2Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 3Improvement of Foaming and Emulsifying Properties of Gluten by Conjugation with Fructose through Maillard Reaction显示文摘Gluten has poor emulsifying and foaming ability due to its amino acid composition. In this study, Maillard reaction was used to improve the emulsifying and foaming properties of gluten. The processing conditions for the preparation of gluten-fructose conjugates were optimized by using Box-Behnken model to achieve optimum foaming and emulsifying activity, respectively.The results showed that glycated gluten exhibited enhanced emulsifying activity compared to native control. The processing conditions for the preparation of gluten-fructose conjugates with optimum emulsifying activity were as follows: the temperature was 48℃, reaction time was 72 h, and maltose/gluten(W/W) ratio was 125%. Under such condition, the average emulsifying activity was 66.54%, being improved by about 2.5 times compared with that of native control. The Foaming properties of gluten also increased significantly by glycation modification. The optimum conditions of response were as below: the temperature was 48℃, reaction time was 66 h, and maltose/gluten ratio(W/W) was 110%. 4Aquaporin 5 is degraded by autophagy in diabetic submandibular gland显示文摘Autophagy is a catabolic process which is involved in the development of many diseases including diabetes mellitus and its complications. Hyposalivation is a common complication of diabetes mellitus, whereas its mechanism remains unclear. Here, we observed that the stimulated salivary flow rate of SMG was significantly decreased in db/db mice, a diabetic mice model. The expressions of aquaporin 5(AQP5), a water channel protein, were decreased, whereas the m RNA level of AQP5 was increased in SMGs of both diabetic patients and mice. Under transmission electron microcope, more autophagosomes were detected in diabetic SMGs. Expressions of autophagy related proteins LC3 II, Beclin-1 and ATG5 were increased, meanwhile autophagy substrate p62 was decreased in SMGs of diabetic patients and mice, indicating that autophagy was activated in diabetic SMG.Double immunofluorescence staining showed that the colocalization of AQP5 and LC3 was increased in SMGs of diabetic mice.In cultured SMG-C6 cells, high glucose(HG), but not high osmotic pressure, reduced AQP5 protein expression and induced autophagy. Moreover, inhibition of autophagy by 3-methyladenin, an autophagy inhibitor, or by autophagy-related gene 5 siRNA, decreased HG-induced AQP5 reduction in SMG-C6 cells. Additionally, the expression of p-p85, p-Akt and p-mTOR were decreased in HG-treated SMG-C6 cells. Pretreatment with 740 Y-P, a PI3 K agonist, significantly suppressed HG-induced autophagy and AQP5 degradation. Taken together, these results indicate that autophagy plays a crucial role in AQP5 degradation in diabetic SMG via PI3 K/Akt/mTOR signaling pathway, which contributes to the dysfunction of diabetic SMG. 5Pre-moxibustion and moxibustion prevent Alzheimer's disease显示文摘The Alzheimer's disease model in Wistar rats was established by injection of amyloid β-peptide(Aβ1–42) into the hippocampal CA1 region. Rats were treated with suspended moxibustion on Baihui(GV20) and Shenshu(BL23) acupoints. Prior to and post Aβ1–42exposure. Results showed no evidence of apoptosis in hippocampal neurons, a significantly reduced apoptosis rate of neurons and improved learning and memory abilities were observed in the Alzheimer's disease model. In particular, moxibustion prior to Aβ1–42exposure was more effective than moxibustion after Aβ1–42exposure in protecting the neuronal structure and lowering the apoptosis rate. 6GmCRY1s modulate gibberellin metabolism to regulate soybean shade avoidance in response to reduced blue light显示文摘Soybean is an important legume crop that displays the classic shade avoidance syndrome(SAS),including exaggerated stem elongation,which leads to lodging and yield reduction under density farming conditions.Here,we compared the effects of two shade signals,low red light to far-red light ratio(R:FR)and low blue light(LBL),on soybean status and revealed that LBL predominantly induces excessive stem elongation.We used CRISPR-Cas9-engineered Gmcry mutants to investigate the functions of seven cryptochromes(GmCRYs)in soybean and found that the four GmCRY1s overlap in mediating LBL-induced SAS.Lightactivated GmCRY1s increase the abundance of the bZlP transcription factors STF1 and STF2,which directly upregulate the expression of genes encoding GA2 oxidases to deactivate GA1 and repress stem elongation.Notably,GmCRY1b overexpression lines displayed multiple agronomic advantages over the wild-type control under both dense planting and intercropping conditions.Our study demonstrates the integration of GmCRY1-mediated signals with the GA metabolic pathway in the regulation of LBL-induced SAS in soybean.It also provides a promising option for breeding lodging-resistant,high-yield soybean cultivars in the future.Xiangguang Lyu Qican Cheng Chao Qin Yinghui Li Xinying Xu Ronghuan Ji Ruolan Mu Hongyu Li Tao Zhao Jun Liu Yonggang Zhou Haiyan Li Guodong Yang Qingshan Chen Bin Liu 2021Molecular Plant2021,14,2:4
