1Effects of Bacillus coagulans supplementation on the growth performance and gut health of broiler chickens with Clostridium perfringens-induced necrotic enteritis显示文摘Background: The poultry industry is in need of effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE) due to Clostridium perfringens.Methods: This study was conducted to investigate the effects of feeding Bacil us coagulans on the growth performance and gut health of broiler chickens with C. perfringens-induced NE. Two hundred and forty 1-day-old broiler chicks were randomly assigned to a 2 × 2 factorial arrangement with two dietary B. coagulans levels(0 or 4 × 109 CFU/kg of diet) and two disease chal enge statuses(control or NE chal enged).Results: NE-induced reduction in body weight gain was relieved by the addition of B. coagulans into broiler diets compared with the NE-infected birds. NE infection damaged intestinal morphological structure, promoted intestinal C.perfringens growth and liver invasion, and enhanced anti-C. perfringens specific sI gA concentrations in the gut and specific IgG levels in serum compared with the uninfected birds. NE infection significantly(P < 0.05) decreased mucin-2(at 14 d post-infection(DPI), tol-like receptor 2(TLR2, at 7 and 14 DPI), TLR4(at 7 and 14 DPI), tumor necrosis factor super family15(TNFSF15, at 7 and 14 DPI), lysozyme(LYZ, at 14 DPI) and fowlicidin-2(at 7 and 14 DPI) mR NA levels, whereas it dramatical y(P = 0.001) increased IFN-γ mR NA levels at 7 DPI. However, chal enged birds fed diets supplemented with B.coagulans showed a significant(P < 0.01) decrease in gut lesion scores, decreased C. perfringens numbers in the cecum and liver, and an increase in fowlicidin-2 mR NA levels in compared with the uninfected birds. 2Kinetic study of phytotoxicity induced by foliar lead uptake for vegetables exposed to fine particles and implications for sustainable urban agriculture显示文摘At the global scale, foliar metal transfer occurs for consumed vegetables cultivated in numerous urban or industrial areas with a polluted atmosphere. However, the kinetics of metal uptake, translocation and involved phytotoxicity was never jointly studied with vegetables exposed to micronic and sub-micronic particles(PM). Different leafy vegetables(lettuces and cabbages) cultivated in RHIZOtest? devices were, therefore, exposed in a greenhouse for 5, 10 and 15 days to various Pb O PM doses. The kinetics of transfer and phytotoxicity was assessed in relation to lead concentration and exposure duration. A significant Pb accumulation in leaves(up to 7392 mg/kg dry weight(DW) in lettuce) with translocation to roots was observed. Lead foliar exposure resulted in significant phytotoxicity, lipid composition change, a decrease of plant shoot growth(up to 68.2% in lettuce) and net photosynthesis(up to 58% in lettuce). The phytotoxicity results indicated plant adaptation to Pb and a higher sensitivity of lettuce in comparison with cabbage. 4反相高效液相色谱法测定有机氯及含氮杀虫剂在水体·土壤沉积物及鱼体中的残留(摘要)(英文)显示文摘[目的]测定水体、土壤沉积物以及鱼体内有机氯及含氮杀虫剂残留物,为科学评估杀虫剂残留对巴基斯坦地区公共卫生、农业和环境的影响提供依据。[方法]以食用鱼南亚野鲮为材料,设计2组体重,采用高效液相色谱法分别测定水体、土壤沉积物以及南亚野鲮体内α-硫丹、DDE、甲基对硫磷、异丙隆、呋喃丹、阿特拉津等含量。[结果]土壤沉积物中DDE的含量达(2.340±0.025)μg/g,在250 ~750 g的南亚野鲮体内的含量分别为(0.270 ±0.000 6)μg/g,但在水体中未发现DDE残留;不同饲料中农药残留物90%为有机磷、呋喃丹以及有机氯杀虫剂,6%为杀菌剂,仅4%为除草剂,在250 ~750 g以及800 ~1 300 g的南亚野鲮体内硫丹的含量分别达到(0.491±0.000 6)μg/g和(3.050±0.060 8)μg/g,异丙隆的含量分别达到(0.010±0.0003)μg/g和(0.014±0.000 6)μg/g,且随体重上升,其脂肪含量增加,积累的农药残留物则越多;硫丹、甲基对硫磷、阿特拉津和呋喃丹的含量在水体达到最大残留限量水平0.001μg/g。[结论]通过生物积累和在自然界中的运输以及再沉积作用,有机氯及其他杀虫剂的使用给全球的环境造成了严重污染,因此,在巴基斯坦已经禁止使用DDT等有机氯农药。Shahid Mahboob Ghazala S.Sultana Muhammad Rafique Asi S.Nadeem A.S.Chaudhry 2009Agricultural Science & Technology2009,10,5:9
