1Gold nanorods together with HSP inhibitor-VER-155008 micelles for colon cancer mild-temperature photothermal therapy显示文摘Enhancing the heat-sensitivity of tumor cells provides an alternative solution to maintaining the therapeutic outcome of photothermal therapy(PTT). In this study, we constructed a therapeutic system, which was composed of methoxy-polyethylene-glycol-coated-gold-nanorods(MPEG-AuN R) and VER-155008-micelles, to evaluate the effect of VER-155008 on the sensitivity of tumor cells to heat, and further investigate the therapeutic outcome of MPEG-AuN R mediated PTT combined with VER-155008-micelles. VER-155008-micelles downregulate the expression of heat shock proteins and attenuate the heat-resistance of tumor cell. The survival of HCT116 cells treated with VER-155008-micelles under 45 ℃ is equal to that treated with high temperature hyperthermia(55 ℃) in vitro. Furthermore, we proved either the MPEG-AuN R or VER-155008-micelles can be accumulate in the tumor site by photoacoustic imaging and fluorescent imaging. In vivo anti-cancer evaluation showed that tumor size remarkably decreased(smaller than 100 mm^3 or vanished) when treated with combing 45℃ mild PTT system, which contrasted to the tumor size when treated with individual 45℃ mild PTT(around 500nm^3) or normal saline as control(larger than 2000 nm^3). 2Regulation of extracellular signal-regulated kinase 1/2 influences hippocampal neuronal survival in a rat model of diabetic cerebral ischemia显示文摘Activation of extracellular signal-regulated kinase 1/2 has been demonstrated in acute brain ischemia. We hypothesized that activated extracellular signal-regulated kinase 1/2 can protect hippocampal neurons from injury in a diabetic model after cerebral ischemia/reperfusion. In this study, transient whole-brain ischemia was induced by four-vessel occlusion in normal and diabetic rats, and extracellular signal-regulated kinase 1/2 inhibitor(U0126) was administered into diabetic rats 30 minutes before ischemia as a pretreatment. Results showed that the number of surviving neurons in the hippocampal CA1 region was reduced, extracellular signal-regulated kinase 1/2 phosphorylation and Ku70 activity were decreased, and pro-apoptotic Bax expression was upregulated after intervention using U0126. These findings demonstrate that inhibition of extracellular signal-regulated kinase 1/2 activity aggravated neuronal loss in the hippocampus in a diabetic rat after cerebral ischemia/reperfusion, further decreased DNA repairing ability and accelerated apoptosis in hippocampal neurons. 4IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
5Robust RGB-D Camera and IMU Fusion-based Cooperative and Relative Close-range Localization for Multiple Turtle-inspired Amphibious Spherical Robots显示文摘In the narrow, submarine, unstructured environment, the present localization approaches, such as GPS measurement, dead?rcckoning, acoustic positioning, artificial landmarks-based method, are hard to be used for multiple small-scale underwater robots. Therefore, this paper proposes a novel RGB-D camera and Inertial Measurement Unit (IMU) fusion-based cooperative and relative close-range localization approach for special environments, such as underwater caves. Owing to the rotation movement with zero-radius, the cooperative localization of Multiple Turtle-inspired Amphibious Spherical Robot (MTASRs) is realized. Firstly, we present an efficient Histogram of Oriented Gradient (HOG) and Color Names (CNs) fusion feature extracted from color images ofTASRs. Then, by training Support Vector Machine (SVM) classifier with this fusion feature, an automatic recognition method of TASRs is developed. Secondly, RGB-D camerabased measurement model is obtained by the depth map In order to realize the cooperative and relative close-range localization of MTASRs, the MTASRs model is established with RGB-D camera and IMU. Finally, the depth measurement in water is corrected and the efficiency of RGB-D camera for underwater application is validated. 7Resistance Characteristics of Hydraulic Oil through Isodiametric T-type Duct with Sharp Corners显示文摘Rational determination and reduction of local energy loss of oil flow at pipe junctions are of important significance to improve hydraulic pipeline’s work efficiency, especially for complex hydraulic pipeline connected by isodiametric T-type ducts with sharp corners to get combined and divided flow. From this point of view, the formulae of resistance loss for combined flow and divided flow through isodiametric T-type duct with sharp corners as well as the correlations of resistance loss coefficients in the branches of the duct are derived using energy method. On this basis, resistance characteristics of hydraulic oil in the duct are obtained by numerical simulation of different flow modes, which are commonly applied in hydraulic pipelines, using computational fluid dynamics (CFD) method, and the reasons for the resistance loss are analyzed based on the pressure change mechanism in the flow field. A part of simulation results was validated with the reference data. The research shows that for combined flows the resistance loss of symmetrical is lower than that of unsymmetrical to obtain low speed in common branch, but to gain high speed is quite the contrary, for divided flows, the symmetrical is always a reasonable choice to reduce resistance loss. 8Introduction to the Regional Coupled Model WRF4-LICOM: Performance and Model Intercomparison over the Western North Pacific显示文摘Regional coupled modeling is one of the frontiers of regional climate modeling,but intercomparison has not been well coordinated.In this study,a community regional climate model,WRF4,with a resolution of 15 km,was coupled with a