| 1 | The vernalization-induced long non-coding RNA VAS functions with the transcription factor TaRF2b to promote TaVRN1 expression for flowering in hexaploid wheat显示文摘Vernalization is a physiological process in which prolonged cold exposure establishes flowering competence in winter plants. In hexaploid wheat, TaVRN1 is a cold-induced key regulator that accelerates floral transition. However, the molecular mechanism underlying the gradual activation of TaVRN1 during the vernalization process remains unknown. In this study, we identified the novel transcript VAS (TaVRN1 alternative splicing) as a non-coding RNA derived from the sense strand of the TaVRN1 gene only in winter wheat, which regulates TaVRN1 transcription for flowering. VAS was induced during the early period of vernalization, and its overexpression promoted TaVRN1 expression to accelerate flowering in winter wheat. VAS physically associates with TaRF2b and facilitates docking of the TaRF2b-TaRF2a complex at the TaVRN1 promoter during the middle period of vernalization. TaRF2b recognizes the Sp1 motif within the TaVRN1 proximal promoter region, which is gradually exposed along with the disruption of a loop structure at the TaVRN1 locus during vernalization, to activate the transcription of TaVRN1. The tarf2b mutants exhibited delayed flowering, whereas transgenic wheat lines overexpressing TaRF2b showed earlier flowering. | 2 | Genome editing in plants with MAD7 nuclease显示文摘MAD7 is an engineered nuclease of the Class 2 type V-A CRISPR-Cas(Cas12 a/Cpf1)family with a low level of homology to canonical Cas12 a nucleases.It has been publicly released as a royalty-free nuclease for both academic and commercial use.Here,we demonstrate that the CRISPR-MAD7 system can be used for genome editing and recognizes T-rich PAM sequences(YTTN)in plants.Its editing efficiency in rice and wheat is comparable to that of the widely used CRISPR-Lb Cas12 a system.We develop two variants,MAD7-RR and MAD7-RVR that increase the target range of MAD7,as well as an M-AFID(a MAD7-APOBEC fusion-induced deletion)system that creates predictable deletions from 50-deaminated Cs to the MAD7-cleavage site.Moreover,we show that MAD7 can be used for multiplex gene editing and that it is effective in generating indels when combined with other CRISPR RNA orthologs.Using the CRISPR-MAD7 system,we have obtained regenerated mutant rice and wheat plants with up to 65.6%efficiency. | Qiupeng Lin Zixu Zhu Guanwen Liu Chao Sun Dexing Lin Chenxiao Xue Shengnan Li Dandan Zhang Caixia Gao Yanpeng Wang Jin-Long Qiu | 2021 | Journal of Genetics and Genomics2021,48,6: | 4 |
| 3 | Seasonal variations of air-sea heat fluxes and sea surface temperature in the northwestern Pacific marginal seas显示文摘Using a net surface heat flux(Qnet) product obtained from the objectively analyzed air-sea fluxes(OAFlux) project and the international satellite cloud climatology project(ISCCP), and temperature from the simple ocean data assimilation(SODA), the seasonal variations of the air-sea heat fluxes in the northwestern Pacific marginal seas(NPMS) and their roles in sea surface temperature(SST) seasonality are studied. The seasonal variations of Qnet, which is generally determined by the seasonal cycle of latent heat flux(LH), are in response to the advection-induced changes of SST over the Kuroshio and its extension. Two dynamic regimes are identified in the NPMS: one is the area along the Kuroshio and its extension, and the other is the area outside the Kuroshio. The oceanic thermal advection dominates the variations of SST and hence the sea-air humidity plays a primary role and explains the maximum heat losing along the Kuroshio. The heat transported by the Kuroshio leads to a longer period of heat losing over the Kuroshio and its Extension. Positive anomaly of heat content corresponds with the maximum heat loss along the Kuroshio. The oceanic advection controls the variations of heat content and hence the surface heat flux. This study will help us understand the mechanism controlling variations of the coupled ocean-atmosphere system in the NPMS. | 6 | The preliminary study of the high chlorophyll in the central Bohai Sea in summer显示文摘In the central Bohai Sea, both historical data and in situ observations show that the distribution of high chlorophyll concentration is consistent with warm water. The previous studies confirm that the warm water is caused by a joint effect of wave and tidal mixing. In this paper, the authors have coupled the Princeton ocean model (POM) with a biochemical model to further investigate the formation of the high chlorophyll concentration. When the wave mixing is excluded, the water temperature is lower and the chlorophyll concentration is also lower than the observation. However, if the wave mixing is included, strong wave mixing in the upper layer brings the warm water downward and nutrients upward so that both temperature and nutrients tend to be uniformly high from the surface to the bottom. This provides an environment which is favorable for the growth of the phytoplankton. Therefore the chlorophyll concentration, as a symbol of phytoplankton, shows high value in the central Bohai Sea. | 7 | Analyses of the predicted changes of the global oceans under the increased greenhouse gases scenarios显示文摘A new climate model (ECHAM5/MPI-OM1) developed for the fourth assessment report of the Intergovernmental Panel on Climate Change (IPCC) at Max-Planck Institute for Meteorology is used to study the climate changes under the different increased CO2 scenarios (B1, A1B and A2). Based on the corresponding model results, the sea surface temperature and salinity structure, the variations of the thermohaline circulation (THC) and the changes of sea ice in the northern hemisphere are analyzed. It is concluded that from the year of 2000 to 2100, un- der the B1, A1B and A2 scenarios, the global mean sea surface temperatures (SST) would increase by 2.5℃, 3.5℃ and 4.0℃ respectively, especially in the region of the Arctic, the increase of SST would be even above 10.0℃; the maximal negative value of the variation of the fresh water flux is located in the sub- tropical oceans, while the precipitation in the eastern tropical Pacific increases. The strength of THC de- creases under the B1, A1B and A2 scenarios, and the reductions would be about 20%, 25% and 25.1% of the present THC strength respectively.
