1PTEN encoding product:a marker for tumorigenesis and progression of gastric carcinoma显示文摘AIM: To detect the expression of PTEN encoding productin normal mucosa, intestinal metaplasia (IM), dysplasia andcarcinoma of the stomach, and to investigate its clinicalimplication in tumorigenesis and progression of gastriccarcinoma.METHODS: Formalin-fixed paraffin embedded specimens from184 cases of gastric carcinoma, their adjacent normal mucosa,IM and dysplasia were evaluated for PTEN protein expressionby SABC immunohistochemistry. PTEN expression wascompared with tumor stage, lymph node metastasis, Lauren'sand WHO's histological classification of gastric carcinoma.Expression of VEGF was also detected in 60 cases of gastriccarcinoma and its correlation with PTEN was concerned.RESULTS: The positive rates of PTEN protein were 100 %(102/102), 98.5 %(65/66), 66.7 % (4/6) and 47.8 %(88/184)in normal mucosa, IM, dysplasia and carcinoma of the stomach,respectively. The positive rates in dysplasia and carcinomawere lower than in normal mucosa and IM (P<0.01).Advanced gastric cancers expressed less frequent PTEN thanearly gastric cancer (42.9 % v567.6 %, P<0.01). The positiverate of PTEN protein was lower in gastric cancer with thanwithout lymph node metastasis (40.3 % v563.3 %, P<0.01).PTEN was less expressed in diffuse-type than in intestinal-type gastric cancer (41.5 % v557.8 %,P<0.05). Signet ringcell carcinoma showed the expression of PTEN at the lowestlevel (25.0 %, 7/28); less than well and moderatelydifferentiated ones (P<0.01). Expression of PTEN was notcorrelated with expression of VEGF (P>0.05).CONCLUSION: Loss or reduced expression of PTEN proteinoccures commonly in tumorigenesis and progression of gastriccarcinoma. 6Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. 7Decreased osteogenesis of adult mesenchymal stem cel s by reactive oxygen species under cyclic stretch: a possible mechanism of age related osteoporosis显示文摘Age related defect of the osteogenic differentiation of mesenchymal stem cells(MSCs) plays a key role in osteoporosis. Mechanical loading is one of the most important physical stimuli for osteoblast differentiation.Here, we compared the osteogenic potential of MSCs from young and adult rats under three rounds of 2 h of cyclic stretch of 2.5% elongation at 1 Hz on 3 consecutive days. Cyclic stretch induced a significant osteogenic differentiation of MSCs from young rats, while a compromised osteogenesis in MSCs from the adult rats.Accordingly, there were much more reactive oxygen species(ROS) production in adult MSCs under cyclic stretch compared to young MSCs. Moreover, ROS scavenger N-acetylcysteine rescued the osteogenic differentiation of adult MSCs under cyclic stretch. Gene expression analysis revealed that superoxide dismutase 1(SOD1) was significantly downregulated in those MSCs from adult rats. 8Chitosan hydrogel incorporated with dental pulp stem cell-derived exosomes alleviates periodontitis in mice via a macrophage-dependent mechanism显示文摘Periodontitis is caused by host immune-inflammatory response to bacterial insult.A high proportion of proinflammatory macrophages to anti-inflammatory macrophages leads to the pathogenesis of periodontitis.As stem cell-derived exosomes can modulate macrophage phenotype,dental pulp stem cell-derived exosomes(DPSC-Exo)can effectively treat periodontitis.In this study,we demonstrated that DPSC-Exo-incorporated chitosan hydrogel(DPSC-Exo/CS)can accelerate the healing of alveolar bone and the periodontal epithelium in mice with periodontitis.Gene Ontology(GO)term enrichment analysis showed that treatment with DPSC-Exo/CS ameliorated periodontal lesion by suppressing periodontal inflammation and modulating the immune response.Specifically,DPSC-Exo/CS facilitated macrophages to convert from a pro-inflammatory phenotype to an anti-inflammatory phenotype in the periodontium of mice with periodontitis,the mechanism of which could be associated with miR-1246 in DPSC-Exo.These results not only shed light on the therapeutic mechanism of DPSCExo/CS but also provide the basis for developing an effective therapeutic approach for periodontitis.Zongshan Shen Shuhong Kuang Yong Zhang Mingmei Yang Wei Qin Xuetao Shi Zhengmei Lin 2020Bioactive Materials2020,5,4:14