high-resolution(0.1°)North Pacific Ocean model(LICOM_np).The performance of the regional coupled model,WRF4_LICOM,was compared to that of another regional coupled model,RegCM4_LICOM,which was a coupling of version 4 of the Regional Climate Model(RegCM4)with LICOM_np.The analysis focused on the 2005 western North Pacific summer monsoon rainfall.The results showed that the regional coupled models with either RegCM4 or WRF4 as their atmospheric model component simulated the broad features over the WNP reasonably well.Quantitative intercomparison of the regional coupled simulations exhibited different biases for different climate variables.RegCM4_LICOM exhibited smaller biases in its simulation of the averaged June–July–August SST and rainfall,while WRF4_LICOM better captured the tropical cyclone(TC)intensity,the percentage contributions of rainfall induced by TCs to the total rainfall,and the diurnal cycle of rainfall and stratiform percentages,especially over land areas.The different behaviors in rainfall simulated by the two models were partly ascribed to the behaviors in the simulated western North Pacific subtropical high(WNPSH).The stronger(weaker)WNPSH in WRF4_LICOM(RegCM4_LICOM)was driven by overestimated(underestimated)diabatic heating,which peaked at approximately 450 hPa over the region around the Philippines in association with different condensation–radiation processes.Coupling of WRF4 with LIOCM is a crucial step towards the development of the next generation of regional earth system models at the Chinese Academy of Sciences.Liwei ZOU Tianjun ZHOU Jianping TANG Hailong LIU 2020Advances in Atmospheric Sciences2020,37,8:4
918beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3
10Injectable kartogenin and apocynin loaded micelle enhances the alleviation of intervertebral disc degeneration by adipose-derived stem cell显示文摘Cell transplantation has been proved the promising therapeutic effects on intervertebral disc degeneration(IVDD).However,the increased levels of reactive oxygen species(ROS)in the degenerated region will impede the efficiency of human adipose-derived stem cells(human ADSCs)transplantation therapy.It inhibits human ADSCs proliferation,and increases human ADSCs apoptosis.Herein,we firstly devised a novel amphiphilic copolymer PEG-PAPO,which could self-assemble into a nanosized micelle and load lipophilic kartogenin(KGN),as a single complex(PAKM).It was an injectable esterase-responsive micelle,and showed controlled release ability of KGN and apocynin(APO).Oxidative stimulation promoted the esterase activity in human ADSCs,which accelerate degradation of esterase-responsive micelle.Compared its monomer,the PAKM micelle possessed better bioactivities,which were attributed to their synergistic effect.It enhanced the viability,autophagic activation(P62,LC3 II),ECM-related transcription factor(SOX9),and ECM(Collagen II,Aggrecan)maintenance in human ADSCs.Furthermore,it is demonstrated that the injection of PAKM with human ADSCs yielded higher disc height and water content in rats.Therefore,PAKM micelles perform promoting cell survival and differentiation effects,and may be a potential therapeutic agent for IVDD.Chao Yu Dongdong Li Chenggui Wang Kaishun Xia Jingkai Wang Xiaopeng Zhou Liwei Ying Jiawei Shu Xianpeng Huang Haibin Xu Bin Han Qixin Chen Fangcai Li Jianbin Tang Chengzhen Liang Nigel Slater 2021Bioactive Materials2021,6,10:3
11Hybrid integrated low-noise linear chirp frequency-modulated continuous-wave laser source based on self-injection to an external cavity显示文摘A hybrid integrated low-noise linear chirp frequency-modulated continuous-wave(FMCW)laser source with a wide frequency bandwidth is demonstrated.By employing two-dimensional thermal tuning,the laser source shows frequency modulation bandwidth of 10.3 GHz at 100 Hz chirped frequency and 5.6 GHz at 1 kHz chirped frequency.The intrinsic linewidth of 49.9 Hz with 42 GHz continuous frequency tuning bandwidth is measured under static operation.Furthermore,by pre-distortion linearization of the laser source,it can distinguish 3 m length difference at 45 km distance in the fiber length measurement experiment,demonstrating its application potential in ultra-long fiber sensing and FMCW light detection and ranging.LIWEI TANG HONGXIANG JIA SHUAI SHAO SIGANG YANG HONGWEI CHEN MINGHUA CHEN 2021Photonics Research2021,9,10:3

13Carbon coated ultrasmall anatase TiO_2 nanocrystal anchored on N,S-RGO as high-performance anode for sodium ion batteries显示文摘Anatase TiO_2 has been investigated as one of the most promising anode materials for sodium ion batteries(SIBs)with low cost and high theoretical capacity.Herein,a composite material of TiO_2 /N,S-RGO@C with carbon coated ultrasmall anatase TiO_2 anchored on nitrogen and sulfur co-doped RGO matrix was successfully prepared by a rational designed process.The composite structure exhibited ultrasmall crystal size,rich porous structure,homogeneous heteroatoms doping and thin carbon coating,which synergistically resulted in elevated electron and ion transfer.The anode exhibited high rate capacities with good reversibility under high rate cycling.The carbon coating was investigated to be effective to prevent active material falling and lead to long term cycling performance with a high capacity retention of 181 m Ah g^(à1)after 2000cycles at 2 C.Kinetic studies were carried out and the results revealed that the superior performance of the composite material were derived from the decreased charge transfer resistance and elevated ion diffusion.Results suggested that the TiO_2 /N,S-RGO@C composite is a promising anode material for sodium ion batteries.Lingfei Zhao Tong Tang Weihua Chen Xiangming Feng Liwei Mi 2018Green Energy & Environment2018,3,3:2
