1The vernalization-induced long non-coding RNA VAS functions with the transcription factor TaRF2b to promote TaVRN1 expression for flowering in hexaploid wheat显示文摘Vernalization is a physiological process in which prolonged cold exposure establishes flowering competence in winter plants. In hexaploid wheat, TaVRN1 is a cold-induced key regulator that accelerates floral transition. However, the molecular mechanism underlying the gradual activation of TaVRN1 during the vernalization process remains unknown. In this study, we identified the novel transcript VAS (TaVRN1 alternative splicing) as a non-coding RNA derived from the sense strand of the TaVRN1 gene only in winter wheat, which regulates TaVRN1 transcription for flowering. VAS was induced during the early period of vernalization, and its overexpression promoted TaVRN1 expression to accelerate flowering in winter wheat. VAS physically associates with TaRF2b and facilitates docking of the TaRF2b-TaRF2a complex at the TaVRN1 promoter during the middle period of vernalization. TaRF2b recognizes the Sp1 motif within the TaVRN1 proximal promoter region, which is gradually exposed along with the disruption of a loop structure at the TaVRN1 locus during vernalization, to activate the transcription of TaVRN1. The tarf2b mutants exhibited delayed flowering, whereas transgenic wheat lines overexpressing TaRF2b showed earlier flowering. 5Cyclic di-adenosine monophosphate regulates the osteogenic and adipogenic differentiation of hPDLSCs via MAPK and NF-κB signaling显示文摘Cyclic di-adenosine monophosphate(c-di-AMP)is a bacterial second messenger that can be recognized by infected host cells and activate the immunoinflammatory response.The purpose of this study is to demonstrate the effect of c-di-AMP on the differentiation of human periodontal ligament stem cells(hPDLSCs)and its underlying mechanisms.In the present study,we find that the gingival crevicular fluid(GCF)of patients with chronic periodontitis has a higher expression level of c-di-AMP than that of healthy people.In vitro,c-di-AMP influences the differentiation of hPDLSCs by upregulating Toll-like receptors(TLRs);specifically,it inhibits osteogenic differentiation by activating NF-κB and ERK/MAPK and promotes adipogenic differentiation through the NF-κB and p38/MAPK signaling pathways.Inhibitors of TLRs or activated pathways reduce the changes induced by c-di-AMP.Our results establish the potential correlation among bacterial c-di-AMP,periodontal tissue homeostasis and chronic periodontitis pathogenesis.Sirui Chen Zuping Wu Yuying He Li Zhu Jiahe Wang Hengyi Lin Jing Xie Chenchen Zhou Shujuan Zou 2023Acta Biochimica et Biophysica Sinica2023,55,3:1

7Downregulation of MANCR inhibits cancer cell proliferation in mantle cell lymphoma possibly by interacting with RUNX2显示文摘The mitotically associated lncRNA(MANCR)participates in breast cancer cell proliferation,while its involvement in other cancers is still unknown.In this study,we therefore studied the role of MANCR in mantle cell lymphoma(MCL).We found that serum MANCR and Runt-related transcription factor 2(RUNX2)were upregulated in MCL patients when compared with those in healthy controls.A positive correlation between serum MANCR and RUNX2 was found in MCL patients but not in controls.Upregulation of serum MANCR distinguished MCL patients from controls.MANCR overexpression promoted RUNX2 expression in MCL cells,while RUNX2 overexpression failed to significantly change the expression levels of MANCR.MANCR overexpression promoted the proliferation of MCL cells,while MANCR silencing inhibited the proliferation of MCL cells.In addition,RUNX2 overexpression attenuated the inhibitory effects of MANCR silencing on cell proliferation.However,MANCR overexpression and silencing had no significant effects on cell migration and invasion.Further bioinformatics analysis showed that MANCR may sponge miR-218 to upregulate RUNX2.Therefore,we conclude that downregulation of MANCR may inhibit cancer cell proliferation in MCL possibly by interacting with RUNX2.Shujuan Wen Min Zeng Yan Li Xin Hu Shan Li Xiao Liang Lin Zhu Shune Yang 2019Acta Biochimica et Biophysica Sinica2019,51,11:1

