2Are bone marrow regenerative cells ideal seed cells for the treatment of cerebral ischemia?显示文摘Bone marrow cells for the treatment of ischemic brain injury may depend on the secretion of a large number of neurotrophic factors. Bone marrow regenerative cells are capable of increasing the secretion of neurotrophic factors. In this study, after tail vein injection of 5-fluorouracil for 7 days, bone marrow cells and bone marrow regenerative cells were isolated from the tibias and femurs of rats, and then administered intravenously via the tail vein after focal cerebral ischemia. Immunohistological staining and reverse transcription-PCR detection showed that transplanted bone marrow cells and bone marrow regenerative cells could migrate and survive in the ischemic regions, such as the cortical and striatal infarction zone. These cells promote vascular endothelial cell growth factor mRNA expression in the ischemic marginal zone surrounding the ischemic penumbra of the cortical and striatal infarction zone, and have great advantages in promoting the recovery of neurological function, reducing infarct size and promoting angiogenesis. Bone marrow regenerative cells exhibited stronger neuroprotective effects than bone marrow cells. Our experimental findings indicate that bone marrow regenerative cells are preferable over bone marrow cells for cell therapy for neural regeneration after cerebral ischemia. 3Control measures during the COVID-19 outbreak reduced the transmission of hand,foot,and mouth disease显示文摘Control measures during the coronavirus disease 2019(COVID-19)outbreak may have limited the spread of infectious diseases.This study aimed to analyze the impact of COVID-19 on the spread of hand,foot,and mouth disease(HFMD)in China.A mathematical model was established to fit the reported data of HFMD in six selected cities in China's Mainland from 2015 to 2020.The absolute difference(AD)and relative difference(RD)between the reported incidence in 2020,and simulated maximum,minimum,or median incidence of HFMD in 2015-2019 were calculated.The incidence and R effof HFMD have decreased in six selected cities since the outbreak of COVID-19,and in the second half of 2020,the incidence and R effof HFMD have rebounded.The results show that the total attack rate(TAR)in 2020 was lower than the maximum,minimum,and median TAR fitted in previous years in six selected cities(except Changsha City).For the maximum,median,minimum fitted TAR,the range of RD(%)is 42·20-99·20%,36·35-98·41%48·35-96·23%(except Changsha City)respectively.The preventive and control measures of COVID-19 have significantly contributed to the containment of HFMD transmission.Yan Niu Li Luo Jia Rui Shiting Yang Bin Deng Zeyu Zhao Shengnan Lin Jingwen Xu Yuanzhao Zhu Yao Wang Meng Yang Xingchun Liu Tianlong Yang Weikang Liu Peihua Li Zhuoyang Li Chan Liu Jiefeng Huang Tianmu Chen 2021Journal of Safety Science and Resilience2021,2,2:3
4Microwave triggered multifunctional nanoplatform for targeted photothermal-chemotherapy in castration-resistant prostate cancer显示文摘Lacking a precise targeting strategy,castration-resistant prostate cancer(CRPC)is still hard to be treat effectively.Exploring treatment options that can accurately target CPRC is an important issue with urgent need.In this study,a novel nanotechnologybased strategy had been developed for the precise target treatment of CRPC.By combining microwaves and photothermal therapy(PTT),this nanoplatform,cmHSP70-PL-AuNC-DOX,targets tumor tissues with outstanding precision and achieves better anti-tumor activity by simultaneously eliciting photothermal and chemotherapeutic effects.From nanotechnology,cmHSP70-modified and thermo-sensitive liposome-coated AuNC-DOX were prepared and used for CRPC-targeted photothermal ablation and chemotherapy.Doxorubicin(DOX)was selected as the chemotherapeutic agent for cytotoxicity.In terms of the curative scheme,prostate tissues were firstly pre-treated with microwaves to induce the expression of heat shock protein 70(HSP70)and its migration to the cell membrane,which was then targeted by HSP70 antibody(cmHSP70)coated on the nanoparticles to achieve accurate drug delivery.The nanoplatform then achieved precise ablation and controlled release of DOX under external near-infrared(NIR)irradiation.Through the implementation,the targeting,cell killing,and safety of this therapeutical strategy had been verified in vivo and in vitro.This work establishes an accurate,controllable,efficient,non-invasive,and safe treatment platform for targeting CRPC,provides a rational design for CRPC’s PTT,and offers new prospects for nanomedicines with great precision.Pengyuan Liu Yajun Wu Xiaogang Xu Xudong Fan Chuan Sun Xiaojie Chen Jindan Xia Shiting Bai Li Qu Huasong Lu Jing Wu Jun Chen Ji-Gang Piao Zhibing Wu 2023Nano Research2023,16,7:2
