1Increased expression of YAP1 in prostate cancer correlates with extraprostatic extension显示文摘Objective:Yes associated protein 1(YAP1)is a member of the Hippo pathway,acting as a transcriptional coactivator.To elucidate the role of YAP1 and phosphorylated(p)YAP1 in prostate cancer(PCa)tumorigenesis,we investigated their expression in clinical samples of PCa and cell lines.Methods:Fifty-four tumor,adjacent nontumor,and prostate intraepithelial neoplasia(PIN)tissues from patients with PCa after radical prostatectomy were selected from a retrospective cohort and studied using immunohistochemistry(IHC).Protein and m RNA expression levels of YAP1 were evaluated by Western blot analysis and quantitative real-time reverse transcription PCR,respectively,in cancer cell lines.Publicly available gene expression datasets were downloaded to analyze YAP1 m RNA and protein levels in PCa tissue samples.Results:IHC analysis of PCa tissues revealed that YAP1 staining intensities were moderate to weak in the nucleus and cytoplasm of tumor cells,whereas adjacent normal epithelia showed strong staining.We observed that benign prostates were characterized by higher expression levels of both nuclear(P=0.004)and cytosolic(P=0.005)YAP1.p YAP1 staining was weak in the cytoplasm and absent in the nucleus of all the tissues investigated.YAP1 expression was an indicator of extraprostatic extension(EPE).The level of YAP1 was negatively correlated with the level of the androgen receptor(AR)in The Cancer Genome Atlas dataset and Western blot analysis of cell lines.Conclusions:Our study suggested that YAP1 expression is heterogeneous in PCa tissue samples;therefore,YAP1 might play different roles in different aspects of PCa progression.This might involve AR–YAP1 interplay in PCa.Filiz Kisaayak Collak Ummuhan Demir Seyma Ozkanli Esra Kurum Pinar Engin Zerk 2017Cancer Biology & Medicine2017,14,4:8
2Comparison of one and three initial monthly intravitreal ranibizumab injection in patients with macular edema secondary to branch retinal vein occlusion显示文摘AIM: To compare three initial monthly intravitreal ranibizumab(IVR) injections followed by pro re nata(PRN) dosing with one initial monthly IVR injections followed by PRN dosing for macular edema(ME) secondary to branch retinal vein occlusion(BRVO).METHODS: Forty-two eyes of 42 patients who had IVR injections for BRVO were retrospectively studied. Eighteen eyes received 1 initial IVR injection(1+PRN group) and 24 eyes received 3 monthly IVR injections(3+PRN). At 1, 3, 6 and 12mo; spectral-domain optical coherence tomography(SD-OCT) was performed. Central macular thickness(CMT), the integrity of the external limiting membrane(ELM), the presence of subretinal fluid, cyst size, the presence of inner segment/outer segment(IS/OS) defect were determined.RESULTS: At baseline the mean CMT was 521.3±153.2 μm in the 3+PRN group while it was 438.1±162.4 μm in 1+PRN group. At the final visit, mean CMT was 278.3±87.8 μm in the 3+PRN group and 285.2±74.2 μm in the 1+PRN group(P=0.079). The changes in CMT over the entire study period were also comparable in both groups(243±160 μm in the 3+PRN group, and 152.9±175.3 μm in the 1+PRN group; P=0.090). At baseline, best-corrected visual acuity(BCVA) was 0.92±0.60 logarithm of the minimal angle of resolution(logMAR) in the 3+PRN group, while it was 0.72±0.46 logMAR in the 1+PRN group. Final BCVA was 0.42±0.55 logMAR in the 3+PRN group and 0.38±0.50 logMAR in the 1+PRN group(P=0.979). Additionally, the BCVA changes from baseline to final visit were not significantly different(-0.50±0.45 logMAR in the 3+PRN group, and-0.33±0.39 logMAR in the 1+PRN group; P=0.255).CONCLUSION: No significant differences in the anatomical or functional results are found between 3+PRN and 1+PRN regimens in the patients receiving ranibizumab for ME secondary to BRVO. 3Host genetic factors affecting hepatitis B infection outcomes:Insights from genome-wide association studies显示文摘The clinical outcome of hepatitis B virus(HBV) infection depends on the success or failure of the immune responses to HBV,and varies widely among individuals,ranging from asymptomatic self-limited infection,inactive carrier state,chronic hepatitis,cirrhosis,hepatocellular carcinoma,to liver failure,depending on the success or failure of immune response to HBV.Genome-wide association studies(GWAS) identified key genetic factors influencing the pathogenesis of HBV-related traits.In this review,we discuss GWAS for persistence of HBV infection,antibody response