| 5 | Hepatic COX1 loss leads to impaired autophagic flux and exacerbates nonalcoholic steatohepatitis显示文摘The mechanisms underlying autophagic defects in nonalcoholic steatohepatitis(NASH)remain largely unknown.We aimed to elucidate the roles of hepatic cyclooxygenase 1(COX1)in autophagy and the pathogenesis of diet-induced steatohepatitis in mice.Human nonalcoholic fatty liver disease(NAFLD)liver samples were used to examine the protein expression of COX1 and the level of autophagy.Cox1^(Δhepa)mice and their wildtype littermates were generated and fed with 3 different NASH models.We found that hepatic COX1 expression was increased in patients with NASH and diet induced NASH mice models accompanied by impaired autophagy.COX1 was required for basal autophagy in hepatocytes and liver specific COX1 deletion exacerbated steatohepatitis by inhibiting autophagy.Mechanistically,COX1 directly interacted with WD repeat domain,phosphoinositide interacting 2(WIPI2),which was crucial for autophagosome maturation.Adeno-associated virus(AAV)-mediated rescue of WIPI2 reversed the impaired autophagic flux and improved NASH phenotypes in Cox1^(Δhepa)mice,indicating that COX1 deletion-mediated steatohepatitis was partially dependent on WIPI2-mediated autophagy.In conclusion,we demonstrated a novel role of COX1 in hepatic autophagy that protected against NASH by interacting with WIPI2.Targeting the COX1 WIPI2 axis may be a novel therapeutic strategy for NASH. | Qian Yu Chang Li Qinghui Niu Jigang Wang Zhaodi Che Ke Lei He Ren Boyi Ma Yixing Ren Pingping Luo Zhuming Fan Huan Zhang Zhaohui Liu George L.Tipoe Jia Xiao | 2023 | Acta Pharmaceutica Sinica B2023,13,6: | 0 |
| 6 | Higher Education Expansion, Selections Based on Non-Cognitive Abilities and Gender Differences: The Case of China显示文摘We assess how implicit selections based on non-cognitive abilities may have changed in the context of the great higher education expansion in China,applying the classic supply-demand framework and utilizing the Big Five personality model.Using the Chinese General Social Survey(CGSS)data from 2012,difference-in-difference(DID),difference-in-difference-in-difference(DDD)and descriptive analyses establish the following three major findings.First,from the generation entering college before the expansion-post-70s-to the generation entering college after the expansion-post-80s and post-90s-implicit selections in higher education based on personality have weakened with the increased supply of higher education opportunities.Second,selections have significantly weakened in terms of the openness dimension of personality,yet there is some evidence that selections have strengthened in terms of conscientiousness.This reflects that open-mindedness has become a relatively higher supply trait and conscientiousness has become a relatively lower supply trait among members of the post-90s generation.Third,selections have weakened only on the openness dimension for males,but on multiple dimensions beginning with agreeableness for females,reflecting a greatly increased supply of higher education opportunities for females with this dominant trait.The finding on strengthened selections based on conscientiousness has important implications for what and how to educate today’s college students. | YANG Ying NIU Xinchun YANG Fei HU Pingping | 2020 | Frontiers of Education in China2020,15,2: | 0 |
| 7 | An OsPRMT5-OsAGO2/miR1875-OsHXK1 module regulates rice immunity to blast disease显示文摘Rice ARGONAUTE2(OsAGO2)is a core component of the rice RNA-induced silencing complex(RISC),which is repressed by Magnaporthe oryzae(M.oryzae)infection.Whether and how OsAGO2-mediated gene silencing plays a role in rice blast resistance and which sRNAs participate in this process are unknown.Our results indicate that OsAGO2 is a key immune player that manipulates rice defense responses against blast disease.OsAGO2 associates with the 24-nt miR1875 and binds to the promoter region of HEXOKINASE1(OsHXK1),which causes DNA methylation and leads to gene silencing.Our multiple genetic evidence showed that,without M.oryzae infection,OsAGO2/miR1875 RISC promoted OsHXK1 promoter DNA methylation and OsHXK1 silencing;after M.oryzae infection,the reduced OsAGO2/miR1875 led to a relatively activated OsHXK1 expression.OsHXK1 acts as a positive regulator of blast disease resistance that OsHXK1-OE rice exhibited enhanced resistance,whereas Cas9-Oshxk1 rice showed reduced resistance against M.oryzae infection.OsHXK1 may function through its sugar sensor activity as glucose induced defense-related gene expression and reactive oxygen species(ROS)accumulation in Nipponbare and OsHXK1-OE but not in Cas9-Oshxk1 rice.OsAGO2 itself is delicately regulated by OsPRMT5,which senses M.oryzae infection and attenuates OsAGO2-mediated gene silencing through OsAGO2 arginine methylation.Our study reveals an OsPRMT5-OsAGO2/miR1875-OsHXK1 regulatory module that fine tunes the rice defense response to blast disease. | Cong Sheng Xuan Li Shengge Xia Yimai Zhang Ze Yu Cheng Tang Le Xu Zhaoyun Wang Xin Zhang Tong Zhou Pingping Nie Ayesha Baig Dongdong Niu Hongwei Zhao | 2023 | Journal of Integrative Plant Biology2023,65,4: | 0 |
