| 1 | Microstructure,mechanical and corrosion properties of Mg-2Dy-xZn(x=0,0.1,0.5 and 1 at.%)alloys显示文摘Microstructure,mechanical and corrosion properties of as-cast Mg-2Dy-xZn(x=0,0.1,0.5,and 1)(at.%)alloys were investigated.The microstructures of the as-cast Mg-2Dy and Mg-2Dy-0.1Zn alloys mainly consisted ofα-Mg phase and Mg_(24)Dy_(5)eutectic phase.With 0.5 at.%Zn addition,Mg_(12)ZnDy phase with 18R-type long period stacking ordered(LPSO)structure and Mg_(2)Dy phase precipitated at the grain boundaries.When the content of Zn is 1 at.%,only the Mg_(3)Zn_(3)Dy_(2)phase formed in theα-Mg matrix.The electrochemical measurements and immersion testing results indicated that the Mg-2Dy-0.1Zn alloy exhibited the best corrosion resistance.It revealed that the morphology,scale,amount and distribution of the second phase have a great effect on the corrosion resistance of alloy.Additionally,the tensile testing results showed that the Mg-2Dy-0.5Zn alloy exhibited the higher tensile strength and good elongation,especially at 200℃.The improvement of mechanical properties was mainly due to the strengthening of LPSO phase and grain refinement of α-Mg. | Guangli Bi Yuandong Li Shijun Zang Jianbin Zhang Ying Ma Yuan Hao | 2014 | Journal of Magnesium and Alloys2014,2,1: | 13 |
| 2 | Association of KIR Genotypes and Haplotypes with Susceptibility to Chronic Hepatitis B Virus Infection in Chinese Han Population显示文摘Killer immunoglobulin-like receptor (KIR) genes can regulate the activation of NK and T cells upon interaction with HLA class I molecules. Hepatitis B virus (HBV) infection has been regarded as a multi-factorial disorder disease. Previous studies revealed that KIRs were involved in HCV and HIV infection or clearance. The aim of this study was to explore the possibility of the inheritance of KIR genotypes and haplotypes as a candidate for susceptibility to persistent HBV infection or HBV clearance. The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 150 chronic hepatitis B (CHB) patients, 251 spontaneously recovered (SR) controls, and 412 healthy controls. The frequencies of genotype G, M, FZ1 increased in CHB patients compared with healthy control subjects. The frequency of genotype AH was higher in SR controls than that in both CHB patients and healthy controls. The carriage frequencies of genotype G and AH were higher; while, the frequencies of AF and AJ were lower in SR controls than those in healthy control subjects. The frequency of A haplotype was lower, whereas, the frequency of B haplotype was higher in CHB patients and SR controls than those in healthy controls. In healthy controls, haplotype 4 was found lower compared with that in CHB patients and SR controls and the frequency of haplotype 5 was higher in SR controls than that in other two groups. Based on these findings, it seems that the genotypes M and FZ1 are HBV susceptive genotypes; AH, on the other hand, may be protective genotypes that facilitate the clearance of HBV. | 4 | Subcutaneous and intramuscular fat transcriptomes show large differences in network organization and associations with adipose traits in pigs显示文摘Subcutaneous fat(SCF)and intramuscular fat(IMF)deposition is relevant to health in humans,as well as meat production and quality in pigs.In this study,we generated RNA sequence data for 122 SCF,120 IMF,and 87 longissimus dorsi muscle(LDM)samples using 155 F6 pigs from a specially designed heterogeneous population generated by intercrossing four highly selected European commercial breeds and four indigenous Chinese pig breeds.The phenotypes including waist back fat thickness and intramuscular fat content were also measured in the 155 F6 pigs.We found that the genes in SCF and IMF differed largely in both expression levels and network connectivity,and highlighted network modules that exhibited strongest gain of connectivity in SCF and IMF,containing genes that were associated with the immune process and DNA double-strand repair,respectively.We identified 215 SCF genes related to kinase inhibitor activity,mitochondrial fission,and angiogenesis,and 90 IMF genes related to lipolysis and fat cell differentiation,displayed a tissue-specific association with back fat thickness and IMF content,respectively.We found that cis-expression QTL for trait-associated genes in the two adipose tissues tended to have tissuedependent predictability for the two adipose traits.Alternative splicing of genes was also found to be associated with SCF or IMF deposition,but the association was much less extensive than that based on expression levels.This study provides a better understanding of SCF and IMF gene transcription and network organization and identified critical genes and network modules that displayed tissue-specific associations with subcutaneous and intramuscular fat deposition.These features are helpful for designing breeding programs to genetically improve the two adipose traits in a balanced way. | Yifeng Zhang Yingchun Sun Zhongzi Wu Xinwei Xiong Junjie Zhang Junwu Ma Shijun Xiao Lusheng Huang Bin Yang | 2021 | Science China(Life Sciences)2021,64,10: | 5 |

