| 1 | SIRT3-mediated inhibition of FOS through histone H3 deacetylation prevents cardiac fibrosis and inflammation显示文摘Sirtuin 3(SIRT3)is a deacetylase that modulates proteins that control metabolism and protects against oxidative stress.Modulation of SIRT3 activity has been proposed as a promising therapeutic target for ameliorating metabolic diseases and associated cardiac disturbances.In this study,we investigated the role of SIRT3 in inflammation and fibrosis in the heart using male mice with constitutive and systemic deletion of SIRT3 and human cardiac AC16 cells.SIRT3 knockout mice showed cardiac fibrosis and inflammation that was characterized by augmented transcriptional activity of AP-1.Consistent with this,SIRT3 overexpression in human and neonatal rat cardiomyocytes partially prevented the inflammatory and profibrotic response induced by TNF-α.Notably,these effects were associated with a decrease in the mRNA and protein levels of FOS and the DNA-binding activity of AP-1.Finally,we demonstrated that SIRT3 inhibits FOS transcription through specific histone H3 lysine K27 deacetylation at its promoter.These findings highlight an important function of SIRT3 in mediating the often intricate profibrotic and proinflammatory responses of cardiac cells through the modulation of the FOS/AP-1 pathway.Since fibrosis and inflammation are crucial in the progression of cardiac hypertrophy,heart failure,and diabetic cardiomyopathy,our results point to SIRT3 as a potential target for treating these diseases. | Xavier Palomer MSilvia Román-Azcona Javier Pizarro-Delgado Ana Planavila Francesc Villarroya Brenda Valenzuela-Alcaraz Fátima Crispi Álvaro Sepúlveda-Martínez Irene Miguel-Escalada Jorge Ferrer JFrancisco Nistal Raquel García Mercy MDavidson Emma Barroso Manuel Vázquez-Carrera | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7 |
| 2 | Hepatoprotective and in vivo antioxidant activities of the hydroethanolic leaf extract of Mucuna pruriens (Fabaceae) in antitubercular drugs and alcohol models显示文摘AIM: Hepatotoxicity is a significantly increasing health problem worldwide, and the extent of the problem has stimulated interest in the search for hepatotherapeutic agents from plants. This study investigated the hepatoprotective and in vivo antioxidant activities of the hydroethanolic extract of Mucuna pruriens leaves in antitubercular and alcohol-induced hepatotoxicity assays in rats. METHOD: In each of the models used, seven groups were allotted. The different groups received normal saline(10 mL·kg-1, p.o.); hepatotoxicant(isoniazid-rifampicin, INH-RIF, 100 mg·kg-1, i.p. or 20% ethanol 5 g·kg-1, p.o.) and normal saline(10 mL·kg-1, p.o.); hepatotoxicant and extract at doses of 100, 200, and 400 mg·kg-1 p.o.; hepatotoxicant and silymarin 50 mg·kg-1 p.o.; and extract at 400 mg·kg-1 p.o.. On the 21st day of treatment, blood was collected for assessment of serum biochemical parameters and harvested liver samples were assessed for antioxidants. RESULTS: The hepatotoxicants significantly(P < 0.05-0.001) increased the levels of alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), bilirubin, and malondialdehyde(MDA); and reduced the levels of catalase(CAT), superoxide dismutase(SOD), glutathione peroxidase(GPx), and reduced glutathione GSH compared to control. M. pruriens significantly reversed(P < 0.05-0.001) the elevation in the level of ALT, AST, ALP, and bilirubin caused by the hepatotoxicants. The extract(200 and 400 mg·kg-1) significantly reversed(P < 0.05) the diminution in the level of in vivo antioxidants and increased the level of MDA produced by INH-RIF. M. pruriens(100-400 mg·kg-1) elicited significant reduction(P < 0.001) in the level of MDA compared to the alcohol group. Silymarin also reversed the deleterious effects of the hepatotoxicants. | 3 | Socio-cultural determinants of timely and delayed treatment of Buruli ulcer: implications for disease control显示文摘Introduction:Public health programmes recommend timely medical treatment for Buruli ulcer(BU)infection to prevent pre-ulcer conditions from progressing to ulcers,to minimise surgery,disabilities and the socio-economic impact of BU.Clarifying the role of socio-cultural determinants of timely medical treatment may assist in guiding public health programmes to improve treatment outcomes.This study clarified the role of socio-cultural determinants and health system factors affecting timely medical treatment for BU in an endemic area in Ghana.Methods:A semi-structured explanatory model interview based on the explanatory model interview catalogue(EMIC)was administered to 178 BU-affected persons.Based on research evidence,respondents were classified as timely treatment(use of medical treatment 3 months from awareness of disease)and delayed treatment(medical treatment 3 months after onset of disease and failure to use medical treatment).The outcome variable,timely treatment was analysed with cultural epidemiological variables for categories of distress,perceived causes of BU,outside-help and reasons for medical treatment in logistic regression models.The median time for the onset of symptoms to treatment was computed in days.Qualitative phenomenological analysis of respondents’narratives clarified the meaning,context and dynamic features of the relationship of explanatory variables with timely medical treatment.Results:The median time for initiating treatment was 25 days for pre-ulcers,and 204 days for ulcers.Income loss and use of herbalists showed significantly negative associations with timely