12Three Novel Mutations of APC Gene Found in a Chinese Family with Familial Adenomatous Polyposis显示文摘Objective:To identify the causative adenomatous polyposis coli(APC)gene defects associated with a pedigree of familial adenomatous polyposis(FAP).Methods:FAP was diagnosed based on clinical manifestations,family history,as well as endoscopic and pathological examinations.The blood samples of the FAP pedigree members,colonic polyp patients,and normal individuals were collected.Genomic DNA was then extracted from those samples.APC mutation analysis was conducted via direct polymerase chain reaction(PCR)sequencing.Results:Three synonymous mutations and a missense mutation were found:c.5034G>A(p.Glyl678Gly),c.5465T>A(p.Vall 822Asp),c.5880G>A(p.Prol960Pro),and c.5274T>G(p.Serl758Ser)・Among them,the homozygous mutation on APC gene c.5034G>A has been reported,while the other three mutations have not been reported in the Chinese Han population.Individuals with c.5465T>A(p.Vall822ASP)missense mutation eventually suffer from colon cancer and have poor prognosis.We found no mutation in patients with simple intestinal polyp and in normal individuals.In addition,there were homozygous and heterozygous mutations in different patients from the same family.Conclusion:Three new mutations of APC gene were firstly reported in Han population.The missense mutation of c.5465T>A(p.Vall 822Asp)may be the cause of carcinogenesis in this FAP pedigree with poor prognosis.Shujuan Gao Min Lin Yan Jin Zhuona Wang Yunqing Zhu Guisheng Liu Xueyan Guo 2022Journal of Clinical and Nursing Research2022,6,3:0
13Analysis of the effects of antifungal peptide P-1 from Bacillus pumilus HN-10 on energy metabolism of Trichothecium roseum显示文摘This study investigated the antifungal activity and possible mode of action of Bacillus pumilus HN-10 antifungal peptide P-1 against Trichothecium roseum.The results showed that the antifungal peptide P-1 at a concentration of 1.0μg mL^(-1)had strong antifungal activity against T.roseum.P-1 inhibited the tricarboxylic acid cycle(TCA)pathway and the transporter pathway of NADH to coenzyme Q on the electron transport chain.P-1 significantly reduced succinate dehydrogenase(SDH),malate dehydrogenase(MDH),ATPase,mitochondrial complex enzymes I,II and IV enzyme activities on the electron transport chain,and 5'-triphosphate(ATP),5'-diphosphate(ADP),5'-monophosphate(AMP)content,and energy charge(EC);significantly increased 6-phosphofructokinase(PFK)enzyme activity.The release of Ca^(2+)(OD_(680))from the inner mitochondrial membrane and the openness of the mitochondrial permeability transition pore(MPTP)were analysed,and microscopy was performed following staining of mitochondria with JC-1.The results indicated that P-1 significantly increased the release of Ca^(2+) and the openness of MPTP,decreased the mitochondrial membrane potential,and produced green fluorescence;transcriptomics data analysis showed that there were 39 differentially expressed genes(DEGs)related to energy metabolism enzymes.The results verified by qRT-PCR were basically consistent with the transcriptome sequencing results.Thus,P-1 achieved its inhibitory effect mainly by regulating genes related to energy metabolism.Shujuan Wu Jianmin Yun Rui Wang Wenwei Zhang Lin Hao Pengzheng Pei 2022Food Bioscience2022,47,3:0
14SARS-CoV-2-specific antibody response characteristics in COVID-19 patients of different ages显示文摘Age has been found to be one of the main risk factors for the severity and outcome of COVID-19.However,differences in SARS-CoV-2 specific antibody responses among COVID-19 patients of different age groups remain largely unknown.In this study,we analyzed the IgG/IgM responses to 21 SARS-CoV-2 proteins and 197 peptides that fully cover the spike protein against 731 sera collected from 731 COVID-19 patients aged from 1 to 92 years.We show that there is no overall difference in SARS-CoV-2 antibody responses in COVID-19 patients in the 4 age groups.By antibody response landscape maps,we find that the IgG response profiles of SARS-CoV-2 proteins are positively correlated with age.The S protein linear epitope map shows that the immunogenicity of the S-protein peptides is related to peptide sequence,disease severity and age of the COVID-19 