5Vegetation-environment relationships in the forests of Chitral district Hindukush range of Pakistan显示文摘We investigated the composition of plant communities to quantify their relationships with environmental parameters in the Chitral Hindukush range of Pakistan. We sampled tree vegetation using the Point Centered Quarter (PCQ) method while understory vegetation was sampled in 1.5-m circular quadrats. Cedrus deodara is the national symbol of Pakistan and was dominant in the sampled communities. Because environmental variables determine vegetation types, we analyzed and evaluated edaphic and topographic factors. DCA-Ordination showed the major gradient as an amalgam of elevation (p<0.05) and slope (p<0.01) as the topographic factors correlated with species distribution. Soil variables were the factors of environmental significance along DCA axes. However, among these factors, Mg2+ , K + and N2+ contributed not more than 0.054% 0.20% and 0.073%, respectively, to variation along the first ordination axis. We conclude that the principal reason for weak or no correlation with many edaphic variables was the anthropogenic disturbance of vegetation. The understory vegetation was composed of perennial herbs in most communities and was most dense under the tree canopy. The understory vegetation strongly regulates tree seedling growth and regeneration patterns. 6筛选和转化药用野生稻TAC克隆获得耐旱水稻显示文摘【目的】利用分子生物学技术建立一种可有效转移药用野生稻有利基因的方法,为进一步大量开展利用药用野生稻有利基因提供技术基础。【方法】以植物抗性相关转录因子AP2/EREBP和bZIP家族基因保守序列为探针,采用同位素α-32P对探针进行标记,通过Southern杂交对药用野生稻TAC文库进行第一轮筛选;所获得阳性克隆用地高辛标记的探针进行第二轮筛选;最后利用PCR方法对第二轮筛选所获得的阳性克隆进行验证。通过农杆菌介导法将阳性克隆转入栽培稻品种日本晴,利用TAC载体插入片段两端的抗性标记Hpt和SacB,通过PCR扩增检测和和Southern杂交对转化植株进行鉴定。采用PEG胁迫法和生理指标变化对转基因T2植株的芽期、苗期耐旱性进行鉴定。【结果】第一轮TAC文库筛选共获得1 073个阳性克隆,其中,以AP2/EREBP的保守序列为探针筛选出606个、bZIP为探针筛选出467个。对这些阳性克隆进行第二轮筛选,从中共获得147个阳性克隆,其中,AP2/EREBP为探针筛选出95个、bZIP为探针筛选出52个。PCR验证有103个阳性克隆能够扩增出目的片段,分别是利用AP2/EREBP设计的引物筛选出63个克隆和bZIP的40个克隆,阳性克隆得率分别为66.32%和76.92%。通过农杆菌转化,以编号为52D-M16和22D-P2(与AP2/EREBP相关)以及55R-A17、49R-O14和8-A24(与bZIP相关)的克隆转化水稻日本晴均获得转化植株,转化试验结果发现外源插入片段越大则克隆转化的成苗率越低。抗性鉴定、PCR和Southern杂交结果均证明外源片段已转入受体水稻品种日本晴中。芽期抗旱性鉴定表明R12-23、R15-41和R1-8 3份材料的耐旱性优于受体日本晴,苗期鉴定显示M63-9和R12-23的耐旱性较强。综合认为,R12-23具有较好的耐旱性。【结论】通过构建药用野生稻TAC文库并结合特异克隆筛选和遗传转化方法,可以进行有利基因的转移,结合进一步的育种,有望实现药用野生稻在育种上的利用。刘蕊 张欢欢 陈志雄 SHAHID Muhammad Qasim 傅雪琳 刘耀光 刘向东 卢永根 2014中国农业科学2014,47,8:8

8基于ETA模型的配筋措施对于高拱坝变形损伤指标的影响显示文摘为了评价不同强度地震作用下,配筋措施对混凝土拱坝抗震性能增强的效果,基于耐震时程分析(ETA)法,研究了不同地震强度下,高拱坝是否采用配筋抗震措施对坝体损伤变形指标的影响。首先,介绍了ETA方法、坝体材料本构、横缝模型及地震动输入方法;然后,分别建立高拱坝采用配筋抗震措施和未采用配筋抗震措施拱坝的数值模型,并基于ETA方法生成不同地震强度的耐震时程。分析在不同地震强度下,高拱坝是否采用配筋抗震措施的损伤分布、横缝开度及变形等响应,提出高拱坝损伤体积比、平均横缝开度及拱冠梁变形等坝体损伤变形指标。通过对比高拱坝配筋和未配筋的情况下得出的动力响应指标,研究配筋措施对高拱坝响应及损伤变形指标趋势、相关性及离散性的影响。计算结果表明:在不同的地震动强度条件下,配筋措施对减小高拱坝的整体损伤是有效的,对横缝开度及位移影响有限,配筋措施可以减小地震随机性导致的损伤变形指标离散。同时,归一的损伤变形指标表明损伤变形指标有一定的相关性。徐强 张天然 陈健云 SHAHID Muhammad 李静 2021工程科学与技术2021,53,3:6
9Strategies on Sample Size Determination and Qualitative and Quantitative Traits Integration to Construct Core Collection of Rice (Oryza sativa)显示文摘The development of a core collection could enhance the utilization of germplasm collections in crop improvement programs and simplify their management. Selection of an appropriate sampling strategy is an important prerequisite to construct a core collection with appropriate size in order to adequately represent the genetic spectrum and maximally capture the genetic diversity in available crop collections. The present study was initiated to construct nested core collections to determine the appropriate sample size to represent the genetic diversity of rice landrace collection based on 15 quantitative traits and 34 qualitative traits of 2 262 rice accessions. The results showed that 50-225 nested core collections, whose sampling rate was 2.2%-9.9%, were sufficient to maintain the maximum genetic diversity of the initial collections. Of these, 150 accessions (6.6%) could capture the maximal genetic diversity of the initial collection. Three data types, i.e. qualitative traits (QT1), quantitative traits (QT2) and integrated qualitative and quantitative