9Dysregulation of the miR-146a-Smad4 axis impairs osteogenesis of bone mesenchymal stem cells under inflammation显示文摘Osteoporosis is a common disease that affects patient quality of life, especially among the elderly population.Although inflammation contributes significantly to osteoporosis, the underlying mechanism is unclear. In this study, we found that tumor necrosis factor(TNF)-α, an inflammatory environment mimic, inhibits osteogenesis of bone mesenchymal stem cells(BMSCs), induces mi R-146 a and decreases Smad4. Moreover,overexpression of mi R-146 a inhibited the osteogenic ability of BMSCs, whereas blocking mi R-146 a partially rescued the osteogenesis deficiency under TNF-α treatment. Molecularly, mi R-146 a decreased Smad4 expression at the protein level by binding to an element located in the Smad4 30-untranslated region, and restoration of Smad4 reversed the inhibitory effects of mi R-146 a on osteogenesis. 11Adaptive subsequence adjustment with evolutionary asymmetric path-relinking for TDRSS scheduling显示文摘Due to the limited transmission resources for data relay in the tracking and data relay satellite system(TDRSS), there are many job requirements in busy days which will be discarded in the conventional job scheduling model. Therefore, the improvement of scheduling efficiency in the TDRSS can not only help to increase the resource utilities, but also to reduce the scheduling failure ratio.A model of nonhomogeneous parallel machines scheduling problems with time window(NPM-TW) is firstly built up for the TDRSS,considering the distinct features of the variable preparation time and the nonhomogeneous transmission rates for different types of antennas on each tracking and data relay satellite(TDRS). Then,an adaptive subsequence adjustment(ASA) framework with evolutionary asymmetric path-relinking(Ev APR) is proposed to solve this problem, in which an asymmetric progressive crossover operation is involved to overcome the local optima by the conventional job inserting methods. 12TNF-α and IL-1RA Polymorphisms and Silicosis Susceptibility in Chinese Workers Exposed to Silica Particles:A Case-Control Study显示文摘Objective To assess the association of TNF- α and IL-1RA SNPs with the risk of silicosis in Chinese workers exposed to silica particles. Methods Case-control study design was used to enroll 68 silicotic patients induced by silica particles and 68 healthy workers matched for length of silica particle exposure as controls. Both cases and controls were from the same company in southwest China, and each of them was requested to complete a questionnaire. Blood samples were drawn for genomic DNA extraction from each participant. The genotyping of TNF-α (-238 and -308) and IL-1RA (+2018) was performed using polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP) and SYBR green-based quantitative polymerase chain reaction (qPCR), respectively. Unconditional logistic regression model was used to estimate odds ratios (ORs) and their 95% confidential intervals (CI) for SNPs. Results No significant differences were found between cases and controls in particles exposure length, body mass index (BMI), and status of smoking and alcohol consumption except for age (P=0.001) and blood type (P=0.042). The frequencies of TNF-α (-238) and IL-1RA (+2018) genotypes in cases were significantly different from those in controls, (P=0.001 and P=0.002, respectively), while a borderline significant difference was found in the frequencies of TNF-α (-308) between cases and controls (P=0.063). The variants of three SNPs increased the risk of silicosis in the Chinese workers exposed to silica particles. The adjusted ORs of TNF-α (-308), TNF-α (-238) and IL-1RA (+2018) were 2.8 (95% CI: 1.1-7.5), 20.9 (95% CI: 1.8-236.4) and 4.0 (95% CI: 1.6-10.1), respectively. Conclusion It is suggested that cytokine polymorphisms of TNF- α (-238, -308) and IL-1RA (+2018) are associated with the risk of silicosis in the Chinese workers exposed to silica particles. 