3Impacts of Types and Degree of Obesity on Non-Alcoholic Fatty Liver Disease and Related Dyslipidemia in Chinese School-Age Children?显示文摘Objective To explore the impacts of types and degree of obesity on non‐alcoholic fatty liver disease (NAFLD) and related lipids disturbance in Chinese school‐age children. Methods A total of 1 452 school‐age Children of 7 to 17 years were recruited in Beijing with representative cluster sampling method. Data of anthropometric measurements including weight, height and waist circumference were collected from March to May of 2007. Body mass index(BMI)was calculated. Blood samples were obtained and lipid profiles including triglyceride (TG), total cholesterol (TC), high‐density lipoprotein cholesterol (HDL‐C) and low‐density lipoprotein cholesterol (LDL‐C) were measured, while glutamate‐pyruvate transaminase (ALT) and glutamic‐oxalacetic transaminase (AST) were determined to evaluate liver function. The liver was also scanned by sonography, and abnormal hepatic sonograms were documented. NAFLD was diagnosed according to the criteria recommended by the Fatty Liver and Alcoholic Liver Disease Study Group under the Chinese Liver Disease Association. Analysis of covariance (ANOVA), Chi‐square test for trend and binary logistic regression analysis were performed. Results The dyslipidemia and ultrasonographic fatty liver deteriorated with the degree of obesity defined either by BMI or waist circumference. Compared with BMI, waist circumference contributed more to the development of dyslipidemia, fatty liver and NAFLD. The highest levels of TG, TC, LDL‐C, and lowest level of HDL‐C were seen in the mixed obese group followed by abdominal obese, peripheral obese and non‐obese ones. Adjusted for gender and age, the odds ratios (ORs) and their 95% confidence intervals of peripheral obesity, abdominal obesity and mixed obesity were 0, 10.93 (0.98‐121.96) and 79.16 (10.95‐572.44) for predicting NAFLD; 12.61 (1.24‐127.78), 19.39 (5.23‐71.85), and 93.21 (29.56‐293.90) for predicting ultrasonographic fatty liver; 1.78 (0.59‐5.44), 3.01 (1.91‐4.77), and 4.64 (3.52‐6.12) for predicting dyslipidemia, respectively compared with the non‐obese control group. The trend of hazards over groups was statistically significant (P<0.01). Conclusion The levels of lipid profile and the prevalence of NAFLD and dyslipidemia increased in parallel with the degree of obesity; As compared with the non‐obese control, the mixed obesity had the strongest association with NAFLD and dyslipidemia, followed by abdominal obesity and peripheral obesity in Chinese school‐age Children.MENG LingHui LUO Na MI Jie 2011Biomedical and Environmental Sciences2011,24,1:13

5Acellular ostrich corneal stroma used as scaffold for construction of tissue-engineered cornea显示文摘AIM:To assess acellular ostrich corneal matrix used as a scaffold to reconstruct a damaged cornea.· METHODS:A hypertonic saline solution combined with a digestion method was used to decellularize the ostrich cornea.The microstructure of the acellular corneal matrix was observed by transmission electron microscopy(TEM) and hematoxylin and eosin(H&E) staining.The mechanical properties were detected by a rheometer and a tension machine.The acellular corneal matrix was also transplanted into a rabbit cornea and cytokeratin 3 was used to check the immune phenotype.·RESULTS:The microstructure and mechanical properties of the ostrich cornea were well preserved after the decellularization process.In vitro,the methyl thiazolyl tetrazolium results revealed that extracts of the acellular ostrich corneas(AOCs) had no inhibitory effects on the proliferation of the corneal epithelial or endothelial cells or on the keratocytes.The rabbit lamellar keratoplasty showed that the transplanted AOCs were transparent and completely incorporated into the host cornea while corneal turbidity and graft dissolution occurred in the acellular porcine cornea(APC) transplantation.The phenotype of the reconstructed cornea was similar to a normal rabbit cornea with a high expression of cytokeratin 3 in the superficial epithelial cell layer.· CONCLUSION:We first used AOCs as scaffolds to reconstruct damaged corneas.Compared with porcine corneas,the anatomical structures of ostrich corneas are closer to those of human corneas.In accordance with the principle that structure determines function,a xenograft lamellar keratoplasty also confirmed that the AOC transplantation generated a superior outcome compared to that of the APC graft.Xian-Ning Liu Xiu-Ping Zhu Jie Wu Zheng-Jie Wu Yong Yin Xiang-HuaXiao Xin Su Bin Kong Shi-Yin Pan Hua Yang Yan Cheng Na An Sheng-Li Mi 2016International Journal of Ophthalmology(English edition)2016,9,3:6
