1The Influence of Phacoemulsification on Corneal Endothelia Cells at Varying Blood Glucose Levels显示文摘Purpose:To investigate the influence of phacoemulsification on corneal endothelial cells in diabetes patients and normal controls. Methods:Phacoemulsification and intraocular lens implantation were performed on 75 patients with diabetic cataract (126 eyes) who were divided into two groups: Trial group 1 (Glu≤6 mmol/L); Trial group 2 (Glu 6-10 mmol/L) and 65 nondiabetic controls (112 eyes).The density and percentage of hexagonal cells and coefficient of variation of the corneal endothelia were measured before surgery and 1 day,1 month and 3 months postoperatively. Results: There was no statistical difference in the density and percentage of hexagonal cells and coefficient of variation of the corneal endothelia prior to phacoemulsification.(P>0.05) between the three groups.The density and percentage of hexagonal cells of the corneal endothelia decreased significantly after surgery in all three groups(P<0.05),while the Coefficient of variation increased in all groups(P<0.05). The rate of loss of corneal endothelial cells in the diabetic groups was significantly higher than for the control group (P<0.05), the percentage of hexagonal cells in the diabetic groups was significantly lower than for the control group (P<0.05), and the coefficient of variation in the diabetic groups was significantly higher than for the control group (P<0.05) at 1 day, 1 month and 3 months post-operatively. There was no statistical difference between the two diabetic groups in terms of these post-operative measurements. Conclusion:The corneal endothelium of diabetic patients is more prone to damage from phacoemulsification. 2Risk factors and outcomes of acute kidney injury after intracoronary stent implantation显示文摘BACKGROUND:Acute kidney injury following percutaneous coronary intervention(PCI) is associated with a worse outcome.However,the risk factors and outcomes of acute kidney injury(AKI) in patients after intracoronary stent implantation are still unknown.METHODS:A retrospective case control study was done in 325 patients who underwent intracoronary stent implantation from January 2010 to March 2011 at the Drum Tower Hospital of Nanjing University School of Medicine.Those were excluded from the study if they had incomplete clinical data.The patients were divided into a normal group and a AKI group according to the standard of post-operation day 7 to identify AKI.The parameters of the patients included:1) pre-operative ones:age,gender,hypertension,diabetes mellitus,cerebrovascular disease,left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate(eGFR),hyperuricemia,proteinuria,emergency operation,hydration,medications(ACEI/ARBs,statins);2) intraoperative ones:dose of contrast media,operative time,hypotension;and 3) postoperative one:hypotension.The parameters were analyzed with univariate analysis and multivariate logistical regression analysis.RESULTS:Of the 325 patients,51(15.7%) developed AKI.Hospital day and in-hospital mortality were increased significantly in the AKI-group.Univariate analysis showed that age,pre-operative parameters(left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate,hyperuricemia,proteinuria,hydration),emergency operation,intraoperative parameters(operative time,hypotension) and postoperative hypotension were significantly different.However,multivariate logistic regression analysis revealed that increased age(OR=0.253,95%CI=0.088-0.727),pre-operative proteinuria(OR=5.351,95%CI=2.128-13.459),pre-operative left ventricular insufficiency(OR=8.704,95%CI=3.170-23.898),eGFR<60 ml/min/1.73 m^2(OR=6.677,95%CI = 1.167-38.193),prolonged operative time,intraoperative hypotension(OR=25.245,95%CI=1.001-1.034) were independent risk factors of AKI.CONCLUSIONS:AKI is a common complication and associated with ominous outcome following intracoronary stent implantation.Increased age,pre-operative proteinuria,pre-operative left ventricular insufficiency,pre-operative low estimated glomerular filtration rate,prolonged operative time,intraoperative hypotension were the significant risk factors of AKI.Fei He Jun Zhang Zhong-qiu Lu Qing-ling Gao Du-juan Sha Li-gang Pei Guo-feng Fan 2012World Journal of Emergency Medicine2012,3,3:11

