1Molecular immune pathogenesis and diagnosis of COVID-19显示文摘Coronavirus disease 2019(COVID-19)is a kind of viral pneumonia which is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The emergence of SARS-CoV-2 has been marked as the third introduction of a highly pathogenic coronavirus into the human population after the severe acute respiratory syndrome coronavirus(SARS-CoV)and the Middle East respiratory syndrome coro-navirus(MERS-CoV)in the twenty-first century.In this minireview,we provide a brief introduction of the general features of SARS-CoV-2 and discuss current knowledge of molecular immune pathogenesis,diagnosis and treatment of COVID-19 on the base of the present understanding of SARS-CoV and MERS-CoV infections,which may be helpful in offering novel insights and potential therapeutic targets for combating the SARS-CoV-2 infection.Xiaowei Li Manman Geng Yizhao Peng Liesu Meng Shemin Lu 2020Journal of Pharmaceutical Analysis2020,10,2:31
2Effects of dietary supplementation of Artemisia argyi aqueous extract on antioxidant indexes of small intestine in broilers显示文摘The present study was conducted to investigate the effect of Artemisia argyi aqueous extract(AAE) on antioxidant indexes in the small intestine. A total of 192 Arbor Acre broiler chickens(one-day-old) were randomly divided into 4 treatments with 6 replicates of 8 chickens. These 4 diets were formulated by adding 0, 500,1,000 and 2,000 mg/kg AAE to the basal diet. The results showed as follows: 1) compared with the control, the total antioxidant capacity(T-AOC) in ileum for the 2,000 mg/kg treatment group was significantly increased at 21 days of age(P < 0.05); the T-AOC levels in jejunum and ileum were significantly increased in broilers supplemented with 500 mg/kg AAE at 42 days of age(P < 0.05), and the T-AOC levels in jejunum and ileum were significantly improved in 1,000 mg/kg treatment group(P < 0.01), 2) At 21 days of age, supplementation of 500 mg/kg AAE significantly increased the catalase(CAT) activity of small intestine, and the glutathione peroxidase(GSH-Px) activity of jejunum was improved(P < 0.01), meanwhile, the GSH-Px activity of duodenum and the total superoxide dismutase(T-SOD) activity of duodenum and jejunum were significantly higher than those of the control group(P < 0.05); supplementation of 1,000 mg/kg AAE significantly increased the CAT activity of duodenum and ileum and the GSH-Px activity of duodenum and jejunum(P < 0.05), and the ileum GSH-Px activity was significantly increased(P < 0.01); supplementation of 2,000 mg/kg AAE significantly increased the CAT activity of duodenum and ileum(P < 0.05). At 42 days of age, supplementation of 500 mg/kg AAE significantly increased the GSH-Px activity of ileum and the T-SOD activity of duodenum(P < 0.05),meanwhile, the T-SOD activity of jejunum was significantly increased(P < 0.01); supplementation of1,000 mg/kg AAE significantly increased the CAT activity of jejunum and the T-SOD activity of ileum(P < 0.01), and the GSH-Px activity of jejunum was significantly increased(P < 0.05); supplementation of2,000 mg/kg AAE significantly increased the T-SOD activity of ileum(P < 0.05), but significantly decreased the CAT activity of ileum and the GSH-Px activity of jejunum(P < 0.05). 3) The malondialdehyde(MDA) levels of 3 AAE supplementation groups were significantly decreased at 21 and 42 days of age(P < 0.05). 3Sleep Deprivation Selectively Down-Regulates Astrocytic 5-HT2B Receptors and Triggers Depressive-Like Behaviors via Stimulating P2X7 Receptors in Mice显示文摘Chronic loss of sleep damages health and disturbs the quality of life.Long-lasting sleep deprivation(SD)as well as sleep abnormalities are substantial risk factors for major depressive disorder,although the underlying mechanisms are not clear.Here,we showed that chronic SD in mice promotes a gradual elevation of extracellular ATP,which activates astroglial P2X7 receptors(P2X7Rs).Activated P2X7Rs,in turn,selectively down-regulated the expression of 5-HT2B receptors(5-HT2BRs)in astrocytes.Stimulation of P2X7Rs induced by SD selectively suppressed the phosphorylation of AKT and FoxO3 a in astrocytes,but not in neurons.The overexpression of FoxO3a in astrocytes inhibited the expression of 5-HT2BRs.Down-regulation of 5-HT2BsRs instigated by SD suppressed the activation of STAT3 and relieved the inhibition of Ca2+-dependent phospholipase A2.This latter cascade promoted the release of arachidonic acid and prostaglandin E2.The depression-like behaviors induced by SD were alleviated in P2X7R-KO mice.Our study reveals the mechanism underlying chronic SD-induced depression-like behaviors and suggests 5-HT2BRs as a key target for exploring therapeutic strategies aimed at the depression evoked by sleep disorders.Maosheng Xia Zexiong Li Shuai Li Shanshan Liang Xiaowei Li Beina Chen Manman Zhang Chengyi Dong Alexei Verkhratsky Dawei Guan Baoman Li 2020Neuroscience Bulletin2020,36,11:10