6Streptococcus mutans activates the AIM2, NLRP3 and NLRC4 inflammasomes in human THP-1 macrophages显示文摘Streptococcus mutans(S. mutans), a major aetiologic agent of dental caries, is involved in systemic diseases, such as bacterial endocarditis, if it enters the bloodstream through temporary bacteraemia. Interleukin(IL)-1β, a proinflammatory cytokine, is related to the host defences against pathogens, and its synthesis, maturation, and secretion are tightly regulated by the activation of the inflammasome, an inflammatory signalling complex. This study examined the signalling mechanism of IL-1β secretion and the inflammasome pathway induced by S. mutans to explain the molecular mechanism through which systemic infection by oral streptococci can occur. After infection of THP-1 cells with S. mutans, the expression of inflammasome components was detected using various methods. S. mutans induced IL-1β secretion via caspase-1 activation, and S. mutans-induced IL-1β secretion required absent in melanoma(AIM2), NLR family pyrin domain-containing 3(NLRP3) and NLR family CARD domain-containing 4(NLRC4)inflammasome activation. In particular, the S. mutans-induced NLRP3 inflammasome was mediated by adenosine triphosphate(ATP) release, potassium depletion and lysosomal damage. 7Association between hepatocellular carcinoma and tumor necrosis factor alpha polymorphisms in South Korea显示文摘AIM: To investigate associations between the tumor necrosis factor alpha(TNF-α)-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A polymorphisms and HCC in Korea.METHODS: Hepatocellular carcinoma(HCC) cases were diagnosed at CHA Bundang Medical Center from June 1996 to August 2008. The association between TNF-α polymorphisms and HCC was analyzed in 157HCC patients and 201 controls using a polymerase chain reaction-restriction fragment length polymorphism assay. We investigated five TNF-α polymorphisms,which are TNF-α-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A. The TNF-α genotype frequencies,genotype combinations and haplotypes were analyzed to disclose the association with HCC.RESULTS: None of the TNF-α polymorphisms was significantly associated with HCC. However,nine genotype combinations had associations with increased likelihood of HCC. Among them,TNF-α-1031/-857/-238 TT/CC/GA(AOR = 18.849,95%CI: 2.203-161.246,P = 0.007),TNF-α-1031/-308/-238 TT/GG/GA(AOR = 26.956,95%CI: 3.071-236.584,P = 0.003),and TNF-α-1031/-238 TT/GA(AOR = 21.576,95%CI: 2.581-180.394,P = 0.005) showed marked association with HCC. There were five haplotypes of TNF-α polymorphisms which were significantly associated with HCC. They are TNF-α-1031/-863/-857/-308/-238 T-C-C-G-A(OR = 25.824,95%CI: 1.491-447.223,P = 0.0005),TNF-α-1031/-857/-308/-238 T-C-G-A(OR = 12.059,95%CI: 2.747-52.950,P < 0.0001),TNF-α-1031/-857/-238 T-C-A(OR = 10.696,95%CI: 2.428-47.110,P = 0.0001),TNF-α-1031/-308/-238 T-G-A(OR = 7.556,95%CI: 2.173-26.280,P = 0.0002) and TNF-α-1031/-238 T-A(OR = 10.865,95%CI: 2.473-47.740,P = 0.0001). Moreover,HCC Okuda stage Ⅲ cases with the TNF-α-1031 CC genotype had better survival than those with the TT genotype(AOR = 5.795,95%CI: 1.145-29.323). CONCLUSION: Although no single TNF-α polymorphism is associated with HCC in this study,some TNF-α genotype combinations and haplotypes are associated with HCC. 8Effects of Outdoor Temperature on Blood Pressure in a Prospective Cohort of Northwest China显示文摘Objective The relationship between outdoor temperature and blood pressure(BP) has been inconclusive. We analyzed data from a prospective cohort study in northwestern China to investigate the effect of outdoor temperature on BP and effect modification by season.Methods A total of 32,710 individuals who participated in both the baseline survey and the first follow-up in 2011–2015 were included in the study. A linear mixed-effect model and generalized additive mixed model(GAMM) were applied to estimate the