5Highly sensitive detection of melamine based on reversed phase liquid chromatography mass spectrometry显示文摘In this work, we developed a highly sensitive method to detect melamine based on reversed phase liquid chromatography mass spectrometry. A mass spectrometry compatible ion pair, heptafluorobu-tyric acid(HFBA), was used to separate melamine by reversed phase liquid chromatography prior to electrospray mass spectrometry. The incorporation of isotope internal standard and multiple reaction monitoring improved the accuracy and linearity of quantification. Based on this strategy, the method limit of quantification was 0.1 ng/g. The limits of quantification were 8 ng/g for liquid milk and 15 ng/g for dry milk powder. 6Orthogonal design to sift the optimal parameter of Neiguan acupuncture for cerebral infarction显示文摘The individual difference and non-repeatability in acupuncture have not only restricted the development of acupuncture,but have also affected the specificity of acupoints.The present study used instruments to control needle depth,lifting and thrusting frequency,and the duration of acupuncture.Effects of the quantified acupuncture were observed at Neiguan(PC6)with different stimulation parameters.A frequency of 1,2,or 3 Hz and duration of 5,60,or 180 seconds were used to observe cerebral blood flow and ratio of infarct volume recovery.Results showed that stimulation at Neiguan with a frequency of 1 Hz and long duration of 180 seconds or 2/3 Hz and long duration of 5/60seconds significantly increased cerebral blood flow and decreased the ratio of infarct volume.Interactions between frequency and duration play a critical role in quantified acupuncture therapy.Yanan Zhang Sha Yang Xiaonong Fan Shu Wang Nina He Lingxin Li Ding Luo Xuemin Shi 2013Neural Regeneration Research2013,8,28:7
7ERK1/2 Activities Are Dispensable for Oocyte Growth but Are Required for Meiotic Maturation and Pronuclear Formation in Mouse显示文摘Previous studies revealed that extracellular regulated kinase-1 and-2(ERK1/2) cascade plays pivotal roles in regulating oocyte meiotic cell cycle progression. However, most knowledge about the in vivo function of ERK1/2 in mammalian oocytes was indirectly obtained from analyzing the phenotypes of Mos knockout mice. In this study, we knocked out Erk1 and Erk2 in mouse oocytes as early as the primordial follicle stage using the well-characterized Gdf9-Cre mouse model, and for the first time directly investigated the in vivo function of ERK1/2 in mouse oocytes. In this novel mouse model, we observed that ERK1/2 activities in oocyte are dispensable for primordial follicle maintenance,activation and follicle growth. Different from the Mos null oocytes, the ERK1/2-deleted oocytes had well-assembled spindles at metaphase Ⅰ(MⅠ), extruded polar body-1(PB1) with normal sizes, and did not undergo a full parthenogenetic activation characterized for pronuclear formation. However, the ovulated ERK1/2-deleted oocytes had poorly-assembled metaphase Ⅱ(MⅡ) spindles, spontaneously released polar body-2(PB2), and were arrested at another metaphase called metaphase Ⅲ(MⅢ). In addition, ERK1/2 deletion prevented male pronuclear formation after fertilization, and caused female infertility. 8Design, Synthesis, and Biological Activity of Novel Aromatic Amide Derivatives Containing Sulfide and Sulfone Substructures显示文摘In recent years,the damage caused by soil nematodes has become increasingly serious;however,the varieties and structures of the nematicides available on the market are deficient.Fluopyram,a succinate dehydrogenase inhibitor(SDHI)fungicide developed by Bayer AG in Germany,has been widely used in the prevention and control of soil nematodes due to its high efficiency and novel mechanism of action.In this paper,two series of novel target compounds were designed and synthesized with nematicidal and fungicidal fluopyram as the molecular skeleton in order to introduce sulfide and sulfone substructures.The structures were identified and characterized by 1H nuclear magnetic resonance(NMR),13C NMR,and high-resolution mass spectrometer(HRMS).The bioassays revealed that most of the compounds showed excellent nematicidal activities at 200 lgmL-1 in comparison with fluopyram,while the nematode mortality rate dropped sharply at 100μg·mL-1,except for compounds I-11 and II-6.In terms of fungicidal activity,compound I-9 was discovered to have an excellent inhibitory rate,and a molecular docking simulation was performed that can provide important guidance for the design and exploration of efficient fungicidal lead compounds.Xuewen Hua Nannan Liu Sha Zhou Leilei Zhana Hao Yin Guiqing Wang Zhijin Fan Yi Mac 2020Engineering2020,6,5:4
