4Targeting ferroptosis suppresses osteocyte glucolipotoxicity and alleviates diabetic osteoporosis显示文摘Diabetic osteoporosis(DOP) is the leading complication continuously threatening the bone health of patients with diabetes. A key pathogenic factor in DOP is loss of osteocyte viability. However, the mechanism of osteocyte death remains unclear. Here, we identified ferroptosis, which is iron-dependent programmed cell death, as a critical mechanism of osteocyte death in murine models of DOP. The diabetic microenvironment significantly enhanced osteocyte ferroptosis in vitro, as shown by the substantial lipid peroxidation, iron overload, and aberrant activation of the ferroptosis pathway. RNA sequencing showed that heme oxygenase-1(HO-1) expression was notably upregulated in ferroptotic osteocytes. Further findings revealed that HO-1 was essential for osteocyte ferroptosis in DOP and that its promoter activity was controlled by the interaction between the upstream NRF2 and c-JUN transcription factors. Targeting ferroptosis or HO-1 efficiently rescued osteocyte death in DOP by disrupting the vicious cycle between lipid peroxidation and HO-1 activation, eventually ameliorating trabecular deterioration. 5Brucine Inhibits Bone Metastasis of Breast Cancer Cells by Suppressing Jagged1/Notch1 Signaling Pathways显示文摘Objective: To examine the effects of brucine on the invasion, migration and bone resorption of receptor activator of nuclear factor-kappa B ligand(RANKL)-induced osteoclastogenesis. Methods: The osteoclastogenesis model was builded by co-culturing human breast tumor MDA-MB-231 and mouse RAW264.7 macrophages cells. RANKL(50 ng/m L) and macrophage-colony stimulating factor(50 ng/m L) were added to this system, followed by treatment with brucine(0.02, 0.04 and 0.08 mmol/L), or 10 μmol/L zoledronic acid as positive control. The migration and bone resorption were measured by transwell assay and in vitro bone resorption assay. The protein expressions of Jagged1 and Notch1 were investigated by Western blot. The expressions of transforming growth factor-β1(TGF-β1), nuclear factor-kappa B(NF-κB) and Hes1 were determined by enzyme-linked immunosorbent assay. Results: Compared with the model group, brucine led to a dose-dependent decrease on migration of MDA-MB-231 cells, inhibited RANKL-induced osteoclastogenesis and bone resorption of RAW264.7 cells(P <0.01). Furthermore, brucine decreased the protein levels of Jagged1 and Notch1 in MDA-MB-231 cells and RAW264.7 cells co-cultured system as well as the expressions of TGF-β1, NF-κB and Hes1(P<0.05 or P<0.01). 7Full-thickness tissue engineered skin constructed with autogenic bone marrow mesenchymal stem cells显示文摘To explore the feasibility of repairing clinical cutaneous deficiency, autogenic bone marrow mesen-chymal stem cells (BMSCs) were isolated and differentiated into epidermal cells and fibroblasts in vitro supplemented with different inducing factors and biomaterials to construct functional tissue- engineered skin. The results showed that after 72 h induction, BMSCs displayed morphologic changes such as typical epidermal cell arrangement, from spindle shape to round or oval; tonofibrils, melano-somes and keratohyaline granules were observed under a transmission electronic microscope. The differentiated cells expressed epidermal stem cell surface marker CK19 (59.66% ± 4.2%) and epidermal cells differentiation marker CK10. In addition, the induced epidermal cells acquired the anti-radiation capacity featured by lowered apoptosis following exposure to UVB. On the other hand, the collagen microfibrils deposition was noticed under a transmission electronic microscope after differentiating into dermis fibroblasts; RT-PCR identified collagen type I mRNA expression in differentiated cells; radioimmunoassay detected the secretion of interleukin-6 (IL-6) and interleukin-8 (IL-8) (up to 115.06 pg/mL and 0.84 ng/mL, respectively). Further in vivo implanting BMSCs with scaffold material short-ened skin wound repair significantly. 8Overexpression of HMGB1 A-box reduced lipopolysaccharide-induced intestinal inflammation via HMGB1/TLR4 signaling in vitro显示文摘AIM: To investigate the inhibitory effects and mechanism of high mobility group box(HMGB)1 A-box in lipopolysaccharide(LPS)-induced intestinal inflammation.METHODS: Overexpression of HMGB1 A-box in human intestinal epithelial cell lines(SW480 cells) was achieved using the plasmid p EGFP-N1. HMGB1 A-box-overexpressing SW480 cells were stimulated with LPS and co-culturing with human monocyte-like cell lines(THP-1 cells) using a Transwell system, compared with another HMGB1 inhibitor ethyl pyruvate(EP). The m RNA and protein levels of HMGB1/toll-like receptor(TLR) 4 signaling pathways [including HMGB1, TLR4, myeloid differentiation factor88(MYD88), Phosphorylated Nuclear Factor κB(p NF-κB) p65] in the stimulated cells were determined by realtime polymerase chain reaction and Western blotting. The levels of the proinflammatory mediators [including HMGB1, interleukin(IL)-1β, IL-6 and tumor necrosis factor(TNF)-α] in the supernatants of the stimulated cells were determined by ELISA.RESULTS: EP downregulated the m RNA and protein levels of HMGB1, inhibited the TLR4 signaling pathways(TLR4, MYD88 and p NF-κB p65) and reduced the secretion of proinflammatory mediators(HMGB1, IL-1β, IL-6 and TNF-α) in the SW480 and THP-1 cells activated by LPS but not in the unstimulated cells. Activated by LPS, the overexpression of HMGB1 A-box in the SW480 cells also inhibited the HMGB1/TLR4 signaling pathways and reduced the secretion of these proinflammatory mediators in the THP-1 cells but not in the transfected and unstimulated cells. 