2Study on multi-size seed-metering device for vertical plate soybean precision planter显示文摘Aiming to solve the problem of small range of the appropriately sowing seeds existing in a vertical disc seed-metering device,the planter plate series with four sizes were developed according to the variety and size distribution of all soybeans in China.The structure and working principle of the vertical disc soybean seed-metering device were detailed,and the influence of the diameter of soybean on the working performance of the seed-metering device was analyzed through the software EDEM virtual simulation,so as to achieve the goal of covering the soybean seeds with all sizes by the minimum planter plate series as well as to obtain the most appropriate operating speed of each planter plate by optimization.For the planter plates with the hole diameter of 7,9,12,16 mm,the appropriate size ranges of sowing seeds are 4.5-6.0,6.0-8.0,8.0-10.5,10.5-13.0 mm,respectively,and the appropriate operating speeds are 9,8,7,6 km/h,respectively.The results show that this planter plate series can meet the requirements of seeding with all sizes of soybeans at the range of the most appropriate operating speed.The study method can provide a reference for design and optimization of precision planters.Liu Hongxin Guo Lifeng Fu Lulu Tang Shifa 2015International Journal of Agricultural and Biological Engineering2015,8,1:13

4Study on hydrometallurgical process and kinetics of manganese extraction from low-grade manganese carbonate ores显示文摘In order to utilize low-grade manganese ore resources effectively, a hydrometallurgical process was developed for manganese extraction in dilute sulfuric acid medium, and the kinetics of leaching manganese was also investigated. At room temperature, manganese from low-grade manganese carbonate ores was extracted by sulfuric acid leaching without reductants. During the extracting process, single-factor analysis method was used to evaluate the effects of grinding fineness, sulfuric acid concentration,liquid-to-solid ratio, agitation rate and leaching time on the leaching efficiencies of Mn and Fe. The optimal leaching conditions are determined as coarse particles of below 2 mm size(without ball-milling),sulfuric acid concentration of 0.86 mol/L, liquid-to-solid ratio of 5:1, agitation rate of 150 r/min and leaching for 180 min at room temperature. Under the optimal conditions, the leaching efficiencies of Mn and Fe are 96.21% and 13.35%, respectively. In addition, through the experiments at different temperatures, it is found that the leaching process follows the shrinking core model under the conditions of changing acid concentration and intermittent reaction device. 5TLR9 agonist enhances radiofrequency ablation-induced CTL responses, leading to the potent inhibition of primary tumor growth and lung metastasis显示文摘Radiofrequency ablation(RFA)is the most common approach to thermal ablation for cancer therapy.Unfortunately,its efficacy is limited by incomplete ablation,and further optimization of RFA is required.Here,we demonstrate that incubation at 65°C triggers more EG7 tumor cell death by necrosis than treatment at 45°C,and the 65°C-treated cells are more effective at inducing antigen-specific CD8^(+)cytotoxic T lymphocyte(CTL)responses after injection in mice than the 45°C-treated ones.Dendritic cells(DCs)that phagocytose 65°C-treated EG7 cells become mature with upregulated MHCII and CD80 expression and are capable of efficiently inducing effector CTLs in mouse tumor models.RFA(65°C)therapy of EG7 tumors induces large areas of tumor necrosis and stimulates CTL responses.This leads to complete regression of small(~100 mm^(3))tumors but fails to suppress the growth of larger(~350 mm^(3))tumors.The administration of the Toll-like receptor-9(TLR9)agonist unmethylated cytosine-phosphorothioate-guanine oligonucleotide(CpG)to DCs phagocytosing 65°C-treated EG7 cells enhances the expression of MHCII and CD40 on DCs as well as DC-induced stimulation of CTL responses.Importantly,the intratumoral administration of CpG following RFA also increases the frequencies of tumor-associated immunogenic CD11b−CD11c^(+)CD103^(+)DC2 and CD11b+F4/80+MHCII+M1 macrophages and increases CD4^(+)and CD8^(+)T-cell tumor infiltration,leading to enhanced CD4^(+)T cell-dependent CTL responses and potent inhibition of primary RFA-treated or distant untreated tumor growth as well as tumor lung metastasis in mice bearing larger tumors.Overall,our data indicate that CpG administration,which enhances RFA-induced CTL responses and ultimately potentiates the inhibition of primary tumor growth and lung metastasis,is a promising strategy for improving RFA treatment,which may assist in optimizing this important cancer therapy.Aizhang Xu Lifeng Zhang Jingying Yuan Fatma Babikr Andrew Freywald Rajni Chibbar Michael Moser Wenjun Zhang Bing Zhang Zhaoying Fu Jim Xiang 2019Cellular & Molecular Immunology2019,16,10:3
6Structural Basis of SARS-CoV-2 Polymerase Inhibition by Favipiravir显示文摘The outbreak of severe acute respiratory syndrome coronavirus 2(SARSCoV-2)has developed into an unprecedented global pandemic.Nucleoside analogs,such as Remdesivir and Favipiravir,can serve as the firstline broad-spectrum antiviral drugs by targeting the viral polymerases.However,the underlying mechanisms for the antiviral efficacies of these drugs are far from well understood.Here,we reveal that Favipiravir,as a pyrazine derivative,could be incorporated into the viral RNA products by mimicking both adenine and guanine nucleotides.This drug thus inhibits viral replication mainly by inducing mutations in progeny RNAs,different from Remdesivir or other RNA-terminating nucleoside analogs that impair the elongation of RNA products.We further determined the cryo-EM structure of Favipiravir bound to the replicating polymerase complex of SARSCoV-2 in the pre-catalytic state.This structure provides a missing snapshot for visualizing the catalysis dynamics of coronavirus polymerase,and reveals an unexpected base-pairing pattern between Favipiravir and pyrimidine residues that may explain its capacity for mimicking both adenine and guanine nucleotides.These findings shed light on the mechanism of coronavirus polymerase catalysis and provide a rational basis for developing antiviral drugs to combat the SARS-CoV-2 pandemic.Qi Peng Ruchao Peng Bin Yuan Min Wang Jingru Zhao Lifeng Fu Jianxun Qi Yi Shi 2021The Innovation2021,2,1:1

