142,573 cases of hepatectomy in China: a multicenter retrospective investigation显示文摘Hepatectomy is currently routinely performed in most hospitals in China. China owns the largest population of liver diseases and the biggest number of liver resection cases. A nationwide multicenter retrospective investigation involving 112 hospitals was performed, and focused on liver resection for patients with hepatocellular carcinoma(HCC). 42,573 cases of hepatectomy were enrolled, and 18,275 valid cases of liver resection for HCC patients were selected for statistical analysis. The epidemiology of HCC, distribution of hepatectomy, postoperative complications and prognosis were finally analyzed. In the 18,275 HCC patients,81% had hepatitis B virus infection and 10% had hepatitis C virus infection. 38% of the HCC patients had normal Alphafetoprotein(AFP) level, and other 35% had an AFP level lower than 400 ng mL^(-1). In the study period, 97% of the hepatectomy for HCC were treated with open surgery, and 23.81% had vascular exclusion techniques. The operation time was(191.7±105.6) min,the blood loss was(546.0±562.8) m L, and blood transfusion was(543.0±1,035.2) m L. The median survival for HCC patients was 631 days, with 1-, 3-, and 5-year overall survival of 73.2%, 28.8% and 19.6%, respectively. 2The beneficial effects of intermittent fasting:an update on mechanism,and the role of circadian rhythm and gut microbiota显示文摘Importance:There is accumulating evidence that intermittent fasting(IF)is connected to improved health condition and longevity time-restricted feeding(TRF)is the most recognized and extensively studied model of IF.Objective:To investigate the underlying mechanism of pleiotropic benefits of IF and hint the most advantageous feeding pattern for humans.Evidence review:We searched MEDLINE,EMBASE,Cochrane Library and Google Scholar by 2020 April for publications on IF or TRF and their mechanisms.Studies include animal models and human cohorts.Findings:One important mechanism is that IF allows certain period of fasting time,in which our bodies activate pathways of repair and rejuvenation.Moreover,the advantages of IF,especially TRF over total caloric restriction(CR)provided bases for various animal and human studies which suggested that the feeding-fasting rhythm stimulates the fluctuation of our gut microbiota and a series of subsequent molecular alterations,which in turn restored a healthier circadian clock that resembled our inherent clock formed throughout millions of years of homo sapiens history.Conclusions and Relevance for Reviews:Complete understanding of the mechanism leading to the beneficial effects of IF paves the way for tailored dietary regimen to combat a wide range of diseases and ill health conditions.Dandan Hu Zhibo Xie Yuqian Ye Suhad Bahijri Minshan Chen 2020Hepatobiliary Surgery and Nutrition2020,9,5:5
3Nonlinear Optimization of Orthotropic Steel Deck System Based on Response Surface Methodology显示文摘The steel bridge deck system,directly subjected to the vehicle load,is an important component to be considered in the optimization design of the bridges.Due to its complex structure,the design parameters are coupled with each other,and many fatigue details in the system result in time-consuming calculation during structure optimization.In view of this,a nonlinear optimization method based on the response surface methodology(RSM)is proposed in this study to simplify the design process and to reduce the amount of calculations during optimization.The optimization design of the steel bridge deck system with two-layer pavement on the top of the steel deck plate is taken as an example,the influence of eight structural parameters is considered.The Box-Behnken design is used to construct a sample space in which the eight structural parameters can be distributed evenly to reduce the calculation workload.The finite element method is used to model the mechanical responses of the steel bridge deck system.From the regression analysis by the RSM,the explicit relationships between the fatigue details and the design parameters can be obtained,based on which the nonlinear optimization design of the bridge deck system is conducted.The influence of constraint functions,objective functions,and optimization algorithms is also analyzed.The method proposed in this study is capable of considering the influence of different structural parameters and different optimization objectives according to the actual needs,which will effectively simplify the optimization design of the steel bridge deck system.Wei Huang Minshan Pei Xiaodong Liu Chuang Yan Ya Wei 2020Research2020,,1:4

