| 1 | Blockade of high mobility group box-1 protein attenuates experimental severe acute pancreatitis显示文摘AIM: To examine the effects of anti-high mobility group box 1 (HMGB1) neutralizing antibody in experimental se-vere acute pancreatitis (SAP).METHODS: SAP was induced by creating closed duode-nal loop in C3H/HeN mice. SAP was induced immediately after intraperitoneal injection of anti-HMGB1 neutralizing antibody (200 μg). Severity of pancreatitis, organ injury (liver, kidney and lung), and bacterial translocation to pancreas was examined 12 h after induction of SAP.RESULTS: Anti-HMGB1 neutralizing antibody significant-ly improved the elevation of the serum amylase level and the histological alterations of pancreas and lung in SAP. Anti-HMGB1 antibody also significantly ameliorated the elevations of serum alanine aminotransferase and cre-atinine in SAP. | 4 | Factors associated with the overall survival of elderly patients with hepatocellular carcinoma显示文摘AIM:To identify the factors associated with overall survival of elderly patients with hepatocellular carcinoma(HCC).METHODS:A total of 286 patients with HCC(male/female:178/108,age:46-100 years),who were diagnosed and treated by appropriate therapeutic procedures between January 2000 and December 2010,were enrolled in this study.Patients were stratified into two groups on the basis of age:Elderly(≥ 75 years old) and non-elderly(< 75 years old).Baseline clinical characteristics as well as cumulative survival rates were then compared between the two groups.Univariate and multivariate analyses were used to identify the factors associated with prolonged overall survival of patients in each group.Cumulative survival rates in the two groups were calculated separately for each modified Japan Integrated Stage score(mJIS score) category by the Kaplan-Meier method.In addition,we compared the cumulative survival rates of elderly and non-elderly patients with good hepatic reserve capacity(≤ 2 points as per mJIS).RESULTS:In the elderly group,the proportion of female patients,patients with absence of hepatitis B or hepatitis C viral infection,and patients with coexisting extrahepatic comorbid illness was higher(56.8% vs 31.1%,P < 0.001;27.0% vs 16.0%,P = 0.038;33.8% vs 22.2%,P = 0.047;respectively) than that in the nonelderly group.In the non-elderly group,the proportion of hepatitis B virus(HBV)-infected patients was higher than that in the elderly group(9.4% vs 0%,P = 0.006).The cumulative survival rates in the elderly group were 53.7% at 3 years and 32.9% at 5 years,which were equivalent to those in the non-elderly group(55.9% and 39.4%,respectively),as shown by a log-rank test(P = 0.601).In multivariate analysis,prolonged survival was significantly associated with the extent of liver damage and stage(P < 0.001 and P < 0.001,respectively),but was not associated with patient age.However,on individual evaluation of factors in both groups,stage was significantly(P < 0.001) associated with prolonged survival.Regarding mJIS scores of ≤ 2,the rate of female patients with this score was higher in the elderly group when compared to that in the non-elderly group(P = 0.012) and patients ≥ 80 years of age tended to demonstrate shortened survival.CONCLUSION:Survival of elderly HCC patients was associated with liver damage and stage,but not age,except for patients ≥ 80 years with mJIS score ≤ 2. | Hideki Fujii Yoshito Itoh Naoki Ohnishi Masafumi Sakamoto Tohru Ohkawara Yoshihiko Sawa Koichi Nishida Yasuo Ohkawara Kanji Yamaguchi Masahito Minami Takeshi Okanoue | 2012 | World Journal of Gastroenterology2012,18,16: | 4 |
