| 1 | Enhancement of acellular cartilage matrix scaffold by Wharton’s jelly mesenchymal stem cell-derived exosomes to promote osteochondral regeneration显示文摘Articular cartilage defect repair is a problem that has long plagued clinicians.Although mesenchymal stem cells(MSCs)have the potential to regenerate articular cartilage,they also have many limitations.Recent studies have found that MSC-derived exosomes(MSC-Exos)play an important role in tissue regeneration.The purpose of this study was to verify whether MSC-Exos can enhance the reparative effect of the acellular cartilage extracellular matrix(ACECM)scaffold and to explore the underlying mechanism.The results of in vitro experiments show that human umbilical cord Wharton’s jelly MSC-Exos(hWJMSC-Exos)can promote the migration and proliferation of bone marrow-derived MSCs(BMSCs)and the proliferation of chondrocytes.We also found that hWJMSC-Exos can promote the polarization of macrophages toward the M2 phenotype.The results of a rabbit knee osteochondral defect repair model confirmed that hWJMSC-Exos can enhance the effect of the ACECM scaffold and promote osteochondral regeneration.We demonstrated that hWJMSC-Exos can regulate the microenvironment of the articular cavity using a rat knee joint osteochondral defect model.This effect was mainly manifested in promoting the polarization of macrophages toward the M2 phenotype and inhibiting the inflammatory response,which may be a promoting factor for osteochondral regeneration.In addition,microRNA(miRNA)sequencing confirmed that hWJMSC-Exos contain many miRNAs that can promote the regeneration of hyaline cartilage.We further clarified the role of hWJMSC-Exos in osteochondral regeneration through target gene prediction and pathway enrichment analysis.In summary,this study confirms that hWJMSC-Exos can enhance the effect of the ACECM scaffold and promote osteochondral regeneration. | Shuangpeng Jiang Guangzhao Tian Zhen Yang Xiang Gao Fuxin Wang Juntan Li Zhuang Tian Bo Huang Fu Wei Xinyu Sang Liuqi Shao Jian Zhou Zhenyong Wang Shuyun Liu Xiang Sui Quanyi Guo Weimin Guo Xu Li | 2021 | Bioactive Materials2021,6,9: | 11 |
| 2 | Progress of regional oceanography study associated with western boundary current in the South China Sea显示文摘Recent progress of physical oceanography in the South China Sea (SCS) associated with the western boundary current (WBC) and eddies is reviewed in this paper. It includes Argo observations of the WBC, eddy detection in the WBC based on satellite images, cross-continental shelf exchange in the WBC, eddy-current interaction, interannual variability of the WBC, air-sea interaction, the SCS throughflow (SCSTF), among others. The WBC in the SCS is strong, and its structure, variability and dynamic processes on seasonal and interannual time scales are yet to be fully understood. In this paper, we summarize progresses on the variability of the WBC, eddy-current interaction, air-sea interaction, and the SCSTF achieved in the past few years. Firstly, using the drifting buoy observations, we point out that the WBC becomes stronger and narrower after it reaches the central Vietnam coast. The possible mechanisms influencing the ocean circulation in the northern SCS are discussed, and the dynamic mechanisms that induce the countercurrent in the region of northern branch of WBC in winter are also studied quantitatively using momentum balance. The geostropic component of the WBC was diagnosed using the ship observation along 18°N, and we found that the WBC changed significantly on interannual time scale. Secondly, using the ship observations, two anti-cyclonic eddies in the winter of 2003/2004 in the northern SCS, and three anti-cyclonic eddies in the summer of 2007 along 18°N were studied. The results show that the two anti-cyclonic eddies can propagate southwestward along the continental shelf at the speed of first Rossby wave (~0.1 ms-1 ) in winter, and the interaction between the three anti-cyclonic eddies in summer and the WBC in the SCS is preliminarily revealed. Eddies on the continental shelf of northern SCS propagated southeastward with a maximum speed of 0.09 ms-1 , and those to the east of