4Action of aluminum on high voltage-dependent calcium current and its modulation by ginkgolide B显示文摘Aim: To investigate the effect of aluminum (Al) on high voltage-dependent calcium current (IHVA) and its modulation by ginkgolide B (Gin B). Methods: The whole-cell, patch-clamp technique was used to record IHVA from acutely isolated hippocampal CA1 pyramydal neurons in rats. Results: A1 0.1mmol/L (low concentration) reduced IHVA; Al 0.75 and 1.0mmol/L (high concentrations) increased IHVA, and Al decreased and increased IHVA at intermediate concentrations of 0.25 and 0.5mmol/L. The increase of IHVA by Al 1.0mmol/L was enhanced by the adenylyl cyclase (AC) agonist forskolin and was partly abolished by the cyclic adenosine monophosphate (cAMP)-dependent protein kinase A (PKA) antagonist H-89, whereas the decrease observed with Al 0.1mmol/L was neither reversed by forskolin nor affected by H-89. Gin B had no effect on IHVA in normal neurons,but canceled the increase in IHVA by 1.0mmol/L Al. Conclusion: The results indicate that the mechanism of Al affecting IHVA differs at different concentrations,and this may be attributed to its complex actions. 5Thermal influence of ponding and buried warm-oil pipelines on permafrost: a case study of the China-Russia Crude Oil Pipeline显示文摘Buried pipelines are widely used for transporting oil in remote cold regions. However, the warm oil can induce considerable thermal influence on the surrounding frozen soils and result in severe maintenance problems. This paper presents a case study of the thermal influence of ponding and buried warm-oil pipelines on permafrost along the China-Russia Crude Oil Pipeline(CRCOP) in Northeast China. Since its operation in 2011, the operation of the warm-oil pipelines has led to rapid warming and thawing of the surrounding permafrost and development of sizable ponding along the pipeline route,which, in return, exacerbates the permafrost degradation. A field study was conducted along a 400-km long segment of the CRCOP in permafrost regions of Northeast China to collect the location and size information of ponding. A two-dimensional heat transfer model coupled with phase change was established to analyze the thermal influence of ponding and the operation of warm-oil pipelines on the surrounding permafrost. In-situ measured ground temperatures from a monitoring site were obtained to validate the numerical model. The simulation results show that ponding accelerates the development of the thaw bulb around the pipeline. The maximum thaw depth below the pipeline increases from 4 m for the case without ponding to 9 m for the case with ponding after 50 years of operation, and ponding directly above the pipe induces the maximum thaw depth. 6Deformation behavior of in situ permafrost on the Qinghai-Tibetan Plateau显示文摘Creep is an important mechanical behavior of frozen soils, one which can cause many problems for the infrastructures in permafrost regions on the Qinghai-Tibetan Plateau. To access the natural creep properties of in situ permafrost for explaining the engineering instability and predicting long-term deformation, conducting field tests is necessary. The paper reports on a group of plate loading tests we carried out over many years on the Qinghai-Tibetan Plateau. The results show that the ground temperature at the loading plates ranged from-0.29 °C to-3.03 °C, and the mean annual ground temperature increased year by year in a linear fashion. Affected by the ground-temperature variations, two forms of deformation curves, a step-form and a wave-form occurred, depending on the relative extent of settlement in warm seasons and frost heave in cold seasons. Overall, the deformations of permafrost were characterized by settlement. The deformation magnitudes and curve styles of permafrost are different at different locations attributing to the influence of ground temperature and moisture content. Due to the existence of much unfrozen water in warm frozen soils, consolidation resulting from migration of unfrozen water along seepage channels may play a significant role in the settlement of permafrost. 10Muscarinic cholinergic regulation of L-type calcium channel in heart of embryonic mice at different developmental stages显示文摘AIM: To investigate the muscarinic regulation of L-type calcium current (ICa-L) during development. METHODS:The whole cell patch-clamp technique was used to record ICa-L in mice embryonic cardiomyocytes at different stages (the early developmental stage, EDS; the intermediate developmental stage, IDS; and the late developmental stage, LDS). Carbachol (CCh) was used to stimulate M-receptor in the embryonic cardiomyocytes of mice.RESULTS: The expression of lCa.L density did not change in different developmental stages (P>0.05). There was no difference in the sensitivity of ICa-L to CCh during development (P>0.05). This inhibitory action of CCh was mediated by inhibition of cyclic AMP since 8-bromo-cAMP completely reversed the muscarinic inhibitory action. IBMX, a non-selective inhibitor of phosphodiesterase (PDE), reversed the inhibitory action of M-receptor on ICa-L current by 71.2 %±9.2% (n=8) and 11.3%±2.5% (n=9) in EDS and LDS respectively. However forskolin, an agonist of adenylyl cyclase (AC), reversed the action of CCh by 14.5%±3.5% (n=5) and 82.7%± 10.4% (n=7) in EDS and LDS respectively. CONCLUSION: The inhibitory action of CCh on lca.L current was mediated in different pathways: in EDS, the inhibitory action of M-receptor on ICa-L channel mainly depended on the stimulation of PDE. 12Developmental changes in functional expression andβ-adrenergic regulation of If in the heart of mouse embryo显示文摘The hyperpolarization-activated current (If) plays an important role in determining the spontaneousrate of cardiac pacemaker cells. The automatic rhythmicity also exists in working cells of embryonic heart,therefore we studied developmental changes in functional expression and β-adrenergic regulation of If inembryonic mouse heart. The expression of If is high in early developmental stage (EDS) (10.5 d after coitus)ventricular myocytes, low in intermediate developmental stage (IDS) (13.5 d) atrial or ventricular myocytesand even lower in late developmental stage (LDS) (16.5 d) atrial or ventricular myocytes, indicating thatthese cells of the EDS embryonic heart have some properties of pacemaker cells. β-adrenergic agonistisoproterenol (ISO) stimulates If in LDS but not in EDS cardiomyocytes, indicating that theβ-adrenergicregulation of If is not mature in EDS embryonic heart. But forskolin (a direct activator of adenylate cyclase)and 8-Br-cAMP (a membrane-permeable analogue of cAMP) increase the amplitude of If in EDS cells,indicating that adenylate cyclase and cAMP function fairly well at early stage of development. 