Role of the intestinal barrier in inflammatory bowel disease显示文摘A critical function of the intestinal mucosa is to form a barrier that separates luminal contents from the interstitium. The single layer of intestinal epithelial cells (IECs) serves as a dynamic interface between the host and its environment. Cell polarity and structural properties of the epithelium is complex and is important in the development of epithelial barrier function. Epithelial cells associate with each other via a series of intercellular junctions. The apical most intercellular junctional complex referred to as the Apical Junction Complex (AJC) is important in not only cell-cell recognition, but also in the regulation of paracellular movement of fluid and solutes. Defects in the intestinal epithelial barrier function have been observed in a number of intestinal disorders such as inflammatory bowel disease (IBD). It is now becoming evident that an aberrant epithelial barrier function plays a central role in the pathophysiology of IBD. Thus, a better understanding of the intestinal epithelial barrier structure and function in healthy and disease states such as IBD will foster new ideas for the development of therapies for such chronic disorders. | Mike G Laukoetter Porfirio Nava Asma Nusrat | 2008 | World Journal of Gastroenterology2008,14,3: | 18
Ventilator associated pneumonia following liver transplantation:Etiology, risk factors and outcome显示文摘AIM: To determine the incidence, etiology, risk factors and outcome of ventilator-associated pneumonia(VAP) in patients undergoing orthotopic liver transplantation(OLT).METHODS: This retrospective study considered 242 patients undergoing deceased donor OLT. VAP was diagnosed according to clinical and microbiological criteria. RESULTS: VAP occurred in 18(7.4%) patients, with an incidence of 10 per 1000 d of mechanical ventilation(MV). Isolated bacterial etiologic agents were mainly Enterobacteriaceae(79%). Univariate logistic analysis showed that model for end-stage liver disease(MELD) score, pre-operative hospitalization, treatment with terlipressin, Child-Turcotte-Pugh score, days of MV and red cell transfusion were risk factors for VAP. Multivariateanalysis, considering significant risk factors in univariate analysis, demonstrated that pneumonia was strongly associated with terlipressin usage, pre-operative hospitalization, days of MV and red cell transfusion. Mortality rate was 22% in the VAP group vs 4% in the group without VAP. CONCLUSION: Our data suggest that VAP is an important cause of nosocomial infection during postoperative period in OLT patients. MELD score was a significant risk factor in univariate analysis. Multiple transfusions, treatment with terlipressin, preoperative hospitalization rather than called to the hospital while at home and days of MV constitute important risk factors for VAP development. | Antonio Siniscalchi Lucia Aurini Beatrice Benini Lorenzo Gamberini Stefano Nava Pierluigi Viale Stefano Faenza | 2016 | World Journal of Transplantation2016,6,2: | 9
Effect of human disturbance on the structure and regeneration of forests in the Nevado de Toluca National Park, Mexico显示文摘Sample plots were established in the principal forest types in the the Nevado de Toluca National Park, Mexico including those dominated by Pinus hartwegii, Abies religiosa, Quercus laurina and Alnus jorullensis. The vertical structure was defined by three strata in the coniferous forests and two strata in the broadleaved forests. Timber harvesting in Abies religiosa and Quercus laurina forests and fires generated by humans in Pinus hartwegii forests impeded the recruitment of saplings. Mature trees were also heavily impacted by logging in Pinus hartwegii forests. On the contrary, Alnus jorullensis forests were increasing due to the disturbance of Pinus and Quercus forests, as well abandoned crop lands within the park. A combination of logging, uncontrolled fire, and grazing appears to be compromising the recruitment of important tree species in this national park. These factors, together with human settlements, have also increased the proportion of early successional species. Changes in forest structure from human disturbance indicate a need to control these activities if conservation goals are not to be compromised. | Angel Rolando Endara Agramont Sergio Franco Maass Gabino Nava Bernal Juan Ignacio Valdez Hernández Todd S.Fredericksen | 2012 | Journal of Forestry Research2012,23,1: | 5
Alzheimer’s disease and type 2 diabetes mellitus:Pathophysiologic and pharmacotherapeutics links显示文摘At present,Alzheimer’s disease(AD)and type 2 diabetes mellitus(T2DM)are two highly prevalent disorders worldwide, especially among elderly individuals.T2DM appears to be associated with cognitive dysfunction, with a higher risk of developing neurocognitive disorders, including AD.These diseases have been observed to share various pathophysiological mechanisms, including alterations in insulin signaling, defects in glucose transporters(GLUTs),and mitochondrial dysfunctions in the brain. Therefore, the aim of this review is to summarize the current knowledge regarding the molecular mechanisms implicated in the association of these pathologies as well as recent therapeutic alternatives. In this context, the hyperphosphorylation of tau and the formation of neurofibrillary tangles have been associated with the dysfunction of the phosphatidylinositol 3-kinase and mitogen-activated protein kinase pathways in the nervous tissues as well as the decrease in the expression of GLUT-1 and GLUT-3 in the different areas of the brain, increase in reactive oxygen species, and production of mitochondrial alterations that occur in T2DM.These findings have contributed to the implementation of overlapping pharmacological interventions based on the use of insulin and antidiabetic drugs,or,more recently, azeliragon, amylin, among others, which have shown possible beneficial effects in diabetic patients diagnosed with AD. | Milagros Rojas Mervin Chávez-Castillo Jordan Bautista Ángel Ortega Manuel Nava Juan Salazar Edgar Díaz-Camargo Oscar Medina Joselyn Rojas-Quintero Valmore Bermúdez | 2021 | World Journal of Diabetes2021,12,6: | 5