6A new choice of stent for transjugular intrahepatic portosystemic shunt creation: Viabahn ePTFE covered stent/bare metal stent combination显示文摘Objectives:To compare the clinical outcomes in terms of structure and function between the insertion of a transjugular intrahepatic portosystemic shunt(TIPS) created with the Viabahn ePTFE covered stent/bare metal stent(BMS) combination and the Fluency ePTFE covered stent/BMS combination.Methods:A total of 101 consecutive patients who received a TIPS from February 2016 to August 2018 in our center were retrospectively analyzed.Sixty-four subjects were enrolled in the Viabahn group and 37 were enrolled in the Fluency group.The geometry characteristics of the TIPS were calculated,and the associated occurrence of shunt dysfunction,survival,overt hepatic encephalopathy,and variceal rebleeding were evaluated.Results:The technical success rate was 100%.After the insertion of the TIPS,the rate of shunt dysfunction during the first 3 months was significantly different between the Viabahn and Fluency groups(1.6% and 13.5%,respectively;p=0.024).Multivariate analysis indicated that the angle of portal venous inflow(α) was the only independent risk factor for shunt dysfunction(hazard ratio=1.060,95% confidence interval=1.009-1.112,p=0.020).In addition,3 months after the TIPS insertion,the a angle distinctly increased from 20.9°±14.3°-26.9°±20.1°(p=0.005) in the Fluency group but did not change significantly in the Viabahn group(from 21.9°±15.1°-22.9°± 17.6°,p=0.798).Conclusions:Shunt dysfunction was related to the a angle owing to the slight effect on the a angle after the implantation of the TIPS.The Viabahn ePTFE covered stent/BMS combination was more stable in structure and promised higher short-term stent patency compared with the Fluency ePTFE covered stent/BMS combination.Jiacheng Liu Jie Meng Chen Zhou Qin Shi Chongtu Yang Jinqiang Ma Manman Chen Bin Xiong 2021Journal of Interventional Medicine2021,4,1:9

8Microbial community structure and diversity in deep-sea hydrothermal vent sediments along the Eastern Lau Spreading Centre显示文摘The aim of this study is to investigate microbial structures and diversities in five active hydrothermal fields' sediments along the Eastern Lau Spreading Centre (ELSC) in the Lau Basin (southwest Pacific). Microbial communities were surveyed by denatured gradient gel electrophoresis (DGGE) and clone library analysis of 16S rRNA genes. The differences in microbial community structures among sediment samples from the five deep-sea hydrothermal sites were revealed by DGGE profiles. Cluster analysis of DGGE profiles separated the five hydrothermal samples into two groups. Four different 16S rRNA gene clone libraries, representing two selected hydrothermal samples (19-4TVG8 and 19-4TVG11), were constructed. Twenty-three and 32 phylotypes were identified from 166 and 160 bacterial clones respectively, including Proteobacteria, Bacteroidetes, Firmicutes, Nitrospirae and Planctomycetes. The phylum Proteobacteria is dominant in both bacterial libraries with a predominance of Gamma-Proteobacteria. A total of 31 and 25 phylotypes were obtained from 160 and 130 archaeal clones respectively, including Miscellaneous Crenarchaeotic Group, Marine Group Ⅰ and Ⅲ, Marine Benthic Group E, Terrestrial Hot Spring Crenarchaeota and Deep-sea Hydrothermal Vent Euryarchaeota. 9Pharmacokinetics of H002, a novel S1PR_1 modulator, and its metabolites in rat blood using liquid chromatography–tandem mass spectrometry显示文摘A rapid and sensitive liquid chromatography–tandem mass spectrometry(LC–MS/MS) method was developed and validated for the simultaneous determination of H002 and its phosphorylated metabolite, H002-P and hydroxylated metabolite H002-M, in rat blood. H001, an analogue of H002, was used as the internal standard.Blood samples were prepared by simple protein precipitation. The analytes and internal standard were separated on a Zorbax SB-C18 column with a gradient mobile phase consisting of methanol and water containing 0.1% formic acid at a flow rate of 0.2 mL /min with an operating temperature of 20 1C. The detection was performed on a triple quadrupole tandem mass spectrometer with positive electrospray ionization in multiple-reaction monitoring mode.Linear detection responses were obtained from 0.2–100 ng/mL for H002 and H002-M, while 0.5–100 ng/mL for H002-P. The intra- and inter-day precision(RSD%) was within 11.76%, with the accuracy(RE%) ranging from –9.84% to 9.12%. 