| 1 | A SPE-HPLC-MS/MS method for the simultaneous determination of pr ior itised pharmaceuticals and EDCs with high environmental risk potential in freshwater显示文摘This work describes the development,optimisation and validation of an analytical method for the rapid determination of 17 priority pharmaceutical compounds and endocrine disrupting chemicals(EDCs).Rather than studying compounds from the same therapeutic class,the analyses aimed to determine target compounds with the highest risk potential(with particular regard to Scotland),providing a tool for further monitoring in different water matrices.Prioritisation was based on a systematic environmental risk assessment approach,using consumption data;wastewater treatment removal efficiency;environmental occurrence;toxicological effects;and pre-existing regulatory indicators.This process highlighted 17 compounds across various therapeutic classes,which were then quantified,at environmentally relevant concentrations,by a single analytical methodology.Analytical determination was achieved using a single-step solid phase extraction(SPE)procedure followed by high-performance liquid chromatography with tandem mass spectrometry(HPLC-MS/MS).The fully optimised method performed well for the majority of target compounds,with recoveries>71%for 15 of 17 analytes.The limits of quantification for most target analytes(14 of 17)ranged from 0.07 ng/L to 1.88 ng/L in river waters.The utility of this method was then demonstrated using real water samples associated with a rural hospital/setting.Eight compounds were targeted and detected,with the highest levels found for the analgesic,paracetamol(at up to 105,910 ng/L in the hospital discharge).This method offers a robust tool to monitor high priority pharmaceutical and EDC levels in various aqueous sample matrices. | Yuan Li MarkATaggart Craig McKenzie Zulin Zhang Yonglong Lu Sabolc Pap Stuart WGibb | 2021 | Journal of Environmental Sciences2021,33,2: | 4 |

| 3 | Comparative transcriptomics of Central Asian Vitis vinifera accessions reveals distinct defense strategies against powdery mildew显示文摘Grape powdery mildew(PM),caused by the biotrophic ascomycete Erysiphe necator,is a devastating fungal disease that affects most Vitis vinifera cultivars.We have previously identified a panel of V.vinifera accessions from Central Asia with partial resistance to PM that possess a Ren1-like local haplotype.In this study,we show that in addition to the typical Ren1-associated late post-penetration resistance,these accessions display a range of different levels of disease development suggesting that alternative alleles or additional genes contribute to determining the outcome of the interaction with the pathogen.To identify potential Ren1-dependent transcriptional responses and functions associated with the different levels of resistance,we sequenced and analyzed the transcriptomes of these Central Asian accessions at two time points of PM infection.Transcriptomes were compared to identify constitutive differences and PM-inducible responses that may underlie their disease resistant phenotype.Responses to E.necator in all resistant accessions were characterized by an early up-regulation of 13 genes,most encoding putative defense functions,and a late down-regulation of 32 genes,enriched in transcriptional regulators and protein kinases.Potential Ren1-dependent responses included a hotspot of co-regulated genes on chromosome 18.We also identified 81 genes whose expression levels and dynamics correlated with the phenotypic differences between the most resistant accessions‘Karadzhandahal’,DVIT3351.27,and O34-16 and the other genotypes.This study provides a first exploration of the functions associated with varying levels of partial resistance to PM in V.vinifera accessions that can be exploited as sources of genetic resistance in grape breeding programs. | Katherine C H Amrine Barbara Blanco-Ulate Summaira Riaz Dániel Pap Laura Jones Rosa Figueroa-Balderas M Andrew Walker Dario Cantu | 2015 | Horticulture Research2015,2,1: | 3 |
| 4 | Genetic mapping and survey of powdery mildew resistance in the wild Central Asian ancestor of cultivated grapevines in Central Asia显示文摘Cultivated grapevines(Vitis vinifera)lack resistance to powdery mildew(PM)with few exceptions.Resistance to this pathogen within V.vinifera has been reported in earlier studies and identified as the Ren1 locus in two Central Asian table grape accessions.Other PM-resistant cultivated varieties and accessions of the wild ancestor V.vinifera subsp.sylvestris were soon identified raising questions regarding the origin of the resistance.In this study,F1 breeding populations were developed with a PM susceptible V.vinifera subsp.vinifera breeding line and a PM-resistant subsp.sylvestris accession.Genotyping was carried out with five Ren1 locus linked SSR markers.A PM resistance locus explaining up to 96%of the phenotypic variation was identified in the same genomic position,where the Ren1 locus was previously reported.New SSR marker alleles linked with the resistance locus were identified.We report results of PM resistance in multiple accessions of subsp.sylvestris collected as seed lots or cuttings from five countries in the Caucasus and Central Asia.A total of 20 females from 11 seed lots and 19 males from nine seed lots collected from Georgia,Armenia,and Azerbaijan were resistant to PM.Three male and one female plant collected as cuttings from Afghanistan and Iran were also resistant to PM.Allelic analysis of markers linked with the Ren1 locus in conjunction with disease evaluation data found a high diversity of allelic haplotypes,which are only possible via recombination events occurring over a long time period.Sequence analysis of two alleles of the SSR marker that cosegregates with the resistance found SNPs that were present in the wild progenitor and in cultivated forms.Variable levels of PM resistance among the tested accessions were also observed.These lines of evidence suggest that the powdery mildew fungus may have been present in Asia for a longer time than currently thought,giving the wild progenitor V.vinifera subsp.sylvestris time to coevolve with and develop resistance to this pathogen. | Summaira Riaz Cristina M.Menéndez Alan Tenscher Daniel Pap M.Andrew Walker | 2020 | Horticulture Research2020,7,1: | 2 |
| 5 | CMYKIR红外防伪图像技术显示文摘印刷油墨在近红外波长区域的响应是不同的,不能用CIELAB、sRGB或HSB这些基于可见光颜色理论建立的色彩系统参数来描述。用来描述人眼接收图像的RGB加色呈色系统与可以覆盖几乎全部可见光谱内印刷品的C_0M_0Y_0色料减色呈色系统相对应。加入对红外波谱有着极佳响应的黑色油墨,配上CMY减色油墨,就可以使每一种色调有着无数种配比方法。CMYK印刷油墨的特性是得到独立图像两种外观表现响应算法的基础,一种是日光下的图像外观,一种是红外光下的图像外观。预设算法基于两个图像,即原始图像X_0和在蒙版F基础上生成的印刷图像X。此举的目的是通过对红外图像检测实现文件防伪。在仪器帮助下,700~1000nm波长范围内的图像元素为可见,使只用原色油墨印刷图像进行信息选择成为可能。每个原色油墨的特性用于向为生成红外图像而进行的RGB/CMYK图像分离过程中引入新的标准。这种图像分离称为CMYKIR图像分离,并用其来设计红外响应图像的油墨用量,这种红外响应包括IR效果的产生和消除。CMYKIR印刷的目的在于,引入可见光谱下无法观察到的红外图像。 | V.Ziljak K.