traits (QTT), were compared for their efficiency in constructing core collections based on the weighted pair-group average method combined with stepwise clustering and preferred sampling on adjusted Euclidean distances. Every combining scheme constructed eight rice core collections (225, 200, 175, 150, 125, 100, 75 and 50). The results showed that the QTT data was the best in constructing a core collection as indicated by the genetic diversity of core collections. A core collection constructed only on the information of QT1 could not represent the initial collection effectively. 11Transcranial magnetic stimulation in animal models of neurodegeneration显示文摘Brain stimulation techniques offer powerful means of modulating the physiology of specific neural structures. In recent years, non-invasive brain stimulation techniques, such as transcranial magnetic stimulation(TMS) and transcranial direct current stimulation, have emerged as therapeutic tools for neurology and neuroscience. However, the possible repercussions of these techniques remain unclear, and there are few reports on the incisive recovery mechanisms through brain stimulation. Although several studies have recommended the use of non-invasive brain stimulation in clinical neuroscience, with a special emphasis on TMS, the suggested mechanisms of action have not been confirmed directly at the neural level. Insights into the neural mechanisms of non-invasive brain stimulation would unveil the strategies necessary to enhance the safety and efficacy of this progressive approach. Therefore, animal studies investigating the mechanisms of TMSinduced recovery at the neural level are crucial for the elaboration of non-invasive brain stimulation. Translational research done using animal models has several advantages and is able to investigate knowledge gaps by directly targeting neuronal levels. 12草甘膦对转基因抗草甘膦大豆的安全性研究显示文摘采用草甘膦种子和茎叶处理研究转基因抗草甘膦大豆的安全性,为减少草甘膦用量和转基因抗草甘膦大豆的安全应用提供理论依据。以转基因抗草甘膦大豆为试材,采用田间随机区组设计方法,测定转基因大豆的生理指标和杂草防除效果。结果表明,562. 5 g. a. i./hm^2草甘膦种子处理和1 125 g. a. i./hm^2草甘膦茎叶处理后,除莽草酸含量不受影响,转基因大豆的株高、鲜质量、干质量、叶绿素含量以及光合速率各项生理指标均在施药初期受到抑制,茎叶处理后28 d、种子处理播种后61 d时各生理指标均可恢复正常;草甘膦茎叶处理对杂草具有显著的防除效果,草甘膦施用后7 d,杂草防效为94. 67%,28 d,杂草防效为72. 33%,且对大豆安全。草甘膦种子和茎叶处理对转基因抗草甘膦大豆均具有较好的安全性,且茎叶处理能有效控制杂草。陈银竹 丁伟 刘胜男 Muhammad Shahid khan 2018江苏农业科学2018,46,16:4
13Gastric juice for the diagnosis of H pylori infection in patients on proton pump inhibitors显示文摘AIM: To determine the efficacy of gastric juice polymerase chain reaction (PCR) for the detection of H pylori infection in comparison with histology and gastric antral biopsy PCR in patients on a proton pump inhibitor (PPI). METHODS: Eighty-five consecutive patients with dyspeptic symptoms were enrolled. Gastric biopsies for histology, PCR and gastric juice were collected at endoscopy for PCR of the H pylori urease C gene (ure C). Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), accuracy, positive and negative likelihood ratio for PCR of gastric juice for the H pylori ure C gene was compared to histology and gastric antral biopsy H pylori ure C PCR in patients with and without PPI. RESULTS: Gastric juice PCR was positive in 66 (78%) patients. Histology showed H pylori associated gastritis in 57 (67%). Gastric biopsy PCR was positive in 72 (85%). In patients not taking PPI, the sensitivity, specificity, PPV, NPV, accuracy and positive and negative likelihood ratio for gastric juice PCR were 89%, 72%, 91%, 67%, 90%, 85%, 3.1 and 0.1 respectively. In patients on PPI these values were 86%, 100%%, 100%, 29%, 86%, 9.5 and 1.4, respectively. CONCLUSION: Gastric juice PCR for the diagnosis of H pylori infection has increased sensitivity compared to histology with PPI. 15Phyto-Toxicity of Chromium in Maize: Oxidative Damage, Osmolyte Accumulation, Anti-Oxidative Defense and Chromium Uptake显示文摘Agricultural production systems are immensely