| 2 | Function identification of miR482b, a negative regulator during tomato resistance to Phytophthora infestans显示文摘Tomato is an important horticultural and economic crop cultivated worldwide.As Phytophthora infestans becomes a huge threat to tomato production,it is necessary to study the resistance mechanisms of tomato against P.infestans.Our previous research has found that miR482 might be involved in tomato–P.infestans interaction.In this study,miR482b precursor was cloned from Solanum pimpinellifolium“L3708”and miR482b was shown to decrease in abundance in tomato following P.infestans infection.Compared to wild-type tomato plants,tomato plants that overexpressed miR482b displayed more serious disease symptoms after P.infestans infection,with more necrotic cells,longer lesion diameters,and increased P.infestans abundance.Meanwhile,silencing of miR482b was performed by short tandem target mimic(STTM),resulting in enhancement of tomato resistance to P.infestans.Using miRNA and degradome data sets,NBS–LRR disease-resistance genes targeted by miR482b were validated.Negative correlation between the expression of miR482b and its target genes was found in all miR482b-overexpressing and-silencing tomato plants.Our results provide insight into tomato miR482b involved in the response to P.infestans infection,and demonstrate that miR482b–NBS–LRR is an important component in the network of tomato–P.infestans interaction. | Ning Jiang Jun Meng Jun Cui Guangxin Sun Yushi Luan | 2018 | Horticulture Research2018,5,1: | 13 |
| 3 | Tomato lncRNA23468 functions as a competing endogenous RNA to modulate NBS-LRR genes by decoying miR482b in the tomato-Phytophthora infestans interaction显示文摘Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans. | Ning Jiang Jun Cui Yunsheng Shi Guanglei Yang Xiaoxu Zhou Xinxin Hou Jun Meng Yushi Luan | 2019 | Horticulture Research2019,6,1: | 12 |

| 8 | Antimicrobial peptide LL-37 forms complex with bacterial DNA to facilitate blood translocation of bacterial DNA and aggravate ulcerative colitis显示文摘Bacterial DNA(bacDNA) is frequently found in serum of patient with ulcerative colitis(UC) and Crohn's disease, even blood bacterial culture is negative. How bacDNA evades immune elimination and is translocated into blood remain unclear. Here, we showed that bacDNA avoids elimination and disables bacteriakilling function of antimicrobial peptide LL-37(Cramp in mice) by forming complex with LL-37, which is inducible after culture with bacteria or bacterial products. Elevated LL-37-bacDNA complex was found in plasma and lesions of patients with UC. LL-37-bacDNA promoted inflammation by inducing Th1, Th2 and Th17 differentiation and activating toll-like receptor-9(TLR9). The complex also increased paracellular permeability, which possibly combines its inflammatory effects to promote local damage and bacDNA translocation into blood. Cramp-bacDNA aggravated mouse colitis severity while interference with the complex ameliorated the disease. The study identifies that inflammatogenic bacDNA utilizes LL-37 as a vehicle for blood translocation and to evade immune elimination. | 10 | The Influence of Diabetes, Hypertension, and Hyperlipidemia on the Onset of Age-Related Macular Degeneration in North China: The Kailuan Eye Study显示文摘Objective To analyze the prevalence of dry and wet age-related macular degeneration(AMD)in patients with diabetes,hypertension and hyperlipidemia,and to analyze the risk factors for AMD.Methods A population-based cross-sectional epidemiologic study was conducted involving 14,440 individuals.We assessed the prevalence of dry and wet AMD in diabetic and non-diabetic subjects and analyzed the risk factors for AMD.Results The prevalence of wet AMD in diabetic and non-diabetic patients was 0.3%and 0.5%,respectively,and the prevalence of dry AMD was 17%and 16.4%,respectively.The prevalence of wet AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 0.5%,0.3%,0.2%,and 0.7%,respectively.The prevalence of dry AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 16.6%,16.2%,15.2%,and 17.2%,respectively.Age,sex,body mass index,and use of hypoglycemic drugs or lowering blood pressure drugs were corrected in the risk factor analysis of AMD.Diabetes,diabetes/hypertension,diabetes/hyperlipidemia,and diabetes/hypertension/hyperlipidemia were analyzed.None of the factors analyzed in the current study increased the risk for the onset of AMD.Conclusion There was no significant difference in the prevalence of wet and dry AMD among diabetic and non-diabetic subjects.Similarly,there was no significant difference in the prevalence of wet and dry AMD among subjects with hypertension and hyperlipidemia.Diabetes co-existing with hypertension and hyperlipidemia were not shown to be risk factors for the onset of dry AMD. | ZHANG Yong Peng WANG Ya Xing ZHOU Jin Qiong WANG Qian YAN Yan Ni YANG Xuan YANG Jing Yan ZHOU Wen Jia WANG Ping SHEN Chang YANG Ming LUAN Ya Nan WANG Jin Yuan WU Shou Ling CHEN Shuo Hua WANG Hai Wei FANG Li Jian WAN Qian Qian ZHU Jing Yuan NIE Zi Han CHEN Yu Ning XIE Ying JONAS JB WEI Wen Bin | 2022 | Biomedical and Environmental Sciences2022,35,7: | 2 |
