| 1 | Patterns of local recurrence in rectal cancer after a multidisciplinary approach显示文摘Improvements in surgery and the application of combined approaches to fight rectal cancer have succeeded in reducing the local recurrence (LR) rate and when there is LR it tends to appear later and less often in isolation. Moreover, a subtle change in the distribution of LRs with respect to the pelvis has been observed. In general terms, prior to total mesorectal excision the most common LRs were central types (perianastomotic and anterior) while lateral and posterior forms (presa-cral) have become more common since the growth in the use of combined treatments. No differences have been reported in the current pattern of LRs as a function of the type of approach used, that is, neo-adjuvant therapies (short-term or long-course radiotherapy, orchemoradiotherapy versus extended lymphadenectomy, though there is a trend towards posterior or presacral LR in patients in the Western world and lateral LR in Asia. Nevertheless, both may arise from the same mechanism. Moreover, as well as the mode of treatment, the type of LR is related to the height of the initial tumor. Nowadays most LRs are related to the advanced nature of the disease. Involvement of the circumferential radial margin and spillage of residual tumor cells from lymphatic leakage in the pelvic side wall are two plausible mechanisms for the genesis of LR. The patterns of pelvic recurrence itself (pelvic subsites) also have important implications for prognosis and are related to the potential success of salvage curative approach. The re-operability for cure and prognosis are generally better for anastomotic and anterior types than for presacral and lateral recurrences. | 2 | Identification of colorectal cancer metastasis markers by an angiogenesis-related cytokine-antibody array显示文摘AIM:To investigate the angiogenesis-related protein expression profile characterizing metastatic colorectal cancer (mCRC) with the aim of identifying prognostic markers.METHODS:The expression of 44 angiogenesissecreted factors was measured by a novel cytokine antibody array methodology.The study evaluated vascular endothelial growth factor (VEGF) and its soluble vascular endothelial growth factor receptor (sVEGFR)-1 protein levels by enzyme immunoassay (EIA) in a panel of 16 CRC cell lines.mRNA VEGF and VEGF-A isoforms were quantified by quantitative reverse-transcription polymerase chain reaction (Q-RT-PCR) and vascular endothelial growth factor receptor (VEGFR)-2 expression was analyzed by flow cytometry.RESULTS:Metastasis-derived CRC cell lines expressed a distinctive molecular profile as compared with those isolated from a primary tumor site.Metastatic CRC cell lines were characterized by higher expression of angiogenin-2 (Ang-2),macrophage chemoattractant proteins-3/4 (MCP-3/4),matrix metalloproteinase-1 (MMP-1),and the chemokines interferon γ inducible T cell α chemoattractant protein (I-TAC),monocyte chemoattractant protein I-309,and interleukins interleukin (IL)-2 and IL-1α,as compared to primary tumor cell lines.In contrast,primary CRC cell lines expressed higher levels of interferon γ (IFN-γ),insulin-like growth factor-1 (IGF-1),IL-6,leptin,epidermal growth factor (EGF),placental growth factor (PlGF),thrombopoietin,transforming growth factor β1 (TGF-β1) and VEGF-D,as compared with the metastatic cell lines.VEGF expression does not significantly differ according to the CRC cellular origin in normoxia.Severe hypoxia induced VEGF expression up-regulation but contrary to expectations,metastatic CRC cell lines did not respond as much as primary cell lines to the hypoxic stimulus.In CRC primary-derived cell lines,we observed a twofold increase in VEGF expression between normoxia and hypoxia as compared to metastatic cell lines.CRC cell lines express a similar pattern of VEGF isoforms (VEGF 121,VEGF 165 and VEGF 189) despite variability in VEGF expression,where the major transcript was VEGF 121.No relevant expression of VEGFR-2 was found in CRC cell lines,as compared to that of human umbilical vein endothelial cells and sVEGFR-1 expression did not depend on the CRC cellular origin.CONCLUSION:A distinct angiogenesis-related expression pattern characterizes metastatic CRC cell lines.Factors other than VEGF appear as prognostic markers and intervention targets in the metastatic CRC setting. | Ana Abajo Nerea Bitarte Ruth Zarate Valentina Boni Ines Lopez Marisol Gonzalez-Huarriz Javier Rodriguez Eva Bandres Jesus Garcia-Foncillas | 2012 | World Journal of Gastroenterology2012,18,7: | 8 |