Vietnam coast had the largest kinetic energy, both of which imply strong interaction between eddy activity and WBC in the SCS. Thirdly, strong intraseasonal variability (ISV) of sea surface temperature (SST) near the WBC regions was found, and the ISV signal of SST in winter weakens the ISV signal of latent heat flux by 20%. | 7 | A comparison of the spontaneous firing patterns between principal cells and fast spiking interneurons from dentate gyrus in waking guinea pigs显示文摘Objective To explore the possible mechanisms that cause the dentate gyrus (DG) neurons to play different roles in information coding. Methods In vivo extracellular single unit recording was performed on 22 waking female guinea pigs, which were positioned in a sound-attenuated recording chamber without any muscular relaxants. The spontaneous firing patterns of the DG neurons were detected and compared. Results There were two different electrophysiological populations in the DG of guinea pigs, principal cells (PCs) and fast spiking interneurons (INs). Of the PCs, 1.3% discharged regularly, 48.1% irregularly and 50.6% in bursts; in contrast, of the INs units, 64.1% discharged regularly, 2.6% irregularly and 33.3% in bursts. The spontaneous firing patterns of PCs were significantly different from those of INs (P<0.01). In addition, the differences of several interspike interval (ISI) parameters also have been observed: (1) the ISI coefficients of variation of PCs (3.39±3.56) were significantly higher than those of INs (1.08±0.46) (P<0.01); (2) the ISI asymmetric indexes of PCs (0.047±0.059) were significantly lower than those of INs (0.569±0.238) (P<0.01). Conclusion In the DG, the spontaneous firing patterns of PCs were significantly different from those of INs. The former were prone to fire in bursts, the latter were prone to fire regularly. | 8 | Immunogenicity and Efficacy of Liposome-encapsulated Influenza Split Vaccine in BALB/c Mice显示文摘The cellular immunity of current influenza split vaccine is relatively low. It is necessary to develop a novel vaccine to improve the cellular immunity. Thes of this study prepared liposome-encapsulated influenza split vaccine and tested it in BALB/c mice. The mice were immunized once with 4 μg of haemagglutinin of monovalent A/New Caledonia/20/99(H1N1) encapsulated with liposomes or the split virus vaccine only through intrastomach injection. | 11 | Hierarchical macro-microporous WPU-ECM scaffolds combined with Microfracture Promote in Situ Articular Cartilage Regeneration in Rabbits显示文摘Tissue engineering provides a promising avenue for treating cartilage defects.However,great challenges remain in the development of structurally and functionally optimized scaffolds for cartilage repair and regeneration.In this study,decellularized cartilage extracellular matrix(ECM)and waterborne polyurethane(WPU)were employed to construct WPU and WPU-ECM scaffolds by water-based 3D printing using low-temperature deposition manufacturing(LDM)system,which combines rapid deposition manufacturing with phase separation techniques.The scaffolds successfully achieved hierarchical macro-microporous structures.After adding ECM,WPU scaffolds were markedly optimized in terms of porosity,hydrophilia and bioactive components.Moreover,the optimized WPU-ECM scaffolds were found to be more suitable for cell distribution,adhesion,and proliferation than the WPU scaffolds.Most importantly,the WPU-ECM scaffold could facilitate the production of glycosaminoglycan(GAG)and collagen and the upregulation of cartilage-specific genes.These results indicated that the WPU-ECM scaffold with hierarchical macro-microporous structures could recreate a favorable microenvironment for cell adhesion,proliferation,differentiation,and ECM production.In vivo studies further revealed that the hierarchical macro-microporous WPU-ECM scaffold combined with the microfracture procedure successfully regenerated hyaline cartilage in a rabbit model.Six months after implantation,the repaired cartilage showed a similar histological structure and mechanical performance to that of normal cartilage.In conclusion,the hierarchical macro-microporous WPU-ECM scaffold may be a promising candidate for cartilage tissue engineering applications in the future. | Mingxue Chen YangYang Li Shuyun Liu Zhaoxuan Feng Hao Wang Dejin Yang Weimin Guo Zhiguo Yuan Shuang Gao Yu Zhang Kangkang Zha Bo Huang Fu Wei Xinyu Sang Qinyu Tian Xuan Yang Xiang sui Yixin Zhou Yufeng Zheng Quanyi Guo | 2021 | Bioactive Materials2021,6,7: | 1 |

