1Proteomic Analysis of Macrophages:A Potential Way to Identify Novel Proteins Associated with Activation of Macrophages for Tumor Cell Killing显示文摘One major mechanism through which macrophages effectively kill tumor cells requires cell to cell contact,indicating that certain molecules expressed on cell surface of activated macrophages may mediate the tumoricidal capability. Tumor necrosis factor (TNF) and nitric oxide (NO) are the two classical mediators of tumor cell death. However,evidence of discrepancy is accumulating indicating these known mediators do not appear to account for the broad and potent tumoricidal activity of macrophages. To obtain a full repertoire of tumoricidal activation-associated membrane proteins,we combined one-dimensional SDS-PAGE with capillary liquid chromatography-tandem mass spectrometry (LC-MS/MS). Using this technique,we identified 454 activated macrophage specifically expressed proteins with extremely high confidence,including most known activation markers of macrophages,such as NO synthase (iNOS),Ym1,cyclooxygenase,etc. Membrane bound TNF-α was also identified on activated macrophages. However,it was also detected on thioglycolate elicited macrophages,indicating this molecule may not play a key role in conjugation-dependent tumor cell killing. In contrast,although NO has not been assigned as an effector molecule of conjugation-dependent tumoricidal pathway,iNOS was identified from membrane fraction of activated macrophages,suggesting NO may be involved in conjugation-dependent tumoricidal mechanism,because iNOS association with plasma membrane is ideally suited to deliver NO directly into the contacted tumor cells. 2Histopathology and Ultrastructural Pathology of Cyprinid Herpesvirus Ⅱ(CyHV-2) Infection in Gibel Carp, Carassius auratus gibelio显示文摘Cyprinid herpesvirus II(Cy HV-2) infection is identified in cultured gibel carp, Carassius auratus gibelio, with high mortality in China in recent years. Histological pathology includes acute hepatocellular necrosis, splenic necrosis, kidney necrosis, hyperplasia of the secondary lamellae with focal necrosis. Acute necrotic myocarditis and granulocytes are prominent within the cardiac lumen in infected fish. In addition, necrosis is observed in the submucosa and mucosa epithelium of intestinal tract. Edemas are observed in renal glomerulus, submucosa and mucosa epithelium of intestinal tract, myocardial cells and neurons. Transmission electron microscopy indicates the cytoplasmic inclusions in splenocytes, glomerulus cells and hematopoietic tissue cells of kidney, epithelial cells of gills and brain cells. 3Inhibition of Herpes Simplex Virus-1 Replication by Natural Compound Honokiol显示文摘Honokiol is a pleiotropic natural compound isolated from Magnolia and has multiple biological and clinically relevant effects,including anticancer and antimicrobial function.However,the antiviral activity of honokiol has not yet been well studied.Here we showed that honokiol had no effect on herpes simplex virus-1(HSV-1)entry,but inhibited HSV-1 viral DNA replication,gene expression and the production of new progeny viruses.The combination of honokiol and clinical drug acyclovir augmented inhibition of HSV-1 infection.Our results illustrate that honokiol could be a potential new candidate for clinical consideration in the treatment of HSV-1 infection alone or combination with other therapeutics.Shuai Liu Long Li Lingbing Tan Xiaozhen Liang 2019Virologica Sinica2019,34,3:4