| 11 | LNP-CpG ODN-adjuvanted varicella-zoster virus glycoprotein E induced comparable levels of immunity with Shingrix^(TM) in VZV-primed mice显示文摘Latent varicella-zoster virus(VZV)may be reactivated to cause herpes zoster,which affects one in three people during their lifetime.The currently available subunit vaccine Shingrix^(TM) is superior to the attenuated vaccine Zostavax®in terms of both safety and efficacy,but the supply of its key adjuvant component QS21 is limited.With ionizable lipid nanoparticles(LNPs)that were recently approved by the FDA for COVID-19 mRNA vaccines as carriers,and oligodeoxynucleotides containing CpG motifs(CpG ODNs)approved by the FDA for a subunit hepatitis B vaccine as immunostimulators,we developed a LNP vaccine encapsulating VZV-glycoprotein E(gE)and CpG ODN,and compared its immunogenicity with Shingrix^(TM) in C57BL/6J mice.The results showed that the LNP vaccine induced comparable levels of gE-specific IgG antibodies to Shingrix^(TM) as determined by enzymelinked immunosorbent assay(ELISA).Most importantly,the LNP vaccine induced comparable levels of cellmediated immunity(CMI)that plays decisive roles in the efficacy of zoster vaccines to Shingrix^(TM) in a VZVprimed mouse model that was adopted for preclinical studies of Shingrix^(TM) .Number of IL-2 and IFN-γsecreting splenocytes and proportion of T helper 1(Th1)cytokine-expressing CD4^(+)T cells in LNP-CpG-adjuvanted VZV-gE vaccinated mice were similar to that of Shingrix^(TM) boosted mice.All of the components in this LNP vaccine can be artificially and economically synthesized in large quantities,indicating the potential of LNP-CpGadjuvanted VZV-gE as a more cost-effective zoster vaccine. | Ning Luan Han Cao Yunfei Wang Kangyang Lin Cunbao Liu | 2022 | Virologica Sinica2022,37,5: | 2 |
| 12 | Measurement of the neutron total cross section of carbon at the Back-n white neutron beam of CSNS显示文摘To verify the performance of the neutron total cross-sectional spectrometer, the neutron total cross section of carbon is initially measured in the energy range of 1 eV to 20 MeV using the time-of-flight method. The measurement is performed at the Back-n white neutron source with a 76-m time-of-flight path using the China Spallation Neutron Source. A multilayer fast fission chamber with 235U and 238U is employed as the neutron detector. The diameter and thickness of the natural graphite sample are 70 mm and 40 mm, respectively. Signal waveforms are collected using a data acquisition system. Off-line data processing was used to obtain the neutron time-of-flight spectra and transmissions. The uncertainty of the counting statistics is generally approximately 3% for each bin in the energy range of 1–20 MeV. It is determined that the results for the neutron total cross section of carbon obtained using ^235U cells are in good agreement with the results obtained using 238U cells within limits of statistical uncertainty. | 13 | Genetic diversity center of cultivated soybean(Glycine max) in China——New insight and evidence for the diversity center of Chinese cultivated soybean显示文摘Information on the center of genetic diversity of soybean(Glycine max) will be helpful not only for designing efficient strategies for breeding programs, but also for understanding the domestication and origin of this species. Here, we describe an analysis of genetic diversity based on simple-sequence repeat(SSR) variations within a core collection of 2 111 accessions of Chinese soybean landraces. Prior to the diversity assessment, the geographic origin of each accession was mapped. The map was then divided into grids each 2.5° in latitude and 5° in longitude. We found two regions that had higher number of alleles(NA) and greater polymorphic information content(PIC) values than the others. These regions are adjacently located within grid position of 30°–35°N×105°–110°E, which includes the valley of the middle and lower reaches of the Wei River, and the valley of the upper reaches of the Hanjiang River. It was also observed that in many regions, genetic diversity decreased with the increase in distance from the center. Another region, in northern Hebei Province(115°–120°E×40°–42.5°N), was observed having higher diversity than any surrounding regions, indicating that this is a sub-center of soybean diversity.