8Development of a rapid GC-FID method to simultaneously determine triethylamine, diisopropylamine, and 1,1,3,3-tetramethylguanidine residues in an active pharmaceutical ingredient显示文摘A rapid GC-FID method was developed to simultaneously determine residual levels of triethylamine(TEA), 1,1,3,3-tetramethylguanidine(TMG), and diisopropylamine(DIPA) in the synthetic route of an active pharmaceutical ingredient(API). Due to the severe absorption of amines on GC stationary phases,GC columns with various stationary phases were evaluated for optimal peak shape and reproducibility.The final conditions used the Agilent CP-Volamine column to resolve the three amines in 12 min. Various inlet liners were also screened to further improve the sensitivity of the analysis. The Restek Siltek~? liner was selected to achieve the desired detectability for the method. The quantitation limits were 4, 3, and 4 mg/mL for TEA, DIPA, and TMG in the presence of API, respectively. All three amines showed good linearity(r > 0.999) and recoveries(> 90%) over the concentration range of 3 to 16 mg/mL. The testing of residual amines was initially performed at the penultimate stage of the synthesis. However, this work demonstrates that TMG can act as a proton sponge to react with salicylic acid, the counter ion of the penultimate, to form a volatile component that elutes at a different retention time. Consequently, in the final method, these three amines were monitored in the final API to circumvent the matrix interference.Key parameters of the method were qualified per method validation requirements in ICH guidelines. 15Maternal high salt-diet increases offspring's blood pressure with dysfunction of NO/PKGI signaling pathway in heart tissue显示文摘Background:High salt-diets have become significant threats to human health,resulting in hypertension and cardiovascular diseases.Hypertensive disorders during pregnancy are complicated,since the maternal cardiovascular system undergoes extensive physiological changes during pregnancy.High-salt diets during pregnancy can disturb the intrauterine environment and negatively affect fetal development.Therefore,we explored how high-salt diets during pregnancy could affect the offspring.Methods:Rats were divided into three groups and fed with low,normal,and high salt diets.The offspring were separated into three groups after weaning based on dietary salt concentration.The blood pressure and urine protein content of both dams and offspring were measured.To evaluate cardiac function,we used Masson staining and immunodetection to confirm the fibrosis status.Finally,we extracted protein from cardiac tissue to test the expression levels of the Nitric Oxide(NO)/cGMP-dependent protein kinase I(PKGI)pathway and the angiotensin receptor.Results:High-salt diets increased blood pressure,and offspring previously exposed to high-salt environments were predisposed to hypertension.High-salt diets were also found to induce cardiac fibrosis and exacerbate fibrosis in offspring and alter the epithelial-mesenchymal transition(EMT).Under these conditions,the NO/PKGI pathway was activated in cardiac tissue and the type-1angiotensin II receptor(AT1R)was upregulated,though the type-2 angiotensin II receptor(AT2R)had the opposite effect.Conclusion:High-salt diets induce high blood pressure and increase predisposition to hypertension in offspring.They are accompanied by cardiac fibrosis,which could be caused by the activation of NO/PKGI and upregulation of AT1R.Minshan Huang Xiuying Li Luwen Ren Lin Huang Jiahong Pan Jinlin Yao Lili Du Dunjin Chen Jingsi Chen 2022Gynecology and Obstetrics Clinical Medicine2022,2,2:1

18Disulfiram enhances the antitumor activity of cisplatin by inhibiting the Fanconi anemia repair pathway显示文摘A series of chemotherapeutic drugs that induce DNA damage,such as cisplatin(DDP),are standard clinical treatments for ovarian cancer,testicular cancer,and other diseases that lack effective targeted drug therapy.Drug resistance is one of the main factors limiting their application.Sensitizers can overcome the drug resistance of tumor cells,thereby enhancing the antitumor activity of chemotherapeutic drugs.In this study,we aimed to identify marketable drugs that could be potential chemotherapy sensitizers and explore the underlying mechanisms.We found that the alcohol withdrawal drug disulfiram(DSF)could significantly enhance the antitumor activity of DDP.JC-1 staining,propidium iodide(PI)staining,and western blotting confirmed that the combination of DSF and DDP could enhance the apoptosis of tumor cells.Subsequent RNA sequencing combined with Gene Set Enrichment Analysis(GSEA)pathway enrichment analysis and cell biology studies such as immunofluorescence suggested an underlying mechanism:DSF makes cells more vulnerable to DNA damage by inhibiting the Fanconi anemia(FA)repair pathway,exerting a sensitizing effect to DNA damaging agents including platinum chemotherapy drugs.Thus,our study illustrated the potential mechanism of action of DSF in enhancing the antitumor effect of DDP.This might provide an effective and safe solution for combating DDP resistance in clinical treatment.Meng YUAN Qian WU Mingyang ZHANG Minshan LAI Wenbo CHEN Jianfeng YANG Li JIANG Ji CAO 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,3:1