| 3 | Antenna-assisted picosecond control of nanoscale phase transition in vanadium dioxide显示文摘Nanoscale devices in which the interaction with light can be configured using external control signals hold great interest for next-generation optoelectronic circuits.Materials exhibiting a structural or electronic phase transition offer a large modulation contrast with multi-level optical switching and memory functionalities.In addition,plasmonic nanoantennas can provide an efficient enhancement mechanism for both the optically induced excitation and the readout of materials strategically positioned in their local environment.Here,we demonstrate picosecond all-optical switching of the local phase transition in plasmonic antenna-vanadium dioxide(VO_(2))hybrids,exploiting strong resonant field enhancement and selective optical pumping in plasmonic hotspots.Polarization-and wavelength-dependent pump–probe spectroscopy of multifrequency crossed antenna arrays shows that nanoscale optical switching in plasmonic hotspots does not affect neighboring antennas placed within 100 nm of the excited antennas.The antenna-assisted pumping mechanism is confirmed by numerical model calculations of the resonant,antenna-mediated local heating on a picosecond time scale.The hybrid,nanoscale excitation mechanism results in 20 times reduced switching energies and 5 times faster recovery times than a VO_(2) film without antennas,enabling fully reversible switching at over two million cycles per second and at local switching energies in the picojoule range.The hybrid solution of antennas and VO_(2) provides a conceptual framework to merge the field localization and phase-transition response,enabling precise,nanoscale optical memory functionalities. | Otto L Muskens Luca Bergamini Yudong Wang Jeffrey M Gaskell Nerea Zabala CH de Groot David W Sheel Javier Aizpurua | 2016 | Light(Science & Applications)2016,5,1: | 5 |
| 4 | Pharmacogenomics in colorectal cancer: The first step for individualized-therapy显示文摘Interindividual differences in the toxicity and response to anticancer therapies are currently observed in practically all available treatment regimens. A goal of cancer therapy is to predict patient response and toxicity to drugs in order to facilitate the individualization of patient treatment. Identification of subgroups of patients that differ in their prognosis and response to treatment could help to identify the best available drug therapy according the genetic profile. Several mechanisms have been suggested to contribute to chemo-therapeutic drug resistance: amplification or overexpression of membrane transporters, changes in cellular proteins involved in detoxification or in DNA repair, apoptosis and activation of oncogenes or tumor suppressor genes. Colorectal cancer (CRC) is regarded as intrinsically resistant to chemotherapy. Several molecular markers predictive of CRC therapy have been included during the last decade but their results in different studies complicate their application in practical clinical. The simultaneous testing of multiple markers predictive of response could help to identify more accurately the true role of these polymorphisms in CRC therapy. During the last several years, research has been primarily based on the study of expression profiles using microarray technology. But now, investigators are putting into practice proteomic analyses of cancer tissues and cells to identify new diagnostic or therapeutic biomarkers for this cancer. Because the proteome reflects the state of a cell, tissue or organism more accurately, much is expected from proteomics to yield better tumor markers for disease diagnosis and therapy monitoring. | 6 | Diagnostic performance of serum interleukin-6 in pediatric acute appendicitis:a systematic review显示文摘Background Despite the radiological and analytical advances of the last decades,there is still a significant rate of diagnostic error in pediatric acute appendicitis.In recent years,multiple biomarkers have emerged as potential diagnostic tools.This study aimed to examine the diagnostic performance of serum interleukin-6 in pediatric acute appendicitis.Methods We conducted a systematic review of the literature that involved an extensive search in the main databases of medical bibliography(Medline,PubMed,Web of Science and SciELO).Two independent reviewers selected the relevant articles based on the previously defined inclusion and exclusion criteria.Methodological quality of the selected article was rated using the QUADAS2 index.Data extraction was performed by two independent reviewers.Results The research in