5High prevalence of pathogenic Leptospira in wild and domesticated animals in an endemic area of China显示文摘Objective:To assess the prevalence of Leptospira detected in wildlife and domesticated animals in Jiangxi Province,China,in 2009.Methods:Urine samples from 28 buffaloes and kidney samples from 50 pigs,50 dogs and 38 rats were collected from Fuliang and Shangrao County,Jiangxi Province,China,in October 2009.Polymerase chain reaction(PCR) and culture analyses were used to detect Leptospira.The cultured isolates were typed using the microscopic agglutination lest(MAT).Results:The results showed that rats potentially serve as the main reservoir of leptospiral infection,followed by dogs.Although 16%of rats(6/38) were positive using culture analysis,PCR analysis using the diagnostic primers G1/G2 and B64Ⅰ/B64Ⅱor lipLS2 showed identification as 50%and 24%,respectively,of the rat samples as positive for the presence of leptospiral DNA.Conclusions:PCR-based detection of leptospiral DNA in infected kidney tissues of reservoirs is more efficient when using G1/G2 primers than lipL32 primers. 6MEB-YOLO: An Efficient Vehicle Detection Method in Complex Traffic Road Scenes显示文摘Rapid and precise vehicle recognition and classification are essential for intelligent transportation systems,and road target detection is one of the most difficult tasks in the field of computer vision.The challenge in real-time road target detection is the ability to properly pinpoint relatively small vehicles in complicated environments.However,because road targets are prone to complicated backgrounds and sparse features,it is challenging to detect and identify vehicle kinds fast and reliably.We suggest a new vehicle detection model called MEB-YOLO,which combines Mosaic and MixUp data augmentation,Efficient Channel Attention(ECA)attention mechanism,Bidirectional Feature Pyramid Network(BiFPN)with You Only Look Once(YOLO)model,to overcome this problem.Four sections make up this model:Input,Backbone,Neck,and Prediction.First,to improve the detection dataset and strengthen the network,MixUp and Mosaic data improvement are used during the picture processing step.Second,an attention mechanism is introduced to the backbone network,which is Cross Stage Par-tial Darknet(CSPDarknet),to reduce the influence of irrelevant features in images.Third,to achieve more sophisticated feature fusion without increasing computing cost,the BiFPN structure is utilized to build the Neck network of the model.The final prediction results are then obtained using Decoupled Head.Experiments demonstrate that the proposed model outperforms several already available detection methods and delivers good detection results on the University at Albany DEtection and TRACking(UA-DETRAC)public dataset.It also enables effective vehicle detection on real traffic monitoring data.As a result,this technique is efficient for detecting road targets.Yingkun Song Shunhe Hong Chentao Hu Pingan He Lingbing Tao Zhixin Tie Chengfu Ding 2023Computers, Materials & Continua2023,,6:2

12Germ cell sex prior to meiosis in the rainbow trout显示文摘Germ cells make two major decisions when they move from an indeterminate state to their final stage of gamete production.One decision is sexual commitment for sperm or egg production,and the other is to maintain mitotic division or entry into meiosis.It is unclear whether the two decisions are made as a single event or separate events,because there has been no evidence for the presence of germ cell sex prior to meiosis.Here we report direct evidence in the fish rainbow trout that gonia have distinct sexuality.We show that dazl expression occurs in both male and female gonia but exhibits differential intracellular distribution.More strikingly,we show that boule is highly expressed in male gonia but absent in female gonia.Therefore,mitotic gonia possess sex,sperm/egg decision and mitosis/meiosis decision are two independent events,and sperm/egg decision precedes mitosis/meiosis decision in rainbow trout,making this organism a unique vertebrate model for mechanistic understanding of germ cell sex differentiation and relationship between the two decisions.Mingyou Li Qian Shen Foong Mei Wong Hongyan Xu Ni Hong Lingbing Zeng Lin Liu Qiwei Wei Yunhan Hong 2011Protein & Cell2011,2,1:1

20Learning to Complete Knowledge Graphs with Deep Sequential Models显示文摘Knowledge graph (KG) completion aims at filling the missing facts in a KG, where a fact is typically represented as a triple in the form of (head, relation, tail). Traditional KG completion methods compel two- thirds of a triple provided (e.g., head and relation) to predict the remaining one. In this paper, we propose a new method that extends multi-layer recurrent neural networks (RNNs) to model triples in a KG as sequences. It obtains state-of-the-art performance on the common entity prediction task, i.e., giving head (or tail) and relation to predict the tail (or the head), using two benchmark data sets. Furthermore, the deep sequential characteristic of our method enables it to predict the relations given head (or tail) only, and even predict the whole triples.
