2Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies.However,few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases.In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms.Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes,like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors,including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes.In summary,our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications.Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu 2021Acta Pharmaceutica Sinica B2021,11,11:15
3Phytoplankton communities and its controlling factors in summer and autumn in the southern Yellow Sea, China显示文摘The phytoplankton water samples were collected in two multidisciplinary investigations which were carried out during summer(June) and autumn(November to December) of 2011. Phytoplankton species composition and abundance data were accomplished by Uterm?hl method. The phytoplankton community was dominated by diatoms and dinoflagellates in the southern Yellow Sea(YS) in summer and autumn. In summer, Paralia sulcata and Prorocentrum dentatum were the predominated species, the cell abundance ranged from 0.074 to 107.733×103 cells/L with an average of 9.057×103 cells/L. Two phytoplankton high abundance appeared in northwest part of the survey area and the Changjiang River Estuary, respectively. In autumn, Par. sulcata became the predominant species, and the phytoplankton cell abundance ranged from 1.035×103 to 8.985×103 cells/L, the average was 3.263×103 cells/L. The phytoplankton abundance in surface layer presented the homogeneous distributions. Canonical Correspondence Analysis(CCA) method was applied for discovering the relationship between environmental factors and the common found phytoplankton species. The responses of phytoplankton to nutrients were varied between summer and autumn. 5Selective hydrogenation of benzene to cyclohexene over Ce-promoted Ru catalysts显示文摘Ru-Ce catalysts were prepared by a co-precipitation method.The effects of Ce precursors with different valences and Ce contents on the catalytic performance of Ru-Ce catalysts were investigated in the presence of ZnSO4.The Ce species in the catalysts prepared with different valences of the Ce precursors all exist as CeO2 on the Ru surface.The promoter CeO2alone could not improve the selectivity to cyclohexene of Ru catalysts.However,almost all the CeO2 in the catalysts could react with the reaction modifier ZnSO4 to form(Zn(OH)2)3(ZnSO4)(H2O)3 salt.The amount of the chemisorbed salt increased with the CeO2 loading,resulting in the decrease of the activity and the increase of the selectivity to cyclohexene of Ru catalyst.The Ru-Ce catalyst with the optimum Ce/Ru molar ratio of 0.19 gave a maximum cyclohexene yield of 57.4%.Moreover,this catalyst had good stability and excellent reusability.Haijie Sun Yajie Pan Shuaihui Li Yuanxin Zhang Yingying Dong Shouchang Liu Zhongyi Liu 2013Journal of Energy Chemistry2013,22,5:4
6Mullite whiskers prepared by molten salt method using Si powders显示文摘Mullite whiskers were prepared from Si powders in molten Al2(SO4)3-Na2SO4 mixture salts with different Al/Si molar ratio(R)of raw materials.The resulting mullite whiskers,had been investigated using X-ray diffraction(XRD),scanning electron microscopy(SEM),energy dispersive spectrometer(EDS)and infrared spectroscopy analysis(FT-IR).Mullite phase was obtained in molten salts when the temperature reached at 850℃.SEM and EDS results revealed that two kinds of microstructures were formed in the final product,Al-rich mullite pellets and clusters of tiny mullite crystals,and the content of Al-rich mullite pellets increased with the higher Al/Si molar ratio(R)adopted in raw materials.A new oxidation-dissolution mechanism was proposed to explain mullite whiskers growth.According to thermodynamic analysis,mullite phase might be spontaneously formed as the temperature reached the decomposition temperature of aluminum sulfate(1023 K).Wei WANG Hongwei LI Yajie GUO Qiang SUN Chen CHEN 2012Journal of Advanced Ceramics2012,1,4:2
