1阿尔茨海默病神经细胞凋亡与小胶质细胞活化的关系显示文摘目的 探讨阿尔茨海默病 (Alzheimer′sdisease ,AD)神经细胞凋亡与小胶质细胞活化的关系及活化的小胶质细胞在AD老年斑形成和进展中的作用。方法 收集 10例AD尸检材料及 4例非神经系统疾病死亡的老年尸检材料作为对照 ,进行白细胞介素 1α(IL 1α)免疫组织化学和原位DNA末端转移酶技术 (TUNEL)双重标记及淀粉样 β蛋白免疫组织化学和TUNEL双重标记。 结果 AD组静止的小胶质细胞数与对照组差异无显著性 (P >0 0 5 ) ,但活化的小胶质细胞数明显比对照组高 (P <0 0 1)。活化的小胶质细胞增大、变形 ,可进一步分为初级活化的小胶质细胞、增大的小胶质细胞及巨噬细胞 3种亚型。上述 3种活化的小胶质细胞数分别为 (5 4 0± 0 87)、(11 5 0± 1 2 5 )、(3 4 0± 0 32 )个 /mm2 ,占所有活化的小胶质细胞的比例分别为 2 6 6 0 % ,5 6 6 5 % ,16 75 %。AD组TUNEL阳性的凋亡的神经细胞数明显比对照组高 (P <0 0 5 ) ,且凋亡的神经细胞与活化的小胶质细胞之间具有相关性 (P <0 0 1)。活化的小胶质细胞在 4种类型老年斑中的分布比例不同 ,许多初级活化的小胶质细胞 (4 2 3% ) ,大多数增大的小胶质细胞和巨噬细胞 (分别是 5 6 2 %、70 6 % )分布在弥漫性神经突斑中。结论 小胶质细胞增生。刘冬戈 何淑蓉 张伟 崔娣 Yue kui Li W. 2Exogenous abscisic acid significantly affects proteome in teaplant (Camellia sinensis) exposed to drought stress显示文摘Tea[Camellia sinensis(L.)O.Kuntze]is an important economic crop,and drought is the most important abiotic stress affecting yield and quality.Abscisic acid(ABA)is an important phytohormone responsible for activating drought resistance.Increased understanding of ABA effects on tea plant under drought stress is essential to develop drought-tolerant tea genotypes,along with crop management practices that can mitigate drought stress.The objective of the present investigation is evaluation of effects of exogenous ABA on the leaf proteome in tea plant exposed to drought stress.Leaf protein patterns of tea plants under simulated drought stress[(polyethylene glycol(PEG)-treated]and exogenous ABA treatment were analyzed in a time-course experiment using two-dimensional electrophoresis(2-DE),followed by matrix-assisted laser desorption/ionization time-of-flight(MALDI-TOF)mass spectrometry(MS).Among the 72 protein spots identified by MALDI-TOF MS,16 proteins were downregulated and two were upregulated by exogenous ABA.The upregulated proteins have roles in glycolysis and photosystem II stabilization.Twenty-one protein spots were responsive to drought stress and most participate in carbohydrate and nitrogen metabolism,control of reactive oxygen species(ROS),defense,signaling or nucleic acid metabolism.The combined treatments of exogenous ABA and drought showed upregulation of 10 protein spots at 12 h and upregulation of 11 proteins at 72 h after initiation of drought stress.The results support the importance of the role that ABA plays in the tea plant during drought stress,by improving protein transport,carbon metabolism and expression of resistance proteins.Lin Zhou Hui Xu Sue Mischke Lyndel W Meinhardt Dapeng Zhang Xujun Zhu Xinghui Li Wanping Fang 2014Horticulture Research2014,1,1:12
3Acupuncture for cerebral ischemia/reperfusion injury Does it reduce the inflammatory reaction?显示文摘BACKGROUND:Nuclear factor-κB (NF-κB), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) in brain tissue can participate in inflammatory reactions after cerebral ischemia.Acupuncture treatment for acute cerebral ischemia produces abnormal protein expression.OBJECTIVE:To investigate the effects of acupuncture on NF-κB, ICAM-1, and VCAM-1 mRNA and protein expression in the brain tissue of rats with cerebral ischemia/reperfusion injury.DESIGN, TIME AND SETTING:Randomized, controlled, animal experiments were performed at the Laboratory of Department of Neurosurgery, Xiangya Hospital, Central South University, China between December 2008 and October 2009.MATERIALS:Rabbit anti-NF-κB polyclonal antibody, rabbit anti-ICAM-1 polyclonal antibody, and rabbit anti-VCAM-1 polyclonal antibody were purchased from Santa Cruz Biotechnology, USA.METHODS:A total of 46 healthy, Sprague Dawley rats were randomly assigned to sham surgery (n=10), model (n=12), acupuncture pretreatment (n=12), and acupuncture intervention (n=12) groups.Models of middle cerebral artery occlusion were established by right common carotid artery ligation.In the acupuncture pretreatment group, rats received acupuncture for 3 consecutive days, and then models were established.In the acupuncture intervention group, rats received acupuncture for 3 consecutive days at Waiguan (SJ 5), Sanyinjiao (SP 6), and Dazhui (DU 14) acupoints following model establishment.MAIN OUTCOME MEASURES:Somatosensory asymmetry and forelimb use asymmetry were tested, as well as NF-κB, ICAM-1, and VCAM-1 mRNA and protein expression in the frontal and parietal cortex at 4, 12, 24, 48 and 72 hours following cerebral ischemia/reperfusion injury.RESULTS:Acupuncture improved neurological function and significantly decreased NF-κB, ICAM-1, and VCAM-1 mRNA and protein expression in the frontal and parietal cortex of rats with cerebral ischemia/reperfusion injury (P<0.05).CONCLUSION:Acupuncture can improve neurological function, potentially via inhibition of NF-κB, ICAM-1, and VCAM-1 mRNA and protein expression in the frontal and parietal cortex of rats with cerebral ischemia/reperfusion injury.Zhenyan Li Guangwei Zhong Sujuan Huang Yunsheng Liu Sue Wang Wei Li 2010Neural Regeneration Research2010,5,14:4