18Relationship between ponding and topographic factors along the China-Russia Crude Oil Pipeline in permafrost regions显示文摘The original landform along the China Russia Crude Oil Pipeline(CRCOP,line 2)was disturbed during installation of pavement for the pipeline.Forest and vegetation coverage is dense,and runoff develops along the pipe.Since the opera tion of the CRCOP(line 2)began in 2018,ponding has appeared on both sides of the pipeline.If there is no drainage,ponding can hardly dissipate,due to the low permeability of the permafrost layer.With the supply of surface flow and the transportation of oil at positive temperatures,ponding promotes an increase in temperature and changes the boundary ther mal conditions of the pipeline.Meanwhile,when the ponding freezes and thaws,frost heave threatens operational safety of the pipeline.Furthermore,the ponding can affect the thermal condition of line 1.In this paper,the distribution of pond ing along the CRCOP was obtained by field investigation.The type and cause of ponding were summarized,and the catch ment and stream order were extracted by the Digital Elevation Model(DEM).According to the statistical results in attri butes for topographic factors,it is known that ponding along the pipeline is relative to elevation,slope,aspect,and the Topographic Wetness Index(TWI).Water easily accumulates at altitudes of 300450 m,slopes within 3°5°,aspect in the northeast or south,TWI within 1316,flow direction in north east south,and flow length within 90150 km.This paper proposes a theoretical basis for the cause and characteristics of ponding along the pipeline.MingTang Chai YanHu Mu GuoYu Li Wei Ma Fei Wang 2019Research in Cold and Arid Regions2019,11,6:0
19Boosting the dimensionality of frequency entanglement using a reconfigurable microring resonator显示文摘Integrated quantum frequency combs(QFCs)based on microring resonators supplies as an essential resource for expanding the Hilbert-space dimensionality for high-dimensional quantum computing and information processing.In this work,we propose and demonstrate a reconfigurable ring resonator with tunable quality factors to efficiently increase the dimensionality of frequency entanglement,simultaneously,ensuring a high on-chip pair generation rate(PGR)and coincidence-to-accidental ratio(CAR).Our method exploits the asymmetric Mach-Zehnder interferometer instead of the traditional straight waveguide as the coupler of resonators which offer a tunable external coupling coefficient to modulate the quality factor to enlarge the QFCs’bandwidth and thus increase the dimensionality of frequency entanglement.We measured the QFCs’joint spectral intensity of 28 frequency pairs under various quality factors ranging from 16.6×10^(4) to 3.4×10^(4).Meanwhile,the measured Schmidt number increased from 11.01 to 24.77,denoting a huge expansion of the Hilbert-space dimensionality from 121 to a record number of 613 dimensions,which agrees well with our theoretical calculations.In addition,the PGR and CAR-another two key parameters for high-quality QFCs-were all measured under different quality factors to verify that our method can significantly increase the Schmidt number and CAR while maintaining a high PGR.In fact,bright QFCs with a total PGR of 4.3 MHz under a 0.48 mW pump power and a mean CAR of 1578 were simultaneously obtained at the highest Schmidt number.This method is widely applicable to other material-based ring resonators and can act as a general solution for high-dimensional QFCs.Chao Wu Qilin Zheng Yingwen Liu Yang Wang Jiangfang Ding Pingyu Zhu Shichuan Xue Miaomiao Yu Weihong Luo Kaikai Zhang Anqi Huang Mingtang Deng Junjie Wu Ping Xu 2023Science China(Physics,Mechanics & Astronomy)2023,66,5:0
20Seepage influence of supra-permafrost groundwater on thermal field of embankment on Qinghai-Tibet Plateau,China显示文摘As a unique hydro-geological phenomenon in permafrost regions,the seepage of supra-permafrost groundwater will carry a large amount of heat and cause differential settlement in the embankment.This paper presents the results of a field study monitoring the supra-permafrost groundwater levels on both sides of an embankment in permafrost regions.It describes a two-dimensional coupled hydro-thermal model and uses it to analyze the influence of seepage on its temperature field considering climate warming.The results show that seepage exacerbates permafrost thawing and thickens the active layer.The thermal influence on the sunny side of the embankment toe is more significant than that on the shady side,which will cause differential settlement in the embankment.After 50 years of operation,the embankment soil temperature with seepage during freezing is 0.2C warmer than that without seepage,and the thermal influence diminished with the increase in depth.Additionally,seepage influences the thermal regime in vertical and horizontal directions of the embankment.During freezing seasons,the thaw depth increases,and the horizontal thaw range decreases.During thawing seasons,the thaw range grows both vertically and horizontally.MingTang Chai Yuan Luo Yu Gao Wei Ma YanHu Mu 2023Research in Cold and Arid Regions2023,15,3:0
