1Identification of colorectal cancer metastasis markers by an angiogenesis-related cytokine-antibody array显示文摘AIM:To investigate the angiogenesis-related protein expression profile characterizing metastatic colorectal cancer (mCRC) with the aim of identifying prognostic markers.METHODS:The expression of 44 angiogenesissecreted factors was measured by a novel cytokine antibody array methodology.The study evaluated vascular endothelial growth factor (VEGF) and its soluble vascular endothelial growth factor receptor (sVEGFR)-1 protein levels by enzyme immunoassay (EIA) in a panel of 16 CRC cell lines.mRNA VEGF and VEGF-A isoforms were quantified by quantitative reverse-transcription polymerase chain reaction (Q-RT-PCR) and vascular endothelial growth factor receptor (VEGFR)-2 expression was analyzed by flow cytometry.RESULTS:Metastasis-derived CRC cell lines expressed a distinctive molecular profile as compared with those isolated from a primary tumor site.Metastatic CRC cell lines were characterized by higher expression of angiogenin-2 (Ang-2),macrophage chemoattractant proteins-3/4 (MCP-3/4),matrix metalloproteinase-1 (MMP-1),and the chemokines interferon γ inducible T cell α chemoattractant protein (I-TAC),monocyte chemoattractant protein I-309,and interleukins interleukin (IL)-2 and IL-1α,as compared to primary tumor cell lines.In contrast,primary CRC cell lines expressed higher levels of interferon γ (IFN-γ),insulin-like growth factor-1 (IGF-1),IL-6,leptin,epidermal growth factor (EGF),placental growth factor (PlGF),thrombopoietin,transforming growth factor β1 (TGF-β1) and VEGF-D,as compared with the metastatic cell lines.VEGF expression does not significantly differ according to the CRC cellular origin in normoxia.Severe hypoxia induced VEGF expression up-regulation but contrary to expectations,metastatic CRC cell lines did not respond as much as primary cell lines to the hypoxic stimulus.In CRC primary-derived cell lines,we observed a twofold increase in VEGF expression between normoxia and hypoxia as compared to metastatic cell lines.CRC cell lines express a similar pattern of VEGF isoforms (VEGF 121,VEGF 165 and VEGF 189) despite variability in VEGF expression,where the major transcript was VEGF 121.No relevant expression of VEGFR-2 was found in CRC cell lines,as compared to that of human umbilical vein endothelial cells and sVEGFR-1 expression did not depend on the CRC cellular origin.CONCLUSION:A distinct angiogenesis-related expression pattern characterizes metastatic CRC cell lines.Factors other than VEGF appear as prognostic markers and intervention targets in the metastatic CRC setting.Ana Abajo Nerea Bitarte Ruth Zarate Valentina Boni Ines Lopez Marisol Gonzalez-Huarriz Javier Rodriguez Eva Bandres Jesus Garcia-Foncillas 2012World Journal of Gastroenterology2012,18,7:8

8Gastric intestinal metaplasia development in African American predominant United States population显示文摘BACKGROUND Gastric cancer significantly contributes to cancer mortality globally.Gastric intestinal metaplasia(GIM)is a stage in the Correa cascade and a premalignant lesion of gastric cancer.The natural history of GIM formation and progression over time is not fully understood.Currently,there are no clear guidelines on GIM surveillance or management in the United States.AIM To investigate factors associated with GIM development over time in African American-predominant study population.METHODS This is a retrospective longitudinal study in a single tertiary hospital in Washington DC.We retrieved upper esophagogastroduodenoscopies(EGDs)with gastric biopsies from the pathology department database from January 2015 to December 2020.Patients included in the study had undergone two or more EGDswith gastric biopsy.Patients with no GIM at baseline were followed up until they developed GIM or until the last available EGD.Exclusion criteria consisted of patients age<18,pregnancy,previous diagnosis of gastric cancer,and missing data including pathology results or endoscopy reports.The study population was divided into two groups based on GIM status.Univariate and multivariate Cox regression was used to estimate the hazard induced by patient demographics,EGD findings,and Helicobacter pylori(H.pylori)status on the GIM status.RESULTS Of 2375 patients who had at least 1 EGD with gastric biopsy,579 patients were included in the study.138 patients developed GIM during the study follow-up period of 1087 d on average,compared to 857 d in patients without GIM(P=0.247).The average age of GIM group was 64 years compared to 56 years in the non-GIM group(P<0.001).In the GIM group,adding one year to the age increases the risk for GIM formation by 4%(P<0.001).Over time,African Americans,Hispanic,and other ethnicities/races had an increased risk of GIM compared to Caucasians with a hazard ratio(HR)of 2.12(1.16,3.87),2.79(1.09,7.13),and 3.19(1.5,6.76)respectively.No gender difference was observed between the study populations.Gastritis was associated with an increased risk for GIM development with an HR of 1.62(1.07,2.44).On the other hand,H.pylori infection did not increase the risk for GIM.CONCLUSION An increase in age and non-Caucasian race/ethnicity are associated with an increased risk of GIM formation.The effect of H.pylori on GIM is limited in low prevalence areas.Akram I Ahmad Arielle Lee Claire Caplan Colin Wikholm Ioannis Pothoulakis Zaynab Almothafer NishthaRaval Samantha Marshall Ankit Mishra Nicole Hodgins In Guk Kang Raymond K Chang Zachary Dailey Arvin Daneshmand Anjani Kapadia Jae Hak Oh Brittney Rodriguez Abhinav Sehgal Matthew Sweeney Christopher B Swisher Daniel F Childers Corinne O'Connor Lynette M Sequeira Won Cho 2022World Journal of Gastrointestinal Endoscopy2022,14,10:1

15Improving surface integrity when drilling CFRPs and Ti-6Al-4V using sustainable lubricated liquid carbon dioxide显示文摘In the quest for decreasing fuel consumption and resulting gas emissions in the aeronautic sector,lightweight materials such as Carbon Fiber Reinforced Polymers(CFRPs)and Ti-6Al-4V alloys are being used.These materials,with excellent weight-to-strength ratios,are widely used for structural applications in aircraft manufacturing.To date,several studies have been published showing that the use of metalworking fluids(MWFs),special tool geometries,or advanced machining techniques is required to ensure a surface quality that meets aerospace component standards.Conventional MWFs pose a number of environmental and worker health hazards and also degrade the mechanical properties of CFRPs due to water absorption in the composite.Therefore,a transition to more environmentally friendly cooling/lubrication techniques that prevent moisture problems in the composite is needed.This research shows that lubricated LCO_(2) is a viable option to improve the quality of drilled CFRP and titanium aerospace components compared to dry machining,while maintaining clean work areas.The results show that the best combination of tool geometry and cooling conditions for machining both materials is drilling with Brad point drills and lubricated LCO_(2).Drilling under these conditions resulted in a 90%improvement in fiber pullout volume compared to dry machined CFRP holes.In addition,a 33% reduction in burr height and a 15% improvement in surface roughness were observed compared to dry drilling of titanium.In˜igo RODRIGUEZ Pedro Jose´ARRAZOLA Mikel CUESTA Luka STERLE Franci PUSˇAVEC 2023Chinese Journal of Aeronautics2023,36,7:0