11Controlling the coal dust at transshipment point: A study of the foam-sol foaming device显示文摘In order to effectively control the dust at the transshipment point with foam-sol, this paper attempted to study the characteristics of dust diffusion at transshipment point and the foam-sol foaming device with diffusion outlet was also designed in this paper. To study the diffusion rules of coal dust, fluent discrete phase model was utilized in the numerical simulation, as the coal dust was thrown down at a horizontal velocity of 2.5 m/s. A foam-sol foaming device was designed, through which foaming agent could be automatically sucked into the Venturi by the negative pressure. The automatic controller was also equipped,which could transform the energy of the compressed air into the constant pressure difference so that the gelling agent could be qualitatively added into the gel container. The diffusion outlet that could spray out foam-sol in a continuous, conical and 3D manner was also designed. Moreover, this paper also carried out the contrast experiments on dust removal efficiency among water, aqueous foam and foam-sol. The results clearly show that the symmetrical whirlpools appeared below the inlet where the largest whirlpool diameter was 0.52 m, and the horizontal distance from swirl range to the inlet was approximately0.69 m. By using the self-designed foaming device, the foaming was multiplied by 30 times and the volume ratio with water and foaming agent reached 95%:5%. In this context, the gas pressure was controlled at 0.3 MPa, with gas flow at 15 m3/h and water flow at 0.5 m3/h, with water pressure controlled between0.34 and 0.36 MPa. 12Application of multi-stage dynamic magnetizing roasting technology on the utilization of cryptocrystalline oolitic hematite:A review显示文摘A large number of studies have shown that oolitic hematite is an iron ore that is extremely difficult to utilize because of its fine disseminated particle size, high harmful impurity content and oolitic structure.To recover iron from oolitic hematite, we developed a novel multistage dynamic magnetizing roasting technology. Compared with traditional magnetizing roasting technologies, this novel technology has the following advantages: firstly, the oolitic hematite is dynamically reduced in a multi-stage roasting furnace, which shortens the reduction time and avoids ringing and over-reduction;secondly, the novel dynamic magnetizing roasting technology has strong raw material adaptability, and the size range of raw materials can be as wide as 0–15 mm;thirdly, the roasting furnace adopts a preheating-heating process, and the low-calorific value blast furnace gas can be used as the fuel and reductant, which greatly reduces the cost. The actual industrial production data showed that the energy consumption in the roasting process can be less than 35 kg of standard coal per ton of raw ore. 13Which Inspection Approach Is Better to Prevent Drug Fraud: Announced or Unannounced?显示文摘In recent years,there are many crimes related drug fraud occuring in China and many experts think that the main cause is that China Food and Drug Administration (CFDA)adopts announced inspection (AI).In order to circumvent this difficulty,CFDA has exploited unannounced inspection (UI)since 2014.In this paper,the authors study the problem of which one performs better, AI or UI.Specifically,the authors consider a supervisor,which decides the inspection approach,inspection strength and punishment to force the firm to put self-inspection effort to meet the requirements of Good Manufacturing Practice,and a firm,which produces a drug and decides its self-inspection effort. The authors use game theory to model this problem,characterize the equilibrium policies under AI, and compare the effects of the two approaches on preventing drug fraud under complete and incomplete information.The results show that under the complete information,UI performs better if the firm's technical level and the inspection cost are low and AI performs better otherwise.When the supervisor doesn't know the firm's technical level,if the low technical level is high,AI performs better.Otherwise, UI performs better if the inspection cost is low and AI performs better if the inspection cost is high.ZHANG Manman ZHANG Juliang CHENG T.C.E. 14Accumulation of carotenoids and expression of carotenoid biosynthesis genes in fruit flesh during fruit development in two Cucurbita maxima inbred lines显示文摘Mesocarp color is an important agronomic trait of Cucurbita maxima and is determined mainly by the contents and compositions of