7Near-Term Projections of Global and Regional Land Mean Temperature Changes Considering Both the Secular Trend and Multidecadal Variability显示文摘Near-term climate projections are needed by policymakers; however, these projections are difficult because internally generated climate variations need to be considered. In this study, temperature change scenarios in the near-term period 2017-35 are projected at global and regional scales based on a refined multi-model ensemble approach that considers both the secular trend(ST) and multidecadal variability(MDV) in the Coupled Model Intercomparison Project Phase 5(CMIP5) simulations. The ST and MDV components are adaptively extracted from each model simulation by using the ensemble empirical mode decomposition(EEMD) filter, reconstructed via the Bayesian model averaging(BMA) method for the historical period 1901-2005, and validated for 2006-16. In the simulations of the 'medium' representative concentration pathways scenario during 2017-35, the MDV-modulated temperature change projected via the refined approach displays an increase of 0.44℃(90% uncertainty range from 0.30 to 0.58℃) for global land, 0.48℃(90% uncertainty range from 0.29 to 0.67℃) for the Northern Hemispheric land(NL), and 0.29℃(90% uncertainty range from 0.23 to 0.35℃) for the Southern Hemispheric land(SL). These increases are smaller than those projected by the conventional arithmetic mean approach. The MDV enhances the ST in 13 of 21 regions across the world. The largest MDV-modulated warming effect(46%) exists in central America. 8Cigarette smoke promotes oral leukoplakia via regulating glutamine metabolism and M2 polarization of macrophage显示文摘Oral immunosuppression caused by smoking creates a microenvironment to promote the occurrence and development of oral mucosa precancerous lesions.This study aimed to investigate the role of metabolism and macrophage polarization in cigarette-promoting oral leukoplakia.The effects of cigarette smoke extract(CSE)on macrophage polarization and metabolism were studied in vivo and in vitro.The polarity of macrophages was detected by flow cytometric analysis and q PCR.Liquid chromatography-mass spectrometry(LC-MS)was used to perform a metabolomic analysis of Raw cells stimulated with CSE.Immunofluorescence and flow cytometry were used to detect the polarity of macrophages in the condition of glutamine abundance and deficiency.Cell Counting Kit-8(CCK-8),wound-healing assay,and Annexin V-FITC(fluorescein isothiocyanate)/PI(propidium iodide)double-staining flow cytometry were applied to detect the growth and transferability and apoptosis of Leuk-1 cells in the supernatant of Raw cells which were stimulated with CSE,glutamine abundance and deficiency.Hyperkeratosis and dysplasia of the epithelium were evident in smoking mice.M2 macrophages increased under CSE stimulation in vivo and in vitro.In total,162 types of metabolites were detected in the CSE group.The metabolites of nicotine,glutamate,arachidic acid,and arginine changed significantly.The significant enrichment pathways were also selected,including nicotine addiction,glutamine and glutamate metabolism,and arginine biosynthesis.The results also showed that the supernatant of Raw cells stimulated by CSE could induce excessive proliferation of Leuk-1 and inhibit apoptosis.Glutamine abundance can facilitate this process.Cigarette smoke promotes oral leukoplakia via regulating glutamine metabolism and macrophage M2 polarization.Yanan Zhu Shuo Zhang Jiahui Sun Tingting Wang Qin Liu Guanxi Wu Yajie Qian Weidong Yang Yong Wang Wenmei Wang 2021International Journal of Oral Science2021,13,3:2
9In Vivo Two-photon Calcium Imaging in Dendrites of Rabies Virus-labeled V1 Corticothalamic Neurons显示文摘Monitoring neuronal activity in vivo is critical to understanding the physiological or pathological functions of the brain.Two-photon Ca^(2+)imaging in vivo using a cranial window and specific neuronal labeling enables realtime,in situ,and long-term imaging of the living brain.Here,we constructed a recombinant rabies virus containing the Ca^(2+)indicator GCaMP6 s along with the fluorescent protein DsRed2 as a baseline reference to ensure GCaMP6 s signal reliability.This functional tracer was applied to retrogradely label specific V1-thalamus circuits and detect spontaneous Ca^(2+)activity in the dendrites of V1 corticothalamic neurons by in vivo two-photon Ca^(2+)imaging.Notably,we were able to record single-spine spontaneous Ca2+activity in specific circuits.Distinct spontaneous Ca^(2+)dynamics in dendrites of V1 corticothalamic neurons were found for different V1-thalamus circuits.Our method can be applied to monitor Ca^(2+)dynamics in specific input circuits in vivo,and contribute to functional studies of defined neural circuits and the dissection of functional circuit connections.Yajie Tang Liang Li Leqiang Sun Jinsong Yu Zhe Hu Kaiqi Lian Gang Cao Jinxia Dai 2020Neuroscience Bulletin2020,36,5:2