patients.Furthermore,the enrichment analysis indicates that low S1 IgG responses are enriched in patients aged<50 and high S1 IgG responses are enriched in mild COVID-19 patients aged>60.In addition,high responses of non-structural/accessory proteins are enriched in severe COVID-19 patients aged>70.These results suggest the distinct immune response of IgG/IgM to each SARS-CoV-2 protein in patients of different age,which may facilitate a deeper understanding of the immune responses in COVID-19 patients.Linfang Lu Siqi Yu Min Liu Yang Li Qing Lei Mingxi Lin Danyun Lai Shujuan Guo Hewei Jiang Hongyan Hou Yunxiao Zheng Xuening Wang Mingliang Ma Bo Zhang Hong Chen Junbiao Xue Hainan Zhang Huan Qi Ziyong Sun Feng Wang Xionglin Fan Zhaowei Xu 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
15Two-dimensional blue-phase CX(X=S,Se)monolayers with high carrier mobility and tunable photocatalytic water splitting capability显示文摘Photocatalytic water splitting utilizing solar energy is considered as one of the most ideal strategies for solving the ene rgy and environmental issues.Recently,two-dimensional(2 D)materials with an intrinsic dipole show great chance to achieve excellent photocatalytic performance.In this work,blue-phase monolayer carbon monochalcogenides(CX,X=S,Se)are constructed and systematically studied as photocatalysts for water splitting by performing first-principles calculations based on density functional theory.After confirming the great dynamical,thermal,and mechanical stability of CX monolayers,we observe that they possess moderate band gaps(2.41 eV for CS and 2.46 eV for CSe)and high carrier mobility(3.23×10^(4)cm^(2)V^(^(-1))s^(-1)for CS and 4.27×10^(3)cm^(2)V^(-1)s^(-1)for CSe),comparable to those of many recently reported 2 D photocatalysts.Moreover,these two monolayer materials are found to have large intrinsic dipole(0.43 D for CS and 0.51 D for CSe),thus the build-in internal electric field can be selfintroduced,which can effectively drive the separation of photongenerated carriers.More importantly,the well-aligned band edge as well as rather pronounced optical absorption in the visible-light and ultraviolet regions further ensure that our proposed CX monolayers can be used as high efficient photocatalysts for water splitting.Additionally,the effects of external strain on the electronic,optical and photocatalytic properties of CX monolayers are also evaluated.These theoretical predictions will stimulate further work to open up the energy-related applications of CX monolayers.Shengcai Li Mengyao Shi Jiahui Yu Shujuan Li Shulai Lei Liangxu Lin Jiajun Wang 2021Chinese Chemical Letters2021,32,6:0
16Structural insights of catalytic intermediates in dialumene based CO_(2) capture: Evidences from theoretical resonance Raman spectra显示文摘CO_(2) capture is considered as one of the most ideal strategies for solving the environmental issues and against global warming.Recently,experimental evidence has suggested that aluminum double bond(dialumene) species can capture CO_(2) and further convert it into value-added products.However,the catalytic application of these species is still in its infancy.Both the dynamics mechanism of CO_(2) fixation and the detailed structures of catalytic intermediates are not well understood.In this work,we investigate the structure dependent resonance Raman(RR) signals for different reaction intermediates.Ab-initio simulations of spontaneous resonance Raman(spRR) and time-domain stimulated resonance Raman(stRR) give spectral signatures correlated to the existence of different intermediates during the CO_(2)-dialumene binding process.The unique Raman vibronic feature s contain rich structural information with high temporal resolution,enabling to monitor the transient catalytic intermediates under reaction conditions.Our work shows that RR can be used to monitor intermediates during the dialumene based CO_(2) capture reaction.The spectral features not only provide insight into the structural information of intermediate species,but also allow a deeper understanding of the dynamical details of this kind of catalytic process.Baoling Tian Shujuan Li Shulai Lei Liangxu Lin Wenyue Guo Hao Ren 2021Chinese Chemical Letters2021,32,8:0
