| 1 | Knockdown of OsHox33, a member of the class III homeodomain-leucine zipper gene family, accelerates leaf senescence in rice显示文摘The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants. | LUAN WeiJiang SHEN Ao JIN ZhiPing SONG SuSheng LI ZhengLong SHA AiHua | 2013 | Science China(Life Sciences)2013,56,12: | 13 |
| 2 | Evapotranspiration and Its Energy Exchange in Alpine Meadow Ecosystem on the Qinghai-Tibetan Plateau显示文摘To understand the water and energy exchange on the Qinghai-Tibetan Plateau, we explored the characteristics of evapotranspiration (ET) and energy fluxes from 2002 to 2005 over a Kobresia meadow ecosystem using the eddy covariance method. The ratio of annual ET to precipitation (P) of meadow ecosystem was about 60%, but varied greatly with the change of season from summer to winter. The annual ET/P in meadow was lower than that in shrub, steppe and wetland ecosystems of this plateau. The incident solar radiation (R s ) received by the meadow was obviously higher than that of lowland in the same latitude; however the ratio of net radiation (R n ) to R s with average annual value of 0.44 was significantly lower than that in the same latitude. The average annual ET was about 390 mm for 2002-2005, of which more than 80% occurred in growing season from May to September. The energy consumed on the ET was about 44% of net radiation in growing season, which was lower than that of shrub, steppe and wetland on this plateau. | 4 | Effects of Primary Annealing Condition on Recrystallization Texture in a Grain Oriented Silicon Steel显示文摘The recrystallization texture in grain oriented silicon steel sheets, which were annealed at different primary annealing temperatures with and without an electric field, was investigated. An automated electron backscattered diffraction (EBSD) technique was used to analyze the recrystallization texture. It was found that recovery and application of electric field in primary annealing lead to an increase of {001} component and a decrease of {111} component after annealing at 900℃. | 7 | Computational analysis of antigenic epitopes of avian influenza A (H7N9) viruses显示文摘Influenza virus can rapidly change its antigenicity, via mutation in the hemagglutinin(HA) protein, to evade host immunity. The emergence of the novel human-infecting avian H7N9 virus in China has caused widespread concern. However, evolution of the antigenicity of this virus is not well understood. Here, we inferred the antigenic epitopes of the HA protein from all H7 viruses, based on the five well-characterized HA epitopes of the human H3N2 virus. By comparing the two major H7 phylogenetic lineages, i.e., the Eurasian lineage and the North American lineage, we found that epitopes A and B are more frequently mutated in the Eurasian lineage, while epitopes B and C are more frequently mutated in the North American lineage. Furthermore, we found that the novel H7N9 virus(derived from the Eurasian lineage) isolated in China in the year 2013, contains six frequently mutated sites on epitopes that include site 135, which is located in the receptor binding domain. This indicates that the novel H7N9 virus that infects human may already have been subjected to gradual immune pressure and receptor-binding variation. | 8 | Effects of leaf removal and cluster thinning on berry quality of Vitis vinifera cultivars in the region of Weibei Dryland in China显示文摘Leaf removal and cluster thinning were carried out prior to veraison to evaluate the effects on berry quality of two Vitis vinifera cultivars(Cabernet Sauvignon and Ugni Blanc) in the Weibei Dryland of China in 2013 and 2014, and comprehensive analysis of the chemical and volatile composition in berries was performed. The results showed that content of reducing sugar in both varieties was not affected while total acid was generally decreased by leaf removal and cluster thinning. The pH of berry juice was correspondingly higher in most treatment groups. Meanwhile, promoting effects on accumulation of total phenols, tannin in both varieties and total anthocyanins in Cabernet Sauvignon were found. As for monomeric anthocyanins, percentage of malvidin and its derivatives was decreased by leaf removal and cluster thinning. Besides, cinnamylated anthocyanins decreased with the intensity of cluster thinning. The accumulation of non-anthocyanin phenolics was similarly affected in the two varieties. Notably, cluster thinning was more effective on enhancing the phenolics content than leaf removal. The combination of middle level of leaf removal and cluster thinning was the most favor to the accumulation of phenolic acids. Furthermore, cluster thinning could also significantly enhance the synthesis of flavanols and stilbenes. Lastly, content and variety of aroma compounds in both grape varieties were also significantly affected by the treatments. | 11 | Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects. | 12 | Comparative analysis of the clinical outcomes between wavefront-guided and conventional femtosecond LASIK in myopia and myopia astigmatism显示文摘AIM:To compare the clinical outcomes of wavefront guided femtosecond LASIK(WFG LASIK)and conventional femtosecond LASIK(NWFG LASIK)in eyes with myopia and myopia astigmatism.METHODS:This was a retrospective,nonrandomized,comparative investigation enrolling 236 eyes of 122 patients(18-50 y)with low&moderate and high myopia.The WFG group including 97 eyes(50 patients)undergone WFG LASIK and the NWFG group including 139 eyes(72 patients)undergone conventional LASIK.Mean efficacy index,high order aberrations(HOAs),pupil size and the quality of visual questionnaire were evaluated 6 mo postoperatively.RESULTS:There is no difference between WFG group(-0.054±0.049 in logMAR)and NWFG group(-0.040±0.056)in uncorrected distance visual acuity(UDVA)postoperatively.The myopia astigmatism