12Accelerated testing for automated vehicles safety evaluation in cut-in scenarios based on importance sampling,genetic algorithm and simulation applications显示文摘Purpose–It would take billions of miles’field road testing to demonstrate that the safety of automated vehicle is statistically significantly higher than the safety of human driving because that the accident of vehicle is rare event.Design/methodology/approach–This paper proposes an accelerated testing method for automated vehicles safety evaluation based on improved importance sampling(IS)techniques.Taking the typical cut-in scenario as example,the proposed method extracts the critical variables of the scenario.Then,the distributions of critical variables are statistically fitted.The genetic algorithm is used to calculate the optimal IS parameters by solving an optimization problem.Considering the error of distribution fitting,the result is modified so that it can accurately reveal the safety benefits of automated vehicles in the real world.Findings–Based on the naturalistic driving data in Shanghai,the proposed method is validated by simulation.The result shows that compared with the existing methods,the proposed method improves the test efficiency by 35 per cent,and the accuracy of accelerated test result is increased by 23 per cent.Originality/value–This paper has three contributions.First,the genetic algorithm is used to calculate IS parameters,which improves the efficiency of test.Second,the result of test is modified by the error correction parameter,which improves the accuracy of test result.Third,typical high-risk cut-in scenarios in China are analyzed,and the proposed method is validated by simulation.Yiming Xu Yajie Zou Jian Sun 2018Journal of Intelligent and Connected Vehicles2018,1,1:1

14Inactivated Pseudomonas PE(△Ⅲ)exotoxin fused to neutralizing epitopes of PEDV S proteins produces a specific immune response in mice显示文摘Porcine epidemic diarrhea(PED)caused by the porcine epidemic diarrhea virus(PEDV),is a severe infectious and devastating swine disease that leads to serious economic losses in the swine industry worldwide.An increased number of PED cases caused by variant PEDV have been reported in many countries since 2010.S protein is the main immunogenic protein containing some B-cell epitopes that can induce neutralizing antibodies of PEDV.In this study,the construction,expression and purification of Pseudomonas aeruginosa exotoxin A(PE)without domain Ⅲ(PE△Ⅲ)as a vector was performed for the delivery of PEDV S-A or S-B.PE(△Ⅲ)PEDV S-A and PE(△Ⅲ)PEDV S-B recombinant proteins were confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot analysis.The immunogenicity of PEDV S-A and PEDV S-B subunit vaccines were evaluated in mice.The results showed that PEDV-S-B vaccine could not only induce specific humoral and Th1 type-dominant cellular immune responses,but also stimulate PEDV-specific mucosal immune responses in mice.PEDV-S-B subunit vaccine is a novel candidate mucosal vaccine against PEDV infection.Leqiang Sun Yajie Tang Keji Yan Huanchun Chen Huawei Zhang 2021Animal Diseases2021,1,3:1