association between outdoor temperature and BP after adjusting for confounding variables.Results The mean differences in systolic blood pressure(SBP) and diastolic blood pressure(DBP)between summer and winter were 3.5 mm Hg and 2.75 mm Hg, respectively. After adjusting for individual characteristics, meteorological factors and air pollutants, a significant increase in SBP and DBP was observed for lag 06 day and lag 04 day, a 0.28 mm Hg(95% CI: 0.27–0.30) per 1 °C decrease in average temperature for SBP and a 0.16 mm Hg(95% CI: 0.15–0.17) per 1 °C decrease in average temperature for DBP, respectively. The effects of the average temperature on both SBP and DBP were stronger in summer than in other seasons. The effects of the average temperature on BP were also greater if individuals were older, male, overweight or obese, a smoker or drinker, or had cardiovascular diseases(CVDs), hypertension, and diabetes.Conclusions This study demonstrated a significant negative association between outdoor temperature and BP in a high-altitude environment of northwest China. Moreover, BP showed a significant seasonal variation. 12Influence of gas flow on thermal field and stress during growth of sapphire single crystal using Kyropoulos method显示文摘The professional modeling software package CrysVUn was employed to study the process of a large sapphire single crystal growth using Kyropoulos method. The influence of gas pressure on thermal field, solid-liquid interface shape, gas velocity field and von Mises stress were studied for the first time. It is found that the root of the seed melt when gas pressure equals to one atmosphere or more than one atmosphere, especially during the seeding period, this result is consistent with the experimental observation, and this paper presents three ways to solve this problem. The temperature gradient and stress decreases significantly as the gas pressure increases. The convexity of the solid-liquid interface slightly increases when the gas pressure increases. 13Performance of Weibull function as a diameter distribution model for Pinus thunbergii stands in the eastern coast of South Korea显示文摘This study was carried out to determine the performance of percentile-based Weibull diameter distribution model for Pinus thunbergii stands thriving along the eastern coast of South Korea.The parameter recovery technique was used to estimate the three parameters of the Weibull model.The analysis demonstrated satisfactory results based on the following test statistics for the principal percentile models:fit index(FI) range from 0.501(minimum diameter) to 0.932(50th diameter percentiles) and root mean square error(RMSE) range from 0.112(quadratic mean diameter) to 3.572(minimum diameter).The developed model was further evaluated by determining the mean bias(ē) in trees per ha(TPH) for each diameter class,and the results showed highest over-prediction in the 20 cm,andunder-prediction in the 16 cm and 24 cm diameter classes.The goodness of fit tested by KolmogorovSmirnov(KS) test showed no significant differences(p>0.05) between the observed and predicted diameter distributions for almost all plots.Using site index and aboveground biomass(AGB) models developed for P.thunbergii in South Korea,a model to predict the AGB per ha for each diameter class and subsequently the total AGB of the stand was created.An application guide was also created,which will serve as a decision-support tool for forest managers in quantifying the future total AGB in P.thunbergii stands located in the eastern coast of South Korea and,subsequently,the quantification of potential carbon stocks aside from being a vital input in designing efficient management and protection strategies for these stands.ABINO Azyleah Canizares KIM Sung Yong LUMBRES Roscinto Ian Canicosa JANG Mi Na YOUN Ho doong PARK Ki Hyung LEE Young Jin 2016Journal of Mountain Science2016,13,5:2