14Exosomes: roles and therapeutic potential in osteoarthritis显示文摘Exosomes participate in many physiological and pathological processes by regulating cell–cell communication, which are involved in numerous diseases, including osteoarthritis(OA). Exosomes are detectable in the human articular cavity and were observed to change with OA progression. Several joint cells, including chondrocytes, synovial fibroblasts, osteoblasts, and tenocytes, can produce and secrete exosomes that influence the biological effects of targeted cells. 15Inhibitory effect of Salicornia europaea on the marine alga Skeletonema costatum显示文摘Exploiting the negative biochemical interference between plants and algal species has been suggested as a method to control harmful algal blooms.In this work,we investigated the inhibitory effect of the salt marsh halophyte Salicornia europaea against the marine alga Skeletonema costatum.S.europaea suppressed the growth of S.costatum in a nutrient-sufficient co-culture system,indicating that the inhibition of algal growth was because of the phytotoxic effect of S.europaea,rather than nutrient competition.We tested aqueous and organic extracts from S.europaea roots against S.costatum.The organic extracts inhibited growth and affected the cell size and chlorophyll a content of S.costatum in a dose-dependent manner.Among the three tested organic extracts,the methanol extract had the greatest effects on S.costatum,followed by butanol extract,and then the chloroform extract.Two flavonoids,rutin and quercetin-3-D-glucoside,were identified in the methanol extract by high performance liquid chromatography.The concentration of rutin was much higher than that of quercetin-3-D-glucoside.In an algal bioassay,rutin inhibited the growth of S.costatum and the inhibitory effect increased with increasing rutin concentration and with decreasing initial algal density.Therefore,we concluded that S.europaea negatively affects the growth of S.costatum,and that rutin,a metabolite of S.europaea,may play a role in this inhibitory effect.JIANG Dan HUANG LingFeng LIN YongQing NIE LingLing LV SuLian KUANG TingYun LI YinXin 2012Science China(Life Sciences)2012,55,6:8
16The influence of graded levels of available phosphorus on growth performance,muscle antioxidant and flesh quality of young grass carp(Ctenopharyngodon idella)显示文摘Growth, muscle composition, meat quality characteristics and antioxidant capacity in muscle of young grass carp(Ctenopharyngodon idella)(initial weight 282.9 ± 3.3 g) fed graded levels of phosphorus(1.0,2.5, 3.8, 5.6, 7.8 and 9.5 g/kg diet) for 8 wk were investigated. Results indicated that percentage weight gain, feed intake, feed efficiency, serum phosphorus and alkaline phosphatase were improved with optimal phosphorus supplementations(P < 0.05). Muscle protein content and water holding capacity were significantly elevated, while moisture, lipid and ash contents were significantly decreased with dietary phosphorus to a certain level(P < 0.05). The meat shear force value and hydroxyproline content were not influenced by different levels of phosphorus. Muscle anti-hydroxyl radical, superoxide dismutase, catalase, glutathione S-transferase activities and glutathione content were significantly improved(P < 0.05). Conversely, anti-superoxide anion, glutathione reducase and glutathione peroxidase activities were decreased(P < 0.05) with dietary phosphorus to a certain level. These results indicated that suitable dietary phosphorus improved growth performance, meat quality and muscle antioxidant capacity. 