5Mitochondria-localized cGAS suppresses ferroptosis to promote cancer progression显示文摘A well-established role of cyclic GMP-AMP synthase(cGAS)is the recognition of cytosolic DNA,which is linked to the activation of host defense programs against pathogens via stimulator of interferon genes(STING)-dependent innate immune response.Recent advance has also revealed that cGAS may be involved in several noninfectious contexts by localizing to subcellular compartments other than the cytosol.However,the subcellular localization and function of cGAS in different biological conditions is unclear;in particular,its role in cancer progression remains poorly understood.Here we show that cGAS is localized to mitochondria and protects hepatocellular carcinoma cells from ferroptosis in vitro and in vivo.cGAS anchors to the outer mitochondrial membrane where it associates with dynamin-related protein 1(DRP1)to facilitate its oligomerization.In the absence of cGAS or DRP1 oligomerization,mitochondrial ROS accumulation and ferroptosis increase,inhibiting tumor growth.Collectively,this previously unrecognized role for cGAS in orchestrating mitochondrial function and cancer progression suggests that cGAS interactions in mitochondria can serve as potential targets for new cancer interventions.Shiqiao Qiu Xiuying Zhong Xiang Meng Shiting Li Xiaoyu Qian Hui Lu Jin Cai Yi Zhang Mingjie Wang Zijian Ye Huafeng Zhang Ping Gao 2023Cell Research2023,33,4:2
6Electromyographic evaluation of functional electrical stimulation to injured oculomotor nerve显示文摘Functional electrical stimulation delivered early after injury to the proximal nerve stump has been proposed as a therapeutic approach for enhancing the speed and specificity of axonal regeneration following nerve injury. In this study, the injured oculomotor nerve was stimulated functionally by an implantable electrode. Electromyographic monitoring of the motor unit potential of the inferior oblique muscle was conducted for 12 weeks in two injury groups, one with and one without electric stimulation. The results revealed that, at 2, 4, 6, 8 weeks after functional electric stimulation of the injured oculomotor nerve, motor unit potentials significantly increased, such that amplitude was longer and spike duration gradually shortened. These findings indicate that the injured oculomotor nerve has the potential for regeneration and repair, but this ability is not sufficient for full functional recovery to occur. 10Dorsomorphin induces cancer cell apoptosis and sensitizes cancer cells to HSP90 and proteasome inhibitors by reducing nuclear heat shock factor 1 levels显示文摘Objective: Heat shock factor 1(HSF1), a transcriptional regulator of heat shock proteins(HSPs), is an attractive therapeutic target for cancer. However, only a few HSF1 inhibitors have been identified so far.Methods: The mRNA and protein levels of HSF1, HSPs, cleaved PARP, and phosphorylated HSF1 were examined by real-time PCR and Western blot. Forced expression, RNA interference, and immunofluorescence assay were used for mechanistic studies.Cell viability and apoptosis were measured by WST-8 assay and flow cytometry, respectively. Xenograft studies were performed in nude mice to evaluate the effect of dorsomorphin and an HSP90 inhibitor on tumor growth.Results: Dorsomorphin suppressed multiple stimuli-induced and constitutive HSPs expression in cancer cells. Mechanistic studies revealed that dorsomorphin reduced heat-induced HSP expression independent of adenosine monophosphate activated protein kinase. Dorsomorphin reduced heat-stimulated HSF1 Ser320 phosphorylation and nuclear translocation, as well as resting nuclear HSF1 levels in cancer cells. Dorsomorphin induced cancer cell apoptosis by inhibiting HSF1 expression. A structure-activity study revealed that the 4-pyridyl at the 3-site of the pyrazolo [1, 5-a]pyrimidine ring is critical for the anti-HSF1 activities of dorsomorphin. Dorsomorphin sensitized cancer cells to HSP90 and proteasome inhibitors and inhibited HSP70 expression induced by these inhibitors in vitro. In tumor-bearing nude mice, dorsomorphin enhanced HSP90 inhibitor-induced cancer cell apoptosis, tumor growth inhibition, and HSP70 expression.Conclusions: Dorsomorphin is an HSF1 inhibitor. 