is higher in WFG group than that in NWFG group(P<0.05).However,the mean efficacy index(MEI)in the WFG group(1.09±0.106)is better than that in the NWFG group(1.036±0.124;P<0.001).Increased HOAs were observed in NWFG group(0.30±0.196)than that in WFG group(0.146±0.188;P<0.001).The pupil size is larger in WFG group(5.15±0.76 mm)than that in NWFG group(4.32±0.52 mm).The patients are satisfied with the clinical surgery,yet WFG group showed better visual quality using the questionnaire survey.Meanwhile,high myopia would result in worse MEI,HOAs and visual quality than low&moderate myopia.CONCLUSION:WFG and NWFG FS-LASIK are both effective and safe procedures to correct low&moderate and high myopia,but WFG FS-LASIK gives a better postoperative MEI,aberrometric control and predictable outcome.Meanwhile,WFG FS-LASIK is better than NWFG FS-LASIK in correction of myopia astigmatism.Low&moderate myopia allow better clinical outcomes than high myopia using any surgical method. | Xiao Gui Rui Zhang Sha Li Na Zhao Hao-Rui Zhang Yu-Kun Zhou Chen-Yang Huan Chun-Yan Zhao Hong-Ying Wang Hong-Yuan Song Wei Shen Jian-Hua Zhang | 2021 | International Journal of Ophthalmology(English edition)2021,14,10: | 5 |

| 14 | Redirecting differentiation of mammary progenitor cells by 3D bioprinted sweat gland microenvironment显示文摘Background:Mammary progenitor cells(MPCs)maintain their reproductive potency through life,and their specific microenvironments exert a deterministic control over these cells.MPCs provides one kind of ideal tools for studying engineered microenvironmental influence because of its accessibility and continually undergoes postnatal developmental changes.The aim of our study is to explore the critical role of the engineered sweat gland(SG)microenvironment in reprogramming MPCs into functional SG cells.Methods:We have utilized a three-dimensional(3D)SG microenvironment composed of gelatin-alginate hydrogels and components from mouse SG extracellular matrix(SG-ECM)proteins to reroute the differentiation of MPCs to study the functions of this microenvironment.MPCs were encapsulated into the artificial SG microenvironment and were printed into a 3D cell-laden construct.The expression of specific markers at the protein and gene levels was detected after cultured 14 days.Results:Compared with the control group,immunofluorescence and gene expression assay demonstrated that MPCs encapsulated in the bioprinted 3D-SG microenvironment could significantly express the functional marker of mouse SG,sodium/potassium channel protein ATP1a1,and tend to express the specific marker of luminal epithelial cells,keratin-8.When the Shh pathway is inhibited,the expression of SG-associated proteins in MPCs under the same induction environment is significantly reduced.Conclusions:Our evidence proved the ability of differentiated mouse MPCs to regenerate SG cells by engineered SG microenvironment in vitro and Shh pathway was found to be correlated with the changes in the differentiation.These results provide insights into regeneration of damaged SG by MPCs and the role of the engineered microenvironment in reprogramming cell fate. | Rui Wang Yihui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu | 2019 | Burns & Trauma2019,7,1: | 5 |

The magnet generates a field similar to sinusoid with the maximum of 0.9 T in the channel when it is rotated up by the motor to simulate an AC magnetic field. This moving magnetic field acts on the conductive fluid in the channel, and produces an electromagnetic force to move the fluid in the same direction as that of the magnet rotating. Experiments were carried out to investigate the performance of the pump. Flow velocity in the annular channel was measured for different conductivities and rotating speeds of the magnet. They have been developed from herpes simplex virus 1(HSV-1) strain H129(H129), and have been successfully applied to map diverse neural circuits.Initially, the anterograde polysynaptic tracer H129-G4 was used by many groups. We then developed the first monosynaptic tracer, H129-dTK-tdT, which was highly successful, yet improvements are needed. Now, by inserting another tdTomato expression cassette into the H129-dTK-tdT genome, we have created H129-dTK-T2, an updated version of H129-dTK-tdT that has improved labeling intensity. To help scientists produce and apply our H129-derived viral tracers, here we provide the protocol describing our detailed and standardized procedures. Commonly-encountered technical problems and their solutions are also discussed in detail. | 20 | Role of GA_3,GA_4 and Uniconazole-P in Controlling Gravitropism and Tension Wood Formation in Fraxinus mandshurica Rupr. var. japonica Maxim. Seedlings显示文摘GA3 and GA4(gibberellins) play an important role in controlling gravitropism and tension wood formation in woody angiosperms. In order to improve our understanding of the role of GA3 and GA4 on xylem cell formation and the G-layer,we studied the effect of GA3 and GA4 and uniconazole-P,which is an inhibitor of GA biosynthesis,on tension wood formation by gravity in Fraxinus mandshurica Rupr. var. japonica Maxim. seedlings. Forty seedlings were divided into two groups;one group was placed upright and the other tilted. Each group was further divided into four sub-groups subjected to the following treatments:3.43 × 10-9 μmol acetone as control,5.78 × 10-8 μmol gibberellic acid(GA3),6.21 × 10-8 μmol GA4,and 6.86 × 10-8 μmol uniconazole-P. During the experimental period,GAs-treated seedlings exhibited negative gravitropism,whereas application of uniconazole-P inhibited negative gravitropic stem bending. GA3 and GA4 promoted wood fibers that possessed a gelatinous layer on the upper side,whereas uniconazole-P inhibited wood formation but did not inhibit the differentiation of the gelatinous layer in wood fibers on the upper side.
