping contents are 0.01%,0.05%,0.05%and 0.01%for La-,Ce-,Gd-,and Yb-doped Bi2WO6,respectively.However,light rare earth La and Ce doped composites indicate some difference in visible light adso rption capacity and mineralization on RhB co mpared with heavy rare earth Gd and Yb doped composites.Both La/Bi2WO6 and Ce/Bi2 WO6 possess larger pore size and higher mineralization ability than Gd/Bi2 WO6 and Yb/Bi2WO6 under the same experimental conditions while Gd/Bi2WO6 and Yb/Bi2WO6 show stronger red shift to the visible light due to the more 4 f electrons.The hole oxidation plays a major role in the photodegradation of RhB by all RE/Bi2WO6. | Chunying Wang Chuantao Gu Ting Zeng Qingqing Zhang Xianping Luo | 2021 | Journal of Rare Earths2021,39,1: | 6 |
| 16 | Prediction of microvascular invasion in solitary hepatocellular carcinoma≤5 cm based on computed tomography radiomics显示文摘BACKGROUND Liver cancer is one of the most common malignant tumors,and ranks as the fourth leading cause of cancer death worldwide.Microvascular invasion(MVI)is considered one of the most important factors for recurrence and poor prognosis of liver cancer.Thus,accurately identifying MVI before surgery is of great importance in making treatment strategies and predicting the prognosis of patients with hepatocellular carcinoma(HCC).Radiomics as an emerging field,aims to utilize artificial intelligence software to develop methods that may contribute to cancer diagnosis,treatment improvement and evaluation,and better prediction.AIM To investigate the predictive value of computed tomography radiomics for MVI in solitary HCC≤5 cm.METHODS A total of 185 HCC patients,including 122 MVI negative and 63 MVI positive patients,were retrospectively analyzed.All patients were randomly assigned to the training group(n=124)and validation group(n=61).A total of 1351 radiomic features were extracted based on three-dimensional images.The diagnostic performance of the radiomics model was verified in the validation group,and the Delong test was applied to compare the radiomics and MVIrelated imaging features(two-trait predictor of venous invasion and radiogenomic invasion).RESULTS A total of ten radiomics features were finally obtained after screening 1531 features.According to the weighting coefficient that corresponded to the features,the radiomics score(RS)calculation formula was obtained,and the RS score of each patient was calculated.The radiomics model exhibited a better correction and identification ability in the training and validation groups[area under the curve:0.72(95%confidence interval:0.58-0.86)and 0.74(95%confidence interval:0.66-0.83),respectively].Its prediction performance was significantly higher than that of the image features(P<0.05).CONCLUSION Computed tomography radiomics has certain predictive value for MVI in solitary HCC≤5 cm,and the predictive ability is higher than that of image features. | Peng Liu Xian-Zhen Tan Ting Zhang Qian-Biao Gu Xian-Hai Mao Yan-Chun Li Ya-Qiong He | 2021 | World Journal of Gastroenterology2021,27,17: | 6 |
| 17 | Characterization of integrons and novel cassette arrays in bacteria from clinical isloates in China,2000-2014显示文摘Rapid dissemination of antibiotic resistance genes among bacterial isolates is an increasing problem in China.Integron,a conserved DNA sequence,which is carried on episomal genetic structures,plays a very important role in development of antibiotic resistance.This systematic analysis was based on MEDLINE and EMBASE databases.We summarized the distribution and proportion of different types of gene cassette arrays of integrons(including class 1,2,3 and atypical class 1 integron) from clinical bacteria isolates in China.Fifty-six literatures were included in this study.Most of the strains were Gram-negative bacteria(94.1%,7,364/7,822) while only 5.9% strains were Grampositive bacteria.Class 1 integrons were detected in 54.2%(3956/7295) Gram-negative strains.aadA2 was the most popular gene cassette array detected from 60 Gram-positive bacteria while dfrA17-aadA5 were detected in 426 Gramnegative bacteria.This study identified 12 novel gene cassette arrays which have not been previously found in any species.All the novel gene cassette arrays were detected from Gram-negative bacteria.A regional characteristic of distribution of integrons was presented in this study.The results highlight a need for continuous surveillance of integrons and provide a guide for future research on integron-mediated bacteria resistance. | Wenying Xia Ting Xu Tingting Qin Pengpeng Liu Yun Liu Haiquan Kang Bing Gu Ping Ma | 2016 | The Journal of Biomedical Research2016,30,4: | 5 |

| 20 | Seven-senescence-associated gene signature predicts overall survival for Asian patients with hepatocellular carcinoma显示文摘BACKGROUND Cellular senescence is a recognized barrier for progression of chronic liver diseases to hepatocellular carcinoma(HCC). The expression of a cluster of genes is altered in response to environmental factors during senescence. However, it is questionable whether these genes could serve as biomarkers for HCC patients.AIM To develop a signature of senescence-associated genes(SAGs) that predicts patients' overall survival(OS) to improve prognosis prediction of HCC.METHODS SAGs were identified using two senescent cell models. Univariate COX regression analysis was performed to screen the candidate genes significantly associated with OS of HCC in a discovery cohort(GSE14520) for the least absolute shrinkage and selection operator modelling. Prognostic value of this seven-gene signature was evaluated using two independent cohorts retrieved from the GEO(GSE14520) and the Cancer Genome Atlas datasets, respectively.Time-dependent receiver operating characteristic(ROC) curve analysis was conducted to compare the predictive accuracy of the seven-SAG signature and serum α-fetoprotein(AFP).RESULTS A total of 42 SAGs were screened and seven of them, including KIF18 B, CEP55,CIT, MCM7, CDC45, EZH2, and MCM5, were used to construct a prognostic formula. All seven genes were significantly downregulated in senescent cells andupregulated in HCC tissues. Survival analysis indicated that our seven-SAG signature was strongly associated with OS, especially in Asian populations, both in discovery and validation cohorts. Moreover, time-dependent ROC curve analysis suggested the seven-gene signature had a better predictive accuracy than serum AFP in predicting HCC patients' 1-, 3-, and 5-year OS.CONCLUSION We developed a seven-SAG signature, which could predict OS of Asian HCC patients.