14Vibration characteristics of a multi-block high-temperature superconducting maglev system显示文摘The multi-block high-temperature superconducting (HTS) maglev system has more complicated dynamic characteristics than the single-block HTS maglev system. To study its vibration characteristics, we designed a maglev measurement system. The system responses at the excitation frequencies of 2, 3 and 15 Hz were examined. Results show that the responses under excitation frequencies of 2 and 3 Hz include a 6 Hz component, which means that the maglev system is a critical nonlinear system. Moreover, the 6 Hz component is much stronger than the 2 Hz or 3 Hz components in the response spectra. There is the interaction between excitation and response. 18Rumen microbiota-host transcriptome interaction mediates the protective effects of trans-10,cis-12 CLA on facilitating weaning transition of lambs显示文摘Developing alternatives to antibiotics for prevention of gastrointestinal dysbiosis in early-weaning farmed animals is urgently needed.This study was to explore the potential effects of trans-10,cis-12 conjugated linoleic acid(CLA)on maintaining ruminal homeostasis of young ruminants during the weaning transition period.Thirty neonatal lambs were selected(6 lambs per group)and euthanized for rumen microbial and epithelial analysis.The lambs were weaned at 28 d and experienced the following 5 treatments:euthanized on d 28 as the pre-weaning control(CON0),fed starter feed for 5(CON5)or 21(CON21)d,fed starter feed with 1%of CLA supplemented for 5(CLA5)or 21(CLA21)d.Results showed that the average daily weight gain and dry matter intake were significantly higher in CLA5 than CON5 group.As compared with the CON5 and CON21 group,the relative abundances of volatile fatty acid(VFA)producing bacteria including Bacteroides,Treponema,Parabacteroides and Anaerovibrio,as well as the concentrations of acetate,butyrate and total VFA were significantly increased in CLA5 and CLA21 group,respectively.Integrating microbial profiling and epithelial transcriptome results showed that 7 downregulated inflammatory signaling-related host genes IL2RA,CXCL9,CD4,CCR4,LTB,SPP1,and BCL2A1 with CLA supplementation were significantly negatively correlated with both VFA concentration and VFA producing bacteria,while 3(GPX2,SLC27A2 and ALDH3A1)and 2(GSTM3 and GSTA1)upregulated metabolism-related genes,significantly positively correlated with either VFA concentration or VFA producing bacteria,respectively.To confirm the effects of CLA on epithelial signal transduction,in vitro experiment was further conducted by treating rumen epithelial cells without or with IL-17A+TNF-αfor 12 h after pretreatment of 100μM CLA or not(6 replicates per treatment).The results demonstrated the anti-inflammatory effect of CLA via suppressing the protein expression of NF-кB p-p65/p65 with the activation of peroxisome proliferator-activated receptor gamma(PPARγ).In conclusion,CLA supplementation enhanced the ruminal microbiota-driven transcriptional regulation in healthy rumen epithelial development via rumen VFA production,and CLA may therefore serve as an alternative way to alleviate early-weaning stress and improve physiological and metabolic conditions of young ruminants.Chunlei Yang Xiangfei Deng Peter Lund Haixia Liu Xingwang Ding Zhengwei Fu Naifeng Zhang Jinjun Li Lifeng Dong 2023Animal Nutrition2023,,1:0
19Seismic behavior of precast segmental column bridges under near-fault forward-directivity ground motions显示文摘Precast segmental column bridges exhibit various construction advantages in comparison to traditional monolithic column bridges.However,the lack of cognitions on seismic behaviors has seriously restricted their applications and developments.In this paper,comprehensive investigations are conducted to analyze the dynamic characteristics of precast segmental column bridges under near-fault,forward-directivity ground motions.First,the finite-element models of two comparable bridges with precast segmental columns and monolithic columns are constructed by using OpenSees software,and the nonlinearities of the bridges are considered.Next,three different earthquake loadings are meticulously set up to handle engineering problems,namely recorded near-and far-field ground motions,parameterized pulses,and pulse and residual components extracted from real records.Finally,based on the models and earthquake sets,extensive explorations are carried out.The results show that near-fault forward-directivity ground motions are more threatening than far-field ones;precast segmental column bridges may suffer more pounding impacts than monolithic bridges;the“narrow band”effect caused by near-fault,forward-directivity ground motions may occur in bridges with shorter periods than pulse periods;and pulse and residual components play different roles in seismic responses.Xin Lifeng Li Xiaozhen Fu Peiyao Mu Di 2022Earthquake Engineering and Engineering Vibration2022,21,2:0