15Atomically Dispersed Fe-N_4 Modified with Precisely Located S for Highly Efficient Oxygen Reduction显示文摘Immobilizing metal atoms by multiple nitrogen atoms has triggered exceptional catalytic activity toward many critical electrochemical reactions due to their merits of highly unsaturated coordination and strong metal-substrate interaction.Herein,atomically dispersed Fe-NC material with precise sulfur modification to Fe periphery(termed as Fe-NSC) was synthesized,X-ray absorption near edge structure analysis confirmed the central Fe atom being stabilized in a specific configuration of Fe(N3)(N-C-S).By enabling precisely localized S doping,the electronic structure of Fe-N4 moiety could be mediated,leading to the beneficial adjustment of absorption/desorption properties of reactant/intermediate on Fe center.Density functional theory simulation suggested that more negative charge density would be localized over Fe-N4 moiety after S doping,allowing weakened binding capability to *OH intermediates and faster charge transfer from Fe center to O species.Electrochemical measurements revealed that the Fe-NSC sample exhibited significantly enhanced oxygen reduction reaction performance compared to the S-free Fe-NC material(termed as Fe-NC),showing an excellent onset potential of 1.09 V and half-wave potential of 0.92 V in 0.1 M KOH.Our work may enlighten relevant studies regarding to accessing improvement on the catalytic performance of atomically dispersed M-NC materials by managing precisely tuned local environments of M-Nx moiety.Yin Jia Xuya Xiong Danni Wang Xinxuan Duan Kai Sun Yajie Li Lirong Zheng Wenfeng Lin Mingdong Dong Guoxin Zhang Wen Liu Xiaoming Sun 2020Nano-Micro Letters2020,12,9:1
16Reconstructing avascular necrotic femoral head through a bioactiveβ-TCP system:From design to application显示文摘A variety of techniques have been used for treating avascular necrosis of the femoral head(ANFH),but have frequently failed.In this study,we proposed aβ-TCP system for the treatment of ANFH by boosting revascularization and bone regeneration.The angio-conductive properties and concurrent osteogenesis of the highly interconnected porousβ-TCP scaffold were revealed and quantified through an in vivo model that simulated the ischemic environment of ANFH.Mechanical test and finite element analysis showed that the mechanical loss caused by tissue necrosis and surgery was immediately partially compensated after implantation,and the strength of the operated femoral head was adaptively increased and eventually returned to normal bone,along with continuous material degradation and bone regeneration.For translational application,we further conducted a multi-center open-label clinical trial to assess the efficacy of theβ-TCP system in treating ANFH.Two hundred fourteen patients with 246 hips were enrolled for evaluation,and 82.1%of the operated hips survived at a 42.79-month median follow-up.The imaging results,hip function,and pain scores were dramatically improved compared to preoperative levels.ARCO stage II disease outperformed stage III in terms of clinical effectiveness.Thus,bio-adaptive reconstruction using theβ-TCP system is a promising hip-preserving strategy for the treatment of ANFH.Yajie Lu Xiantao Chen Xiao Lu Changning Sun Minghui Li Guojing Chen Zuoyao Long Yuan Gao Haoqiang Zhang Mengquan Huang Chuanlei Ji Hongbin Fan Dong Liu Yuewen Hao Hong Wang Leilei Zhang Hongmei Zhang Jianxi Lu Zhen Wang Jing Li 2023Bioactive Materials2023,,10:1

20Research on the Signal Reconstruction of the Phased Array Structural Health Monitoring Based Using the Basis Pursuit Algorithm显示文摘The signal processing problem has become increasingly complex and demand high acquisition system,this paper proposes a new method to reconstruct the structure phased array structural health monitoring signal.The method is derived from the compressive sensing theory and the signal is reconstructed by using the basis pursuit algorithm to process the ultrasonic phased array signals.According to the principles of the compressive sensing and signal processing method,non-sparse ultrasonic signals are converted to sparse signals by using sparse transform.The sparse coefficients are obtained by sparse decomposition of the original signal,and then the observation matrix is constructed according to the corresponding sparse coefficients.Finally,the original signal is reconstructed by using basis pursuit algorithm,and error analysis is carried on.Experimental research analysis shows that the signal reconstruction method can reduce the signal complexity and required the space efficiently.Yajie Sun Yanqing Yuan Qi Wang Lihua Wang Enlu Li Li Qiao 2019Computers, Materials & Continua2019,,2:1
