1Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. 2Genetic models of structural traps related to normal faults in the Putaohua Oilfield,Songliao Basin显示文摘The Putaohua Oilfi eld is a fault-prolifi c area and the faults have close relation with structural traps. The genetic models of the structural traps in the Putaohua Oilfi eld can be divided into two types: individual fault model and multi-fault interaction model. This is based on the description of displacement distribution of typical individual normal faults, the geometry of the footwall and hanging wall, and the analysis of the interaction between faults and the corresponding change in geometry when the faults grow. The individual fault model is that the displacement reaches a maximum at or near the center of fault and decreases toward the fault tips, so a half-graben is formed on the hanging wall of the fault and a half-anticline is formed on the footwall because of the isostatic process. The multi-fault interaction model is that during the growth of faults, they overlap and interact with each other, and accommodation zones are formed in the overlapping segments. The accommodation zones are favorable targets for hydrocarbon exploration, and the trap characteristics are dependent on the extent of overlap and occurrence of faults. The multi-fault interaction model can be subdivided into three types: synthetic accommodation zone, convergent accommodation zone and divergent accommodation zone. Hydrocarbon migration and accumulation models of each type have been developed. 3The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway显示文摘Polyphyllin I(PPI)and polyphyllin II(PII)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPI and PII exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepatotoxicity of PPI and PII was associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the cholesterol synthesis,selected as the potential targets,were confirmed by the molecular docking,the overexpression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPI could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPI-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future.Zhiqi Li Qiqi Fan Meilin Chen Ying Dong Farong Li Mingshuang Wang Yulin Gu Simin Guo Xianwen Ye Jiarui Wu Shengyun Dai Ruichao Lin Chongjun Zhao 2023Journal of Pharmaceutical Analysis2023,13,1:2

7Effects of soil organism interactions and temperature on carbon use efficiency in three different forest soils显示文摘Microbial carbon use efficiency(CUE)affects the soil C cycle to a great extent,but how soil organisms and the abiotic environment combine to influence CUE at a regional scale remains poorly understood.In the current study,microcosms were used to investigate how microbial respiration,biomass,and CUE responded to biotic and abiotic factors in natural tropical,subtropical,and temperate forests.Soil samples from the forests were collected,sterilized,and populated with one or a combination of three types of soil organisms(the fungus Botrytis cinerea,the bacterium Escherichia coli,and the nematode Caenorhabditis elegans).The microcosms were then kept at the mean soil temperatures of the corresponding forests.Microbial respiration,biomass,and CUE were measured over one-month incubation period.The results showed that microbial biomass and CUE were significantly higher,but microbial respiration lower in the subtropical and temperate forest soils than in tropical forest soil.Biotic factors mainly affected CUE by their effect on microbial biomass,while temperature affected CUE by altering respiration.Our results indicate that temperature regulates the interactive effects of soil organisms on microbial biomass,respiration,and CUE,which would provide a basis for understanding the soil C cycle in forest ecosystems.Simin Wang Xiaoyun Chen Debao Li Jianping Wu 2021Soil Ecology Letters2021,3,2:1
8Constructed TiO_(2)/WO_(3)heterojunction with strengthened nano-trees structure for highly stable electrochromic energy storage device显示文摘Tungsten trioxide(WO_(3))has been widely regarded as a prospective bifunctional material due to its electrochromic and pseudocapacitive properties,while still facing the dilemma of inadequate cycle stability and trapping-induced degradation.Here,inspired by the trees-strengthening approach,a unique titanium dioxide(TiO_(2))nanorod arrays strengthened WO_(3)nano-trees(TWNTs)heterojunction was rationally designed and constructed.In sharp contrast to the transmittance modulation(ΔT)attenuation of primary WO_(3)nano-trees during cycling,the TWNTs film showed not only excellent electrochromic performance but also fascinating cycle stability(77.35%retention of the initialΔT after 10,000 cycles).Besides,the trapping-induced degradation could be easily rejuvenated by a potentiostatic de-trapping process.An electrochromic energy storage device(EESD)was further assembled based on the TWNTs film to deliver excellentΔT(up to 79.5%at 633 nm),fast switching speed(tc/tb=1.9 s/14.8 s),extremely high coloration efficiency value(443.4 cm^(2)·C^(−1)),and long-term cycle stability(over 10,000 charge/discharge cycles).This innovative study provided in-depth insights into the electrochromism nature and a significant step in the realization of stable electrochromic-energy storage application,paving the way for multifunctional smart windows as well as next-generation optoelectronic devices.Lili Zhao Zhuoan Cai Xiaoyang Wang Wenbo Liao Simin Huang Lingyun Ye Jilie Fang Chunxing Wu Hao Qiu Lei Miao 2023Journal of Advanced Ceramics2023,12,3:1

