1Hydrogen sulfide suppresses transforming growth factor-β1-induced differentiation of human cardiac fibroblasts into myofibroblasts显示文摘In heart disease, transforming growth factor-β1(TGF-β1) converts fibroblasts into myofibroblasts, which synthesize and secrete fibrillar type I and III collagens. The purpose of the present study was to investigate how hydrogen sulfide(H2S) suppresses TGF-β1-induced differentiation of human cardiac fibroblasts to myofibroblasts. Human cardiac fibroblasts were serum-starved in fibroblast medium for 16 h before exposure to TGF-β1(10 ng m L-1) for 24 h with or without sodium hydrosulfide(Na HS, 100 μmol L-1, 30 min pretreatment) treatment. Na HS, an exogenous H2 S donor, potently inhibited the proliferation and migration of TGF-β1-induced human cardiac fibroblasts and regulated their cell cycle progression. Furthermore, Na HS treatment led to suppression of fibroblast differentiation into myofibroblasts, and reduced the levels of collagen, TGF-β1, and activated Smad3 in TGF-β1-induced human cardiac fibroblasts in vitro. We therefore conclude that H2 S suppresses TGF-β1-stimulated conversion of fibroblasts to myofibroblasts by inhibiting the TGF-β1/Smad3 signaling pathway, as well as by inhibiting the proliferation, migration, and cell cycle progression of human cardiac myofibroblasts. 2On the determination and simulation of seawater freezing point temperature under high pressure显示文摘At present,it is believed that the freezing point temperature of seawater is a function of salinity and pressure,and the freezing point is a key parameter in a coupled air-sea-ice system.Generally,empirical formulas or methods are used to calculate the freezing point of seawater.Especially in high-pressure situations,e.g.,under a thick ice sheet or ice shelf,the pressure term must be taken into account in the determination of seawater freezing point temperature.This study summarized various methods that have been used to calculate seawater freezing point with high pressure.The methods that were employed in two ocean-ice models were also assessed.We identified the disadvantages of these methods used in these two models and addressed the corresponding uncertainties of the freezing point temperature formulas.This study provides useful information on the calculation of the freezing point temperature in numerical modeling and indicates a need to investigate the sensitivity of numerical simulations to the uncertainties in the freezing point temperature in future.YAN Liangjun WANG Zhaomin LIU Chengyan WU Yang 2019Advances in Polar Science2019,30,4:2

7Impacts of Ice-Ocean Stress on the Subpolar Southern Ocean:Role of the Ocean Surface Current显示文摘The mechanical influences involved in the interaction between the Antarctic sea ice and ocean surface current(OSC)on the subpolar Southern Ocean have been systematically investigated for the first time by conducting two simulations that include and exclude the OSC in the calculation of the ice-ocean stress(IOS), using an eddy-permitting coupled ocean-sea ice global model. By comparing the results of these two experiments, significant increases of 5%, 27%, and 24%, were found in the subpolar Southern Ocean when excluding the OSC in the IOS calculation for the ocean surface stress,upwelling, and downwelling, respectively. Excluding the OSC in the IOS calculation also visibly strengthens the total mechanical energy input to the OSC by about 16%, and increases the eddy kinetic energy and mean kinetic energy by about38% and 12%, respectively. Moreover, the response of the meridional overturning circulation in the Southern Ocean yields respective increases of about 16% and 15% for the upper and lower branches;and the subpolar gyres are also found to considerably intensify, by about 12%, 11%, and 11% in the Weddell Gyre, the Ross Gyre, and the Australian-Antarctic Gyre, respectively. The strengthened ocean circulations and Ekman pumping result in a warmer sea surface temperature(SST), and hence an incremental surface heat loss. The increased sea ice drift and warm SST lead to an expansion of the sea ice area and a reduction of sea ice volume. 8Chromosome-scale genome assembly of marigold (Tagetes erecta L.):An ornamental plant and feedstock for industrial lutein production显示文摘Marigold(Tagetes erecta L., African marigold) is a widely grown ornamental plant and a main source of the carotenoid lutein for the industrial production of pharmaceuticals, food coloring, and feed additives. To gain a deeper understanding of the genetic mechanism of lutein in marigold, a chromosome-scale assembly of the marigold(T. erecta V-01) genome was completed based on Illumina, PacBio, and Hi-C reads. The707.21-Mb assembled genome consisted of 35 834 annotated protein-coding genes, with 97.7% genomic integrity. We anchored 87.8% of the contigs(covering 621.20 Mb) to 12 pseudochromosomes, bringing the scaffold N50 length to 54.15 Mb. Phylogenetic analysis showed that marigold was closely related to the Asteraceae species bitter vine(Mikania micrantha) and sunflower(Helianthus annuus), all three of which originated in the Americas. Marigold diverged from the sunflower clade 23.57 million years ago(MYA) and from M. micrantha 19.59 MYA.Marigold has undergone three whole-genome duplication events, as well as a recent whole-genome duplication event(WGD-2) common to H.annuus and M. micrantha. Marigold gene families were significantly less expanded than those of M. micrantha or H. annuus, and the marigold genome contained significantly fewer interspersed repeats, which might account for its smaller genome. In addition, a range of candidate genes involved in the lutein biosynthetic pathway were identified. 