the carotenoids.The two inbred lines with significant differences in fruit flesh color were used in the study,the orange’312-1’and white’98-2’.Changes in seven carotenoid contents and compositions in the flesh of fruit produced by inbred lines’312-1’and’98-2’were analyzed during fruit development.The expression of eight key carotenoid biosynthesis genes in the fruit flesh were investigated during five fruit development stage in two inbred lines.As the flesh color intensified,the orange flesh color of’312-1’was determined mainly by the increased contents of lutein,β-carotene and zeaxanthin and the lack of carotenoid accumulation led to the formation of white flesh in’98-2’fruit.The expression of the LCY-e and CHYb genes was significantly stronger in’312-1’than in’98-2’,and their expression was strongly correlated with lutein andβ-carotene contents during fruit development.This study provides a deep understanding of the molecular mechanism of carotenoid biosynthesis in fruit flesh and provides a basis for additional studies on the highly refined improvement of squash quality.Yusong Luo Chaojie Wang Manman Wang Yunli Wang Wenlong Xu Hongyu Han Zhichao Wang Yujuan Zhong Hexun Huang Shuping Qu 2021Horticultural Plant Journal2021,7,6:3
15Stimulus-Responsive Room Temperature Phosphorescence Materials:Internal Mechanism,Design Strategy,and Potential Application显示文摘CONSPECTUS:Room temperature phosphorescence(RTP)materials,which could respond to external stimuli,such as force,heat,light,electric filed,etc.,have drawn increasing attention for their broad application prospects,especially in the fields of anticounterfeiting,sensors,data storage,and so on.In comparison with the traditional fluorescence ones,RTP materials show much longer emission lifetimes,which can be even caught by the naked eye.Thus,the change in emission lifetime under an external stimulus for RTP materials can be also a potential monitoring parameter,in addition to emission color and intensity.In other words,the number of visual monitoring parameters could increase from two to three in RTP materials,which would greatly facilitate their practical applications.Until now,RTP materials have been typically limited to metal-containing inorganic materials,particularly rare-earth phosphors.Their emissions are governed by the slow liberation of trapped charge carriers from isolated traps of impurities,defects,or ions through thermal stimulation with low luminescence efficiency.However,these materials suffer from some intrinsic disadvantages,including high cost,potential toxicity,and instability in aqueous environments.In order to solve these problems,the purely organic RTP materials should be a good choice.However,these kinds of materials are really scarce now,especially for the ones with stimulus response characteristic.To develop purely organic RTP materials with a stimulus response effect,we and other scientists have tried a lot.Luckily,some progresses have been made.In this Account,we present our recent progress on the stimulus-responsive room temperature phosphorescence of organic materials,mainly focusing on the internal mechanism and potential applications.First,the fundamental knowledge is described to illustrate the importance and main principles of the stimulus-responsive RTP effect.Then,some typical stimulus-responsive RTP materials based on different internal mechanisms are discussed.Mainly,two kinds of stimulus-responsive RTP materials were introduced,namely,single-component and multicomponent ones.Correspondingly,their dynamic change of the RTP property under external stimulus occurred based on the distinct internal mechanisms.For single-component materials,the changes in molecular structure,packing,or conformation,have played a significant role in their corresponding stimulus-responsive RTP effect.As for multicomponent materials,the changed oxygen concentration in matrix and intermolecular distance between different components were found more during the stimulus-responsive RTP process.Accordingly,different potential applications were explored based on the different stimulus-responsive RTP processes.With the classification of stimulus-responsive RTP materials based on different internal mechanisms,the corresponding design strategy could be well proposed,thus guiding the further development of this research field.Jie Yang Manman Fang Zhen Li 2021Accounts of Materials Research2021,2,8:3