9Effects of transfected adenovirus-mediated transcription factor X-box binding protein 1 on hippocampal-derived neural stem cell proliferation and apoptosis under hypoxia显示文摘BACKGROUND:Neural stem cell(NSC) survival is closely associated with cell apoptosis in ischemic-hypoxic regions following transplantation.Numerous studies have revealed that X-box binding protein 1(XBP1) is a transcription factor during endoplasmic reticulum unfolded protein response and is essential for cell survival,differentiation,and anti-apoptotic effects.OBJECTIVE:To determine the effects of the XBP1 gene on NSC proliferation and apoptosis under hypoxic conditions following XBP1 gene transfection into rat embryonic hippocampal NSCs using recombinant adenovirus vector.DESIGN,TIME AND SETTING:In vitro experiments were performed at the Laboratory of Cell Biology of Jilin University and Laboratory of Proteomics,Department of Neurology,Jilin University,China from September 2008 to November 2009.MATERIALS:Recombinant adenovirus package XBP1 gene and Ad-XBP1-enhanced green fluorescent protein plasmid(Guangzhou Easywin BioMed Technology,China),rabbit anti-XBP1 and its target gene estrogen receptor degradation-enhancing α-mannosidase-like protein(EDEM),glucose-regulated protein 78(GRP78),anti-apoptotic molecule Bcl-2 and proapoptotic molecule Bax polyclonal antibody(Santa Cruz Biotechnology,Inc.,Santa Cruz,CA,USA),and CoCl2(Sigma,St.Louis,MO,USA) were used in the present study.METHODS:Hippocampi from embryonic,Sprague Dawley rats on gestational day 16 were harvested for NSC isolation and cloning,followed by immunofluorescence for Nestin and sub-culturing.The recombinant adenovirus Ad-XBP1-enhanced green fluorescent protein plasmid was transfected into rat embryonic hippocampal NSCs,and then CoCl2 was applied to induce hypoxia.MAIN OUTCOME MEASURES:Cell quantification and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide colorimetric assay were utilized to detect proliferation in XBP1-transfected NSCs for 7 consecutive days.Western blot assay was utilized to quantify XBP1,GRP78,EDEM,Bcl-2,and Bax expression.Flow cytometry was used to measure apoptosis.RESULTS:NSC proliferation was significantly enhanced following XBP1 gene transfection(P < 0.05).Under hypoxic conditions,GRP78,EDEM,and Bcl-2 levels increased,but Bax levels decreased.In addition,NSC apoptosis decreased following transfection(P < 0.05).CONCLUSION:The XBP1 gene was successfully transfected into rat embryonic hippocampal NSCs using a recombinant adenovirus vector.NSC proliferation following transfection,as well as anti-apoptotic effects under hypoxia,was significantly increased.Ying Sha Baohua Liu Qun Liu Lei Song Jia Fan Yong Liu 2010Neural Regeneration Research2010,5,13:4
10Establishment and Characterization of a New Cell Line from the Kidney of Spotted Halibut Verasper variegates显示文摘A cell line,SHK,was derived from the kidney of spotted halibut Verasper variegates.The cell line was subculturedmore than 40 passages in minimum essential medium(MEM)supplemented with fetal bovine serum(FBS)and 10 ng ml-1basic fi-broblast growth factor(bFGF).Cell morphology from primary culture and subculture was observed continuously by microscopy.TheSHK cell line consisted predominantly of fibroblast-like cells.The cell line was able to grow between 20℃and 30℃with the optimum growth at 24℃and with a reduced growth between 12℃and 20℃.The growth rate of the cells increased as the proportion ofFBS increased from 10%to 20%at 28℃with optimum growth at the concentration of 20%.The