1A bioactive injectable self-healing anti-inflammatory hydrogel with ultralong extracellular vesicles release synergistically enhances motor functional recovery of spinal cord injury显示文摘The repair and motor functional recovery after spinal cord injury(SCI)remains a worldwide challenge.The inflammatory microenvironment is one of main obstacles on inhibiting the recovery of SCI.Using mesenchymal stem cells(MSCs)derived extracellular vesicles to replace MSCs transplantation and mimic cell paracrine secretions provides a potential strategy for microenvironment regulation.However,the effective preservation and controlled release of extracellular vesicles in the injured spinal cord tissue are still not satisfied.Herein,we fabricated an injectable adhesive anti-inflammatory F127-polycitrate-polyethyleneimine hydrogel(FE)with sustainable and long term extracellular vesicle release(FE@EVs)for improving motor functional recovery after SCI.The orthotopic injection of FE@EVs hydrogel could encapsulate extracellular vesicles on the injured spinal cord,thereby synergistically induce efficient integrated regulation through suppressing fibrotic scar formation,reducing inflammatory reaction,promoting remyelination and axonal regeneration.This study showed that combining extracellular vesicles into bioactive multifunctional hydrogel should have great potential in achieving satisfactory locomotor recovery of central nervous system diseases.Chenggui Wang Min Wang Kaishun Xia Jingkai Wang Feng Cheng Kesi Shi Liwei Ying Chao Yu Haibin Xu Shining Xiao Chengzhen Liang Fangcai Li Bo Lei Qixin Chen 2021Bioactive Materials2021,6,8:9

4Injectable kartogenin and apocynin loaded micelle enhances the alleviation of intervertebral disc degeneration by adipose-derived stem cell显示文摘Cell transplantation has been proved the promising therapeutic effects on intervertebral disc degeneration(IVDD).However,the increased levels of reactive oxygen species(ROS)in the degenerated region will impede the efficiency of human adipose-derived stem cells(human ADSCs)transplantation therapy.It inhibits human ADSCs proliferation,and increases human ADSCs apoptosis.Herein,we firstly devised a novel amphiphilic copolymer PEG-PAPO,which could self-assemble into a nanosized micelle and load lipophilic kartogenin(KGN),as a single complex(PAKM).It was an injectable esterase-responsive micelle,and showed controlled release ability of KGN and apocynin(APO).Oxidative stimulation promoted the esterase activity in human ADSCs,which accelerate degradation of esterase-responsive micelle.Compared its monomer,the PAKM micelle possessed better bioactivities,which were attributed to their synergistic effect.It enhanced the viability,autophagic activation(P62,LC3 II),ECM-related transcription factor(SOX9),and ECM(Collagen II,Aggrecan)maintenance in human ADSCs.Furthermore,it is demonstrated that the injection of PAKM with human ADSCs yielded higher disc height and water content in rats.Therefore,PAKM micelles perform promoting cell survival and differentiation effects,and may be a potential therapeutic agent for IVDD.Chao Yu Dongdong Li Chenggui Wang Kaishun Xia Jingkai Wang Xiaopeng Zhou Liwei Ying Jiawei Shu Xianpeng Huang Haibin Xu Bin Han Qixin Chen Fangcai Li Jianbin Tang Chengzhen Liang Nigel Slater 2021Bioactive Materials2021,6,10:3
5Generation of transgenic wheat resistant to wheat yellow mosaic virus and identification of gene silence induced by virus infection显示文摘The plasmid containing the promoter Actl, the coat protein (cp) gene of wheat yellow mosaic virus (WYMV) and the selectable bar gene, was delivered via particle bombardment, directly into immature embryos of a wheat culti-vars. PCR and PCR-RFLP were employed to screen the existence of the cp gene in T0 and T1 generations. Seeds from the positive T1 plants were sowed in fields heavily contaminated with WYMV to detect their resistance. In field trial of virus infection, one of the transgenic wheat lines, P8-T2, exhibited highly disease-resistance. Western blot and RT-PCR analysis showed that the expression level of cp gene in the resistant transgenic line was reduced greatly compared to those susceptible to WYMV infection. 9Ore-forming age and tectonic setting of the Linjiasandaogou gold deposit in the Liaodong Peninsula,northeast China:constraints from geochronology and geochemistry显示文摘The Linjiasandaogou gold deposit is located in the Qingchengzi Orefield,North China Craton,China,and has not attracted many studies.We present geochronological,whole-rock geochemical,and Sr–Nd–Hf isotopic data to constrain the age and tectonic setting of the mineralization.U–Pb dating of zircon from pre-and post-ore dikes indicates the Linjiasandaogou Au deposit formed at ca.227–226 Ma.The granite porphyry(ca.227 Ma)and quartz diorite porphyry(ca.228 Ma),which are slightly older than the mineralization,have(;Sr/;Sr)i-=0.7127–0.7162,εNd(t)=–13.7 to–17.0,andεHf(t)=–14.6 to–16.9,and display enrichment in light rare earth elements and large ion lithophile elements and depletion in high field strength elements.Two lamprophyres(226 and225 Ma),which are slightly younger than the mineralization,have higher(;Sr/;Sr)i(0.7165–0.7216),negativeεNd(t)(–11.2 to–14.3)andεHf(t)(–15.6 to–18.6)values,and are enriched in light rare earth elements but depleted in high field strength elements(Nb and Ta).The geochemical characteristics of the granitoid and lamprophyres indicate a lower crustal and enriched mantle source,respectively.We infer that the Linjiasandaogou Au deposit formed in a postcollisional tectonic setting,following the collision between the North China,Yangtze craton,and Central Asian Orogeny in Triassic.Xiaofeng Yao Tingjie Yan Zhicheng Lü Chenggui Lin Kuifeng Mi Shenghui Li Yang Li Wange Du 2022Acta Geochimica2022,41,2:1