| 7 | Ascorbate peroxidase 1 confers resistance to southern corn leaf blight in maize显示文摘Southern corn leaf blight (SCLB), caused by Bipolarismaydis, is one of the most devastatingdiseases affecting maize production. However,only one SLCB resistance gene, conferring partialresistance, is currently known, underscoring theimportance of isolating new SCLB resistancerelatedgenes. Here, we performed a comparativeproteomic analysis and identified 258 proteinsshowing differential abundance during the maizeresponse to B. maydis. These proteins included anascorbate peroxidase (Zea mays ascorbate peroxidase1 (ZmAPX1)) encoded by a gene locatedwithin the mapping interval of a previously identifiedquantitative trait locus associated with SCLBresistance. ZmAPX1 overexpression resulted inlower H_(2)O_(2) accumulation and enhanced resistanceagainst B. maydis. Jasmonic acid (JA)contents and transcript levels for JA biosynthesisand responsive genes increased in ZmAPX1-overexpressing plants infected with B. maydis,whereas Zmapx1 mutants showed the oppositeeffects. We further determined that low levels of H_(2)O_(2) are accompanied by an accumulation of JAthat enhances SCLB resistance. These resultsdemonstrate that ZmAPX1 positively regulatesSCLB resistance by decreasing H_(2)O_(2) accumulationand activating the JA-mediated defensesignaling pathway. This study identified ZmAPX1as a potentially useful gene for increasing SCLBresistance. | 8 | Mycobacterium tuberculosis latency-associated antigen Rv1733c SLP improves the accuracy of differential diagnosis of active tuberculosis and latent tuberculosis infection显示文摘Background: Differential diagnosis of active tuberculosis (ATB) and latent tuberculosis infection (LTBI) has been a challenge for clinicians in high TB burden countries. The purpose of this study was to improve the accuracy of differential diagnosis of ATB and LTBI by using fluorescent immunospot (FluoroSpot) assay to detect specific Th1 cell immune responses. The novelmycobacterium tuberculosis (MTB) latency-associated antigens Rv1733c and synthetic long peptides derived from Rv1733c (Rv1733c SLP) were used based on virulence factors early secreting antigen target-6 (ESAT-6) and culture filtrate protein-10 (CFP-10).Methods: Fifty-seven ATB cases, including 20 pathogen-confirmed ATB and 37 clinically diagnosed ATB, and 36 LTBI cases, were enrolled between January and December 2017. FluoroSpot assay was used to detect the interferon γ (IFN-γ) and interleukin 2 (IL-2) secreted by the specific T cells after being stimulated with MTB virulence factors ESAT-6 and CFP-10, MTB latency-associated antigens Rv1733c and Rv1733c SLP. The receiver operating characteristic (ROC) curve was used to define the best cutoff value of latency-associated antigens in the use of differentiating ATB and LTBI. The sensitivity, specificity, predictive value, and likelihood ratio of ESAT-6 and CFP-10-FluoroSpot combined with latency-associated antigen in the differential diagnosis of ATB and LTBI were also calculated.Results: Following the stimulation with Rv1733c and Rv1733c SLP, the frequency of single IL-2-secreting T cells stimulated by Rv1733c SLP had the largest area under the ROC curve, which was 0.766. With a cutoff value of 1 (spot-forming cells [SFCs]/2.5 × 105 peripheral blood mononuclear cells) for frequency, the sensitivity and specificity of distinguishing ATB from LTBI were 72.2% and 73.7%, respectively. ESAT-6 and CFP-10-FluoroSpot detected the frequency and proportion of single IFN-γ-secreting T cells;the sensitivity and specificity of distinguishing ATB from LTBI were 82.5% and 66.7%, respectively. | 9 | An imputed whole-genome sequence-based GWAS approach pinpoints causal mutations for complex traits in a specific swine population显示文摘Sequencing-based genome-wide association studies(GWAS) have facilitated the identification of causal associations between genetic variants and traits in diverse species. However, it is cost-prohibitive for the majority of research groups to sequence a large number of samples. Here, we carried out genotype imputation to increase the density of single nucleotide polymorphisms in a large-scale Swine F;population using a reference panel including 117 individuals, followed by a series of GWAS analyses. The imputation accuracies reached 0.89 and 0.86 for allelic concordance and correlation, respectively. A quantitative trait nucleotide(QTN) affecting the chest vertebrate was detected directly, while the investigation of another QTN affecting the residual glucose failed due to the presence of similar haplotypes carrying wild-type and mutant allelesin the reference panel used in this study. A high imputation accuracy was confirmed by Sanger sequencing technology for the most significant loci. Two candidate genes,CPNE5 and MYH3, affecting meat-related traits were proposed. | 11 | A Fragment Substitution in Promoter of MS92/PTC1 Causes Male Sterility in Rice显示文摘Persistent tapetal cell1(PTC1) plays a curial role in pollen development, and is thought to function as a transcriptional activator in rice. However, the molecular mechanism of PTC1 in regulating pollen development and its cis-elements are not well understood. We identified a novel weak male sterility mutant(ms92) which exhibited expanded tapetum and shrink pollen grains. Map-based cloning and allelic analysis suggested