| 1 | Chromosome-level Genomes Reveal the Genetic Basis of Descending Dysploidy and Sex Determination in Morus Plants显示文摘Multiple plant lineages have independently evolved sex chromosomes and variable karyotypes to maintain their sessile lifestyles through constant biological innovation.Morus notabilis,a dioecious mulberry species,has the fewest chromosomes among Morus spp.,but the genetic basis of sex determination and karyotype evolution in this species has not been identified.In this study,three high-quality genome assemblies were generated for Morus spp.[including dioecious M.notabilis(male and female)and Morus yunnanensis(female)]with genome sizes of 301–329 Mb and were grouped into six pseudochromosomes.Using a combination of genomic approaches,we found that the putative ancestral karyotype of Morus species was close to 14 protochromosomes,and that several chromosome fusion events resulted in descending dysploidy(2n=2x=12).We also characterized a~6.2-Mb sex-determining region on chromosome 3.Four potential male-specific genes,a partially duplicated DNA helicase gene(named MSDH)and three Ty3_Gypsy long terminal repeat retrotransposons(named MSTG1/2/3),were identified in the Y-linked area and considered to be strong candidate genes for sex determination or differentiation.Population genomic analysis showed that Guangdong accessions in China were genetically similar to Japanese accessions of mulberry.In addition,genomic areas containing selective sweeps that distinguish domesticated mulberry from wild populations in terms of flowering and disease resistance were identified.Our study provides an important genetic resource for sex identification research and molecular breeding in mulberry. | Zhongqiang Xia Xuelei Dai Wei Fan Changying Liu Meirong Zhang Peipei Bian Yuping Zhou Liang Li Baozhong Zhu Shuman Liu Zhengang Li Xiling Wang Maode Yu Zhonghuai Xiang Yu Jiang Aichun Zhao | 2022 | Genomics, Proteomics & Bioinformatics2022,20,6: | 1 |

| 4 | miR-4530 inhibits the malignant biological behaviors of human glioma cells by directly targeting RTEL1显示文摘Human glioma is the most com m on prim ary brain tumor and is associated with high morbidity and mortality.Aberrant expressions of microRNAs(miRNAs)are involved in gliom a progression.In the present study,we aimed to elucidate the roles of miR-4530 in the pathogenesis of gliomas.miR-4530 expression was examined in human gliom a clinical tissues and cell lines including U251 and T98G.The target gene of miR-4530,RTEL1,was predicted with online tools and validated by luciferase reporter assay.Lentivirus infection,transfection of plasmids,and miRNA m im ics were used to m anipulate gene expression.Cell proliferation was determined using the CCK-8 method,and m igration and invasion assays were determ ined w ith transwell experim ents.Colony form ation was measured by crystal violet staining,while apoptosis was determined by Annexin V/PI staining.The anti-tumor effects of miR-4530 were evaluated in nude mice xenografted using U251 cells.Our results showed that miR-4530 was significantly down-regulated in human gliom a tissues and cell iines.miR-4530 over-expression inhibited the malignant behaviors of U251 and T98G cells,including reduced proliferation,dim inished colony form ation,migration and invasion,and increased apoptosis.Further mechanistic investigations revealed that RTEL1 is a direct functional target of miR-4530 in gliom as,and its over-expression rem arkably reverses the effects of miR-4530 m im ics on inhibiting these m alignant behaviors.In addition,miR-4530 over-expression inhibited the growth of xenografted U251 gliom a in nude mice.Therefore,miR-4530 acts as atum or-suppressor gene and inhibits the m alignant biological behaviors of human glioma cells,which is associated w ith directly targeting RTEL1.The miR-4530/RTEL1 axis is a potential therapeutic target for gliomas. | Tuo Wang Yan Zhang Bo Cui Maode Wang Ya Li Ke Gao | 2020 | Acta Biochimica et Biophysica Sinica2020,52,12: | 1 |

