1Ki-67 as a prognostic marker according to breast cancer molecular subtype显示文摘Objective: Ki-67 plays an important function in cell division, but its exact role is still unknown. Moreover, few works regarding its overall function were published. The present study evaluated the clinical significance of Ki-67 index as a prognostic marker and predictor of recurrence in different molecular subtypes of breast cancer. The relationship of Ki-67 index with different clinicopathological factors was also analyzed.Methods: Ki-67 index was measured in 107 cases of primary breast cancer from 2010-2012. These patients were evaluated for estrogen receptor, progesterone receptor, and HER2. Ki-67 was divided according to percentage levels: < 15% and > 15%. Followup ranged from 32 months up to 6 years.Results: Approximately 44, 23, 15, and 25 cases were grouped as luminal A, luminal B, HER2 subtype, and triple-negative(TN),respectively. No luminal A patients showed Ki-67 level higher than 15%, and their recurrence was 20%. In luminal B group, Ki-67 level higher than 15% was observed in 69% of patients, and recurrence was 39%. In HER2 subtype, Ki-67 was higher than 15% in34% of cases, and recurrence was 40%. In triple-negative cases, Ki-67 was higher than 15% in 60% of cases, and recurrence was detected in 32% of patients. Patients with Ki-67 less than 15% displayed better overall survival than those with Ki-67 higher than15%(P = 0.01). 2Decreased apoptosis in advanced-stage/high-grade hepatocellular carcinoma complicating chronic hepatitis C is mediated through the downregulation of p21 ras显示文摘Objective and background: Although p21 ras has been reported to be upregulated in hepatocellular carcinoma complicating chronic hepatitis C type I, p21 ras has a different role in advanced stages, as it has been found to be downregulated. The goal of this study was to investigate the status of p21 ras in early-stage/low-grade and late-stage/high-grade hepatocellular carcinoma and its possible link to apoptosis. Material and methods: Thirty-five cases each of chronic HCV hepatitis type 4 (group I) and cirrhosis with hepatocellular carcinoma (HCC) complicating chronic HCV hepatitis (groups II and III) were immunohistochemically evaluated using a p21 ras polyclonal antibody. The apoptotic index was determined in histologic sections using the terminal deoxynucleotidyl transferase-mediated d-UTP biotin nick end labeling (TUNEL) assay. Results: Significant differences (P=0.001) were detected in p21 ras protein expression between the three groups. A near 2-fold increase in p21 ras staining was observed in the cirrhotic cases compared to the hepatitis cases, and p21 ras expression was decreased in the HCC group. p21 ras expression correlated with stage (r=0.64, P=0.001) and grade (r=-0.65, P=0.001) in the HCC group and grade in the HCV group (r=0.44, P=0.008). Both p21 ras expression and TUNEL-LI were significantly lower in large HCCs compared to small HCCs (P=0.01 each). The TUNEL values were negatively correlated with stage in the HCC group (r=-0.85, P=0.001). The TUNEL values were also negatively correlated with grade in both the HCV and HCC groups (r=0.89, P=0.001 and r=-0.53, P=0.001, respectively). The p21 ras scores were significantly correlated with the TUNEL-LI values in the HCC group (r=0.63, P=0.001) and HCV group (r=0.88, P=0.001). 3Quantum dots protect against MPP+-induced neurotoxicity in a cell model of Parkinson's disease through autophagy induction显示文摘Autophagy is a basic cellular process that decomposes damaged organelles and aberrant proteins. Dysregulation of autophagy is implicated in pathogenesis of neurodegenerative disorders, including Parkinson's disease(PD). Pharmacological compounds that stimulate autophagy can provide neuroprotection in models of PD. Nanoparticles have emerged as regulators of autophagy and have been tested in adjuvant therapy for diseases. In this present study, we explore the effects of quantum dots(QDs) that can induce autophagy in a cellular model of Parkinson's disease. Cd Te/Cd S/Zn S QDs protect differentiated rat pheochromocytoma PC12 cells from MPP+-induced cell damage, including reduced viability, apoptosis and accumulation of α-Synuclein, a characteristic protein of PD. The protective function of QDs is autophagy-dependent. In addition, we investigate the interaction between quantum dots and autophagic pathways and identify beclin1 as an essential factor for QDs-induced autophagy. 