to hepatitis B vaccine,and HBV-related advanced liver diseases.HBV persistence is associated with multiple genes with diverse roles in immune mechanisms.The strongest associations are found within the classical human leukocyte antigen(HLA) genes,highlighting the central role of antigen presentation in the immune response to HBV.Associated variants affect both epitope binding specificities and expression levels of HLA molecules.Several other susceptibility genes regulate the magnitude of adaptive immune responses,determining immunity vs tolerance.HBV persistence and nonresponse to vaccine share the same risk variants,implying overlapping genetic bases.On the other hand,the risk variants for HBV-related advanced liver diseases are largely different,suggesting different host-virus dynamics in acute vs chronic HBV infections.The findings of these GWAS are likely to pave the way for developing more effective preventive and therapeutic interventions by personalizing the management of HBV infection.Izzet Mehmet Akcay Seyma Katrinli Kamil Ozdil Gizem Dinler Doganay Levent Doganay 2018World Journal of Gastroenterology2018,24,30:6
4Association of human leukocyte antigen DQB1 and DRB1 alleles with chronic hepatitis B显示文摘AIM: To investigate the effect of human leukocyte antigen(HLA) DRB1 and DQB1 alleles on the inactive and advanced stages of chronic hepatitis B.METHODS: Patient records at a single institution's hepatology clinic were reviewed. Demographic data, laboratory results, endoscopy results, virological parameters, biopsy scores and treatment statuses were recorded. In total, 355 patients were eligible for thestudy, of whom 226(63.7%) were male. Overall, 82(23.1%) were hepatitis B early antigen(HBeAg) positive, 87(24.5%) had cirrhosis, and 66(18.6%) had inactive disease. The presence of DQB1 and DRB1 alleles was determined by polymerase chain reaction with sequence-specific primers. The distribution of the genotyped alleles among patients with cirrhosis and patients with chronic active hepatitis was analyzed.RESULTS: The most frequent HLA DQB1 allele was DQB1*03:01(48.2%), and the most frequent HLA DRB1 allele was DRB1*13/14(51.8%). DQB1*05:01 was more frequent in patients with active disease than in inactive patients(27% vs 9.1%; P = 0.002, Pc = 0.026). DRB1*07 was rare in patients with cirrhosis compared with non-cirrhotics(3.4% vs 16%; P = 0.002, Pc = 0.022). Older age(P < 0.001) and male gender(P = 0.008) were the other factors that affected the presence of cirrhosis. In a multivariate logistic regression analysis, DRB1*07 remained a significant negative predictor of cirrhosis(P = 0.015). 11Successful treatment of tubulointerstitial nephritis in immunoglobulin G4-related disease with rituximab: A case report显示文摘BACKGROUND Immunoglobulin G4-related disease (IgG4-RD) is an immune-mediated condition that consisted of disorders that share particular clinical,serologic and pathologic properties.The common presentation of disease includes tumor-like swelling of involved organs and the histopathological findings are a lymphoplasmacytic infiltrate enriched with IgG4-positive plasma cells,and a variable degree of fibrosis that has a characteristic 'storiform' pattern in biopsy specimens of tumorlike masses.Major presentations of this disease,which often affects more than one organ,include autoimmune pancreatitis,salivary gland disease (sialadenitis),orbital disease and retroperitoneal fibrosis.The steroid treatment is essential for the treatment of the disease however,other immunosuppressive drugs including cyclophosphamide or rituximab could be an option in resistant cases.CASE SUMMARY Herein,we reported a 34-year-old woman whom previously had diagnosed with asthma,rheumatoid arthritis and Sj?gren’s syndrome (SS) referred our nephrology department due to acute kidney failure development at the last rheumatology visit.After kidney biopsy she has been diagnosed with IgG4-RD and tubuluointerstitial nephritis.She had been accepted resistant to steroid,mycophenolate mofetil,methotrexate and azathioprine therapies due to receiving in last two years.She refused to receive cyclophosphamide due to potential gonadotoxicity of the drug.Thus,rituximab therapy was considered.She received 1000 mg infusion,15 d apart and 6 mo later it has been administered same protocol.After one year from the last rituximab dose serum creatinine decreased from 4.4 mg/dL to 1.6 mg/dL,erythrocyte sedimentation rate decreased from 109 mm/h to 13 mm/h [reference range (RR) 0-20],and Creactive protein decreased from 55.6 mg/L to 5 mg/L (RR 0–6).All pathologic lymph nodes and masses were also disappeared.CONCLUSION Patients with IgG4-RD usually misdiagnosed with rheumatologic diseases including systemic lupus erythematous or SS and also they were screened for the presence of malignancy.Rituximab could be an important treatment option in cases with steroid resistant tubulointerstitial nephritis in IgG4-RD.Eray Eroglu Murat Hayri Sipahioglu Soner Senel Sule Ketenci Ertas Seyma Savas Figen Ozturk Ismail Kocyigit Bulent Tokgoz Oktay Oymak 2019World Journal of Clinical Cases2019,7,16:1