12Establishment of a beagle dog model of oculomotor nerve injury显示文摘The oculomotor nerves of beagle dogs received electrical stimulation at 0.3-2.0 V. After recording compound muscle action potentials of the inferior oblique muscle, the oculomotor nerve was quickly cut off and a direct end-to-end anastomosis was then performed. As a result, the stimulating elec-trode was smoothly inserted and placed, and ideal bioelectrical signals of the interior oblique muscle were acquired. After oculomotor nerve injury, compound muscle action potentials of the inferior oblique muscle were significantly decreased in beagle dogs. 13Discrepancies between Nonalcoholic and Metabolic-associated Fatty Liver Disease by Multiple Steatosis Assessment显示文摘Background and Aims:The redefinition of metabolic-as-sociated fatty liver disease(MAFLD)from nonalcoholic fat-ty liver disease(NAFLD)has caused a revolution in clinical practice,and the characteristics of patients with steatosis but not MAFLD remain unclear.The aims were to compare the diagnosis rate of MAFLD in NAFLD using different steato-sis methods and explore the features of non-MAFLD-NAFLD and MAFLD-non-NAFLD.Methods:A cross-sectional study enrolling consecutive individuals was conducted at three medical centers in southern China from January 2015 to September 2020.Steatosis was evaluated by liver biopsy or magnetic resonance imaging-based proton density fat frac-tion(MRI-PDFF),ultrasound,controlled attenuation param-eter(CAP),and fatty liver index(FLI).Fibrosis was assessed by the NAFLD fibrosis score,transient elastography,or shear wave elastography.Results:The study enrolled 14,985 Chi-nese adults.The agreement of MAFLD and NAFLD diagnoses were 83%for FLI,95%for ultrasound,94%for both CAP and MRI-PDFF,and 95%for liver biopsy.The body mass index,blood pressure and lipid levels among non-MAFLD-NAFLD pa-tients were similar metabolic parameters(p>0.05 for all),but not the alanine aminotransferase and the proportion of pa-tients with insulin resistance,which were significantly higher in non-MAFLD-NAFLD with significant fibrosis.Conclusions:The new MAFLD definition ruled out 5-17%of NAFLD cases.NAFLD and MAFLD-NAFLD involved more severe metabolic abnormalities than MAFLD and MAFLD-non-NAFLD.Non-MAFLD-NAFLD patients with significant fibrosis had more se-vere liver injury and increased glycemic dysregulation within the normal range.Attention should be paid to its progression.Congxiang Shao Junzhao Ye Xin Li Yansong Lin Shiting Feng Bing Liao Wei Wang Xiaorong Gong Bihui Zhong 2022Journal of Clinical and Translational Hepatology2022,10,6:0

15Comparison of clinical efficacy between LAPS and ALPPS in the treatment of hepatitis B virus-related hepatocellular carcinoma显示文摘Background:Insufficient post-operative future liver remnant(FLR)limits the feasibility of hepatectomy for patients.Staged hepatectomy is an effective surgical approach that can improve the resection rate of hepatocellular carcinoma(HCC).This study aimed to compare the safety and efficacy of laparoscopic microwave ablation and portal vein ligation for staged hepatectomy(LAPS)and classical associating liver partition and portal vein ligation for staged hepatectomy(ALPPS)in the treatment of hepatitis B virus(HBV)-related HCC.Methods:Clinical data of patients with HBV-related HCC who underwent LAPS or ALPPS in our institute between January 2017 and May 2022 were retrospectively analysed.Results:A total of 18 patients with HBV-related HCC were retrospectively analysed and divided into the LAPS group(n=9)and ALPPS group(n=9).Eight patients in the LAPS group and eight patients in the ALPPS group proceeded to a similar resection rate(88.9%vs 88.9%,P=1.000).The patients undergoing LAPS had a lower total comprehensive complication index than those undergoing ALPPS but there was not a significant different between the two groups(8.66 vs 35.87,P=0.054).The hypertrophy rate of FLR induced by ALPPS tended to be more rapid than that induced by LAPS(24.29 vs 13.17 mL/d,P=0.095).The 2-year recurrence-free survival(RFS)was 0%for ALPPS and 35.7%for LAPS(P=0.009),whereas the 2-year overall survival for ALPPS and LAPS was 33.3%and 100.0%(P=0.052),respectively.Conclusions:LAPS tended to induce lower morbidity and FLR hypertrophy more slowly than ALPPS,with a comparable resection rate and better long-term RFS in HBV-related HCC patients.Zebin Chen Shunli Shen Wenxuan Xie Junbin Liao Shiting Feng Shaoqiang Li Jiehui Tan Ming Kuang 2023Gastroenterology Report2023,11,1:0
16Material patterning on substrates by manipulation of fluidic behavior显示文摘Patterned materials on substrates are of great importance for a wide variety of applications.In solution-based approaches to material patterning;fluidic flow is inevitable.Here we demonstrate not only the importance of fluidic behavior but also the methodology of engineering the flow pattern to guide the material crystallization and assembly.We show by both experiment and simulation that substrate heating;which is generally used to accelerate evaporation,produces irregular complex vortexes.Instead;a top-heating-bottom-cooling(THBC)set-up offers an inverse temperature gradient and results in a single Marangoni vortex,which is desired for ordered nanomaterial patterning near the contact line.We then realize the fabrication of large-scale patterns of iodide perovskite crystals on different substrates under THBC conditions.We further demonstrate that harnessing the flow behavior is a general strategy with great feasibility to pattern various functional materials ranging from inorganic;organic,hybrid to biological categories on different substrates,presenting great potential for practical applications.Yitan Li Hao Wang Henglu Xu Shiting Wu Xuemei Li Jiapeng Yu Chaoyu Huang Zeyao Zhang Hao Sun Lu Han Meihui Li Anyuan Cao Zhenhai Pan Yan Li 2019National Science Review2019,6,4:0