4Residual effects of tobacco biochar along with different fixing agents on stabilization of trace elements in multi-metal contaminated soils显示文摘The residual effect of tobacco biochar(TB≥500°C)mono and co-application with Cahydroxide(CH),Ca-bentonite(CB)and natural zeolite(NZ)on the bio-availability of trace elements TE(s)in alkaline soils has not been deeply studied yet.A pot study that had earlier been investigated TB mono and blended with CH,CB and NZ on the immobilization of Pb,Cu Cd,and Zn by Chinese cabbage.Maize crop in the rotation was selected as test plant to assess the residual impact of amendments on stabilization of Pb,Cu Cd,and Zn in mine polluted(M-P),smelter heavily and low polluted(S-HP and S-LP,respectively)soils.The obtained results showed that stabilization of Pb,Cd,Cu and Zn reached 63.84% with TB+CB,61.19% with TB+CH,83.31% with TB+CH and 35.27% with TB+CH for M-P soil,36.46% with TB+NZ,38.46% with TB+NZ,19.40% with TB+CH and 62.43% with TB+CH for S-LP soil,52.94%TB+NZ,57.65% with TB+NZ,52.94% with TB+NZ,and 28.44% with TB+CH for S-LP soil.Conversely,TB+CH and TB alone had mobilized Pb and Zn up to19.29% and 34.96% in M-P soil.The mobility of Zn reached 8.38% with TB+CB and 66.03% with TB for S-HP and S-LP soils.The uptake and accumulation of Pb,Cd,Cu and Zn in shoot and root were reduced in three polluted soils.Overall,the combination of TB along with CH,CB and NZ has been proven to be effective in Pb,Cd,Cu and Zn polluted mine/smelter soils restoration.Altaf Hussain Lahori Monika Mierzwa-Hersztek Muhammad Rashid Shahmir Ali Kalhoro Mehrunisa Memon Zobia Naheed Muneer Ahmed Zengqiang Zhang 2020Journal of Environmental Sciences2020,32,1:3
5Label-free SARS-CoV-2 detection and classification using phase imaging with computational specificity显示文摘Efforts to mitigate the COVID-19 crisis revealed that fast,accurate,and scalable testing is crucial for curbing the current impact and that of future pandemics.We propose an optical method for directly imaging unlabeled viral particles and using deep learning for detection and classification.An ultrasensitive interferometric method was used to image four virus types with nanoscale optical path-length sensitivity.Pairing these data with fluorescence images for ground truth,we trained semantic segmentation models based on U-Net,a particular type of convolutional neural network.The trained network was applied to classify the viruses from the interferometric images only,containing simultaneously SARS-CoV-2,H1N1(influenza-A virus),HAdV(adenovirus),and ZIKV(Zika virus).Remarkably,due to the nanoscale sensitivity in the input data,the neural network was able to identify SARS-CoV-2 vs.the other viruses with 96%accuracy.The inference time for each image is 60 ms,on a common graphic-processing unit.This approach of directly imaging unlabeled viral particles may provide an extremely fast test,of less than a minute per patient.As the imaging instrument operates on regular glass slides,we envision this method as potentially testing on patient breath condensates.The necessary high throughput can be achieved by translating concepts from digital pathology,where a microscope can scan hundreds of slides automatically.Neha Goswami Yuchen R.He Yu-Heng Deng Chamteut Oh Nahil Sobh Enrique Valera Rashid Bashir Nahed Ismail Hyunjoon Kong Thanh H.Nguyen Catherine Best-Popescu Gabriel Popescu 2021Light(Science & Applications)2021,10,9:3
