| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Expression of calreticulin is associated with infiltration of T-cells in stage ⅢB colon cancer显示文摘AIM: To investigate the correlation between expression of calreticulin and infiltration of lymphocytes in stage ⅢB colon cancer. METHODS: Sixty-eight pathologically-confirmed speci-mens were obtained from stage ⅢB (T3N1M0) colon cancer patients who underwent radical resection between January 1999 and May 2002 at the Cancer Center of Sun Yat-Sen University, Guangzhou, China. Immuno-histochemical analysis was performed to show infiltration of lymphocytes and expression of calreticulin in colon cancer. Association between calreticulin expression, infiltration of lymphocytes, and 5-year survival rate of patients was assessed. RESULTS: The expression level of calreticulin was lower in cancer nest than in its adjacent normal epithelium since 61.8% (42/68) of the samples were stained with calreticulin in colon cancer. The expression of calreticulin in colon cancer was associated with the infi ltration of CD45RO+ cells rather than with that of CD3+ cells. In addition, the stronger expression of calreticulin and the higher infiltration of CD3+ and CD45RO+ cells in colon cancer were associated with the higher 5-year survival rate of patients. | 4 | Effect of controlled low central venous pressure on renal function in major liver resection显示文摘Objective: To investigate the effects of low central venous pressure (LVCP) on blood loss and evaluate its influ- ence on renal function in patients undergoing hepatectomy. Methods: Forty-six patients, ASA classification I–III, undergoing liver resection were randomized into LCVP group (n = 23) and control group (n = 23). In LCVP group, CVP was maintained at 2–4 mmHg and MBP above 60 mmHg during hepatectomy, while in control group hepatectomy was performed routinely without lowering CVP. Volume of blood loss during hepatectomy, volume of blood transfusion, and changes of renal functions were compared between the two groups. Results: There were no significant differences in demographics, ASA score, type of hepatectomy, duration of inflow occlusion, operation time, weight of resected liver tissues, and renal functions between the two groups. LCVP group had a significantly lower volume of total intraoperative blood loss (P < 0.01) and RBC transfusion (P < 0.05). | 6 | Evaluation of a scientific CMOS camera for astronomical observations显示文摘We evaluate the performance of the first generation scientific CMOS (sC-MOS) camera used for astronomical observations. The sCMOS camera was attached to a 25 cm telescope at Xinglong Observatory, in order to estimate its photometric capabilities. We further compared the capabilities of the sCMOS camera with that of full-frame and electron multiplying CCD cameras in laboratory tests and observations. | 7 | Parkin deficiency accentuates chronic alcohol intake-induced tissue injury and autophagy defects in brain,liver and skeletal muscle显示文摘Alcoholism leads to organ injury including mitochondrial defect and apoptosis with evidence favoring a role for autophagy dysregulation in alcoholic damage.Parkin represents an autosomal recessive in herited gene for Parkins on's disease and an important member of selective autophagy for mitoch on dria.The associati on between Parkins on's disease and alcoholic injury remains elusive.This study aimed to examine the effect of parki n deficiency on chr onic alcohol in takeinduced organ injury in brain,liver and skeletal muscle(rectus femoris muscle).Adult parkinknockout(PRK-/-)and wild-type mice were placed on Liber-De Carli alcohol liquid diet(4%)for 12 weeks prior to assessment of liver enzymes,intraperitoneal glucose toleranee,protein carbonyl content apoptosis,hematoxylin and eosin morphological staining,and mitochondrial respiration(cytochrome c oxidase,NADH:cytochrome c reductase and succinate:cytochrome c reductase).Autophagy protein markers were monitored by western blot analysis.Our data revealed that chronic alcohol intake imposed liver injury as evidenced by elevated aspartate aminotransferase and alanine transaminase,glucose intoleranee,elevated protein carbonyl formation,apoptosis,focal inflammation,necrosis,microvesiculation,autophagy/mitophagy failure and dampened mitochondrial respiration(complexⅣ,complexesⅠandⅢ,and complexesⅡandⅢ)in the brain,liver and rectus femoris skeletal muscle.Although parkin ablation itself did not generate any notable effects on liver enzymes,insulin sensitivity,tissue carbonyl damage,apoptosis,tissue morphology,autophagy or mitochondrial respiration,it accentuated alcohol intake-induced tissue damage,apoptosis,morphological change,autophagy/mitophagy failure and mitochondrial injury without affecting insulin sensitivity.These data suggest that parkin plays an integral role in the preservation against alcohol-induced organ injury,apoptosis and mitochondrial damage. | Hu Peng Xing Qin Sainan Chen Asli E Ceylan Maolong Dong Zhaofen Lin Jun Ren | 2020 | Acta Biochimica et Biophysica Sinica2020，52，6： | 3 |
| 8 | Simulating global soil-CO_2 flux and its response to climate change显示文摘It has been argued that increased soil respiration would become a major atmospheric source of CO\-2 in the event of global warming. The simple statistical models were developed based on a georeferenced database with 0 5°×0 5° longitude/latitude resolution to simulate global soil\|CO\-2 fluxes, to investigate climatic effects on these fluxes using sensitivity experiments, and to assess possible responses of soil\|CO\-2 fluxes to various climate change scenarios. The statistical models yield a value of 69 PgC/a of global soil\|CO\-2 fluxes for current condition. Sensitivity experiments confirm that the fluxes are responsive to changes in temperature, precipitation and actual evapotranspiration, but increases in temperature and actual evapotranspiration affect soil\|CO 2 fluxes more than increases in precipitation. Using climatic change projections from four global circulation models, each corresponding to an equilibrium doubling of CO 2, it can be found that the largest increases in soil\|CO 2 fluxes were associated with the boreal and tundra regions. The globally averaged soil\|CO 2 fluxes were estimated to increase by about 35% above current values, providing a positive feedback to the greenhouse effect. | PENG Chang\|hui 1* , Michael J. | 9 | Oral delivery of capsaicin using MPEG-PC nanoparticles显示文摘Aim: To prepare a biodegradable polymeric carrier for oral delivery of a water-insoluble drug capsaicin (CAP) and evaluate its quality. Methods: CAP-loaded methoxy poly (ethylene glycol)-poly(c-caprolactone) nanoparticles (CAP/NPs) were prepared using a modified emulsification solvent diffusion technique. The quality of CAP/NPs were evaluated using transmission electron microscopy, powder X-ray diffraction, differential scanning calorimetry and Fourier transform infrared techniques. A dialysis method was used to analyze the in vitro release profile of CAP from the CAP/NPs. Adult male rats were orally administered CAP/NPs (35 mg/kg); and the plasma concentrations of CAP were measured with a validated HPLC method. The morphology of rat gastric mucosa was studied with HE staining. Results: CAP/NPs had an average diameter of 82.54±0.51 nm, high drug-loading capacity of 14.0%±0.13% and high stability. CAP/NPs showed a biphasic release profile in vitro: the burst release was less than 25% of the loaded drug within 12 h followed by a more sustained release for 60 h. The pharmacokinetics study showed that the mean maximum plasma concentration was observed 4 h after oral administered of CAP/NPs, and approximately 90 ng/mL of CAP was detected in serum after 36 h. The area under the curve for the CAP/NPs group was approximately 6-fold higher than that for raw CAP suspension. Histological studies showed that CAP/NPs markedly reduced CAP-caused gastric mucosa irritation.
