| 1 | HPV16 E5蛋白免疫优势肽段分析及其多克隆抗体的制备显示文摘目的预测人乳头瘤病毒16型(human papillomavirus 16,HPV16)E5蛋白的免疫优势肽段,并制备其多克隆抗体。方法以人HPV16 E5蛋白全长氨基酸序列为基础,采用Hopp&Woods的亲水性方案、Zimmerman极性参数、Jameson-Wolf抗原指数方案和Emini表面可及性方案及抗原性指数(antigenic index,AI)等生物信息学方法,综合分析E5蛋白免疫优势肽段。通过免疫日本大耳白兔进行E5蛋白免疫优势肽段的免疫原性验证并制备多克隆抗体;进一步采用ELISA法和间接免疫荧光试验分别检测多克隆抗体的效价及特异性。结果HPV16 E5蛋白N-端1~17氨基酸序列(MTNLDTASTTLLACFLL)为免疫优势肽段,可诱导兔产生高水平的E5蛋白特异性IgG抗体,抗体滴度随免疫次数增加而升高;间接细胞免疫荧光试验结果显示,制备的多克隆抗体可特异性识别TC-1-E5细胞胞浆内表达的E5蛋白。结论HPV16 E5蛋白氨基酸肽段(MTNLDTASTTLLACFLL)具有良好的免疫原性;成功制备了HPV16 E5蛋白多克隆抗体,为进一步研究HPV16 E5蛋白的生物学特性奠定了基础。 | 唐婉林 王路得 顾美萍 Kamara SAIDU 汪琪 李明洋 陈韶 张丽芳 | 2021 | 中国生物制品学杂志2021,34,6: | 4 |
| 2 | 新型冠状病毒E蛋白结构与免疫优势表位的信息学预测分析显示文摘目的:应用生物信息学方法预测和分析新型冠状病毒(SARS-CoV-2)包膜蛋白(E蛋白)的结构及免疫优势表位。方法:从NCBI GenBank数据库中检索SARS-CoV-2 E蛋白的氨基酸序列,通过Expas y服务器上的软件分析预测其理化性质、二级结构、跨膜区域;并结合其亲水性、表面可及性、极性、抗原性和柔韧性等综合分析,预测其可能的B细胞表位;利用Net CTL1.2 server预测CTL表位;通过Vector NTI对冠状病毒属E蛋白进行同源性分析;同时应用Swiss Model对E蛋白进行三维结构同源建模及功能结构域预测分析。结果:SARS-CoV-2 E蛋白由75个氨基酸残基组成,为跨膜蛋白,跨膜区域以α螺旋为主;E蛋白可能含有2个B细胞表位和6个CTL表位,其免疫优势表位区域分别位于其N端16-34aa和C端50-70aa区段;SARS-CoV-2与SARS-CoV和Bat-SARS-like-CoV E蛋白的免疫优势表位存在极高的同源性;其疏水性跨膜结构和PDZ结合基序分别位于N端12-34aa和C端72-75aa区段。结论:E蛋白为跨膜蛋白,其氨基酸序列的16-34和50-70区段可能为免疫优势表位。 | 李明洋 SAIDU Kamara 汪琪 郭艳茹 李青峰 朱珊丽 陈俊 蒋朋飞 张丽芳 | 2021 | 温州医科大学学报2021,51,3: | 3 |

| 10 | 小鼠PD-1及其配体PD-L1胞外区基因的原核表达及亲和性显示文摘目的:制备小鼠PD-1胞外区(mPD-1)与其配体PD-L1胞外区(mPD-L1)原核表达蛋白,并制备mPDL1兔多克隆抗体,在体外对mPD-L1和mPD-1的结合亲和性进行验证。方法:通过分子克隆方法分别构建重组质粒pET21a/mPD-L1和p GEX4T-1/mPD-1,并通过原核表达系统制备相应的重组蛋白并纯化,通过Western blot进行鉴定;将纯化的mPD-L1蛋白免疫健康日本大耳白兔制备兔多克隆血清抗体,用ELISA、Western blot分析得到的多克隆抗体的效价和特异性,并且将该多克隆抗体作为免疫荧光的阳性对照。最后应用ELISA和免疫荧光验证mPD-1与其配体mPD-L1蛋白的亲和性。结果:SDS-PAGE和Western blot结果显示原核表达的重组蛋白与预期大小一致,mPD-L1和mPD-1分别为25 kDa和40 kDa。经mPD-L1蛋白免疫后的日本大耳白兔能够产生特异性抗体,抗体效价在免疫后第6周达到高峰,Western blot检测结果显示多克隆血清可特异性识别mPD-1。ELISA结果表明,原核表达的mPD-L1蛋白能够和mPD-1蛋白结合,实验组OD450值明显高于对照组。免疫荧光结果显示,在小鼠PD-L1阳性细胞B16(小鼠黑色素瘤细胞株)的胞膜区出现绿色荧光团块。结论:利用原核细胞表达并纯化后获得mPD-L1和mPD-1蛋白,并在体外成功验证了mPD-L1和mPD-1之间的结合特性,为后期利用mPD-1和PD-L1的生物学特性研究提供了基础。 | 吕开绩 陈旭东 程开 王路得 朱进顺 Kamara Saidu 张丽芳 朱冠保 | 2018 | 温州医科大学学报2018,48,6: | 1 |