15A combo technology of autotrophic and heterotrophic denitrification processes for groundwater treatment显示文摘In this study,a sequential process(heterotrophic up-flow column and completely mixed membrane bioreactors)was proposed combining advantages of the both processes.The system was operated for 249 days with simulated and real groundwater for nitrate removal at concentrations varying from 25 to 145 mg·L^(-1) NO_(3)^(-)-N.The contribution of heterotrophic process to total nitrate removal in the system was controlled by dozing the ethanol considering the nitrate concentration.By this way,sulfur based autotrophic denitrification rate was decreased and the effluent sulfate concentrations were controlled.The alkalinity requirement in the autotrophic process was produced in the heterotrophic reactor,and the system was operated without alkalinity supplementation.Throughout the study,the chemical oxygen demand in the heterotrophic reactor effluent was(23.7±22)mg L^(-1) and it was further decreased to(7.5±7.2)mg·L^(-1) in the system effluent,corresponding to a 70%reduction.In the last period of the study,the real groundwater containing 145 mg·L^(-1) NO_(3)^(-)-N was completely removed.Membrane was operated without chemical washing in the first 114 days.Between days 115-249 weekly chemical washing was required.Cemile Seyma Arzum Yapıcı Dilan Toprak Müjgan Yıldız Sinan Uyanık Yakup Karaaslan Deniz Uçar 2021Chinese Journal of Chemical Engineering2021,34,9:0

17The in-vivo assessment of Turkish propolis and its nano form on testicular damage induced by cisplatin显示文摘Objective: Chemotherapeutic drugs, such as cisplatin(CP), which are associated with oxidative stress and apoptosis, may adversely affect the reproductive system. This study tests whether administration of propolis and nano-propolis(NP) can alleviate oxidative stress and apoptosis in rats with testicular damage induced by CP.Methods: In this study, polymeric nanoparticles including propolis were synthesized with a green sonication method and characterized using Fourier transform-infrared spectroscopy, Brunauer-EmmettTeller, and wet scanning transmission electron microscopy techniques. In total, 56 rats were divided into the following seven groups: control, CP, propolis, NP-10, CP + propolis, CP + NP-10, and CP + NP-30.Propolis(100 mg/kg), NP-10(10 mg/kg), and NP-30(30 mg/kg) treatments were administered by gavage daily for 21 d, and CP(3 mg/kg) was administered intraperitoneally in a single dose. After the experiment,oxidative stress parameters, namely, malondialdehyde(MDA), glutathione(GSH), glutathione peroxidase(GPx), and catalase(CAT), and apoptotic pathways including B cell leukemia/lymphoma-2 protein(Bcl-2)and Bcl-2-associated X protein(Bax) were measured in testicular tissues. Furthermore, sperm quality and weights of the testis, epididymis, right cauda epididymis, seminal vesicles and prostate were evaluated.Results: Propolis and NP(especially NP-30) were able to preserve oxidative balance(decreased MDA levels and increased GSH, CAT, and GPx activities) and activate apoptotic pathways(decreased Bax and increased Bcl-2) in the testes of CP-treated rats. Sperm motility in the control, CP, and CP + NP-30 groups were 60%, 48.75%, and 78%, respectively(P<0.001). 