1S100A8 promotes epithelial-mesenchymal transition and metastasis under TGF-β/USF2 axis in colorectal cancer显示文摘Background:The transforming growth factor-β(TGF-β)pathway plays a pivotal role in inducing epithelial-mesenchymal transition(EMT),which is a key step in cancer invasion and metastasis.However,the regulatory mechanism of TGF-βin inducing EMT in colorectal cancer(CRC)has not been fully elucidated.In previous studies,it was found that S100A8 may regulate EMT.This study aimed to clarify the role of S100A8 in TGF-β-induced EMT and explore the underlying mechanism in CRC.Methods:S100A8 and upstream transcription factor 2(USF2)expression was detected by immunohistochemistry in 412 CRC tissues.Kaplan-Meier survival analysis was performed.In vitro,Western blot,and migration and invasion assays were performed to investigate the effects of S100A8 and USF2 on TGF-β-induced EMT.Mouse metastasis models were used to determine in vivo metastasis ability.Luciferase reporter and chromatin immunoprecipitation assay were used to explore the role of USF2 on S100A8 transcription.Results:During TGF-β-induced EMT in CRC cells,S100A8 and the transcription factor USF2 were upregulated.S100A8 promoted cell migration and invasion and EMT.USF2 transcriptionally regulated S100A8 expression by directly binding to its promoter region.Furthermore,TGF-βenhanced the USF2/S100A8 signaling axis of CRC cells whereas extracellular S100A8 inhibited the USF2/S100A8 axis of CRC cells.S100A8 expression in tumor cells was associated with poor overall survival in CRC.USF2 expression was positively related to S100A8 expression in tumor cells but negatively related to S100A8-positive stromal cells.Conclusions:TGF-βwas found to promote EMT and metastasis through the USF2/S100A8 axis in CRC while extracellular S100A8 suppressed the USF2/S100A8 axis.USF2 was identified as an important switch on the intracellular and extracellular S100A8 feedback loop.Si Li Jun Zhang Senmi Qian Xuesong Wu Liang Sun Tianyi Ling Yao Jin Wenxiao Li Lichao Sun Maode Lai Fangying Xu 2021Cancer Communications2021,41,2:5

3Power savings in software defined data center networks via modified hybrid genetic algorithm显示文摘In modern data centers, power consumed by network is an observable portion of the total energy budget and thus improving the energy efficiency of data center networks(DCNs) truly matters. One effective way for this energy efficiency is to make the size of DCNs elastic along with traffic demands by flow consolidation and bandwidth scheduling, i.e., turning off unnecessary network components to reduce the power consumption. Meanwhile, having the instinct support for data center management, software defined networking(SDN) provides a paradigm to elastically control the resources of DCNs. To achieve such power savings, most of the prior efforts just adopt simple greedy heuristic to reduce computational complexity. However, due to the inherent problem of greedy algorithm, a good-enough optimization cannot be always guaranteed. To address this problem, a modified hybrid genetic algorithm(MHGA) is employed to improve the solution's accuracy, and the fine-grained routing function of SDN is fully leveraged. 5Packet delay analysis on IEEE 802.11 DCF under finite load traffic in multi-hop ad hoc networks显示文摘In this paper, the average packet delay on IEEE 802.11 DCF under finite load traffic in multi-hop ad hoc networks is analyzed. We employ a Markov chain model to analyze the probability of transmission at each node in an arbitrary slot and derive the channel access delay. We model each node using an M/G/1 queue and derive the queueing delay. The model is extended from analyzing the single-hop average packet delay to evaluating the end-to-end packet delay in multi-hop ad hoc net-works without assuming the traffic to be in a saturation state. To validate our ana-lytic results, we have done extensive simulation. 8Chromosome-level Genomes Reveal the Genetic Basis of Descending Dysploidy and Sex Determination in Morus Plants显示文摘Multiple plant lineages have independently evolved sex chromosomes and variable karyotypes to maintain their sessile lifestyles through constant biological innovation.Morus notabilis,a dioecious mulberry species,has the fewest chromosomes among Morus spp.,but the genetic basis of sex determination and karyotype evolution in this species has not been identified.In this study,three high-quality genome assemblies were generated for Morus spp.[including dioecious M.notabilis(male and female)and Morus yunnanensis(female)]with genome sizes of 301–329 Mb and were grouped into six pseudochromosomes.Using a combination of genomic approaches,we found that the putative ancestral karyotype of Morus species was close to 14 protochromosomes,and that several chromosome fusion events resulted in descending dysploidy(2n=2x=12).We also characterized a~6.2-Mb sex-determining region on chromosome 3.Four potential male-specific genes,a partially duplicated DNA helicase gene(named MSDH)and three Ty3_Gypsy long terminal repeat retrotransposons(named MSTG1/2/3),were identified in the Y-linked area and considered to be strong candidate genes for sex determination or differentiation.Population genomic analysis showed that Guangdong accessions in China were genetically similar to Japanese accessions of mulberry.In addition,genomic areas containing selective sweeps that distinguish domesticated mulberry from wild populations in terms of flowering and disease resistance were identified.Our study provides an important genetic resource for sex identification research and molecular breeding in mulberry.Zhongqiang Xia Xuelei Dai Wei Fan Changying Liu Meirong Zhang Peipei Bian Yuping Zhou Liang Li Baozhong Zhu Shuman Liu Zhengang Li Xiling Wang Maode Yu Zhonghuai Xiang Yu Jiang Aichun Zhao 2022Genomics, Proteomics & Bioinformatics2022,20,6:1

16miR-4530 inhibits the malignant biological behaviors of human glioma cells by directly targeting RTEL1显示文摘Human glioma is the most com m on prim ary brain tumor and is associated with high morbidity and mortality.Aberrant expressions of microRNAs(miRNAs)are involved in gliom a progression.In the present study,we aimed to elucidate the roles of miR-4530 in the pathogenesis of gliomas.miR-4530 expression was examined in human gliom a clinical tissues and cell lines including U251 and T98G.The target gene of miR-4530,RTEL1,was predicted with online tools and validated by luciferase reporter assay.Lentivirus infection,transfection of plasmids,and miRNA m im ics were used to m anipulate gene expression.Cell proliferation was determined using the CCK-8 method,and m igration and invasion assays were determ ined w ith transwell experim ents.Colony form ation was measured by crystal violet staining,while apoptosis was determined by Annexin V/PI staining.The anti-tumor effects of miR-4530 were evaluated in nude mice xenografted using U251 cells.Our results showed that miR-4530 was significantly down-regulated in human gliom a tissues and cell iines.miR-4530 over-expression inhibited the malignant behaviors of U251 and T98G cells,including reduced proliferation,dim inished colony form ation,migration and invasion,and increased apoptosis.Further mechanistic investigations revealed that RTEL1 is a direct functional target of miR-4530 in gliom as,and its over-expression rem arkably reverses the effects of miR-4530 m im ics on inhibiting these m alignant behaviors.In addition,miR-4530 over-expression inhibited the growth of xenografted U251 gliom a in nude mice.Therefore,miR-4530 acts as atum or-suppressor gene and inhibits the m alignant biological behaviors of human glioma cells,which is associated w ith directly targeting RTEL1.The miR-4530/RTEL1 axis is a potential therapeutic target for gliomas.Tuo Wang Yan Zhang Bo Cui Maode Wang Ya Li Ke Gao 2020Acta Biochimica et Biophysica Sinica2020,52,12:1