| 13 | Protein quality of four indigenous edible insect species in Nigeria显示文摘Food security is a serious concern particularly for developing countries.To overcome hunger and malnutrition there is a need for increased research towards finding alternative and cheaper sources of nutrients.Insects have been reported to be rich in protein and could be alternative sources of protein.This work was therefore designed to determine the protein quality of moth caterpillar,termite,cricket and grasshopper and the effects of diets supplemented with these insects on some biochemical and haematological indices of rats.The amino acid compositions of the insects were determined using standard analytical methods.Five iso-nitrogenous and isocaloric diets were formulated on a 10%protein basis with the insects and casein.A Nitrogen free diet was also formulated as control.Thirty six(36)young weanling albino rats(21–28 days old)were divided into 6 groups and fed with the diets ad libitum for 28 days.Their weekly weight gain and daily feed intake were recorded,urine and faeces were collected for nitrogen determination using Kjedahl method and the data obtained used to calculate the various protein quality indices.After the feeding trial periods,the rats were sacrificed,blood samples collected and organs excised for various analyses.Cricket was found to have the highest amino acid score(0.91),protein efficiency ratio(1.78),net protein ratio(3.04)biological value(93.02%)and protein digestibility corrected for amino acid score(0.73)as compared to other insect proteins analysed.The organ body weight ratios of the liver,spleen,lung and heart of rats placed on the insect supplemented diets were not significantly different(p>0.05)from those fed with casein and basal diets.Serum total protein concentrations in rats fed with cricket diet were not significantly different(p>0.05)from those fed casein diet while serum LDL cholesterol concentration was lowest in rats fed cricket diet.The finding in this work that the selected insects are nutritious and safe for consumption may alleviate the fear of entomophagy thereby reducing the overdependence on conventional animal proteins. | Oibiokpa Florence Inje Akanya Helmina Olufunmilayo Jigam Ali Audu Saidu Abubakar Ndaman Egwim Evans Chidi | 2018 | Food Science and Human Wellness2018,7,2: | 1 |

| 15 | The Establishment and Application of Mobile Electronic Surveillance System for Infectious Diseases with the Help of China—Sierra Leone,2016-Present显示文摘Introduction:Infectious disease surveillance has long been a challenge for low-income countries like Sierra Leone.Traditional approaches based on paper and Short Message Service(SMS)were subject to severe delays in obtaining,transmitting,and analyzing information.Methods:During the China aid operation for fighting Ebola since the end of 2014,a mobile electronic surveillance system for infectious diseases(MESSID)was developed in collaboration with the Republic of Sierra Leone Armed Forces(RSLAF),which comprised an Android-based reporting system and a complementary web-based program designed by Active Server Page.NET(ASP.NET)with the main functions including surveillance,real-time reporting,and risk assessment of infectious diseases.Results:MESSID was successfully registered in June 2016 and had been used by all medical and health institutions in RSLAF.From June 1,2016 to July 5,2021,34,419 cases were diagnosed with 47 infectious diseases of 5 categories,with a total of 42 clinical symptoms.Compared to traditional approaches based on paper and SMS,the MESSID showed flexibility,high efficiency,convenience,and acceptability.Discussion:MESSID is an accessible tool for surveillance of infectious diseases in Sierra Leone and possibly in other African countries with similar needs,capable of improving timeliness of disease reporting,thus rendering a timely outbreak detection and response. | Guangyu Zhao Haorong Chen Yanfeng Yan Jiafu Jiang Lei Lin Baogui Jiang Foday Sahr Stephen Sevalie Qiang Xu Jinjin Chen Henry Saidu Bangura Kandeh Bassie Kargbo Yajun Song Wei Liu Liqun Fang Yansong Sun | 2021 | China CDC weekly2021,3,36: | 1 |
