1N末端B型利钠肽原对射血分数保留的心力衰竭患者全因死亡的预测价值显示文摘目的 探讨入院基线N末端B型利钠肽原(NT-proBNP)对射血分数保留的心力衰竭(HFpEF)患者全因死亡的预测价值.方法 入选中国住院患者心力衰竭注册研究(CN-HF)中符合HFpEF诊断标准的患者,根据随访期间是否死亡分为死亡组和存活组.从CN-HF中获得研究对象的人口学信息、入院时体格检查信息、入院首次超声心动图检查结果、实验室检查结果、合并症情况、用药情况和临床结局等资料.通过单因素Cox回归模型对可能预测预后的变量进行筛选,将单因素分析中与全因死亡有统计学意义的协变量纳入多因素Cox回归模型,进而分析基线NT-proBNP对全因死亡的预测价值.采用Spearman相关分析分析NT-proBNP与估算的肾小球滤过率(eGFR)的关系,并进一步探讨肾功能不全与NT-proBNP预测全因死亡的交互作用.鉴于NT-proBNP不服从二元正态分布,本研究中NT-proBNP作连续变量分析时均取自然对数(LnNT-proBNP).结果 共1 846例患者纳入本研究,平均年龄71.5岁,男性1 017例(55.1%),NT-proBNP中位数860 ng/L,eGFR中位数73.9 ml·min^-1· 1.73m^-2.本研究中位随访时间34(24~42)个月,随访期间全因死亡213例(11.5%)被纳入死亡组,存活1 633例(88.5%)被纳入存活组.与存活组比较,死亡组患者年龄较大,纽约心脏协会(NYHA)心功能Ⅲ~Ⅳ级者比例较高,住院时间较长,血钾、NT-proBNP较高,合并糖尿病、心律失常、心房颤动者较多,服用血管紧张Ⅱ受体阻滞剂(ARB)、盐皮质激素受体拮抗剂(MRA)、利尿剂和地高辛者较多(P均<0.05).与存活组比较,死亡组患者体重指数(BMI)、舒张压、左心室射血分数(LVEF)较低,血红蛋白、血清总胆固醇(TC)、血清甘油三酯(TG)、eGFR较低,服用血管紧张素转化酶抑制剂(ACEI)、他汀类药物和阿司匹林者较少(P均<0.05).单因素Cox回归分析结果显示NT-proBNP是HFpEF患者全因死亡的预测因素(HR=2.522,95 %CI 2.040~3.119,P<0.001).多因素Cox回归分析结果显示,校正了年龄、BMI、舒张压、LVEF、血红蛋白、血钾、血钠、TC、高密度脂蛋白胆固醇、TG、eGFR、心房颤动以及ACEI/ARB 、MRA、利尿剂、地高辛使用情况后,NT-proBNP仍是HFpEF患者全因死亡的独立预测因素(HR=1.230,95%CI 1.049~1.442,P=0.011).Spearman相关分析结果显示,LnNT-proBNP与eGFR呈负相关(r=-0.361,P<0.001).而校正了混杂因素后,多因素Cox回归分析结果显示肾功能不全与NT-proBNP预测HFpEF患者全因死亡无交互作用(P>0.05).结论 入院基线NT-proBNP是HFpEF患者全因死亡的独立预测因素.曹娟 金雪娟 周俊 陈桢玥 许顶立 杨新春 董蔚 黎励文 罗洁 陈丽 Fu Michael 周京敏 葛均波 2019中华心血管病杂志2019,47,11:21

3On-chip plasmonic spin-Hall nanograting for simultaneously detecting phase and polarization singularities显示文摘Phase and polarization singularities are important degrees of freedom for electromagnetic field manipulation.Detecting these singularities is essential for modern optics,but it is still a challenge,especially in integrated optical systems.In this paper,we propose an on-chip plasmonic spin-Hall nanograting structure that simultaneously detects both the polarization and phase singularities of the incident cylindrical vortex vector beam(CVVB).The nanograting is symmetry-breaking with different periods for the upper and lower parts,which enables the unidirectional excitation of the surface plasmon polariton depending on the topological charge of the incident optical vortex beam.Additionally,spin-Hall meta-slits are integrated onto the grating so that the structure has a chiral response for polarization detection.We demonstrate theoretically and experimentally that the designed structure fully discriminates both the topological charges and polarization states of the incident beam simultaneously.The proposed structure has great potential in compact integrated photonic circuits.Fu Feng Guangyuan Si Changjun Min Xiaocong Yuan Michael Somekh 2020Light(Science & Applications)2020,9,1:11
4Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11