doubling time of the cells was de-termined to be 44.8 h.Chromosome analysis revealed that 52%of the SHK cells maintained a normal diploid chromosome number(2n=46).The cells were successfully transfected with green fluorescent protein(GFP)reporter plasmids and the expression of GFPgene in the cells indicated the possible utility of the cells in gene expression studies.The cells were infected by lymphosystis diseasevirus(LCDV)and found to be susceptible to the virus in cytopathic effect(CPE)observation.The infection was confirmed by PCRand electron microscopy experiments,which proved the existence of the viral particles in the cytoplasm of the virus-infected cells.WANG Xianli CHEN Songlin SHA Zhenxia FAN Tingjun WANG Na 2010Journal of Ocean University of China2010,9,2:4

13A Distributed Approach of Big Data Mining for Financial Fraud Detection in a Supply Chain显示文摘Supply Chain Finance(SCF)is important for improving the effectiveness of supply chain capital operations and reducing the overall management cost of a supply chain.In recent years,with the deep integration of supply chain and Internet,Big Data,Artificial Intelligence,Internet of Things,Blockchain,etc.,the efficiency of supply chain financial services can be greatly promoted through building more customized risk pricing models and conducting more rigorous investment decision-making processes.However,with the rapid development of new technologies,the SCF data has been massively increased and new financial fraud behaviors or patterns are becoming more covertly scattered among normal ones.The lack of enough capability to handle the big data volumes and mitigate the financial frauds may lead to huge losses in supply chains.In this article,a distributed approach of big data mining is proposed for financial fraud detection in a supply chain,which implements the distributed deep learning model of Convolutional Neural Network(CNN)on big data infrastructure of Apache Spark and Hadoop to speed up the processing of the large dataset in parallel and reduce the processing time significantly.By training and testing on the continually updated SCF dataset,the approach can intelligently and automatically classify the massive data samples and discover the fraudulent financing behaviors,so as to enhance the financial fraud detection with high precision and recall rates,and reduce the losses of frauds in a supply chain.Hangjun Zhou Guang Sun Sha Fu Xiaoping Fan Wangdong Jiang Shuting Hu Lingjiao Li 2020Computers, Materials & Continua2020,,8:3
14The Altitudinal Belts of Subalpine Virgin Forest on Mt.Gongga Simulated by a Succession Model显示文摘How to accurately simulate the distribution of forest species based upon their biological attributes has been a traditional biogeographical issue.Forest gap models are very useful tools for examining the dynamics of forest succession and revealing the species structure of vegetation.In the present study,the GFSM(Gongga Forest Succession Model) was developed and applied to simulate the distribution,composition and succession process of forests in 100 m elevation intervals.The results indicate that the simulated results of the tree species,quantities of the different types of trees,tree age and differences in DBH(diameter at breast height) composition were in line with the actual situation from 1400 to 3700 MASL(meters above sea level) on the eastern slope of Mt.Gongga.Moreover,the dominant species in the simulated results were the same as those in the surveyed database.Thus,the GFSM model can best simulate the features of forest dynamics and structure in the natural conditions of Mt.Gongga.The work provides a new approach to studying the structure and distribution characteristics of mountain ecosystems in varied elevations.Moreover,the results of this study suggest that the biogeochemistry mechanism model should be combined with the forestsuccession model to facilitate the ecological model in simulating the physical and chemical processes involved.CHENG Gen-wei SUN Jian SHA Yu-kun FAN Ji-hui 2014Journal of Mountain Science2014,11,6:3