16Three-dimensional observation of the phase structure of high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend by laser scanning confocal microscopy显示文摘In this paper,high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend (50/50 wt%) was prepared through solution blending and then compression molding,and subsequently examined by laser scanning confocal microscopy (LSCM). The PEB used in this experiment was labeled with a small quantity of a fluorescein derivative to render fluorescence. The initial films showed uniform dye dis-tribution and no indication of phase separation within the resolution of optical microscopy. Sample films annealing at 140℃ followed by rapid cooling to room temperature showed obvious phase sepa-ration and bicontinuous structure. 18Functional analysis of beet necrotic yellow vein virus (BNYVV) RNA4 in fungal transmission显示文摘Full-length RNA4 of beet necrotic yellow vein virus (BNYVV) and its mutants, including a frame-shift and deletions within the coding region, were inserted into a transcription plasmid. Transcripts derived from these plasmids and RNAs extracted from a BNYVV isolate containing only RNAs 1, 2 and 3 were coinoculated to Tetragona expansa. Then these recombinant isolates were inoculated to sugar beet plants grown in a sand culture system containing Po-lymyxa betae that had been freed of virus particles to test efficiency of BNYVV transmission. The result showed that the efficiency of BNYVV transmission among the plants by Polymyxa betae was highly decreased by different deletions in the coding region of RNA4, but not affected by the frame-shift mutant with four nucleotides insertion. 19A conductive supramolecular hydrogel creates ideal endogenous niches to promote spinal cord injury repair显示文摘The current effective method for treatment of spinal cord injury(SCI)is to reconstruct the biological microenvironment by filling the injured cavity area and increasing neuronal differentiation of neural stem cells(NSCs)to repair SCI.However,the method is characterized by several challenges including irregular wounds,and mechanical and electrical mismatch of the material-tissue interface.In the current study,a unique and facile agarose/gelatin/polypyrrole(Aga/Gel/PPy,AGP3)hydrogel with similar conductivity and modulus as the spinal cord was developed by altering the concentration of Aga and PPy.The gelation occurred through non-covalent interactions,and the physically crosslinked features made the AGP3 hydrogels injectable.In vitro cultures showed that AGP3 hydrogel exhibited excellent biocompatibility,and promoted differentiation of NSCs toward neurons whereas it inhibited over-proliferation of astrocytes.The in vivo implanted AGP3 hydrogel completely covered the tissue defects and reduced injured cavity areas.In vivo studies further showed that the AGP3 hydrogel provided a biocompatible microenvironment for promoting endogenous neurogenesis rather than glial fibrosis formation,resulting in significant functional recovery.RNA sequencing analysis further indicated that AGP3 hydrogel significantly modulated expression of neurogenesis-related genes through intracellular Ca2+signaling cascades.Overall,this supramolecular strategy produces AGP3 hydrogel that can be used as favorable biomaterials for SCI repair by filling the cavity and imitating the physiological properties of the spinal cord.Biao Yang Chengzhen Liang Di Chen Feng Cheng Yuang Zhang Shaoke Wang Jiawei Shu Xianpeng Huang Jingkai Wang Kaishun Xia Liwei Ying Kesi Shi Chenggui Wang Xuhua Wang Fangcai Li Qian Zhao Qixin Chen 2022Bioactive Materials2022,7,9:0
20A Study of the Factors that Affect Safe Full Heading of Machine-transplanted Rice Seedlings in the Temperate and Cool Rice-growing Areas显示文摘This paper studies the factors that affect the safe full heading of machine-transplanted rice seedlings in the temperate and cool ricegrowing areas,and explores the influence of machine-transplanted rice seedling varieties,seedling age,nitrogen fertilizer application rate and management on the safe full heading of machine-transplanted rice seedlings. Results show that for the mechanized production in the temperate and cool rice-growing areas,we should select the mid-early maturity cold-resistant varieties with growth period of not exceeding 180 d,control the seedling age to be 30- 35 d and total amount of pure nitrogen to be not more than 300 kg / ha,and moderately reduce the nitrogen fertilizer postponing ratio.