19Serum concentrations of insulin-like growth factor-binding protein 5 in Crohn's disease显示文摘AIM:To investigate serum insulin-like growth factorbinding protein 5(IGFBP-5)levels and intestinal IGFBP-5expression in patients with Crohn’s disease(CD).METHODS:We analyzed the serum concentrations and intestinal expression of IGFBP-5 in 42 patients with CD,of whom 26 had endoscopically or radiologically proven stricture formation.Nine of the 42 patients had active disease,with a Crohn’s disease activity index>150.Serum IGFBP-5 levels were analyzed in 20 healthy controls matched by sex and age to the CD patients.Serum IGFBP-5 was measured using an enzyme-linked immunosorbent assay.Intestinal tissue was obtained from patients through endoscopic biopsies.IGFBP-5expression was detected using immunohistochemistry and was scored semiquantitatively.RESULTS:The median serum IGFBP-5 concentrations of CD patients were significantly lower compared with healthy controls[median 7.2(IQR:5.5-11.3)ng/mL vs 11.3(8.0-44.6)ng/mL,P<0.001].There was no significant difference between median serum IGFBP-5levels in CD patients with or without stricture formation[6.9(5.5-11.3)ng/mL vs 7.8(5.3-10.1)ng/mL,P=0.815].The serum IGFBP-5 levels were not significantly different between patients with active disease and inactive disease[7.2(6.5-7.6)ng/mL vs 7.2(5.5-11.3)ng/mL,P=0.890].However,a significant correlation was observed between serum IGFBP-5 levels and platelet count(PLT)(r=0.319,P=0.0395).No significant correlation was found between tissue IGFBP-5 immunohistochemical staining intensity scores and serum IGFBP-5 levels.No significant difference was found when comparing the serum IGFBP-5 levels among the patients with different tissue IGFBP-5 staining scores(absent/very weak,weak,moderate or strong).There was a significant correlation between tissue IGFBP-5staining scores and white blood cell count(r=0.391,P=0.01)and PLT(r=0.356,P=0.021).CONCLUSION:Our results indicate that serum IGFBP-5 concentrations were lower in CD patients compared to healthy controls regardless of disease activity or the presence of stricture formation.Gupse Adali Elif Yorulmaz Seyma Ozkanli Celal Ulasoglu Baris Bayraktar Alev Orhun Yasar Colak Ilyas Tuncer 2013World Journal of Gastroenterology2013,19,47:0
20Increased serum soluble lectin-like oxidized low-density lipoprotein receptor-1 levels in patients with biopsy-proven nonalcoholic fatty liver disease显示文摘AIM: To analyze the relationship between the serum lectin-like oxidized low-density lipoprotein receptor-1(LOX-1) levels and clinical and histopathological features of biopsy-confirmed nonalcoholic fatty liver disease(NAFLD) patients.METHODS: Fifty-three consecutive,biopsy-proven NAFLD patients(31 males and 22 females,mean age 42.5 ± 9.6 years) and 26 age- and gender-matched,healthy controls(14 males and 12 females,mean age 39 ± 10.7 years) were included.The patientswith NAFLD were consecutive patients who had been admitted to the hepatology outpatient clinic within the last year and had been diagnosed with NAFLD as the result of liver biopsy.The healthy controls were individuals who attended the outpatient clinic for routine health control and had no known chronic illnesses.The histological evaluation was conducted according t o t he N AF LD ac t ivi ty scoring syst em recommended by The National Institute of Diabetes and Digestive and Kidney Diseases Nonalcoholic Steatohepatitis Clinical Research Network.The serum LOX-1 levels were measured using an ELISA kit(Life Science Inc.USCN.Wuhan,Catalog No.E1859Hu) in both patients and healthy controls.A receiver operating characteristic(ROC) curve analysis was used to identify the optimal cutoff value of LOX-1 and thereby distinguish between patients with nonalcoholic steatohepatitis(NASH) and healthy controls.A P-value < 0.05 was considered statistically significant.RESULTS: NAFLD and healthy control groups were similar in terms of age and sex.NAFLD patients consisted of 8 patients with simple steatosis(15%),27 with borderline NASH(51%) and 18 with definitive NASH(34%).Metabolic syndrome was found in 62.2% of the patients with NAFLD.The mean serum LOX-1 level in biopsy-proven NAFLD patients was 8.49 ± 6.43 ng/m L compared to 4.08 ± 4.32 ng/m L in healthy controls(P = 0.001).The LOX-1 levels were significantly different between controls,simple steatosis and NASH(borderline+definite) cases(4.08 ± 4.32 ng/m L,6.1 ± 6.16 ng/m L,8.92 ± 6.45 ng/m L,respectively,P = 0.004).When the cut-off value for the serum LOX-1 level was set at 5.35 ng/m L,and a ROC curve analysis was performed to distinguish between steatohepatitis patients and controls; the sensitivity and specificity of the serum LOX-1 level were 69.8% and 69.2%,respectively.CONCLUSION: The serum LOX-1 levels were significantly higher in NAFLD patients than in healthy controls.Additionally,the serum LOX-1 levels could differentiate between steatohepatitis patients and healthy controls.Oguzhan Ozturk Yasar Colak Ebubekir Senates Yusuf Yilmaz Celal Ulasoglu Levent Doganay Seyma Ozkanli Yasemin Musteri Oltulu Ender Coskunpinar Ilyas Tuncer 2015World Journal of Gastroenterology2015,21,26:0