11Chemo-drugs in cell microparticles reset antitumor activity of macrophages by activating lysosomal P450 and nuclear hnRNPA2B1显示文摘Macrophages in tumors(tumor-associated macrophages,TAMs),a major population within most tumors,play key homeostatic functions by stimulating angiogenesis,enhancing tumor cell growth,and suppressing antitumor immunity.Resetting TAMs by simple,efficacious and safe approach(s)is highly desirable to enhance antitumor immunity and attenuate tumor cell malignancy.Previously,we used tumor cell-derived microparticles to package chemotherapeutic drugs(drug-MPs),which resulted in a significant treatment outcome in human malignant pleural effusions via neutrophil recruitments,implicating that drug-MPs might reset TAMs,considering the inhibitory effects of M2 macrophages on neutrophil recruitment and activation.Here,we show that drug-MPs can function as an antitumor immunomodulator by resetting TAMs with M1 phenotype and IFN-βrelease.Mechanistically,drug molecules in tumor MPs activate macrophage lysosomal P450 monooxygenases,resulting in superoxide anion formation,which further amplifies lysosomal ROS production and pH value by activating lysosomal NOX2.Consequently,lysosomal Ca^(2+)signaling is activated,thus polarizing macrophages towards M1.Meanwhile,the drug molecules are delivered from lysosomes into the nucleus where they activate DNA sensor hnRNPA2B1 for IFN-βproduction.This lysosomal-nuclear machinery fully arouses the antitumor activity of macrophages by targeting both lysosomal pH and the nuclear innate immunity.These findings highlight that drug-MPs can act as a new immunotherapeutic approach by revitalizing antitumor activity of macrophages.This mechanistic elucidation can be translated to treat malignant ascites by drug-MPs combined with PD-1 blockade.Keke Wei Huafeng Zhang Shuaishuai Yang Yuxiao Cui Bingxia Zhang Jincheng Liu Liang Tang Yaoyao Tan Simin Liu Shiqi Chen Wu Yuan Xiao Luo Chen Chen Fei Li Junwei Liu Jie Chen Pingwei Xu Jiadi Lv Ke Tang Yi Zhang Jingwei Ma Bo Huang 2023Signal Transduction and Targeted Therapy2023,8,2:0
12Improving current and mitigating energy loss in ternary organic photovoltaics enabled by two well-compatible small molecule acceptors显示文摘Ternary organic photovoltaic(OPV)strategy is an effective but facile approach to enhance the photovoltaic performance for single-junction devices.Herein,a series of ternary OPVs were fabricated by employing a wide bandgap donor(PBDB-TF)and two acceptor-donor-acceptor(A-D-A)-type nonfullerene small molecule acceptors(NF-SMAs,called F-2 Cl and 3 TT-OCIC).As the third component,the near-infrared SMA,3 TT-OCIC,has complementary absorption spectrum,narrow bandgap and wellcompatible crystallization property to the host acceptor(F-2 Cl)for efficient ternary OPVs.With these,the optimal ternary devices yield significantly enhanced power conversion efficiency of 15.23%,one of the very few examples with PCE higher than15%other than Y6 systems.This is mainly attributed to the increased short-circuit current density of 24.92 m A cm^(-2) and dramatically decreased energy loss of 0.53 e V.This work presents a successful example for simultaneously improving current,minimizing energy loss and together with modifying the morphology of active layers in OPVs,which will contribute to the further construction of high performance ternary OPVs.Yanna Sun Huan-Huan Gao Simin Wu Lingxian Meng Xiangjian Wan Mengyang Li Zaifei Ma Ziqi Guo Shitong Li Hongtao Zhang Chenxi Li Yongsheng Chen 2021Science China Chemistry2021,64,4:0

14分化型甲状腺癌患者停用甲状腺素后短期甲减对血脂的影响(英文)显示文摘Objective: The aim of the study was to investigate the effects of short-term overt hypothyroidism on lipids after thyroxine-withdrawal in patients with iatrogenically induced hypothyroidism before radioiodine treatment for differentiated thyroid carcinoma (DTC). Methods: Thirty patients with a history of differentiated thyroid carcinoma on thyroid-stimulating hormone (TSH)-suppressive thyroxine replacement therapy were studied. Blood sample were taken before and 4 weeks after withdrawal of thyroxine substitution. Venous blood was drawn after an overnight fast and analyzed for serum free T4 (FT4), free T3 (FT3), thyroid-stimulating hormone (TSH), total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and apolipoprotein A1 (ApoA1), apolipoprotein B (ApoB). Fifty healthy people matched for age and gender were controls. Their blood samples were taken only once. Results: After thyroxine-withdrawal, the patients presented with increased serum TSH and low serum FT4 and FT3 levels compared with controls. Serum TG, TC, LDL, HDL, ApoB and Lp(a) increased after thyroxine withdrawal, reaching statistical significant differences with previous evaluation. However, when compared to euthyroid controls, only TC, LDL and ApoB were increased when patients were hypothyroidism. No changes were observed in ApoA1 in patients during thyroxine withdrawal, or when comparing the values observed in patients to those of euthyroid controls. 