exposed to different environmental stresses in which heavy metal stress receives serious concerns. This study was conducted to explore the deleterious effects of different chromium(Cr) stress levels, i.e., 0, 30, 6090, 120, and 150 μmol L-1, on two maize genotypes, Wandan 13 and Runnong 35. Both genotypes were evaluated by measuring their growth and yield characteristics, Cr accumulation in different plant tissues, alterations in osmolyte accumulation, generation o reactive oxygen species(ROS), and anti-oxidative enzyme activity to scavenge ROS. The results showed that Cr stress decreased the leaf area, cob formation, 100-grain weight, shoot fresh biomass, and yield formation, while Cr accumulation in different maize tissues was found in the order of roots > leaves > stem > seeds in both genotypes. The increased Cr toxicity resulted in higher free proline soluble sugars and total phenolic contents, and lower soluble protein contents. However, enhanced lipid peroxidation was noticed in the forms of malondialdehyde, hydrogen peroxide(H2O2) and thiobarbituric acid reactive substance accumulation, and electrolyte leakage. The hyperactivity of superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, especially glutathione peroxidase and glutathione reductase indicated that these anti-oxidative enzymes had a central role in protecting maize from Cr toxicity, especially for Wandan 13. 17Boron Application Improves Growth,Yield and Net Economic Return of Rice显示文摘A field trial was conducted to evaluate the role of boron (B) application at different growth stages in improving the growth,yield and net economic return of rice at farmer's fields during summer season,2009.Boron was soil applied (1.5 kg/hm2) at the transplanting,tillering,flowering and grain formation stages of rice;foliar applied (1.5% B solution) at the tillering,flowering and grain formation stages of rice,and dipped seedling roots in 1.5% B solution before transplanting;while control plots did not apply any B.Boron application (except dipping of seedling roots in B solution,which caused toxicity and reduced the number of tillers and straw yield than control) substantially improved the rice growth and yield.However,soil application was better in improving the number of grains per panicle,1000-grain weight,grain yield,harvest index,net economic income and ratio of benefit to cost compared with the rest of treatments.Overall,for improving rice performance and maximizing the net economic returns,B might be applied as soil application at flowering.Mubshar HUSSAIN M.Ayaz KHAN M.Bismillah KHAN Muhammad FAROOQ Shahid FAROOQ 2012Rice science2012,19,3:3

20Identification of an S5^n allele in Oryza rufipogon Griff. and its effect on embryo sac fertility显示文摘Oryza rufipogon exhibits abundant genetic diversity and contains many elite genes. In this work, a total of 441 accessions of O. rufipogon were genetically examined using the functional marker S5n, revealing that 18 accessions carried the S5n allele with a heterozygous genotype (S5nS5i/j). Of these accessions, 14 were from Guangdong Province, 2 from Guangxi Zhuang Autonomous Region, 1 from Hainan Province and 1 from Jiangxi Province. Further sequence analysis spanning both sides of the deletion in S5n allele of O. rufipogon confirmed that this allele is identical with that of 02428, a wide-compatibility variety of domesticated rice. The segregation ratio of the 3 genotypes (S5iS5i/S5jS5j:S5nS5i/j:S5nS5n) among 15 self-bred progenies of O. rufipogon significantly deviated from the Mendelian expectation (1:2:1) in that the proportion of plants exhibiting S5n heterozygous and homozygous genotypes was much lower in the progeny. Sequence analysis of full-length S5n alleles indicated that a few base pairs were different between O. rufipogon and O. sativa, and it was presumed that S5n may be an ancient allele.