| 5 | Identification of Naturally Occurring Polyamines as Root-Knot Nematode Attractants显示文摘Root-knot nematodes(RKNs;genus Meloidogyne)are a class of plant parasites that infect the roots of many plant species.It is believed that RKNs target certain signaling molecules derived from plants to locate their hosts;however,currently,no plant compound has been unambiguously identified as a universal RKN attractant.To address this question,we screened a chemical library of synthetic compounds for Meloidogyne incognita attractants.The breakdown product of aminopropylam ino-anthraquinone,1,3-diam inopropane,as well as its related compounds,putrescine and cadaverine,were found to attract M.incognita.After exam ining various polyamines,M.incognita were found to be attracted specifically by natural com pounds that possess three to five methylene groups between two terminal amino groups.Using cryo-TO F-SIM S/SEM,cadaverine was indeed detected in soybean root cortex cells and the surrounding rhizosphere,establishing a chemical gradient.In addition to cadaverine,putrescine and 1,3-diam inopropane were also detected in root exudate by HPLC-MS/MS.Furtherm ore,exogenously applied cadaverine is sufficient to enhance M.incognita infection of Arabidopsis seedlings.These results suggest that M.incognita is likely attracted by polyam ines to locate the appropriate host plants,and the naturally occurring polyamines have potential applications in agriculture in developing protection strategies for crops from RKN infection. | Morihiro Oota Allen Yi-Lun Tsai Dan Aoki Yasuyuki Matsushita Syuuto Toyoda Kazuhiko Fukushima Kentaro Saeki Kei Toda Laetitia Perfus-Barbeoch Bruno Favery Hayato Ishikawa Shinichiro Sawa | 2020 | Molecular Plant2020,13,4: | 4 |
| 6 | Regulation of Root-Knot Nematode Behavior by Seed-Coat Mucilage-Derived Attractants显示文摘Seed exudates influence the behavior of soil organisms,but howthis occurs remains unclear,particularly for multicellular animals.Here we show that compounds associated with Arabidopsis seed-coat mucilage regulate the behavior of soil-borne animals,specifically root-knot nematodes (RKNs).Infective RKN J2 larvae actively travel toward Arabidopsis seeds through chemotaxis.Analysis of Arabidopsis mucilage mutants demonstrated that the attraction of RKNs toArabidopsis seeds requires the synthesis and extrusion of.seed-coat mucilage.Extracted mucilage alone is not sufficient to attract RKNs,but seed-surface carbohydrates and proteins are required for this process.These findings suggest that the RKN chemoattractant is synthesized de novo upon mucilage extrusion but may be highly unstable.RKNs attracted by thismucilage-dependent mechanism can infect the emerging seedling.However,the attraction signal from seedling roots likely acts independently of the seed-coat signal and may mask the attraction to seed-coat mucilage after germination.Multiple RKN species are attracted byArabidopsis seeds,suggesting that this mechanism is conserved in RKNs.These findings indicate that seed exudate can regulate the behavior of multicellular animals and highlight the potential roles of seed-coat mucilage in biotic interactions with soil microorganisms. | Allen Yi-Lun Tsai Takumi Higaki Chinh-Nghia Nguyen Laetitia Perfus-Barbeoch Bruno Favery Shinichiro Sawa | 2019 | Molecular Plant2019,12,1: | 3 |

| 10 | Case of autoimmune hepatitis with markedly enlarged hepatoduodenal ligament lymph nodes显示文摘Autoimmune hepatitis(AIH) is a necroinflammatory liver disease of unknown etiology.The disease is characterized histologically by interface hepatitis,biochemically by increased aspartate aminotransferase and alanine aminotransferase levels,and serologically by increased autoantibodies and immunoglobulin G levels.Here we discuss AIH in a previously healthy 37-year-old male with highly elevated serum levels of soluble interleukin-2 receptor and markedly enlarged hepatoduodenal ligament lymph nodes(HLLNs,diameter,50 mm).Based on these observations,the differential diagnoses were AIH,lymphoma,or Castleman's disease.Liver biopsy revealed the features of interface hepatitis without bridging fibrosis along with plasma cell infiltration which is the typical characteristics of acute AIH.Lymph node biopsy revealed lymphoid follicles with inflammatory lymphocytic infiltration;immunohistochemical examination excluded the presence of lymphoma cells.Thereafter,he was administered corticosteroid therapy:after 2 mo,the enlarged liver reached an almost normal size and the enlarged HLLNs reduced in size.We could not find AIH cases with such enlarged lymph nodes(diameter,50 mm) in our literature review.Hence,we speculate that markedly enlarged lymph nodes observed in our patient may be caused by a highly activated,humoral immune response in AIH. | Hideki Fujii Naoki Ohnishi Kazuho Shimura Masafumi Sakamoto Tohru Ohkawara Yoshihiko Sawa Koichi Nishida Yasuo Ohkawara Tatsuro Kobata Kanji Yamaguchi Yoshito Itoh | 2013 | World Journal of Gastroenterology2013,19,11: | 3 |