16Photodegradation of Hexafluoropropylene Oxide Trimer Acid under UV Irradiation显示文摘As a novel alternative to traditional perfluoroalkyl substances(PFASs),including perfluorooctanoic acid(PFOA)and perfluorooctane sulfonate(PFOS),hexafluoroproplyene oxide trimer acid(HFPO-TA)has been detected worldwide in surface water.Moreover,recent researches have demonstrated that HFPO-TA has stronger bioaccumulation potential and higher hepatotoxicity than PFOA.To treat these contaminants e.g.PFOA and PFOS,some photochemical techniques by adding exogenous substances had been reported.However,there is still no report for the behavior of HFPO-TA itself under direct UV irradiation.The current study investigated the photo-transformation of HFPO-TA under UV irradiation in aqueous solution.After 72 hr photoreaction,75%degradation ratio and 25%defluorination ratio were achieved under ambient condition.Reducing active species,i.e.,hydrated electrons and active hydrogen atoms,generated from water splitting played dominant roles in degradation of HFPO-TA,which was confirmed by different effects of reaction atmospheres and quenching experiments.A possible degradation pathway was proposed based on the products identification and theoretical calculations.In general,HFPO-TA would be transformed into shorter-chain PFASs,including hexafluoropropylene oxide dimer acid(HFPODA),perfluoropropionic acid(PFA)and trifluoroacetate(TFA).This research provides basic information for HFPO-TA photodegradation process and is essential to develop novel remediation techniques for HFPO-TA and other alternatives with similar structures.Chen Li Na Mi Zhanghao Chen Cheng Gu 2020Journal of Environmental Sciences2020,32,11:2
17Human umbilical cord mesenchymal stem cells for psoriasis:a phase 1/2a,single-arm study显示文摘Psoriasis is a common,chronic immune-mediated systemic disease that had no effective and durable treatment.Mesenchymal stem cells(MSCs)have immunomodulatory properties.Therefore,we performed a phase 1/2a,single-arm clinical trial to evaluate the safety and efficacy of human umbilical cord-derived MSCs(UMSCs)in the treatment of psoriasis and to preliminarily explore the possible mechanisms.Seventeen patients with psoriasis were enrolled and received UMsC infusions.Adverse events,laboratory parameters,PASl,and PGA were analyzed.We did not observe obvious side effects during the treatment and 6-month follow-up.A total of 47.1%(8/17)of the psoriasis patients had at least 40%improvement in the PASl score,and 17.6%(3/17)had no sign of disease or minimal disease based on the PGA score.And the efficiency was 25%(2/8)for males and 66.7%(6/9)for females.After UMSC transplantation(UMSCT),the frequencies of Tregs and CD4^(+)memory T cells were significantly increased,and the frequencies of T helper(Th)17 and CD4^(+)naive T cells were significantly decreased in peripheral blood(PB)of psoriasis patients.And all responders showed significant increases in Tregs and CD4^(+)memory T cells,and significant decreases in Th17 cells and serum IL-17 level after UMsCT.And baseline level of Tregs in responders were significantly lower than those in nonresponders.In conclusion,allogeneic UMSCT is safe and partially effective in psoriasis patients,and level of Tregs may be used as a potent biomarker to predict the clinical efficacy of UMSCT.Trial registration Clinical Trials NCT03765957.Lamei Cheng Siqi Wang Cong Peng Xiao Zou Chao Yang Hua Mei Chuang Li Xian Su Na Xiao Qi Ouyang Mi Zhang Qiaolin Wang Yan Luo Minxue Shen Qun Qin Honglin Wang Wu Zhu Guangxiu Lu Ge Lin Yehong Kuang Xiang Chen 2022Signal Transduction and Targeted Therapy2022,7,9:2
18Perforated duodenal ulcer presenting with massive hematochezia in a 30-month-old child显示文摘Peptic ulcer disease is uncommon in children and rarely suspected as a cause of abdominal complaints in this age group; the diagnosis is therefore made almost exclusively when complications develop. Peptic ulcer disease is usually not considered in the differential diagnosis of pediatric patients. We present the case of a 30-month-old boy with duodenal perforation due to a peptic ulcer without a known etiology. The patient was admitted through the emergency department due to severe hematochezia and ongoing anemia; he presented with neither abdominal pain nor abdominal distension. There were no medical problems, and no drugs, such as corticosteroids or nonsteroidal anti-inflammatory drugs, had been prescribed or administered recently. We tried to control the active bleeding by medical treatment including arterial embolization, but the active bleeding was not controlled. Finally, an exploratory laparotomy was performed. A discrete anterior perforation with active bleeding of the duodenal wall was found.