17Transmissibility of tuberculosis among students and non-students:an occupational-specific mathematical modelling显示文摘Background: Recently, despite the steady decline in the tuberculosis (TB) epidemic globally, school TB outbreaks have been frequently reported in China. This study aimed to quantify the transmissibility ofMycobacterium tuberculosis (MTB) among students and non-students using a mathematical model to determine characteristics of TB transmission.Methods: We constructed a dataset of reported TB cases from four regions (Jilin Province, Xiamen City, Chuxiong Prefecture, and Wuhan City) in China from 2005 to 2019. We classified the population and the reported cases under student and non-student groups, and developed two mathematical models [nonseasonal model (Model A) and seasonal model (Model B)] based on the natural history and transmission features of TB. The effective reproduction number (R(eff)) of TB between groups were calculated using the collected data.Results: During the study period, data on 456,423 TB cases were collected from four regions: students accounted for 6.1% of cases. The goodness-of-fit analysis showed that Model A had a better fitting effect (P < 0.001). The average R_(eff) of TB estimated from Model A was 1.68 [interquartile range (IQR): 1.20–1.96] in Chuxiong Prefecture, 1.67 (IQR: 1.40–1.93) in Xiamen City, 1.75 (IQR: 1.37–2.02) in Jilin Province, and 1.79 (IQR: 1.56–2.02) in Wuhan City. The average R_(eff) of TB in the non-student population was 23.30 times (1.65/0.07) higher than that in the student population.Conclusions: The transmissibility of MTB remains high in the non-student population of the areas studied, which is still dominant in the spread of TB. TB transmissibility from the non-student-to-student-population had a strong influence on students. 18Multi-source coordinated stochastic restoration for SOP in distribution networks with a two-stage algorithm显示文摘After suffering from a grid blackout, distributed energy resources(DERs), such as local renewable energy and controllable distributed generators and energy storage can be used to restore loads enhancing the system’s resilience. In this study, a multi-source coordinated load restoration strategy was investigated for a distribution network with soft open points(SOPs). Here, the flexible regulation ability of the SOPs is fully utilized to improve the load restoration level while mitigating voltage deviations. Owing to the uncertainty, a scenario-based stochastic optimization approach was employed,and the load restoration problem was formulated as a mixed-integer nonlinear programming model. A computationally efficient solution algorithm was developed for the model using convex relaxation and linearization methods. The algorithm is organized into a two-stage structure, in which the energy storage system is dispatched in the first stage by solving a relaxed convex problem. In the second stage, an integer programming problem is calculated to acquire the outputs of both SOPs and power resources. 19Regenerating nerve fiber innervation of extraocular muscles and motor functional changes following oculomotor nerve injuries at different sites显示文摘In the present study, the oculomotor nerves were sectioned at the proximal (subtentorial) and distal (superior orbital fissure) ends and repaired. After 24 weeks, vestibulo-ocular reflex evaluation confirmed that the regenerating nerve fibers following oculomotor nerve injury in the superior orbital fissure had a high level of specificity for innervating extraocular muscles. The level of functional recovery of extraocular muscles in rats in the superior orbital fissure injury group was remarkably superior over that in rats undergoing oculomotor nerve injuries at the proximal end (subtentorium). Horseradish peroxidase retrograde tracing through the right superior rectus muscle showed that the distribution of neurons in the nucleus of the oculomotor nerve was directly associated with the injury site, and that crude fibers were badly damaged. The closer the site of injury of the oculomotor nerve was to the extraocular muscle, the better the recovery of neurological function was.
