1Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy.Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma 2018Cancer Communications2018,38,1:6

9Constructing metal-free heterophotocatalyst using two-dimensional carbon nitride sheets and violet phosphorene for highly efficient visible-light photocatalysis显示文摘Two-dimensional(2D)carbon nitride sheets(CNs)with nanoscale thickness have great potential in solar energy conversion owing to their intrinsic bandgap.However,their photocatalytic efficiency falls far below the practical requirements because the generated electron-hole pairs rapidly recombine.We construct a metal-free 2D p–n junction heterophotocatalyst from CNs and violet phosphorene(VP)using a simple electrostatic self-assembly method.The experimental results and density functional theory calculations show that the built-in electric field of the p–n junction promotes photogenerated carrier transport at the heterojunction interface,thereby promoting the separation of photogenerated electron-hole pairs.Under visible-light illumination,the visible-light photocatalytic hydrogen and CO production rates of the CNs/VP heterojunction were increased by 5.85 and 1.51 times,respectively,when compared to that of CNs.This study provides new insights into the design of metal-free heterogeneous photocatalysts with high catalytic activity。Yong Wang Chengxin Zeng Liting Wu Yalin Dong Yu Zhang Dingyi Yang Wen Hu Jian Hao Hongzhe Pan Rusen Yang 2023Journal of Materials Science & Technology2023,,15:0
10SARS-CoV-2 spike-specific TFH cells exhibit unique responses in infected and vaccinated individuals显示文摘Long-term humoral immunity to SARS-CoV-2 is essential for preventing reinfection. The production of neutralizing antibody (nAb)and B cell differentiation are tightly regulated by T follicular help (T_(FH)) cells. However, the longevity and functional role of T_(FH) cellsubsets in COVID-19 convalescents and vaccine recipients remain poorly defined. Here, we show that SARS-CoV-2 infection andinactivated vaccine elicited both spike-specific CXCR3^(+) T_(FH) cell and CXCR3^(-) T_(FH) cell responses, which showed distinct responsepatterns. Spike- specific CXCR3^(+) T_(FH) cells exhibit a dominant and more durable response than CXCR3^(-) T_(FH) cells that positivelycorrelated with antibody responses. A third booster dose preferentially expands the spike-specific CXCR3^(+) T_(FH) cell subset inducedby two doses of inactivated vaccine, contributing to antibody maturation and potency. Functionally, spike-specific CXCR3^(+) T_(FH) cellshave a greater ability to induce spike-specific antibody secreting cells (ASCs) differentiation compared to spike-specific CXCR3^(-) T_(FH)cells. In conclusion, the persistent and functional role of spike-specific CXCR3^(+) T_(FH) cells following SARS-CoV-2 infection andvaccination may play an important role in antibody maintenance and recall response, thereby conferring long-term protection. 12Notoginsenoside R1 Protects against Diabetic Nephropathy through TXNIP-NLRP3 Signaling Pathway显示文摘Background:Diabetic nephropathy(DN)is a microvascular complication of diabetes mellitus(DM).DN results from many factors,including changes in glomerular hemodynamics,oxidative stress and inflammation,and in-terstitial fibrosis and tubular atrophy.Panax notoginseng,a commonly used Chinese medicine,has been used in the treatment of kidney disease.Notoginsenoside R1(NGR1),the main compound isolated from P.notoginseng,has been reported to have a renoprotective role in DN.However,the therapeutic effect and mechanism of NGR1 in DN remain unclear.Objective:The present study aimed to investigate the therapeutic effect and mechanism of NGR1 in DN.Methods:In this study,mouse podocytes(MPC-5 cells)and db/db mice were used to investigate the effect of NGR1 on DN in vitro and in vivo,respectively.Blood glucose,renal function,inflammatory factors,and PI3K/AKT-Nrf2-NLRP3 signaling pathway proteins were assessed.Results:The study results indicated that NGR1 reversed cell viability induced by high glucose(HG,30 mM).The related mechanism results showed that NGR1 decreased oxidative stress by inhibiting reactive oxygen species(ROS)level and upregulating the expression of Nrf2,NQO1,and HO-1 via TXNIP targeting.In addition,NLRP3 inflammasome and PI3K/AKT were engaged in NGR1-based protection against HG-stimulated podocytes.In db/db mice,NGR1 significantly decreased blood glucose,urine protein,urine output,blood urea nitrogen,and other parameters as well as reversed kidney injury by inhibiting oxidative stress and proinflammatory response.Conclusion:Taken together,this study revealed that NGR1 exerted a significant therapeutic effect on DN both in vitro and in vivo via a mechanism related to the TXNIP-Nrf2 pathway and NLRP3 inflammasome,suggesting that NGR1 is a potential therapeutic option for DN.Chunting Zhang Renyikun Yuan Siyuan Li Guodong Huang Kaili Sun Jiaping Pan Qiuxia Liu Xiang Gao Zhijing Wang Tongyu Li Shilong Lu Jianzhen Lv Liting Huang Hongwei Gao 2023Clinical Complementary Medicine and Pharmacology2023,3,4:0
13Temperature-regulation liquid gating membrane with controllable gas/liquid separation显示文摘Membrane separation technology with the ability to regulate gas/liquid transport and separation is critical for environmental fields, such as sewerage treatment, multiphase separation, and desalination. Although numerous membranes can dynamically control liquid-phase fluids transport via external stimuli, the transport and separation of gas-phase fluids remains a challenge. Here, we show a temperature-regulation liquid gating membrane that allows in-situ dynamically controllable gas/liquid transfer and multiphase separation by integrating a thermo-wettability responsive porous membrane with functional gating liquid. Experiments and theoretical analysis have demonstrated the temperature-regulation mechanism of this liquid gating system, which is based on thermo-responsive changes of porous membrane surface polarity, leading to changes in affinity between the porous membrane and the gating liquid. In addition, the sandwich configuration with dense Au-coated surfaces and heterogeneous internal components by a bistable interface design enables the liquid gating system to enhance response sensitivity and maintain working stability.