treatment.Respondents’use of herbalists was often motivated by the desire for quick recovery in order to continue with work and because herbalists were relatives and easily accessible.However,drinking unclean water was significantly associated with timely treatment and access to health services encouraged timely treatment(OR 8.5,p=0.012).Findings show that health system factors of access are responsible for non-compliance to treatment regimes.Conclusions:Findings highlight the importance of an integrated approach to BU control and management considering the social and economic features that influence delayed treatment and factors that encourage timely medical treatment.This approach should consider periodic screening for early case-detection,collaboration with private practitioners and traditional healers,use of mobile services to improve access,adherence and treatment outcomes. | Mercy M Ackumey Margaret Gyapong Matilda Pappoe Cynthia Kwakye Maclean Mitchell G Weiss | 2012 | Infectious Diseases of Poverty2012,1,1: | 3 |
| 4 | The influence of moisture on the permeability of crushed shale samples显示文摘Shale cuttings and cores recovered from the subsurface and stored for hours to decades tend to dry out and lose moisture and hydrocarbons,leading to an increase in the effective matrix permeability.Moisture loss in shale samples is a fundamental sample preservation problem which can be solved by applying a standard moisture equilibration procedure to restore lost moisture.Our aim was to investigate the relationship between permeability and variable moisture as-received,as-received moisture-equilibrated and saturated moisture-equilibrated samples.Samples were crushed to a series of particle sizes(0.6-2.0)mm and moisture equilibrated at 97% relative humidity.Results show that moisture equilibration in the samples was achieved after 72 h.The permeability of the saturated moisture-equilibrated and as-received moisture-equilibrated samples decreased exponentially with increase in moisture content.The high correlation coefficient between permeability and particle size(r = 0.96 and 0.97)for moisture-equilibrated samples compared to 0.76 for as-received samples indicates that moisture equilibration improves permeability measurements in crushed shale samples.Furthermore,permeability measurements are repeatable for moisture-equilibrated samples compared to samples that were not equilibrated(as-received).We conclude that moisture content affects permeability and moisture equilibration normalizes and improves the repeatability of permeability measurements in crushed shale. | Mercy Achang Jack C.Pashin Eliot A.Atekwana | 2019 | Petroleum Science2019,16,3: | 3 |

| 7 | Pseudothrombus deposition accompanied with minimal change nephrotic syndrome and chronic kidney disease in a patient with Waldenstrom’s macroglobulinemia: A case report显示文摘BACKGROUND Waldenstrom's macroglobulinemia (WM) is a rare lymphoid neoplasia, which can have renal complications. These rarely occur, and most common renal manifestations are mild proteinuria and microscopic hematuria. Herein we describe a case of WM that presented with pseudothrombi depositing in capillaries associated with minimal change nephrotic syndrome and chronic kidney disease (CKD). CASE SUMMARY A 52-year-old man presented with features suggesting nephrotic syndrome. Extensive workups were done, and there were elevated serum levels of interleukin-6 and vascular endothelial growth factor (VEGF), capillary pseudothrombus accumulation associated with minimal change nephrotic syndrome, CKD, and WM. Treatment was directed at the patient’s WM with bortezomib, thalidomide, and dexamethasone whereby serum immunoglobulin M (IgM) decreased. The damage of IgM on the kidney was corrected;thus, the patient’s proteinuria and serum creatinine had improved. The patient is still under clinical follow-up. | 9 | Phenotypic plasticity of floral traits and pollination adaption in an alpine plant Pedicularis siphonantha D. Don when transplanted from higher to lower elevation in Eastern Himalaya显示文摘Phenotypic plasticity and/or pollinatormediated selection may be responsible for the changes in floral traits of plants when they are forced to live in new conditions. Although the two events could be independent, we hypothesized that phenotypic plasticity in floral traits might help to coordinate plant-pollinator interactions and enhance plant reproductive success in changing habitats. To test this hypothesis, we investigated floral traits and pollination on three natural populations of a lousewort(Pedicularis siphonantha) ranging at different elevations, as well as two downward transplanted populations in Shangeri-La County and Deqin County, northwest Yunnan, China. The results indicated that floral traits, i.e. phenology, longevity,display size, corolla tube length and pollen production differed significantly among populations. Moreover,or the two transplanted populations, floral traits diverged from their original populations, but converged to their host populations. Although the phenotypic plasticity in floral traits might be a rapid response to abiotic factor such as warmer environment, the changes in floral traits were found to be well adapted to pollination environment of the host population. Compared with plants of their original habitats in higher elevation, the transplanted individuals advanced flowering time, shortened flower longevity, reduced floral display size and pollen production, received higher visiting frequency and yielded more seeds.