17Photodimerization of azaanthracene derivatives mediated by cucurbit[10]uril显示文摘Challenges of achieving efficient photodimerization of azaanthracene derivatives remain due to the low selectivity and slow reaction rate.In this paper,cucurbit[10]uril(CB[10]),with the largest rigid and hydrophobic cavity among CB[n]s,was used to affect the photodimerization reaction of four water-soluble 1-(2-)substituted azaanthracene derivatives(1-4).It revealed that 1-4 could form 1:2 host-guest complexes with CB[10]in aqueous solution.Irradiation of 1 in the presence of 0.5 equiv.of CB[10]selectively produced a head-to-tail(anti-HT)photodimer product.As for 2-4,CB[10]acted as a nanoreactor accelerating their photodimerization reaction in water.Our results suggest that photodimerization of azaanthracene derivatives could be promoted by the CB[10]-based host-guest strategy with high efficiency and selectivity.Huaxing Li Xianchen Hu Fengbo Liu Dongdong Sun Yong Wu Simin Liu 2022Chinese Chemical Letters2022,33,12:0
18Prenatal metal mixture exposure and birth weight: A two-stage analysis in two prospective cohort studies显示文摘The understanding of the impact of prenatal exposure to metal mixtures on birth weight is limited.We aimed to identify metal mixture components associated with birth weight and to determine additional pairwise interactions between metals showing such associations.Concentrations of 18 metals were measured using inductively coupled plasma mass spectrometry in urine samples collected in the 3rd trimester from a prenatal cohort(discovery;n=1849)and the Healthy Baby Cohort(replication;n=7255)in Wuhan,China.In the discovery set,we used two penalized regression models,i.e.,elastic net regression for main effects and a lasso for hierarchical interactions,to identify important mixture components associated with birth weight,which were then replicated.We observed that 8 of the 18 measured metals were retained by elastic net regression,with five metals(vanadium,manganese,iron,cesium,and barium)showing negative associations with Z-scores for birth weight and three metals(cobalt,zinc,and strontium)showing positive associations.In replication set,associations remained significant for vanadium(β=-0.035;95%confidence interval[CI],-0.059 to0.010),cobalt(β=-0.073;95%CI,0.049 to 0.097),and zinc(β=-0.040;95%CI,0.016 to 0.065)after Bonferroni correction.We additionally identified and replicated a single pairwise interaction between iron and copper exposure on birth weight(P<0.001).Using a two-stage analysis,we identified and replicated individual metals and additional pairwise interactions-associated birth weight.The approach could be used in other studies estimating the effect of complex mixtures on human health.Jie Hu George D.Papandonatos Tongzhang Zheng Joseph M.Braun Bin Zhang Wenyu Liu Chuansha Wu Aifen Zhou Simin Liu Stephen L.Buka Kunchong Shi Wei Xia Shunqing Xu Yuanyuan Li 2022Eco-Environment & Health2022,1,3:0

20Molten metal-organic complex to synthesize versatile ultrathin nonlayered oxides显示文摘Two-dimensional(2D)materials have attracted a great deal of research interest because of their unique electrical,magnetic,optical,mechanical,and catalytic properties for various applications.To date,however,it is still difficult to fabricate most functional oxides as 2D materials unless they have a layered structure.Herein,we report a one-step universal strategy for preparing versatile non-layered oxide nanosheets by directly annealing the mixture of metal nitrate and dimethyl imidazole(2-MI).The 2-MI plays the key role for 2D oxides since 2-MI owns a very low molten point and sublimation temperature,in which its molten liquid can coordinate with metal ions,forming a metal-organic framework,and easily puffing by its gas molecules.A total of 17 materials were prepared by this strategy,including non-layered metal oxide nanosheets as well as metal/metal oxide loaded nitrogen-doped carbon nanosheets.The as-prepared cobalt particle-loaded nitrogen-doped carbon nanosheets(Co@N/C)exhibit remarkable bifunctional oxygen reduction reaction(ORR)and oxygen evolution reaction(OER)electrocatalytic activity and durability.Besides,the Zn-air battery utilizing a Co@N/C catalyst exhibits high power density of 174.3 mW·cm^(-2).This facile strategy opens up a new way for large-scale synthesis of 2D oxides that holds great potential to push 2D oxides for practical applications.Kaisi Liu Jiabin Wu Qun Li Hongrun Jin Yongxin Luo Bei Qi Simin Dai Jianquan Zhao Liang Huang Jun Zhou 2024Nano Research2024,17,4:0