| 16 | Guanidinoamidized Linear Polyethyleneimine for Gene Delivery显示文摘Guanidine was introduced to low molecular weight linear polyethyleneimine(LPEI) via amide groups, to explore the effect of both guanidine degree and pendant chain length on its transfection behavior. The resulting guanidinoamidized LPEIs(GLPEIs) could dramatically reduce LPEI's toxicity, enhance its DNA-packaging capability, cellular uptake and therefore transfection efficiency. These polyplexes were taken up very efficiently via caveolae-mediated endocytosis and their transfection efficiencies in ovarian cancer cells were significantly improved compared to native LPEI10 k polyplexes. Among these GLPEIs, LPEI-C3-G100 showed higher DNA affinity even than LPEI25 k and the highest transfection efficiency, probably due to the optimization of polymer chain flexibility. | 19 | Interaction of {11■2} twin variants in hexagonal close-packed titanium显示文摘As multiple{11■2}twin variants are often formed during deformation in hexagonal close-packed (hcp)titanium, the twin-twin interaction structure has a profound influence on mechanical properties. In this paper, the twin-twin interaction structures of the{11■2}contraction twin in cold-rolled commercial purity titanium were studied by using electron backscatter diffraction (EBSD) and transmission electron microscopy (TEM). Formation of the{11■2}twin variants was found to deviate the rank of Schmid factor,and the non-Schmid behavior was explained by the high-angle grain boundary nucleation mechanism.All the observed twin-twin pairs manifested a quilted-looking structure, which consists of the incoming twins being arrested by the obstacle twins. Furthermore, the quilted-looking{11■2}twin-twin boundary was revealed by TEM and high resolution TEM observations. De-twinning, lattice rotation and curved twin boundary were observed in the obstacle twin due to the twin-twin reaction with the impinging twin. | 20 | Synoptic mode of Antarctic summer sea ice superimposed on interannual and decadal variability显示文摘In contrast to decreased Arctic sea ice extent,Antarctic sea ice extent shows a somewhat increased trend.There is a large interannual variability of Antarctic sea ice,especially in the Pacific sector of the Southern Ocean.The change and variability of Antarctic sea ice in synoptic timescales in the recent decades remain unclear.We identify synoptic modes of variability of Antarctic summer sea ice by applying the Self Organizing Map(SOM)technique to daily sea ice concentration data for the period 1979–2018.Nearly 40%of the variability is characterized by opposite changes between sea ice cover in the Bellingshausen,Amundsen and western Ross Seas and in the rest of the Antarctic seas,and another 30%by meridional asymmetry in the Weddell,Amundsen,and Ross Seas.Most of these spatial patterns may be explained by the dynamics and thermodynamic processes associated with anomalous atmospheric circulations related to the Southern Annular Mode(SAM)with a structure of strong zonal asymmetry.The interannual variability of the sea ice modes appears to have little connection to SAM,and only a weak relation to ENSO.The annual frequencies of SOM node occurrences also show a great decadal variability.Node 9 appears mainly prior to 1990;while node 1 occurs mainly after 1990.The decadal variability of nodes 1 and 9 is associated with the asymmetrical SAM,which results from two wavetrains excited over northern Australia and the southeastern Indian Ocean.These results further highlight the importance of understanding the role of southern mid-to-high latitude atmospheric intrinsic variability in predicting Antarctic summer sea ice variations from synoptic to decadal timescales. | YU Le-Jiang ZHONG Shi-Yuan SUI Cui-Juan ZHANG Zhao-Ru SUN Bo | 2021 | Advances in Climate Change Research2021,12,2: | 1 |