4Development and evaluation of a safe and effective sugar-free herbal lollipop that kills cavity-causing bacteria显示文摘Dental caries (tooth decay) is caused by a specific group of cariogenic bacteria, like Streptococcus mutans, which convert dietary sugars into acids that dissolve the mineral in tooth structure. Killing cariogenic bacteria is an effective way to control or prevent tooth decay. In a previous study, we discovered a novel compound (Glycyrrhizol A), from the extraction of licorice roots, with strong antimicrobial activity against cariogenic bacteria. In the current study, we developed a method to produce these specific herbal extracts in large quantities, and then used these extracts to develop a sugar-free lollipop that effectively kills cariogenic bacteria like Streptococcus mutans. Further studies showed that these sugar-free lollipops are safe and their antimicrobial activity is stable. Two pilot human studies indicate that a brief application of these lollipops (twice a day for ten days) led to a marked reduction of cariogenic bacteria in oral cavity among most human subjects tested. 5Highly efficient and genotype-independent barley gene editing based on anther culture显示文摘Recalcitrance to tissue culture and genetic transformation is the major bottleneck for gene manipulation in crops.In barley,immature embryos of Golden Promise have typically been used as explants for transformation.However,the genotype dependence of this approach limits the genetic modification of commercial varieties.Here,we developed an anther culture-based system that permits the effective creation of transgenic and gene-edited plants from commercial barley varieties.The protocol was tested in Golden Promise and four Australian varieties,which differed in phenology,callus induction,and green plant regeneration responses.Agrobacterium-mediated transformation was performed on microspore-derived callus to target the HvPDS gene,and T0 albinos with targeted mutations were successfully obtained from commercial varieties.Further editing of three targets was achieved with an average mutation rate of 53%in the five varieties.In 51 analyzed T0 individuals,Cas9 induced a large proportion(69%)of single-base indels and two-base deletions in the target sites,with variable mutation rates among targets and varieties.Both ontarget and off-target activities were detected in T1 progenies.Compared with immature embryo protocols,this genotype-independent platformcan deliver a high editing efficiency and more regenerant plants within a similar time frame.It shows promise for functional genomics and the application of CRISPR technologies for the precise improvement of commercial varieties.Yong Han Sue Broughton Li Liu Xiao-Qi Zhang Jianbin Zeng Xiaoyan He Chengdao Li 2021Plant Communications2021,2,2:3
6A novel inexpensive conductive material:Er-modified maifanite by gaseous penetration technology显示文摘Aiming to solve the problem of corrosion and high cost of the conductive fillers, the powders with the properties of anti-oxidation, corrosion resistance and thermal stability should be developed. A new modification technology, rare earth gaseous penetration, was applied to enhance the conductivity of maifanite in this paper, and the mechanism for improving the conductivity of maifanite effectively was speculated in detail for the variations of the composition, structure and morphology between the original and Er-penetration maifanite powders. The results of the resistivity show that Er-penetration can really improve the conductivity of maifanite powders. When the penetration temperature is 500 ℃, the resistivity of maifanite surprisingly declines to 93.2 Ω·m. Moreover, XRD and XPS patterns display that the cations in maifanite are substituted by Er^(3+) successfully, resulting in the production of electrons in the system, which is the key reason to heighten the conductivity of maifanite.