13Post-stroke pain hypersensitivity induced by experimental thalamic hemorrhage in rats is region-specific and demonstrates limited efficacy of gabapentin显示文摘Intractable central post-stroke pain(CPSP) is one of the most common sequelae of stroke, but has been inadequately studied to date. In this study, we first determined the relationship between the lesion site and changes in mechanical or thermal pain sensitivity in a rat CPSP model with experimental thalamic hemorrhage produced by unilateral intra-thalamic collagenase IV(ITC) injection. Then, we evaluated the efficacy of gabapentin(GBP), an anticonvulsant that binds the voltage-gated Ca2+ channel α2δ and a commonly used anti-neuropathic pain medication. Histological case-by-case analysis showed that only lesions confined to the medial lemniscus and the ventroposterior lateral/medial nuclei of the thalamus and/or the posterior thalamic nucleus resulted in bilateral mechanical pain hypersensitivity. All of the animals displaying CPSP also had impaired motor coordination, while control rats with intra-thalamic saline developed no central pain or motor deficits. GBP had a dose-related anti-allodynic effect after a single administration(1, 10, or 100 mg/kg) on day 7 post-ITC, with significant effects lasting at least 5 hfor the higher doses. However, repeated treatment, once a day for two weeks, resulted in complete loss of effectiveness(drug tolerance) at 10 mg/kg, while effectiveness remained at 100 mg/kg, although the time period of efficacious analgesia was reduced. 14Acute pancreatitis in aging animals:Loss of pancreatitis-associated protein protection?显示文摘AIM:To investigate the effect of age on severity of acute pancreatitis(AP) using biochemical markers,histology and expression of the protective pancreatitisassociated proteins(PAPs).METHODS:AP was induced via intraductal injection of 4% sodium taurocholate in young and old rats.Sera and pancreata were assayed at 24 h for the parameters listed above;we also employed a novel molecular technique to assess bacterial infiltration using polymerase chain reaction to measure bacterial genomic ribosomal RNA.RESULTS:At 24 h after induction of AP,the pancreata of older animals had less edema(mean ± SE histologic score of young vs old:3.11 ± 0.16 vs 2.50 ±-0.11,P < 0.05),decreased local inflammatory response(histologic score of stromal infiltrate:3.11 ± 0.27 vs 2.00 ± 0.17,P < 0.05) and increased bacterial infiltration(174% ± 52% increase from sham vs 377% ± 4%,P < 0.05).A decreased expression of PAP1 and PAP2 was demonstrated by Western blotting analysis and immunohistochemical staining.There were no differences in serum amylase and lipase activity,or tissue myeloperoxidase or monocyte chemotactic protein-1 levels.However,in the most-aged group,serum C-reactive protein levels were higher(young vs old:0.249 ± 0.04 mg/dL vs 2.45 ± 0.68 mg/dL,P < 0.05).CONCLUSION:In older animals,there is depressed PAP expression related to a blunted inflammatory response in AP which is associated with worsened bacterial infiltration and higher C-reactive protein level;this may explain the more aggressive clinical course.Sophia Fu Albert Stanek Cathy M Mueller Nefertti A Brown Chongmin Huan Martin H Bluth Michael E Zenilman 2012World Journal of Gastroenterology2012,18,26:5

18Enhancing Genetic Gain through Genomic Selection: From Livestock to Plants显示文摘Although long-term genetic gain has been achieved through increasing use of modern breeding methods and technologies,the rate of genetic gain needs to be accelerated to meet humanity’s demand for agricultural products.In this regard,genomic selection(GS)has been considered most promising for genetic improvement of the complex traits controlled by many genes each with minor effects.Livestock scientists pioneered GS application largely due to livestock’s significantly higher individual values and the greater reduction in generation interval that can be achieved in GS.Large-scale application of GS in plants can be achieved by refining field management to improve heritability estimation and prediction accuracy and developing optimum GS models with the consideration of genotype-by-environment interaction and non-additive effects,along with significant cost reduction.Moreover,it would be more effective to integrate GS with other breeding tools and platforms for accelerating the breeding process and thereby further enhancing genetic gain.In addition,establishing an open-source breeding network and developing transdisciplinary approaches would be essential in enhancing breeding efficiency for small-and medium-sized enterprises and agricultural research systems in developing countries.New strategies centered on GS for enhancing genetic gain need to be developed.Yunbi Xu Xiaogang Liu Junjie Fu Hongwu Wang Jiankang Wang Changling Huang Boddupalli MPrasanna Michael SOlsen Guoying Wang Aimin Zhang 2020Plant Communications2020,1,1:4

20TLR9 agonist enhances radiofrequency ablation-induced CTL responses, leading to the potent inhibition of primary tumor growth and lung metastasis显示文摘Radiofrequency ablation(RFA)is the most common approach to thermal ablation for cancer therapy.Unfortunately,its efficacy is limited by incomplete ablation,and further optimization of RFA is required.Here,we demonstrate that incubation at 65°C triggers more EG7 tumor cell death by necrosis than treatment at 45°C,and the 65°C-treated cells are more effective at inducing antigen-specific CD8^(+)cytotoxic T lymphocyte(CTL)responses after injection in mice than the 45°C-treated ones.Dendritic cells(DCs)that phagocytose 65°C-treated EG7 cells become mature with upregulated MHCII and CD80 expression and are capable of efficiently inducing effector CTLs in mouse tumor models.RFA(65°C)therapy of EG7 tumors induces large areas of tumor necrosis and stimulates CTL responses.This leads to complete regression of small(~100 mm^(3))tumors but fails to suppress the growth of larger(~350 mm^(3))tumors.The administration of the Toll-like receptor-9(TLR9)agonist unmethylated cytosine-phosphorothioate-guanine oligonucleotide(CpG)to DCs phagocytosing 65°C-treated EG7 cells enhances the expression of MHCII and CD40 on DCs as well as DC-induced stimulation of CTL responses.Importantly,the intratumoral administration of CpG following RFA also increases the frequencies of tumor-associated immunogenic CD11b−CD11c^(+)CD103^(+)DC2 and CD11b+F4/80+MHCII+M1 macrophages and increases CD4^(+)and CD8^(+)T-cell tumor infiltration,leading to enhanced CD4^(+)T cell-dependent CTL responses and potent inhibition of primary RFA-treated or distant untreated tumor growth as well as tumor lung metastasis in mice bearing larger tumors.Overall,our data indicate that CpG administration,which enhances RFA-induced CTL responses and ultimately potentiates the inhibition of primary tumor growth and lung metastasis,is a promising strategy for improving RFA treatment,which may assist in optimizing this important cancer therapy.Aizhang Xu Lifeng Zhang Jingying Yuan Fatma Babikr Andrew Freywald Rajni Chibbar Michael Moser Wenjun Zhang Bing Zhang Zhaoying Fu Jim Xiang 2019Cellular & Molecular Immunology2019,16,10:3