| 14 | A meta-analysis of tight versus conventional glycemia control in critically ill brain-injured adults显示文摘Objective: To evaluate the benefits and risks of tight glycemia control (TGC) versus conventional glucose control (CGC) in critically ill brain-injured adults. Methods: We performed meta-analysis by systematically searching PubMed, EMBASE, OVID, ScienceDirect, Web of Science, CNKI, Wanfang Data, and CQVIP databases to retrieve RCTs in any languages. We used Review Manager to perform meta-analysis. Odds ratios (ORs) or weighted mean differences (WMDs) with 95% confidence intervals (CIs) were calculated in analyses. Results: Twenty-six RCTs with a total of 3,759 participants were included in this meta-analysis. In-hospital mortality showed significant dissimilarity between TGC and CGC groups with OR of 0.76 (95%CI 0.58, 0.99). However, in terms of overall mortality and long-term neurological severity outcome, it didn't show differences with ORs of 0.93 (95%CI 0.79, 1.10) and 1.15 (95%CI 0.96, 1.37). There were also discrepancies in infection rate and ICU length of stay with OR of 0.51 (95%CI 0.42, 0.62) and WMD of -2.37 (95%CI -2.99, -1.74). Significances were observed in hypoglycemia events with ORs of 6.24 (95%CI 4.83, 8.07) and 2.73 (95%CI 2.56, 2.91) using two methods. Our experiments show that the one-step forecasts based on the FARIMA models are better than those based on autoregressive integrated moving average (ARIMA) models. We also propose a prediction-based admission control algorithm in hybrid QoS networks with WiFi access using the FARIMA models. | 20 | Pharmacokinetics of gene recombined angiogenesis inhibitor Kringle 5 in vivo using 131I specific markers and SPECT/CT显示文摘The previous pharmacokinetic methods can be only limited to drug analysis in vitro, which provide less information on the distribution and metabolismof drugs, and limit the interpretation and assessment of pharmacokinetics, the determination of metabolic principles, and evaluation of treatment effect. The objective of the study was to investigate the pharmacokinetic characteristics of gene recombination angiogenesis inhibitor Kringle 5 in vivo. The SPECT/CT and specific^(131)I-Kringle 5 marked by Iodogen method were both applied to explore the pharmacokinetic characteristics of^(131)I-Kringle 5 in vivo, and to investigate the dynamic distributions of^(131)I-Kringle 5 in target organs. Labeling recombinant angiogenesis inhibitor Kringle 5 using131 I with longer half-life and imaging in vivo using SPECT instead of PET,could overcome the limitations of previous methods. When the doses of^(131)I-Kringle 5 were 10.0, 7.5 and5.0 g/kg, respectively, the two-compartment open models can be determined within all the metabolic process in vivo. There were no significant differences in t1/2α, t1/2β, apparent volume of distribution and CL between those three levels. The ratio of AUC(0 1)among three different groups of 10.0, 7.5 and 5.0 g/kg was 2.56:1.44:1.0, which was close to the ratio(2:1.5:1.0). It could be clear that in the range of 5.0–10.0 g/kg, Kringle 5 was characterized by the first-order pharmacokinetics. Approximately 30 min after^(131)I-Kringle 5 was injected,^(131)I-Kringle 5 could be observed to concentrate in the heart, kidneys, liver and other organs by means of planar imaging and tomography. After 1 h of being injected, more radionuclide retained in the bladder, but not in intestinal. It could be concluded that^(131)I-Kringle 5 is mainly excreted through the kidneys. About 2 h after the injection of^(131)I-Kringle 5, the radionuclide in the heart, kidneys,liver and other organs was gradually reduced, while more radionuclide was concentrated in the bladder.The radionuclide was completely metabolized within 24 h, and the distribution of radioactivity in rats was similar to normal levels. In our study, the specific marker^(131)I-Kringle 5 and SPECT/CT were successfully used to explore pharmacokinetic characteristics of Kringle 5 in rats.