9One-pot synthesis of Ag-rich AgPd alloy nanoactiniae and their enhanced electrocatalytic activity toward oxygen reduction显示文摘The electro-catalytic properties can be effectively optimized by designing bimetallic alloy nanoparticles with high-content less-active metal to enhance the competence of more-active noble metal. Herein, a one-pot hydrothermal approach is demonstrated for the controllable synthesis of Ag-rich Ag_9Pd_1 alloy nanoactiniae with obviously enhanced electro-catalytic activity(2.23 mA cm^(-2) at 0.85 V) and stability for oxygen reduction reaction. In alkaline solution, the ORR onset potential and half-wave potential of the Ag_9Pd_1 alloy nanoactiniae can reach a value of 1.02 V and 0.89 V, respectively, which origin from strong ligand and ensemble effects between Pd element and Ag element. The nanocrystals are uniformly alloyed, displaying a Ag_9Pd_1 combination, as displayed by an assembly of X-ray diffraction(XRD) spectrum,energy dispersive X-ray(EDX) analysis, and cyclic voltammetry(CV). 10Cisplatin and paclitaxel co-delivery nanosystem for ovarian cancer chemotherapy显示文摘We have designed and developed an effective drug delivery system using biocompatible polymer of poly(ethylene glycol)-polyaspartic acid(mPEG-PAsp)for co-loading the chemotherapy drugs paclitaxel(PTX)and cisplatin(CP)in one nano-vehicle.This study aimed to improve the anti-cancer effi-cacy of combinations of chemotherapy drugs and reduce their side effects.mPEG-PAsp-(PTX/Pt)nano-micelles disperse well in aqueous solution and have a narrow size distribution(37.863.2 nm)in dynamic light scattering(DLS).Drug release profiles found that CP released at pH 5.5 was signifi-cantly faster than that at pH 7.4.MPEG-PAsp-(PTX/Pt)nano-micelles displayed a significantly higher tumor inhibitory effect than mPEG-PAsp-PTX nano-micelles when the polymer concentrations reached 50 lg/mL.Our data indicated that polymer micelles of mPEG-PAsp loaded with the combined drug exert synergistic anti-tumor efficacy on SKOV3 ovarian cells via different action mechanisms.Results from our studies suggested that mPEG-PAsp-(PTX/Pt)nano-micelles are promising alternatives for carrying and improving the delivery of therapeutic drugs with different water solubilities.Qiaoying Wang Changqiang Wu Xiaoting Li Dixiao Yang Liangjun Shi 2021Regenerative Biomaterials2021,8,3:0
11Self-supported porous heterostructure WC/WO_(3−x)ceramic electrode for hydrogen evolution reaction in acidic and alkaline media显示文摘Tungsten carbide(WC)-based materials are widely considered as the hydrogen evolution reaction(HER)process catalysts due to their“Pt-like”electronic structure.Nonetheless,traditional powder electrodes have a high cost,and display problems related to the process itself and the poor stability over operation time.This paper presented a self-supported asymmetric porous ceramic electrode with WO_(3-x)whiskers formed in situ on the walls of the finger-like holes and membrane surface,which was prepared by combining phase inversion tape-casting,pressureless sintering,and thermal treatment in a CO_(2) atmosphere.The optimized ceramic electrode displayed good catalytic HER activity and outstanding stability at high current densities.More specifically,it demonstrated the lowest overpotentials of 107 and 123 mV and the lowest Tafel slopes of 59.3 and 72.4 mV·dec^(-1)at 10 mA·cm^(-2)in acidic and alkaline media,respectively.This superior performance was ascribed to the structure of the ceramic membrane and the charge transfer efficiency,which was favored by the in situ developed WC/WO_(3-x)heterostructure and the oxygen vacancies.Feihong WANG Binbin DONG Junwei WANG Nianwang KE Chuntian TAN Anding HUANG Yutong WU Luyuan HAO Liangjun YIN Xin XU Yuxi XIAN Simeon AGATHOPOULOS 2022Journal of Advanced Ceramics2022,11,8:0
12Formal Verification Platform as a Service:WebAssembly Vulnerability Detection Application显示文摘In order to realize a general-purpose automatic formal verification platform based on WebAssembly technology as a web service(FVPS),which aims to provide an automated report of vulnerability detections,this work builds a Hyperledger Fabric blockchain runtime model.It proposes an optimized methodology of the functional equivalent translation from source program languages to formal languages.This methodology utilizes an external application programming interface(API)table to replace the source codes in compilation,thereby pruning the part of housekeeping codes to ease code inflation.Code inflation is a significant metric in formal language translation.Namely,minor code inflation enhances verification scale and performance efficiency.It determines the efficiency of formal verification,involving launching,running,and memory usage.For instance,path explosion increases exponentially,resulting in out-of-memory.The experimental results conclude that program languages like golang severely impact code inflation.FVPS reduces the wasm code size by over 90%,achieving two orders of optimization magnitude,from 2000 kilobyte(KB)to 90 KB.That means we can cope with golang applications up to 20 times larger than the original in scale.This work eliminates the gap between Hyperledger Fabric smart contracts and WebAssembly.Our approach is pragmatic,adaptable,extendable,and flexible.Nowadays,FVPS is successfully applied in a Railway-Port-Aviation blockchain transportation system.LiangJun Deng Hang Lei Zheng Yang WeiZhong Qian XiaoYu Li Hao Wu Sihao Deng RuChao Sha WeiDong Deng 2023Computer Systems Science & Engineering2023,45,5:0