17桑叶多糖对早期断奶仔猪生长性能、腹泻、血液指标和肠道菌群的影响显示文摘研究旨在探讨日粮补充桑叶多糖(MLPs)对早期断奶仔猪生长性能、腹泻、血液生化指标和肠道菌群的影响。共选取(28±2)日龄、体重为(9.18±0.46)kg的'杜洛克×长白×约克夏'杂交断奶仔猪150头,随机分成5个处理组,每组6个重复,每个重复5头猪。饮食处理如下:对照组(CT):基础日粮(BD);低剂量桑叶多糖组(LT):0.3 g·kg-1MLPs+BD;中剂量桑叶多糖组(MT):0.6 g·kg-1MLPs+BD;高剂量桑叶多糖组(HT):0.9 g·kg-1MLPs+BD;抗生素处理组(AT):0.15 g·kg-1金霉素+BD。分别在第0、10、21天的早晨,每头猪空腹测量体重并记录采食量,这些数据用来计算平均日增重(ADG)、平均日采食量(ADFI)和料重比(F/G)。每天两次(上午和下午)对每头仔猪的腹泻状况进行检查并记录。在试验结束时,收集血液样本用于生化分析,并收集盲肠、结肠和直肠内容物进行识别和量化相关的肠道菌群。研究结果表明,MLPs处理组间的日增重、平均日采食量和料重比无显著差异(P>0.05),但低剂量组(LT)、中剂量组(MT)、高剂量组(HT)的平均日采食量高于对照组(CT)和抗生素组(AT)。并且,LT、MT和HT组的料重比低于CT和AT组。与CT和AT组相比,LT、MT和HT组仔猪的腹泻发病率也显著降低(P<0.05)。生化分析表明,MLPs各处理组仔猪的血糖(GLU)水平均显著低于CT或AT组(P<0.05)。MT和HT组血液尿素氮(BUN)水平与CT和AT组存在显著差异(P<0.05),HT组T3水平显著高于CT和AT组(P<0.05)。虽然MLPs各处理组的T4水平与AT组并没有显著差异(P>0.05),但MT组的胰岛素样生长因子Ⅰ(IGF-Ⅰ)和生长激素(GH)水平较CT或AT组均显著升高(P<0.05)。LT、MT和HT 3组在抑制肠道大肠杆菌生长和促进肠道乳酸杆菌和双歧杆菌生长的整体效果上均优于CT和AT组。值得注意的是,与CT和AT组相比,HT组(0.9 g·kg-1MLPs)对抑制肠道大肠杆菌以及促进肠道有益菌群具有更显著效果(P<0.05)。结果表明,日粮添加桑叶多糖可以改善早期断奶仔猪肠道菌群的生态环境,降低腹泻率,提高整体生长性能。Zhao X J Lin L Luo Q L Ye M Q Luo G Q Kuang Z S 2015饲料博览2015,0,7:7
18Inhibition of CCL2 by bindarit alleviates diabetes-associated periodontitis by suppressing inflammatory monocyte infiltration and altering macrophage properties显示文摘Diabetes-associated periodontitis(DP)aggravates diabetic complications and increases mortality from diabetes.DP is caused by diabetes-enhanced host immune-inflammatory responses to bacterial insult.In this study,we found that persistently elevated CCL2 levels in combination with proinflammatory monocyte infiltration of periodontal tissues were closely related to DP.Moreover,inhibition of CCL2 by oral administration of bindarit reduced alveolar bone loss and increased periodontal epithelial thickness by suppressing periodontal inflammation.Furthermore,bindarit suppressed the infiltration of proinflammatory monocytes and altered the inflammatory properties of macrophages in the diabetic periodontium.This finding provides a basis for the development of an effective therapeutic approach for treating DP.Zongshan Shen Shuhong Kuang Min Zhang Xin Huang Jiayao Chen Meiliang Guan Wei Qin Hockin H.K.Xu Zhengmei Lin 2021Cellular & Molecular Immunology2021,18,9:6
19Numerical modelling of channel migration with application to laboratory rivers显示文摘The paper presents the development of a morphological model and its application to experimental model rivers.The model takes into account the key processes of channel migration,including bed deformation,bank failure and wetting and drying.Secondary flows in bends play an important role in lateral sediment transport,which further affects channel migration.A new formula has been derived to predict the near-bed secondary flow speed,in which the magnitude of the speed is linked to the lateral water level gradient.Since only non-cohesive sediment is considered in the current study,the bank failure is modelled based on the concept of submerged angle of repose.The wetting and drying process is modelled using an existing method.Comparisons between the numerical model predictions and experimental observations for various discharges have been made.It is found that the model predicted channel planform and cross-sectional shapes agree generally well with the laboratory observations.A scenario analysis is also carried out to investigate the impact of secondary flow on the channel migration process.It shows that if the effect of secondary flow is ignored,the channel size in the lateral direction will be seriously underestimated.Jian SUN Bin-liang LIN Hong-wei KUANG 2015International Journal of Sediment Research2015,30,1:6