16Mechanisms of traditional Chinese medicines in the treatment of allergic rhinitis using a network biology approach显示文摘Objective:To systematically explore the effect and mechanism of traditional Chinese herbs in the treatment of allergic rhinitis.Methods:Magnolia flower,xanthium fruit,astragalus root,paniculate cynanchum,scutellaria root,cicada molting,stephania root,small centipeda herb,and licorice root as traditional Chinese herbs are widely used to treat allergic rhinitis.With multiple database integration and entity grammar systems,a molecular interaction network of traditional Chinese herbs for treating allergic rhinitis was established.Bioinformatics approaches were adopted to integrate relevant data and biological information by multiple databases.Molecular interaction network of Chinese herbs for treating allergic rhinitis was constructed based on entity grammar systems model.The network was then used for elucidating the antiallergic rhinitis mechanism of the Chinese herbs on the molecular level.The subnetworks were also extracted to explicitly display the pathway and targets where effective components acted on.Results:Traditional Chinese herbs could influence various pathological aspects in allergic rhinitis including the production of pro-inflammatory substances,such as histamine and cytokine interleukin-4(IL-4),interleukin-13(IL-13),lowering immunoglobulin E(IgE)level or blocking antigen binding,altering the biological processes of IgE,modulating the balance of T helper(Th)or other cells by cell proliferation and differentiation,mediating cellecell signaling and second-messenger-mediated signaling,stabilizing the cell membrane,and affecting regulation of cellular defense response.Conclusion:The research theoretically confirms the mechanism of anti-allergic rhinitis by traditional Chinese herbs,provides important fundamental research information for treatment of allergic rhinitis,and may serve as a reference for new drug development and effective ingredients of compatibility.Li Du Xiaotong Ye Manman Li Huihui Wang Baixia Zhang Rao Zheng Yun Wang 2021Journal of Traditional Chinese Medical Sciences2021,8,1:3
17AFF3-DNA methylation interplay in maintaining the mono-allelic expression pattern of XIST in terminally differentiated cells显示文摘X chromosome inactivation and genomic imprinting are two classic epigenetic regulatory processes that cause mono-allelic gene expression.In female mammals,mono-allelic expression of the long non-coding RNA gene X-inactive specific transcript(XIST)is essential for initiation of X chromosome inactivation upon differentiation.We have previously demonstrated that the central factor of super elongation complex-like 3(SEC-L3),AFF3,is enriched at gamete differentially methylated regions(DMRs)of the imprinted loci and regulates the imprinted gene expression.Here,we found that AFF3 can also bind to the DMR downstream of the XIST promoter.Knockdown of AFF3 leads to de-repression of the inactive allele of X IST in terminally differentiated cells.In addition,the binding of AFF3 to the XIST DMR relies on DNA methylation and also regulates DNA methylation level at DMR region.However,the KAP1-H3K9 methylation machineries,which regulate the imprinted loci,might not play major roles in maintaining the mono-aUelic expression pattern of XIST in these cells.Thus,our results suggest that the differential mechanisms involved in the X IST DMR and gDMR regulation,which both require AFF3 and DNA methylation.Yue Zhang Chao Wang Xiaoxu Liu Qian Yang Hongliang Ji Mengjun Yang Manman Xu Yunyan Zhou Wei Xie Zhuojuan Luo Chengqi Lin 2019Journal of Molecular Cell Biology2019,11,9:3

19A review of systematic evaluation and improvement in the big data environment显示文摘The era of big data brings unprecedented opportunities and challenges to management research.As one of the important functions of management decision-making,evaluation has been given more functions and application space.Exploring the applicable evaluation methods in the big data environment has become an important subject of research.The purpose of this paper is to provide an overview and discussion of systematic evaluation and improvement in the big data environment.We first review the evaluation methods based on the main analytic techniques of big data such as data mining,statistical methods,optimization and simulation,and deep learning.Focused on the characteristics of big data(association feature,data loss,data noise,and visualization),the relevant evaluation methods are given.Furthermore,we explore the systematic improvement studies and application fields.Finally,we analyze the new application areas of evaluation methods and give the future directions of evaluation method research in a big data environment from six aspects.We hope our research could provide meaningful insights for subsequent research.Feng YANG Manman WANG 2020Frontiers of Engineering Management2020,7,1:3