7Whole-exome sequencing reveals twovariants in thegene in two Chinese patients with left ventricular non-compaction cardiomyopathy显示文摘Importance:Pathogenic variants in theRBM20 gene are associated with aggressive dilated cardiomyopathy(DCM).Recently,RBM20 was found to be associated with left ventricular non-compaction cardiomyopathy(LVNC).Thus far,only five families with LVNC have been reported to carry variants inRBM20.It remains unknown whether the variants inRBM20 associated with DCM can also cause LVNC.Objective:To elucidate the causativeRBM20 variant in two unrelated patients with both LVNC and DCM,and to identify the clinical characteristics associated with variants inRBM20.Methods:Trio whole-exome sequencing(WES)was performed.Variants were filtered and classified in accordance with the guidelines of the American College of Medical Genetics and Genomics(ACMG).Results:We identified two distinctde novo variants inRBM20(one per patient)in these two patients with LVNC.Both variants have been reported in patients with DCM,without the LVNC phenotype.Patient 1 was an 11-year-old girl who had DCM,LVNC,and heart failure;the ratio of noncompacted-to-compacted myocardium was 2.7:1.Ade novo heterozygous variant c.1907G>A(p.Arg636His)in exon 9 was identified in this patient.Patient 2 was a 13-year-old boy who had clinical phenotypes identical to those of Patient 1;the ratio of noncompacted-to-compacted myocardium was 3.2:1 in this patient.WES revealed ade novo heterozygous variant c.1909A>G(p.Ser637Gly)in exon 9.Both variants were previously characterized as pathogenic,and our study classified them as pathogenic variants based on the ACMG guidelines.Interpretation:We found that two patients with LVNC had variants inRBM20.Our results extended the clinical spectrum of the twoRBM20 variants and illustrated that the same variant inRBM20 can cause DCM,with or without the LVNC phenotype.Qiqing Sun Jun Guo Chanjuan Hao Ruolan Guo Xuyun Hu Yuanying Chen Weili Yang Wei Li Yingjun Feng 2020Pediatric Investigation2020,4,1:2

14Evaluation and Hydrological Application of CMADS Reanalysis Precipitation Data against Four Satellite Precipitation Products in the Upper Huaihe River Basin, China显示文摘Satellite-and reanalysis-based precipitation products are important data source for precipitation, particularly in areas with a sparse gauge network. Here, five open-access precipitation products, including the newly released China Meteorological Assimilation Driving Datasets for the Soil and Water Assessment Tool(SWAT) model(CMADS)reanalysis dataset and four widely used bias-adjusted satellite precipitation products [SPPs;i.e., Tropical Rainfall Measuring Mission(TRMM) Multisatellite Precipitation Analysis 3B42 Version 7(TMPA 3B42V7), Climate Prediction Center(CPC) morphing technique satellite–gauge blended product(CMORPH-BLD), Climate Hazards Group Infrared Precipitation with Station Data(CHIRPS), and Precipitation Estimation from Remotely Sensed Information using Artificial Neural Networks–Climate Data Record(PERSIANN-CDR)], were assessed. These products were first compared with the gauge observed data collected for the upper Huaihe River basin, and then were used as forcing data for streamflow simulation by the Xin’anjiang(XAJ) hydrological model under two scenarios with different calibration procedures. The performance of CMADS precipitation product for the Chinese mainland was also assessed. The results show that:(1) for the statistical assessment, CMADS and CMORPH-BLD perform the best, followed by TMPA 3B42V7, CHIRPS, and PERSIANN-CDR, among which the correlation coefficient(CC) and rootmean-square error(RMSE) values of CMADS are optimal, although it exhibits certain significant negative relative bias(BIAS;-22.72%);(2) CMORPH-BLD performs the best in capturing and detecting rainfall events, while CMADS tends to underestimate heavy and torrential precipitation;(3) for streamflow simulation, the performance of using CMADS as input is very good, with the highest Nash–Sutcliffe efficiency(NSE) values(0.85 and 0.75 for calibration period and validation period, respectively);and(4) CMADS exhibits high accuracy in eastern China while with significant negative BIAS, and the performance declines from southeast to northwest. The statistical and hydrological evaluations show that CMADS and CMORPH-BLD have high potential for observing precipitation. As high negative BIAS values showed up in CMADS evaluation, further study on the error sources from original data and calibration algorithms is necessary. 16Promoting hybrid twins structure to reduce yield asymmetry of rolled AZ31 plates by combining side-rolling and torsion显示文摘In this work,an as-rolled AZ31 square bar with c-axis//ND(normal direction)texture was used.Side-rolling and reciprocating torsion were performed to treat the bar.Microstructure evolution and tensile-compressive properties were investigated in detail.Initial rolled AZ31 bar exhibits a large yield asymmetry along the rolling direction(RD).Reciprocating torsion can generate extension twins to introduce twin boundaries and twin-texture.The twin structure can reduce yield asymmetry.However,only limited regions in the rolled AZ31 bar can be twinned during torsion.Pre-side-rolling along the transverse direction(TD)can generate two texture components(c-axis//TD texture and c-axis//ND texture)by introducing profuse{10–12}twins.Such dual texture components help increase the regions which are favorable for twinning during torsion.Finally,combining side-rolling and reciprocating torsion generates hybrid{10–12}twins structure on the entire cross-section,resulting in a remarkably low yield asymmetry.The relevant mechanisms were discussed in detail.Bo Song Meng Wang Ruolan Shi Zhiwen Du Ning Guo Fang Wang Shengfeng Guo 2023Journal of Magnesium and Alloys2023,11,6:1

20CREB is activated in EPO induced HEL cells显示文摘cAMP response element binding protein (CREB) is a transcription factor in nucleus. The activating CREB can specifically bind to the cAMP response element (CRE). The present result showed that erythropoietin (EPO) could induce the phosphorylation of CREB on Serine133 (Pser133), as detected by western blot analysis. in addition, the EPO-dependent activation of CREB binding to CRE element was demonstrated by electrophoretic mobility shift assay. However, the binding of CREB to CRE element could be inhibited by anti-CREB-Pser133 antibody.