Magnesium incorporated chitosan based scaffolds for tissue engineering applications显示文摘Chitosan based porous scaffolds are of great interest in biomedical applications especially in tissue engineering because of their excellent biocompatibility in vivo, controllable degradation rate and tailorable mechanical properties. This paper presents a study of the fabrication and characterization of bioactive scaffolds made of chitosan(CS),carboxymethyl chitosan(CMC)and magnesium gluconate(MgG).Scaffolds were fabricated by subsequent freezing-induced phase separation and lyophilization of polyelectrolyte complexes of CS,CMC and MgG.The scaffolds possess uniform porosity with highly interconnected pores of 50-250 μm size range.Compressive strengths up to 400 kPa,and elastic moduli up to 5 MPa were obtained. The scaffolds were found to remain intact, retaining their original threedimensional frameworks while testing in in-vitro conditions. These scaffolds exhibited no cytotoxicity to 3T3 fibroblast and osteoblast cells. These observations demonstrate the efficacy of this new approach to preparing scaffold materials suitable for tissue engineering applications. | Udhab Adhikari Nava PRijal Shalil Khanal Devdas Pai Jagannathan Sankar Narayan Bhattarai | 2016 | Bioactive Materials2016,1,2: | 2

The Potassium Transporter OsHAK5 Alters Rice Architecture via ATP-Dependent Transmembrane Auxin Fluxes显示文摘Plant HAK/KUP/KT family members function as plasma membrane(PM)H^(+)/K^(+)symporters and may modulate chemiosmotically-driven polar auxin transport(PAT).Here,we show that inactivation of OsHAK5,a rice K^(+)transporter gene, decreased rootward and shootward PAT,tiller number, and the length of both lateral roots and root hairs, while OsHAK5 overexpression increased PAT,tiller number, and root hair length, irrespective of the K^(+)supply.Inhibitors of ATP-binding-cassette type-B transporters,NPA and BUM,abolished the OsHAK5-overexpression effect on PAT.The mechanistic basis of these changes included the OsHAK5-mediated decrease of transmembrane potential(depolarization),increase of extracellular pH,and increase of PM-ATPase activity. These findings highlight the dual roles of OsHAK5 in altering cellular chemiosmotic gradients(generated continuously by PM H^(+)-ATPase)and regulating ATP-dependent auxin transport. Both functions may underlie the prominent effect of OsHAK5 on rice architecture, which may be exploited in the future to increase crop yield via genetic manipulations. | Tianyuan Yang Huimin Feng Song Zhang Huojun Xiao Qingdi Hu Guang Chen Wei Xuan Nava Moran Angus Murphy Ling Yu Guohua Xu | 2020 | Plant Communications2020,1,5: | 2

Hepatic lipid metabolism changes in short-and long-term prehepatic portal hypertensive rats显示文摘AIM: To verify the impairment of the hepatic lipid metabolism in prehepatic portal hypertension. METHODS: The concentrations of free fatty acids, diacylglycerol, triglycerides, and phospholipids were assayed by using D-[U-14C] glucose incorporation in the different lipid fractions and thin-layer chromatography and cholesterol was measured by spectrophotometry, in liver samples of Wistar rats with partial portal vein ligation at short- (1 mo) and long-term (1 year) (i.e. portal hypertensive rats) and the control rats. RESULTS: In the portal hypertensive rats, liver phospholipid synthesis significantly decreased (7.42 ± 0.50 vs 4.70 ± 0.44 nCi/g protein; P < 0.01) and was associated with an increased synthesis of free fatty acids (2.08 ± 0.14 vs 3.36 ± 0.33 nCi/g protein; P < 0.05), diacylglycerol (1.93 ± 0.2 vs 2.26 ± 0.28 nCi/g protein), triglycerides (2.40 ± 0.30 vs 4.49 ± 0.15 nCi/g protein) and cholesterol (24.28 ± 2.12 vs 57.66 ± 3.26 mg/g protein; P < 0.01). CONCLUSION: Prehepatic portal hypertension in rats impairs the liver lipid metabolism. This impairment consists in an increase in lipid deposits (triglycerides, diacylglycerol and cholesterol) in the liver, accompanied by a decrease in phospholipid synthesis. | Maria-Angeles Aller Elena Vara Cruz García Maria-Paz Nava Alejandra Angulo Fernando Sánchez-Patán Ana Calderón Patri Vergara Jaime Arias | 2006 | World Journal of Gastroenterology2006,12,42: | 2