11Effects of viral infection and microbial diversity on patients with sepsis:A retrospective study based on metagenomic next-generation sequencing显示文摘BACKGROUND: The study aims to investigate the performance of a metagenomic next-generationsequencing (NGS)-based diagnostic technique for the identifi cation of potential bacterial and viral infectionsand eff ects of concomitant viral infection on the survival rate of intensive care unit (ICU) sepsis patients.METHODS: A total of 74 ICU patients with sepsis who were admitted to our institution from February1, 2018 to June 30, 2019 were enrolled. Separate blood samples were collected from patients for bloodcultures and metagenomic NGS when the patients’ body temperature was higher than 38 °C. Patients’demographic data, including gender, age, ICU duration, ICU scores, and laboratory results, were recorded.The correlations between pathogen types and sepsis severity and survival rate were evaluated.RESULTS: NGS produced higher positive results (105 of 118;88.98%) than blood cultures(18 of 118;15.25%) over the whole study period. Concomitant viral infection correlated closelywith sepsis severity and had the negative effect on the survival of patients with sepsis. However,correlation analysis indicated that the bacterial variety did not correlate with the severity of sepsis.CONCLUSIONS: Concurrent viral load correlates closely with the severity of sepsis and thesurvival rate of the ICU sepsis patients. 13QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. 15Biochemical Mechanism of Chlorantraniliprole Resistance in the Diamondback Moth, Plutella xylostella Linnaeus显示文摘The insecticide chlorantraniliprole exhibits good efficacy and plays an important role in controlling the diamondback moth, Plutella xylostella Linnaeus. However, resistance to chlorantraniliprole has been observed recently in some field populations. At present study, diamondback moths with resistance to chlorantraniliprole(resistant ratio(RR) was 82.18) for biochemical assays were selected. The assays were performed to determine potential resistance mechanisms. The results showed that the selected resistant moths(GDLZ-R) and susceptible moth could be synergized by known metabolic inhibitors such as piperonyl butoxide(PBO), triphenyl phosphate(TPP) and diethyl-maleate(DEM) at different levels(1.68-5.50-fold and 2.20-2.89-fold, respectively), and DEM showed the maximum synergism in both strains. In enzymes assays, a high level of glutathione-S-transferase(GST) was observed in the resistant moth, in contrast, moths that are susceptible to the insecticide had only 1/3 the GST activity of the resistant moths. The analysis of short-term exposure of chlorantraniliprole on biochemical response in the resistant strain also showed that GST activity was significantly elevated after exposure to a sub-lethal concentration of chlorantraniliprole(about 1/3 LC50, 12 mg L-1) 12 and 24 h, respectively. 17Rationalizing phonon dispersion for lattice thermal conductivity of solids显示文摘Lattice thermal conductivity(κL) is one of the most fundamental properties of solids. The acoustic–elastic-wave assumption, proposed by Debye(Debye P. Ann Phys 1912; 344: 789–839), has led to linear phonon dispersion being the most common approximation for understanding phonon transport over the past century. Such an assumption does not take into account the effect of a periodic boundary condition on the phonon dispersion, originating from the nature of periodicity on atomic arrangements. Driven by modern demands on the thermal functionality of materials, with κLranging from ultra-low to ultra-high,any deviation from the Debye approximation in real materials becomes more and more significant. This work takes into account the periodic boundary condition, and therefore rationalizes the phonon dispersion to be more realistic. This significantly improves the precision for quickly predicting κLwithout any fitting parameters, as demonstrated in hundreds of materials, and offers a theoretical basis rationalizing κLto be lower than the minimum currently accepted based on the Debye dispersion. 20Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments,therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival,proliferation,apoptosis,autophagy of cardiomyocytes,endothelial cells,fibroblasts,monocytes,and stem cells.These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis,for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog,etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence,drug therapy,gene therapy,protein therapy,cell therapy,and exosome therapy have been emerging and are known as novel therapies.In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways,which contribute to inhibiting cardiomyocytes death,attenuating inflammation,enhancing angiogenesis,etc.so as to repair and re-functionalize damaged hearts.Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei 2022Signal Transduction and Targeted Therapy2022,7,4:11
