2LIM矿化蛋白-1抑制破骨前体细胞合成一氧化氮的分子机制研究显示文摘目的 探讨LIM矿化蛋白-1(LIM mineralization protein-1,LMp-1)抑制破骨前体细胞合成一氧化氮(nitric oxide,NO)的分子机制.方法 用不同浓度的TAT-LMP-1(0.05nmol/L,0.1 rimol/L,0.25 nmol/L)处理肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)或脂多糖(lipopolysaccharide,LPS)诱导的破骨前体细胞(RAW 264.7细胞),利用Greiss反应间接检测各组NO的生成量.用上述不同浓度的TAT-LMp-1处理LPS诱导的RAW 264.7细胞,使用逆转录-实时定量PCR和Western blot检测细胞中一氧化氮合酶(inducible NO synthase,iNOS)基因及蛋白的表达,利用Western blot检测细胞中NF-κB亚单位P65和磷酸化IκB含量,利用荧光素酶报告基因检测LMp-1对NF-κB通路转录活性的影响.结果 LMP-1可浓度依赖性抑制TNF-α或LPS诱导的RAW 264.7细胞合成NO.LMP-1可浓度依赖性抑制LPS诱导的RAW 264.7细胞中iNOS基因及蛋白的表达、NF-κB亚单位p65和磷酸化IκB含量升高以及NF-κB通路的转录活性.结论 LMp-1抑制LPS诱导的RAW 264.7细胞合成NO的可能分子机制为通过抑制IκB磷酸化使得p65不能与IκB解离,进而阻止p65核转移,导致NF-κB信号通路介导的iNOS生成障碍,最后引起NO生成减少.刘辉 Bargouti M 郑召民 张奎渤 Zughaier S 刘云山 Sangadala S Boden SD Titus L 2011中华骨科杂志2011,31,5:3