that the male sterility of ms92 was caused by a DNA fragment substitution in the promoter of PTC1. The decreased expression of MS92/PTC1 in ms92 and cis-element analysis indicated that the substituted sequence contained several potential binding cis-element of negative feedback. MS92/PTC1 was specifically expressed in tapetum and microspores at the young microspore stage, and its protein was localized in nucleus. We further found that MS92/PTC1 functions as a transcription activator by recognizing H3K4me3. Transcriptomic analysis revealed that a number of genes involved in tapetum degeneration and pollen wall formation were down-regulated in ms92, which are the potential targets of MS92/PTC1. | 13 | Optimizing soil dissolved organic matter extraction by grey relational analysis显示文摘Dissolved organic matter(DOM)in soil plays an important role in the fate and transport o f contaminants.It is typically composed of many compounds,but the effect of different extraction factors on the abundance of different DOM components is unknown.In this study,DOM was extracted from three soils(paddy field,vegetable field and forest soils)with various extraction time,liquid to solid ratios(LSRs).extractant types,and extractant concentrations.The LSR had a significant effect on DOM content,which increased by 0.5-4.0 times among the three soils when LSR increased from 2:1 to 10:1(P<0.05).Dissolved organic matter content increased by 4%-53%when extraction time increased from 10 to 300 min(P<0.05).Extractant concentration had different effects on DOM content depending on the extractant.Higher concentrations of KC1 promoted DOM extraction,while higher concentrations o f KH2PO4 inhibited DOM extraction.Therefore,grey relational analysis was used to further quantitatively evaluate the effect of extraction time,LSR,and extractant concentration on DOM,using KC1 as an extractant.For the paddy field and forest soils,the impact of these three factors on DOM extraction efficiency was in the following order:KC1 concentration>LSR>extraction time.However,the effect was different for the vegetable field soil:LSR>extraction time>KCI concentration.Taking all these factors into account,1.50 mol L^-1 KC1 and an LSR of 10:1 with a shaking time of 300 min was recommended as the most appropriate method for soil DOM extraction. | Wenming XIE You MA Shijun LI Shanshan ZHANG Lin RUAN Mingyue YANG Weiming SHI Hailin ZHANG Limin ZHANG | 2020 | Pedosphere2020,30,5: | 1 |

| 20 | Do alternative stable states exist in large shallow Taihu Lake,China?显示文摘Regime shifts from submersed macrophyte dominance to phytoplankton dominance have been widely reported in small-to medium-sized shallow lakes.However,alternative stable states in large shallow lakes(surface area>500 km^(2))remain unconfirmed.To understand the alternative stable states and the main influencing factors of submersed macrophytes in large lakes,the ecosystem states from monitoring data from 1959 to 2019 in large shallow Taihu Lake(2338 km^(2)in average depth of 2.12 m)in China were examined.Changes in submersed macrophyte coverage(C_(Mac))and phytoplankton chlorophyll a(Chl a)in the time series and their relationships with environmental factors were analyzed.During the field investigation from August 2018 to May 2019,nutrients and Chl a showed obvious heterogeneity across the lake,being generally higher in the western and northern areas and lower in the southeast area,while C_(Mac)was only observed in the eastern areas,e.g.,East Taihu Lake,Xukou Bay,and Gonghu Bay.During the long-term monitoring from 1959 to 2019 in the Central Region,Meiliang Bay,and East Taihu Lake,Chl a increased significantly in the time series.C_(Mac)varied slightly among different subareas,always at low levels(<10%)in the Central Region and Meiliang Bay but at relatively high levels in East Taihu Lake(10%–90%).Frequency distributions of response variables had no multimodality except for C_(Mac)in East Taihu Lake,with two peaks between 15%and 20%and between 55%and 60%.A dual relationship was found between Chl a and total phosphorus(TP)in the areas with and without macrophytes,while C_(Mac)showed no relationship with TP,and submersed macrophytes did not flourish in the Central Region and Meiliang Bay even when TP was at very low levels(≈10 mg/m3).Taihu Lake had similar algal turbidity(TurbAlg)as small-to mediumsized lakes but generally presented with higher values of nonalgal turbidity(TurbNonAlg),as did their contribution to total turbidity as a percentage.This study suggested that large shallow Taihu Lake may have no alternative stable states,but more evidence is needed for East Taihu Lake,which was dominated by macrophytes,as it remains unknown whether hysteresis occurs between the processes of eutrophication and oligotrophication.Unfavorable conditions caused by wind might be the main reason due to the absence of submersed macrophytes in Taihu Lake.These results demonstrate that stricter nutrient control is needed to maintain a healthy state or to recover from a decayed state for large lakes. | Yan LI Yu MA Haijun WANG Hongzhu WANG Yongde CUI Shijun BIAN Miao ZHANG Mengmei LIU Yexin YU Marc SCHALLENBERG | 2023 | Journal of Oceanology and Limnology2023,41,3: | 0 |