| 11 | Running-in friction of hip joint replacements can be significantly reduced:The effect of surface-textured acetabular cup显示文摘Hip joint replacements represent the most effective way of treatment for patients suffering from joint diseases.Despite the rapid improvement of implant materials over the last few decades,limited longevity associated with wear-related complications persists as the main drawback.Therefore,improved tribological perfor-mance is required in order to extend the service life of replacements.The effect of surface texturing of ultra-high molecular weight polyethylene(UHMWPE)acetabular cup was investigated in the present study.Unique tilling method was utilized for manufacturing the dimples with controlled diameter and depths on the contact surface of the cup.The experiments with four commercial femoral components and two model lubricants were realized.The main attention was paid to a coefficient of friction considering the differences between the original and the dimpled cups.Results showed remarkable lowering of friction,in general.Focusing on the simulated human synovial fluid,friction was reduced by 40%(alumina ceramic),38.8%(zirconia toughened ceramic),25.5%(metal),and 9.9%(oxinium).In addition,the dimples helped to keep the friction stable without fluctuations.To conclude,the paper brings a new insight into frictional behaviour of the hip replacements during running-in phase which is essential for overall implant lifespan.It is believed that proper surface texturing may rapidly improve the life quality of millions of patients and may lead to considerable financial savings. | David NECAS Hatsuhiko USAMI Tatsuya NIIMI Yoshinori SAWAE Ivan KRUPKA Martin HARTL | 2020 | Friction2020,8,6: | 2 |
| 12 | End-stage renal disease is associated with increased post endoscopic retrograde cholangiopancreatography adverse events in hospitalized patients显示文摘AIM To determine if end-stage renal disease (ESRD) is a risk factor for post endoscopic retrograde cholangio-pancreatography (ERCP) adverse events (AEs). METHODS We performed a retrospective cohort study using the Nationwide Inpatient Sample (NIS) 2011-2013. We identified adult patients who underwent ERCP using the International Classification of Diseases 9^(th) Revision (ICD-9-CM). Included patients were divided into three groups: ESRD, chronic kidney disease (CKD), and control. The primary outcome was post-ERCP AEs including pancreatitis, bleeding, and perforation determined based on specific ICD-9-CM codes. Secondary outcomes were length of hospital stay, in-hospital mortality, and admission cost. AEs and mortality were compared using multivariate logistic regression analysis.RESULTS There were 492175 discharges that underwent ERCP during the 3 years. The ESRD and CKD groups contained 7347 and 39403 hospitalizations respectively, whereas the control group had 445424 hospitalizations. Post-ERCP pancreatitis (PEP) was significantly higher in the ESRD group (8.3%) compared to the control group (4.6%) with adjusted odd ratio (aOR) = 1.7 (95% CI: 1.4-2.1, ~aP < 0.001). ESRD was associated with significantly higher ERCP-related bleeding (5.1%) compared to the control group 1.5% (aOR = 1.86, 95%CI: 1.4-2.4, ~aP < 0.001). ESRD had increased hospital mortality 7.1% vs 1.15% in the control OR = 6.6 (95%CI: 5.3-8.2, ~aP < 0.001), longer hospital stay with adjusted mean difference (aMD) = 5.9 d (95% CI: 5.0-6.7 d, ~aP < 0.001) and higher hospitalization charges aMD = $+82064 (95%CI: $68221-$95906, ~aP < 0.001). CONCLUSION ESRD is a risk factor for post-ERCP AEs and is associated with higher hospital mortality.