15Response dynamics of three defense related enzymes in cotton leaves to the interactive stress of Helicoverpa armigera (Hübner) herbivory and omethoate application显示文摘In order to explore the response dynamics of the activities of defense related enzymes in cotton leaves towards the interactive stress of Helicoverpa armigera herbivory and omethoate application, the activities of phenylalanine ammonia-lyase(PAL), lipoxygenase(LOX), and polyphenol oxidase(PPO) were examined from 6 to 126 h after cotton leaves were treated 12 h of H. armigera herbivory, and then sprayed with 800 mg L–1 omethoate. The results showed that the changes in the activities of PAL, LOX and PPO that occured under the interactive stress of H. armigera herbivory and omethoate application reflected the interactive effects of the two stresses on cotton defense. The similarity between the response dynamics of PAL, LOX, and PPO activities in cotton leaves under the interactive stress and that under H. armigera herbivory treatment alone showed that the induction of H. armigera herbivory on the activities of PAL, LOX and PPO in cotton leaves played a leading role in the interactive effects, and the effect of omethoate application played only a minor role. A joint factor analysis was performed according to a method which has been used to analyze the joint toxicity of pesticides; this analysis sought to clarify if there was a synergistic, antagonistic, or additive effect on PAL, LOX, and PPO activity in cotton leaves resulting from the interactive H. armigera herbivory and omethoate treatment. In the interactive effect on the response of PAL activity in cotton leaves, antagonistic effects of the omethoate application towards H. armigera herbivory were observed at 6 and 12 h. Synergistic effects were then observed at 18 and 30 h. Antagonistic effects were observed from 54 to 78 h and synergistic effects were finally observed at 126 h. The correlation between H. armigera herbivory and omethoate application in the interactive effect on cotton defense responses of LOX activity also fluctuated from synergism to antagonism during the time course. In the interactive effect on PPO activity, only antagonism was observed between H. armigera herbivory and omethoate application. In the interactive stress of H. armigera herbivory and omethoate application on cotton defense responses, omethoate affected the defense responses of cotton to H. armigera herbivory by producing antagonistic and synergistic effects. 17Visualizing DC morphology and T cell motility to characterize DC-T cell encounters in mouse lymph nodes under mTOR inhibition显示文摘Mammalian target of rapamycin(mTOR),a serine/threonine kinase orchestrating cellular metabolism,is a crucial immune system regulator.However,it remains unclear how mTOR regulates dendritic cell(DC) function in vivo,especially DC-T cell encounters,a critical step for initiating adaptive immune responses.We dynamically visualized DC-T contacts in mouse lymph node using confocal microscopy and established an encounter model to characterize the effect of mTOR inhibition on DC-T cell encounters using DC morphology.Quantitative data showed mTOR inhibition via rapamycin altered DC shape,with an increased form factor(30.17%) and decreased cellular surface area(20.36%) and perimeter(22.43%),which were associated with Cdc42 protein downregulation(52.71%).Additionally,DCs adopted a similar morphological change with Cdc42 inhibition via ZCL278 as that observed with mTOR inhibition.These morphologically altered DCs displayed low encounter rates with T cells.Time-lapse imaging data of T cell motility supported the simulated result of the encounter model,where antigen-specific T cells appeared to reduce arrest in the lymph nodes of rapamycin-pretreated mice relative to the untreated group.Therefore,mTOR inhibition altered DC morphology in vivo and decreased the DC-T cell encounter rate,as well as Cdc42 inhibition.By establishing an encounter model,our study provides an intuitive approach for the early prediction of DC function through morphological quantification of form factor and area.Qiaoya Lin Zheng Liu Meijie Luo Hao Zheng Sha Qiao Chenlu Han Deqiang Deng Zhan Fan Yafang Lu Zhihong Zhang Qingming Luo 2019Science China(Life Sciences)2019,62,9:2