the medical bibliography databases resulted in 68 articles.We removed 26 duplicates.Among the remaining 42 articles,we excluded 33 following the inclusion and exclusion criteria.Of the final 9 studies included in this review,8 provided measured serum interleukin-6 values,and all of them reported significant differences between groups,but inconsistent results regarding sensitivity and specificity.Conclusions The diagnostic performance of interleukine-6 alone for the diagnosis of acute appendicitis in children is limited.The sensitivity and specificity of interleukine-6 for the diagnosis of non-complicated acute appendicitis in the pediatric population are moderate but increased in complicated appendicitis.There seems to be a direct relationship between serum level of interleukin-6 and the hours of evolution of abdominal pain in children with acute appendicitis.The increasingly widespread use of non-operative management of acute appendicitis warrants further exploration of the classificatory potential of this marker between complicated and uncomplicated appendicitis.We consider that this may be an avenue of research to explore in the future. | Javier Arredondo Montero Carlos Bardaji Pascual Monica Bronte Anaut Natalia Lopez-Andres Giuseppa Antona Nerea Martin-Calvo | 2022 | World Journal of Pediatrics2022,18,2: | 3 |
| 7 | Exploring the factors affecting carbon and nutrient concentrations in tree biomass components in natural forests,forest plantations and short rotation forestry显示文摘Background: Coupling biomass models with nutrient concentrations can provide sound estimations of carbon and nutrient contents, enabling the improvement of carbon and nutrient balance in forest ecosystems. Although nutrient concentrations are often assumed to be constant for some species and specific tree components, at least in mature stands,the concentrations usually vary with age, site index and even with tree density. The main objective of this study was to evaluate the sources of variation in nutrient concentrations in biomass compartments usually removed during harvesting operations, covering a range of species and management conditions: semi-natural forest, conventional forest plantations and short rotation forestry(SRF). Five species(Betula pubescens, Quercus robur, Eucalyptus globulus, Eucalyptus nitens and Populus spp.) and 14 genotypes were considered. A total of 430 trees were sampled in 61 plots to obtain 6 biomass components:leaves, twigs, thin branches, thick branches, bark and wood Aboveground leatos biomass was pooled togetto The concentrations of C, N, K P, Ca, Mg, S; Fe, Mn, Cu, Zn and B were measured and the total biomass of each sampled tree and plot were determined. The data were analysed using boosted regression trees and conventional techniques.Results: The main sources of variation in nutrient concentrations were biomass component > > genotype(species)-age >tree diameter. The concentrations of Ca, Mg and K were most strongly affeaed by genotype and age. The concentrations of P, K Ca, Mg; S and Cu in the wood component decreased with age, whereas C concentrations increased, with a trend to reach 50% in the older trees. In the SRF, interamerican poplar and P. trichocorpa genotypes were comparatively more efficient in terms of Ca and K nutrient assimilation index(NAI)(+65-85%) than eucalypts, mainly because leafless biomass can be removed. In the conventional eucalypt plantations(rotation 15 years), debarking the wood at logging(savings of225% of Ca and 254% of Mg for £ globulus) or the use of selected genotypes(savings of 45% of P and 35% of Ca) will provide wood at a relatively lower nutrient cost. Considering all the E globulus genotypes together, the management for pulp with removal of debarked wood shows NAI values well above(x 1.7-x 3.9) the ones found for poplar or eucalypt SRF and also higher(x 1.6-X4.0) than the ones found for oak and birch managed in medium or long rotations.The annual rates of nutrient removal were low in the native broadleaved species but the rates of available soil nutrients removed were high as compared to poplar or eucalypts. Management of native broadleaved species should consider nutrient stability through selection of the biomass compartments removed.(Continued on next page)(Continued from previous page)Conclusions: The nutrient assimilation index is higher in poplar grown under short rotation forestry management than in the other systems considered. Nutrient management of fast growing eucalyptus plantations could be improved by selecting efficient genotypes and limiting removal of wood.
